Pancreatic cancer is the fourth leading cause of cancer death. Gemcitabine is widely used as a chemotherapeutic agent for the treatment of pancreatic cancer, but the prognosis is still poor. Berberine, an isoquinoline alkaloid extracted from a variety of natural herbs, possesses a variety of pharmacological properties including anticancer effects. In this study, we investigated the anticancer effects of berberine and compared its use with that of gemcitabine in the pancreatic cancer cell lines PANC-1 and MIA-PaCa2. Berberine inhibited cell growth in a dose-dependent manner by inducing cell cycle arrest and apoptosis. After berberine treatment, the G1 phase of PANC-1 cells increased by 10% compared to control cells, and the G1 phase of MIA-PaCa2 cells was increased by 2%. Whereas gemcitabine exerts antiproliferation effects through S-phase arrest, our results showed that berberine inhibited proliferation by inducing G1-phase arrest. Berberine-induced apoptosis of PANC-1 and MIA-PaCa2 cells increased by 7 and 2% compared to control cells, respectively. Notably, berberine had a greater apoptotic effect in PANC-1 cells than gemcitabine. Upon treatment of PANC-1 and MIA-PaCa2 with berberine at a half-maximal inhibitory concentration (IC50), apoptosis was induced by a mechanism that involved the production of reactive oxygen species (ROS) rather than caspase 3/7 activation. Our findings showed that berberine had anti-cancer effects and may be an effective drug for pancreatic cancer chemotherapy.

As an essential trace element, copper can be toxic in mammalian cells when present in excess. Metallothioneins (MTs) are small, cysteine-rich proteins that avidly bind copper and thus play an important role in detoxification. YeastCUP1 is a member of the MT gene family. The aim of this study was to determine whether yeast CUP1 could bind copper effectively and protect cells against copper stress. In this study,CUP1 expression was determined by quantitative real-time PCR, and copper content was detected by inductively coupled plasma mass spectrometry. Production of intracellular reactive oxygen species (ROS) was evaluated using the 2',7'-dichlorofluorescein-diacetate (DCFH-DA) assay. Cellular viability was detected using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, and the cell cycle distribution of CUP1 was analyzed by fluorescence-activated cell sorting. The data indicated that overexpression of yeast CUP1 in HeLa cells played a protective role against copper-induced stress, leading to increased cellular viability (P<0.05) and decreased ROS production (P<0.05). It was also observed that overexpression of yeast CUP1 reduced the percentage of G1 cells and increased the percentage of S cells, which suggested that it contributed to cell viability. We found that overexpression of yeast CUP1 protected HeLa cells against copper stress. These results offer useful data to elucidate the mechanism of the MT gene on copper metabolism in mammalian cells.

As farinhas que são freqüentemente utilizadas em preparos industriais e domésticos de alimentos, podem vir a ser um bom veículo para contaminantes de vários tipos, inclusive as micotoxinas que por ventura não sejam eliminadas durante a moagem. Com o objetivo de avaliar a qualidade das farinhas de trigo (especial, comum, integral) e farinhas de centeio comercializadas na região sul do Rio Grande do Sul, nos anos de 1995 e 1996, foram determinadas a composição química e ocorrência de micotoxinas: aflatoxina B1, B2, G1, G2; ocratoxina A e zearalenona. A caracterização físico-química mostrou que 27% das farinhas comercializadas como especiais não se adequariam aos níveis recomendados pela legislação brasileira. O teste estatístico de contraste (SANEST), indicou que o conteúdo de cinzas pode explicar a presença de micotoxinas nas farinhas. As micotoxinas detectadas foram ocratoxina A e zearalenona, nos níveis de 12 e 53mg.kg-1 respectivamente.

Foi realizado um levantamento de características micotoxicológicas e microscópicas em arroz destinado ao consumo humano, comercializado nas cidades de Pelotas e Rio Grande, RS. As amostras, arroz integral [16], arroz parboilizado [16] e arroz branco polido [24] foram coletadas em supermercados, no período de maio a setembro de 2000. A avaliação de ocorrência de aflatoxinas B1, B2, G1, G2, ocratoxina A, zearalenona, desoxinivalenol e toxina T-2 foi realizada pela técnica de cromatografia de camada delgada (ccd). As duas últimas toxinas foram avaliadas também pela técnica de cromatografia gasosa (cg). A micoflora foi determinada através de plaqueamento em superfície em Ágar Batata Dextrose Acidificado, com incubação a 25ºC por 5 dias. As colônias mais freqüentes foram identificadas através da técnica de microcultivo. Dentre as amostras analisadas, duas apresentaram contaminação por ocratoxina A (104 e 128mig.Kg-1), três por zearalenona (559, 1117 e 1955mig.Kg-1), sendo uma co-contaminação com a primeira. Uma das amostras estava contaminada com desoxinivalenol (266 e 300mig.Kg-1), este quantificado por CCD e CG. Quanto à enumeração de bolores e leveduras, os resultados mostraram que 3,6% das amostras apresentaram valores superiores a 10 4 UFC.g -1 e 11% valores próximos deste. Sujidades microscópicas, acima dos limites da legislação (0,25%), foi detectada em uma amostra de arroz integral.

