In this study a new approach, solid phase micro extraction (SPME), is used in the evaluation of the infinite dilution activity coefficient of the solute in a given solvent. It is the purpose of the current work to demonstrate a different approach to obtain the data needed for studying the solution thermodynamics of binary liquid mixtures as well as for designing multi-component separations. The solutes investigated at the temperature 298.15 K were toluene, ethyl benzene and xylene in the solvent methanol.

This paper proposes an alternate method to detect forbidden doping substances present in biological matrices of horseracing. The method was fully validated for caffeine, identified as the most frequent forbidden substance in the analysis conducted by the Antidoping Laboratory of the Brazilian Jockey Club, which adopts a zero threshold limit according to national and international horseracing practices. The metrological reliability of the method applied to toxicological analysis in biological matrices is discussed. Although the analytical method proposed for detection of a zero threshold level of the doping substance is qualitative, it was validated for the determination of the limiting value (also known as quantification limit value) introducing a criterion that prevents the issuing of incorrect results ("false-positives" and "false-negatives").

The chemical composition of the essential oils from leaves and fruits of Triphasia trifolia was analyzed by GC-FID and GC-MS. The major constituents of oil obtained from leaves were sabinene (35.4%) and myrcene (34.1%), while the prevalent compounds in oil from fruits were sabinene (37.2%), beta-pinene (23.95) and gamma-terpinene (16.3%). Both oils showed moderate antimicrobial activity. The fruit decoction was also investigated leading to the isolation of the coumarins isopimpinelin, (R)-byakangelicin and (S)-mexoticin. From leaves were isolated the coumarins (R)-byakangelicin, aurapten, (S)-mexoticin, isosibiricin, isomerazin and coumurrayin and the flavonoid vitexin. All coumarins showed cholinesterase inhibition on TLC tests.

The effect of microwave (MW) irradiation on the crystalline structure of two natural clays and one commercial clay, Montmorillonite K10, was analyzed comparing the X-ray diffraction, N2 isotherms, NMR-MAS of 27Al and 29Si spectra of the clays before and after MW irradiation. The preparation of dioxolane ketals of isatin was used to analyze the MW effect on the catalyst activation. The yields achieved using catalysts activated by MW irradiation were lower (2 to 5%) than the yields achieved using catalysts activated by heat in a conventional oven.

In this work we describe both a chromatographic purification procedure and a spot test for the enzyme peroxidase (POD: EC 1.11.1.7). The enzyme was obtained from crude extracts of sweet potatoes and the chromatographic enzyme purification procedure resulted in several fractions. Therefore a simple, fast and economic spot test for monitoring peroxidase during the purification procedure was developed. The spot test is based on the reaction of hydrogen peroxide and guaiacol, which is catalyzed by the presence of peroxidase yielding the colored tetraguaiacol.

Most of the snakebite incidents in the Amazon region involve Bothrops atrox, whose venom presents the most potent edematogenic and necrotic activities in the genus. This work describes the studies of isolation of the chemical constituents and antiedematogenic activity of the species Peltodon radicans (Lamiaceae), which is used in the treatment of snakebites and scorpion stings in the region. The extracts presented aliphatic hydrocarbons, 3beta-OH,beta-amirin (1), 3beta-OH,alpha-amirin (2), beta-sitosterol (3), stigmasterol (4), ursolic acid (5), 2alpha,3beta,19alpha-trihydroxy-urs-12-en-28-oic acid (tormentic acid, 6), methyl 3beta-hydroxy,28-methyl-ursolate (7), sitosterol-3-O-beta-D-glucopyranoside (8), and stigmasterol-3-O-beta-D-glucopyranoside (9). The flower extracts presented the higher antiedematogenic activity. This is the first report on the study of the flowers, stem, and roots of this plant.

The essential oils of leaves, aerial and underground stems of Senecio crassiflorus var. crassiflorus were extracted by hydrodistillation and their analyses were performed by GC/MS. Qualitative and quantitative differences have been found between the analyzed parts. The main compounds of the essentials oils of the leaves were found to be oxygenated sesquiterpenes, while the oils from the aerial and underground stems showed sesquiterpene hydrocarbons as major constituents. Furthermore, the essential oils were tested against Staphylocccus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus cereus. The essential oil of the leaves was shown to be the most active.

