Mit Hilfe des zeitgenössischen Romans 'The Circle' von Dave Eggers werden Entgrenzungs- und Verdichtungstendenzen moderner subjektivierter Arbeit illustriert, die einen Zusammenhang mit Burnout- und Erschöpfungsdepressionsdiagnosen plausibilisieren. Angesichts der Uneindeutigkeit der empirischen Evidenz wird vorgeschlagen, die Problematik der modernen Gestalt der Erwerbsarbeit – vor allem für diejenigen am unteren Ende der jeweiligen Hierarchien – nicht nur anhand ihrer Krankheitsträchtigkeit, sondern mit Hilfe des Konzepts der Lebensformen und ihrer normativen Aufladung zu kritisieren: Massiv entgrenzte und verdichtete Arbeit erscheint dann als Bruch des 'Arbeitsversprechens', das sich mit Entstehung der Arbeitsgesellschaft im 19. Jh. herausgebildet hat.

The armadillo family protein plakoglobin (Pg) is a well-characterized component of anchoring junctions, where it functions to mediate cell-cell adhesion and maintain epithelial tissue integrity. Although its closest homolog beta-catenin acts in the Wnt signaling pathway to dictate cell fate and promote proliferation and survival, the role of Pg in these processes is not well understood. Here, we investigate how Pg affects the survival of mouse keratinocytes by challenging both Pg-null cells and their heterozygote counterparts with apoptotic stimuli. Our results indicate that Pg deletion protects keratinocytes from apoptosis, with null cells exhibiting delayed mitochondrial cytochrome c release and activation of caspase-3. Pg-null keratinocytes also exhibit increased messenger RNA and protein levels of the anti-apoptotic molecule Bcl-X(L) compared to heterozygote controls. Importantly, reintroduction of Pg into the null cells shifts their phenotype towards that of the Pg+/- keratinocytes, providing further evidence that Pg plays a direct role in regulating cell survival. Taken together, our results suggest that in addition to its adhesive role in epithelia, Pg may also function in contrast to the pro-survival tendencies of beta-catenin, to potentiate death in cells damaged by apoptotic stimuli, perhaps limiting the potential for the propagation of mutations and cellular transformation.Journal of Investigative Dermatology advance online publication, 16 November 2006; doi:10.1038/sj.jid.5700615.

Die Arbeit stellt sich in die Reihe von Studien, die den Status des lyrischen Subjekts innerhalb der Dichtung „nach Auschwitz“ untersucht haben, versucht aber selbst einen neuen Weg einzuschlagen, indem sie die Problematik der (historischen, ethischen) Subjektbestimmung aus ihrer rhetorischen und linguistischen Natur herleitet. Durch detaillierte formale Lektüre wird die selbstreflexive Funktion der Personalpronomina in den Sonetten Shakespeares sowie in der Übersetzung Celans analysiert. Die übersetzungstheoretischen Implikationen der Selbstreflexivität werden vor allem in Bezug auf die Werke von Antoine Berman und Henri Meschonnic erklärt. Präzise identifiziert und deutet der Verfasser syntaktische, grammatikalische, rhetorische und metrische Merkmale der untersuchten Gedichte, um Celans Verfahren en détail zu beschreiben: Tempus und Modus, Enjambement und Zäsur, Synkope und Anakoluth, Katapher und Chiasmus, Satz- und Ausführungszeichen, Kursivschrift und graphische Elemente. Theoretisch stützt sich die Arbeit auf bestimmte Hauptbegriffe von Jean Bollack (Virtualität, Poetik der Fremdheit, Trennung von den Mitmenschen, Schreiben auf seiten des Todes) und Gilles Deleuze (Virtualität, Differenz und Repetition, Ordnung der leeren Zeit). Der Begriff von Virtualität fungiert in diesem Zusammenhang als trait d'union zwischen den beiden Autoren, in dem Versuch, ihre unterschiedlichen, teilweise entgegengesetzten Positionen in einem Punkt konvergieren zu lassen: der Neudefinition eines gespaltenen Subjektes und seines subversiven Potentials in einem direkten politischen Sinne. Diese Neudefinition der pronominalen Funktionen und die ihres politischen Widerstands werden darüber hinaus als übersetzungstheoretische Kriterien in der Interpretation der Nationalliteratur und der Rezeptionsgeschichte in Deutschland verwendet. Aus dieser Perspektive wird der theoretische Rahmen dieser Arbeit, nach den Hauptfiguren von Bollack und Deleuze, von Foucault Auffassung der Historizität und des politischen Werts des Subjektbegriffs ergänzt. Die Poetik der Übersetzung wird als Vorgang der “Wiederholung” konzipiert, der letztendlich eine “Differenz” hervorbringt, wie auch eine Neubestimmung der Zeitlichkeit als nicht-lineare Erscheinung.

