ABSTRACT After a dengue outbreak, the knowledge on the extent, distribution and mechanisms of insecticide resistance is essential for successful insecticide-based dengue control interventions. Therefore, we evaluated the potential changes to insecticide resistance in natural Aedes aegypti populations to Organophosphates (OP) and Pyrethroids (PY) after chemical vector control interventions. After a Dengue outbreak in 2010, A. aegypti mosquitoes from the urban area of Jacarezinho (Paraná, Brazil) were collected in 2011 and 2012. Insecticide resistance to OP Temephos was assessed in 2011 and 2012 by dose–response bioassays adopting WHO-based protocols. Additionally, in both sampling, PY resistance was also investigated by the Val1016Ile mutation genotyping. In 2011, a random collection of mosquitoes was carried out; while in 2012, the urban area was divided into four regions where mosquitoes were sampled randomly. Bioassays conducted with larvae in 2011 (82 ± 10%; RR95 = 3.6) and 2012 (95 ± 3%; RR95 = 2.5) indicated an incipient altered susceptibility to Temephos. On the other hand, the Val1016IIe mutation analysis in 2011, presented frequencies of the 1016Ilekdr allele equal to 80%. Nevertheless, in 2012, when the urban area of Jacarezinho was analyzed as a single unit, the frequency of the mutant allele was 70%. Additionally, the distribution analysis of the Val1016Ile mutation in 2012 showed the mutant allele frequencies ≥60% in all regions. These outcomes indicated the necessity of developing alternative strategies such as insecticide rotations for delaying the evolution of resistance.

The objective of this work was to screen plants with insecticide activity, in order to isolate, identify and assess the bioactivity of insecticide compounds present in these plants, against Coleoptera pests of stored products: Oryzaephilus surinamensis L. (Silvanidae), Rhyzopertha dominica F. (Bostrichidae) and Sitophilus zeamais Mots. (Curculionidae). The plant species used were: basil (Ocimum selloi Benth.), rue (Ruta graveolens L.), lion's ear (Leonotis nepetifolia (L.) R.Br.), jimson weed (Datura stramonium L.), baleeira herb (Cordia verbenacea L.), mint (Mentha piperita L.), wild balsam apple (Mormodica charantia L.), and billy goat weed or mentrasto (Ageratum conyzoides L.). The insecticide activity of hexane and ethanol extracts from those plants on R. dominica was evaluated. Among them, only hexane extract of A. conyzoides showed insecticide activity; the hexane extract of this species was successively fractionated by silica gel column chromatography, for isolation and purification of the active compounds. Compounds 5,6,7,8,3',4',5'-heptamethoxyflavone; 5,6,7,8,3'-pentamethoxy-4',5'-methilenedioxyflavone and coumarin were identified. However, only coumarin showed insecticide activity against three insect pests (LD50 from 2.72 to 39.71 mg g-1 a.i.). The increasing order of insects susceptibility to coumarin was R. dominica, S. zeamais and O. surinamensis.

The objective of this work was to evaluate the protective effect of different forms of insecticide application on the transmission of yellow dwarf disease in barley cultivars, as well as to determine the production costs and the net profit of these managements. The experiments were carried out during 2011 and 2012 growing seasons, using the following managements at main plots: T1, seed treatment with insecticide (ST) + insecticide on shoots at 15-day interval; T2, just ST; T3, insecticide applied on shoots, when aphid control level (CL) was reached; T4, without insecticide; and T5, ST + insecticide on shoots when CL was reached. Different barley cultivars - BRS Cauê, BRS Brau and MN 6021 - were arranged in the subplots. Insecticides lambda cyhalothrin (pyrethroid) and thiamethoxam (neonicotinoid) were used. There were differences on yellow dwarf disease index in both seasons for the different treatments, while damage to grain yield was influenced by year and aphid population. Production costs and net profit were different among treatments. Seed treatment with insecticide is sufficient to reduce the transmission of yellow dwarf disease in years with low aphid population pressure, while in years with larger populations, the application of insecticide on shoots is also required.

DDT and others organochlorine insecticides are very persistent substances. Clinical symptoms of intoxication have been reported in humans, although the main problem concerning such substances is bioaccumulation and biomagnification along throphic chains, leading to contamination of top predators and humans after them. In this review these characteristics are described, as well as some aspects of the control of vector borne diseases, like leishmaniasis and malaria, which were until recently, controlled by the health authorities using DDT.