In this work, the effectiveness of four screening techniques (three techniques of the diffusion method and one microdilution broth method) were compared. Evaluated were the ethanolic and dichloromethanic extracts of Miconia rubiginosa (Melastomataceae) against six standard bacteria (ATCC). The results showed statistical disagreement among the three diffusion techniques. Among the diffusion techniques, the well technique displayed the best result. However the microdilution broth method demonstrated to be the most adequate method to evaluate the antibacterial activity of plant crude extracts and pure compounds when compared to the other methodologies.

The crude methanol extract and hexane, ethyl acetate, chloroform and methanol fractions from leaves of S. garckeana were examined in vitro for their antiproliferative activity on MCF-7, NCI-ADR, NCI-460, UACC-62, 786-0, OVCAR-03, PCO-3 and HT-29 human cancer cell lines. Among the assayed fractions, the ethyl acetate fraction showed to be cytotoxic against 786-0, UACC-62, OVCAR-03 and NCI-ADR cell lines with IC50 values of 12 µg/mL, 42 µg/mL, 53 µg/mL and 51 µg/mL, respectively. Through fractionation and isolation procedures compound (1) was obtained from the EtOAc fraction and its structure was elucidated by spectral techniques.

The natural quinones lapachol, α-lapachone and β-lapachone, and the synthetic derivative β-lapachone-3-sulfonic-acid were assayed for inhibition of fungal growth (Fusarium oxysporum) and germination of lettuce seeds (Lactuca sativa L.). β-Lapachone has the strongest activity as a germination inhibitor and lapachol shows no effect. β-Lapachone, followed by lapachol, are the most active in reducing fungal growth.

Kefir, a symbiont microorganism suspension, presents benefic effects to health. Some kefir grains were cultivated in brown sugar, allowing to isolate a substance named CSQ. This was evaluated on a biologic essay of mouse foot edema, presenting an inhibitory activity of 30+4 % against carrageenan after the stimulus. It was observed that a cultivation mean containing sucrose, and not the milky mean, lead to the production of different sugar polymeric chains of kefir. The results in vivo suggest that the CSQ exerted an anti-inflammatory activity.

Several reasons motivated the development of new generations of antibiotics, such as their high ability to develop resistance to virtually all kinds of anti-infective agents and the crescent market demand for new drugs to treat special demanding patients. After penicillin discovery, several antibiotics were developed from fungal metabolites, since antibacterial secondary metabolites consists on a fungal endogenous protective mechanism against natural competitors. The aim of this review is to present the structural diversity of antibacterial and antifungal metabolites produced by fungi, mentioning sources of fungal isolates, cultivation process and details on the scope of their antibiotic activity.

From the lichen Parmotrema lichexantonicum were isolated the depsidone salazinic acid, the xanthone lichexanthone, and the depside atranorin. The two major compounds, salazinic acid and lichexanthone, were selected for structure modifications. Salazinic acid afforded O-alkyl salazinic acids, some of them potentially cytotoxic against tumor cell lines (HCT-8, SF-295 and MDA/ MB - 435). From lichexanthone were obtained norlichexanthone, 3-O-methylnorlichexanthone, 3-O-methyl-6-O-prenylnorlichexanthone, 3,6-di-O-prenyl-norlichexanthone, 3,6-bis[(3,3-dimethyloxyran-2-il)methoxy]-1-hydroxy-8-methyl-9H-xanten-9-one and 3,6-bis[3-(dimethylamine)propoxy]-1-hydroxy-8-methyl-9H-xanten-9-one. The last compound was the most active against S. aureus.

Propolis is mostly used as hydroalcoholic extract. Recently there has been a growing number of patents dealing with new solvents for preparing propolis extracts. This study aimed to prepare edible oil propolis extracts and compare their chemical composition and biological activity with ethanolic propolis extracts. ESI-MS and spectrophotometric methods were used for qualitative and quantitative analyses, respectively. Antibacterial activity was evaluated by diffusion in agar. Cytotoxicity was tested by MTT assay using tumor cell lines. The oil is able to extract bioactive compounds from propolis. Further studies are needed to improve extraction efficiency and to characterize the active components.

Yeast cell wall contains polymers glucan and mannan-protein that have received much attention with respect to their biological activities. Conventional isolation process involving treatments with hot alkali and acids cause degradation of these polymers. The aim of this paper was to study a low-degrading process for the isolation of glucan and mannan-protein from S. cerevisiae cell wall comprising physic and enzymatic treatments. Yeast cell glucan was obtained in a purity of 87.4% and a yield of 33.7%. The isolated mannan-protein presented antioxidant activity that was increased after thirty minutes of protease treatment. Antioxidant activity was determined by β-carotene/linoleate model system.