Tomatoes are highly susceptible to fungi contamination in the field, during transportation, processing, and storage. Aspergillus flavus and Aspergillus parasiticus have been isolated from tomatoes and tomato products, and both fungi species can produce aflatoxin, mycotoxin with hepatotoxic, carcinogenic, teratogenic, and mutagenic effects on all animal species tested so far. In order to verify a possible aflatoxin contamination of tomato products commercialized in Brazil, 63 samples of tomato products (pulp, paste, purée, ketchup, dehydrated tomatoes, and dried tomatoes preserved in oil) produced in 5 Brazilian states and 1 imported sample (ketchup), totalizing 29 brands, were analyzed by thin layer chromatography. The analytical method showed an average recovery of 86% for all aflatoxins at two spiking levels. The limits of detection for the aflatoxins B1, B2, G1, and G2 varied with the type of the product ranging from 2 to 7 µg/kg. Aflatoxins were not detected in any evaluated sample indicating that they did not pose a risk to human health since there was no invasion of raw materials by toxigenic fungi or no conditions for toxin production.

Bactérias ácido-láticas (BAL) constituem um grupo diverso, associado à produção de alimentos, incluindo fermentados de peixe. Por meio de processos fermentativos, é possível obter produtos mais saborosos e saudáveis. Estas bactérias também são importantes na medicina por suas atividades probióticas. Neste contexto, nove linhagens de bactérias ácido-láticas foram isoladas de sardinha e guaivira. Destas, duas linhagens foram caracterizadas fisiologicamente, juntamente com Leuconostoc mesenteroides ATCC8293. Lactobacillus homohiochii F3 apresentou ótimo crescimento a 40 ºC, diferente de Lactobacillus intestinalis G1 e Leuconostoc mesenteroides ATCC8293, que tiveram ótimo crescimento a 30 ºC. Em relação à tolerância ao NaCl, L. homohiochii F3 e L. mesenteroides ATCC8293 toleraram uma concentração de 5% de NaCl no meio. A espécie L. intestinalis G1 tolerou uma concentração de 7,5%, podendo ser considerada mais halotolerante que as demais espécies. L. mesenteroides ATCC8293, por causa do aumento da taxa de crescimento, pode ser considerada levemente halofílica. Na presença de concentrações não ótimas de NaCl, a adição de solutos orgânicos foi eficaz na restauração do balanço osmótico das células

The odor and taste profile of cocoa bean samples obtained from trees cultivated in southern Mexico were evaluated by trained panelists. Seven representative samples (groups) of a total of 45 were analyzed. Four attributes of taste (sweetness, bitterness, acidity and astringency), and nine of odor (chocolate, nutty, hazelnut, sweet, acidity, roasted, spicy, musty and off-odor) were evaluated. A sample (G7) with higher scores in sweet taste and sweet and nutty odors was detected, as well as a high association between these descriptors and the sample, analyzed through principal component analysis (PCA). Similarly, samples that showed high scores for non-desired odors in cocoas such as off-odor and musty were identified and related by PCA to roasted odor and astringent taste (G2 and G4). Based on this scores, the samples were listed in descending order by their sensory quality as G7> G5> G6> G3> G1> G4> G2.

AbstractLiterature has unveiled that a paper has not been published yet on using non-parametric stability statistics (NPSSs) for evaluating genotypic stability in dough properties of wheat. Accordingly, the effects of genotype (G), environment (E) and GE interaction (GEI) on alveograph parameters, i.e. dough baking strength (W) and its tenacity (P)/extensibility (L), of 18 wheat (T. aestivum L.) genotypes were studied under irrigated field conditions in an 8-year trial (2006-2014) in central Turkey. Furthermore, genotypic stability for W and P/L was determined using 8 NPSSs viz. RM-Rank mean, RSD-Rank’s standard deviation, RS-Rank Sum, TOP-Ranking, Si(1), Si(2), Si(3) and Si(6) rank statistics. The ANOVA revealed that W and P/L were primarily controlled by E, although G and GEI also had significant effects. Among the 8 NPSSs, only RM, RS and TOP statistics were suitable for detecting the genotypes with high stable and bread making quality (e.g. G1 and G17). In conclusion, using RM, RS and TOP statistics is advisable to select for dough quality in wheat under multi-environment trials (METs).

Introdução: A doença renal crônica (DRC) interfere diretamente na capacidade funcional, na independência e, consequentemente, na qualidade de vida (QV). Objetivo: Comparar a capacidade funcional e a qualidade de vida de doentes renais crônicos em hemodiálise (G1) e pré-dialíticos (G2). Métodos: Estudo transversal descritivo, 54 pacientes com DRC, 27 do G1 (58,15 ± 10,84 anos) e 27 do G2 (62,04 ± 16,56 anos). Verificaramse os fatores de risco cardiovasculares, medidas antropométricas, força muscular respiratória verificada por meio da pressão inspiratória (PImax) e expiratória (PEmax) máximas, teste de caminhada de seis minutos (TC6'), teste cardiopulmonar de exercício, teste de sentar e levantar de um minuto (TSL1') e o Short-Form Questionary (SF-36) para avaliar a QV. Os pacientes apresentavam estadiamento da doença entre 2 a 5. Realizou-se o teste de normalidade Kolmogorov-Smirnov e utilizou-se o teste t (Student) ou o teste U (Mann Whitney) para a comparação das médias das variáveis quantitativas e o teste de Quiquadrado de Pearson e exato de Fischer para as variáveis qualitativas. Para identificar as correlações, foi utilizado o teste de Pearson ou de Spearman. Resultados: Não foi encontrada diferença estatisticamente significativa entre G1 e G2, no VO2pico (p = 0,259), no TC6' (p = 0,433), na PImax (p = 0,158) e somente foi encontrada diferença na PEmax (p = 0,024) para G1. Os escores do questionário SF-36 mostram em ambos os grupos um pior estado de saúde evidenciada pela pontuação baixa nos escores de QV. Conclusão: Os pacientes com DRC apresentaram reduzida capacidade funcional e QV, sendo que a hemodiálise não demonstrou estatisticamente ter repercussão negativa quando comparados com os pacientes pré-dialíticos.