Neste trabalho foram estudados dois sistemas utilizando reações de Fenton para a remediação de um solo contaminado com p,p´ DDT. O primeiro envolveu a formação de uma lama com o solo contaminado, na qual foram adicionados os reagentes de Fenton. No segundo, não houve a formação de lama e os reagentes foram adicionados diretamente ao solo por meio de um "tubo injetor". Foram avaliados alguns parâmetros que podem influenciar a reação de Fenton, tais como minerais de ferro naturalmente presentes no solo e a irradiação solar. A principal espécie mineral de ferro encontrada no solo é a hematita (Fe2O3), que teve pouca influência na degradação do contaminante. Nos experimentos com lama sem adição de ferro solúvel a porcentagem de degradação do p,p´ DDT foi inferior a 25% após 24 horas de reação. Nos ensaios com lama na presença de ferro solúvel, a degradação do p,p´ DDT atingiu 70% em 32 horas, enquanto que nos experimentos sem lama, a porcentagem de degradação foi de 32% no mesmo intervalo de tempo. Em ambos os sistemas, a irradiação solar teve pouca influência na degradação do contaminante. Os resultados indicam que para um processo de remediação deste solo, há necessidade de adição de ferro solúvel e que o sistema em lama resulta em maior eficiência de degradação de p,p´ DDT.

The organophosphorus insecticide Nuvacron (Monocrotophos) is a very toxic agent widely utilized in Brazilian agriculture. To evaluate the clastogenic potential of this insecticide, in vivo and in vitro micronucleus (MN) assay experiments were carried out on Swiss mice and on Chinese hamster ovary (CHO) cells, respectively. Nuvacron administered at doses of 2.5 and 5.0 mg/kg induced a statistically significant increase in the frequencies of MN detected in polychromatic bone marrow erythrocytes from animals (six/group) treated ip 24 h before. Exponentially growing CHAO cells were treated continuously (16h) with Nuvacron diluted in water to final concentrations of 1, 10, 100, 200, and 400 mug/ml. Three experiments were carried out using the cytokinesis-block method and a total of 6000 binucleated cells were scored to determine MN frequencies. A statistically significant increase in the frequencies of MN was observed for the cells treated with 1 and 10 mug/ ml Nuvacron. A marked decrease in cell proliferation rates was observed for CHO cultures treated with higher concentrations. These data demonstrate that Nuvacron has a genotoxic effect on both in vivo and in vitro mammalian test systems.

The cytokinesis-block micronucleus (CBMN) assay is one of the standard cytogenetic tools employed to assess chromosomal damage subsequent to exposure to genotoxic/cytotoxic agents, and is widely applicable to plant, animal and human cells. In the present study, the CBMN assay was used to assess the baseline damage in binuclear human peripheral blood lymphocytes exposed to 25 µg/L p,p'-DDT for 1, 2, 24, and 48 h by measuring the frequency of micronuclei, nucleoplasmic bridges and nuclear buds. These new scoring criteria facilitated the detection of different types of clastogenic and aneugenic effects induced by this type of pollutant. With these criteria, CBMN can also be used to measure nucleoplasmic bridges which are considered to be consequences of chromosome rearrangements and nuclear buds which are biomarkers of altered gene amplification and gene dosage. The total number of micronuclei observed in binuclear human peripheral blood lymphocytes of the exposed samples (ranging from 32 to 47) was significantly greater (P < 0.05) than that detected in the unexposed (0 time) control sample, where the total number of micronuclei was 7. The number of nucleoplasmic bridges and nuclear buds obtained after 24 and 48 h was also significantly (P < 0.05) greater in the samples treated with p,p'-DDT than in the unexposed control samples. Thus, our results confirmed the usefulness of the new criteria applicable for the CBMN assay employed in measuring the DNA damage and its role of a sensitive cytogenetic biomarker.

Development of new technologies, aiming at increasing productivity in different crops, involves constant research on the effectiveness and application of these techniques in seed treatment. In this study, it was aimed at evaluating physiological potential of rice seeds treated with plant growth-promoting rhizobacteria (PGPR) (strains DFs185, DFs223, DFs306, DFs416), or with two dosages of the insecticide thiamethoxam. The variables assessed were: germination (G); first count of germination (FCG); cold test (CT); length of seedlings aerial parts (LAP), root system (LRS), and total length (TL); emergence speed index (ESI); emergence (E), at 14 days; and dry phytomass (DP). Treatments have had a positive effect on percentage of G, FCG and E. The strain DFs185 has promoted increase in percentage emergence, for five of the six lots assessed. The variables: LAP; LRS; TL; ESI; and DP have undergone low or none influence of treatments; and there has been no toxic effect of rhizobacteria or insecticide thiamethoxam. In the cold test, a negative effect of treatments has been detected. Seed treatment with rhizobacteria, as well as with thiamethoxam, improve quality of low quality rice seeds. The strain DFs185 is promising for treating rice seeds, once it stimulates seed germination and emergence.

This study evaluated the effects on the development and predatory capacity of Podisus nigrispinus fed on Spodoptera frugiperda that have ingested different concentrations of neem oil. The predatory capacity of Podisus nigrispinus was assessed, separating nymphs (fourth instar) and adults (males and females). The treatments consisted of S. frugiperda larvae reared in neem oil aqueous solutions (0.077, 0.359 and 0.599%), deltamethrin EC 25 (0.100%) and control arranged in a completely randomized design, with ten replicates. Insects were offered three larval densities (one, three and six), in the third or fourth instars. The predated larvae were examined at 24 and 48 hours after the beginning of the experiment. Biological parameters of Podisus nigrispinus were evaluated in groups of ten second-instar nymphs transferred to pots, in five replicates. Insects were offered 2-6 third and/or fourth-instar larvae reared in the same neem oil concentrations in a completely randomized design. The following parameters were evaluated: duration of each nymph stage (days), nymph mortality (%), weight of fifth-instar nymphs (mg), sex ratio, weight of males and females (mg) and longevity of unfed adults (days). The predatory capacity of nymphs and adults of Podisus nigrispinus was influenced by the neem oil at the concentrations of 0.359% and 0.599% in the highest density. The concentration of 0.359% lengthened the nymphal stage and the concentration of 0.599% reduced the weight of males.

A experiência e os resultados obtidos em erradicação da malária pelo SEMPDC do Estado de São Paulo demonstraram que as medidas adotadas inicialmente e durante tôda a fase de ataque, foram adequadas de acôrdo com a extensão e situação epidemiológica de sua área malárica. No referente a fase de consolidação, foram sempre solucionados os problemas originados pela importação de casos e em algumas ocasiões, o restabelecimento da transmissão em pequenos focos. Atualmente, continua-se com o seguimento dos focos embora sejam inativos. O futuro do programa de erradicação da malária do Estado de São Paulo é favorável em virtude do progresso na cobertura total do programa de erradicação da malária federal. Também contribuirão para acelerar o desaparecimento dos casos de malária a cobertura com DDT em três ciclos anuais e a adoção do tratamento de cura radical "familiar" nas Ilhas situadas nos Rios Paraná e Grande, fronteiras com os Estados de Mato Grosso e Minas Gerais. Tendo em conta as possibilidades do Programa entrar em manutenção e solicitar à OMS em 1971 o Certificado de Malária Erradicada, serão aproveitados os anos de 1969 e 1970 para intensificar a busca de casos, a coordenação com os serviços de saúde, o estabelecimento de áreas de demonstração para integração desses serviços e a malária, e um aprimoramento das operações.

Com objetivo de evidenciar a freqüência e os níveis de resíduos de praguiddas organoclorados, foram analisadas 445 unidades amostrais de matéria prima (gordura bovina) provenientes de carcaças de bovinos de frigoríficos e 132 unidades amostrais de carne bovina já processada (enlatados, tais como "corned beef" e "roast beef"). Apresentaram-se com resíduos de praguicidas acima dos limites de tolerância estipulados, 77 unidades amostrais de gordura bovina e 5 de produto processado. Resíduos de praguiddas, sem contudo ultrapassar os limites de tolerância, foram identificados em, praticamente, todas as amostras, sendo que 27% das unidades amostrais de gordura bovina e 10,6% de produto processado apresentaram resíduos de praguicidas em violação aos limites estabelecidos na legislação. Heptacloro, BHC e Dieldrin foram os praguiddas mais freqüentes e Lindane e DDT os menos evidenciados.

Foram incluídos em rações comerciais, e dados a cães, os seguintes inseticidas: Phoxim, Chlorphoxim, Themephos (Abate), Decametrina, Lindano e DDT. A ação inseticida, no sangue, foi testada com ninfas e adultos de Triatoma infestans. Foram obtidos bons resultados com o Phoxim e o Themephos. O Chlorphoxim e o Lindano, nas doses eficientes, mostraram-se pouco palatáveis e a Decametrina e o DDT, nas concentrações aceitas pelos cães, não tiveram ação letal sobre os insetos.
Contaminación por compuestos organoclorados en algunos alimentos procedentes de una región de México

Fueran determinados los residuos de plaguicidas organoclorados en huevos y quesos de la población agricola de la Villa de Ahorne, Sinaloa, México, mediante cromatografia gas-líquido con detector de captura de eletrones. En los huevos, los compuestos detectados con mayor frecuencia y concentración fueran: epóxido de heptacloro, p, p'-DDT e p, p'-DDE. Simultáneamente fué realizado en la Villa de Ahorne un estudio epidemiológico preliminar en la cual se detectó varias manifestaciones patológicas en las personas entrevistadas y en sus familiares.

An alternative vector control method, using lambda-cyhalothrin impregnated wide-mesh gauze covering openings in the walls of the houses was developed in an area in the Eastern part of the interior of Suriname. Experimental hut observations showed that Anopheles darlingi greatly reduced their biting activity (99-100%) during the first 5 months after impregnation. A model assay showed high mortality both of mosquitoes repelled by the gauze as well as of those that succeeded in getting through it. A field application test in 270 huts showed good acceptance by the population and good durability of the applied gauze. After introducing the method in the entire working area, replacing DDT residual housespraying, the malaria prevalence, of 25-37% before application dropped and stabilized at between 5 and 10% within one year. The operational costs were less than those of the previously used DDT housespraying program, due to a 50% reduction in the cost of materials used. The method using widemesh gauze impregnated with lambdacyhalothrin strongly affects the behavior of An. darlingi. It is important to examine the effect of the method on malaria transmission further, since data indirectly obtained suggest substantial positive results.