Purpose To evaluate the degree of psychological distress in adult childhood cancer survivors in Switzerland and to characterize survivors with significant distress. Methods Childhood cancer survivors who were age younger than 16 years when diagnosed between 1976 and 2003, had survived more than 5 years, and were currently age 20 years or older received a postal questionnaire. Psychological distress was assessed using the Brief Symptom Inventory (BSI). Raw scores were transformed into T scores according to the German norm sample, and the proportion of participants being at increased risk for psychological distress was calculated (case rule: T ≥ 63). t tests and univariable and multivariable logistic regressions were used for statistical analyses. Results One thousand seventy-six survivors (63.% of eligible survivors, 71.9% of contacted survivors) returned the questionnaire, 987 with complete data on BSI. Comparison with the norm populations showed lower T scores (T < 50) in the Global Severity Index (GSI; T = 46.2), somatization (T = 47.6), obsessive-compulsive tendencies (T = 46.9), and anxiety (T = 48.4). However, more childhood cancer survivors (especially women) had increased distress for GSI (14.4%), interpersonal sensitivity (16.5%), depression (13.4%), aggression (16.9%), and psychotic tendencies (15.6%) than the expected 10% from the norm population. Caseness was associated with female sex, being a single child, older age at study, and self-reported late effects, especially psychological problems. Conclusion Results show that childhood cancer survivors, on average, have less psychological distress than a norm population but that the proportion of survivors at risk for high psychological distress is disproportionally large. Monitoring psychological distress in childhood cancer survivors may be desirable during routine follow-up, and psychological support should be offered as needed.

The hERG voltage-gated potassium channel mediates the cardiac I(Kr) current, which is crucial for the duration of the cardiac action potential. Undesired block of the channel by certain drugs may prolong the QT interval and increase the risk of malignant ventricular arrhythmias. Although the molecular determinants of hERG block have been intensively studied, not much is known about its stereoselectivity. Levo-(S)-bupivacaine was the first drug reported to have a higher affinity to block hERG than its enantiomer. This study strives to understand the principles underlying the stereoselectivity of bupivacaine block with the help of mutagenesis analyses and molecular modeling simulations. Electrophysiological measurements of mutated hERG channels allowed for the identification of residues involved in bupivacaine binding and stereoselectivity. Docking and molecular mechanics simulations for both enantiomers of bupivacaine and terfenadine (a non-stereoselective blocker) were performed inside an open-state model of the hERG channel. The predicted binding modes enabled a clear depiction of ligand-protein interactions. Estimated binding affinities for both enantiomers were consistent with electrophysiological measurements. A similar computational procedure was applied to bupivacaine enantiomers towards two mutated hERG channels (Tyr652Ala and Phe656Ala). This study confirmed, at the molecular level, that bupivacaine stereoselectively binds the hERG channel. These results help to lay the foundation for structural guidelines to optimize the cardiotoxic profile of drug candidates in silico.

Background The evaluation of the hepatic parenchyma in patients with chronic liver disease is important to assess the extension, localization and relationship with adjacent anatomical structures of possible lesions. This is usually performed with conventional abdominal ultrasound, CT-scan or magnetic resonance imaging. In this context, the feasibility and the safety of intravascular ultrasound in the liver have not been assessed yet. Methods We tested the safety and performance of an intracardiac echography (ICE) catheter applied by a transjugular approach into the hepatic veins in patients with chronic liver disease undergoing hepatic hemodynamic measurements. Results Five patients were enrolled in this pilot study. The insertion of the ICE catheter was possible into the right and middle, but not into the left hepatic vein. The position of the ICE was followed using fluoroscopy and external conventional ultrasound. Accurate imaging of focal hepatic parenchymal lesions, Doppler ultrasound of surrounding blood vessels and assessment of liver surface and ascites were achieved without complications. Conclusions This study demonstrated that a diagnostic approach using an ICE device inserted in the hepatic veins is feasible, safe and well tolerated. However, it remains for the moment only an experimental investigative tool. Whether ICE adds further information regarding parenchymal lesions and associated vascular alterations as compared to other techniques, needs additional investigation.

In this paper, we use morphological and numerical methods to test the hypothesis that seasonally formed fracture patterns in the Martian polar regions result from the brittle failure of seasonal CO2 slab ice. The observations by the High Resolution Imaging Science Experiment (HiRISE) of polar regions of Mars show very narrow dark elongated linear patterns that are observed during some periods of time in spring, disappear in summer and re-appear again in the following spring. They are repeatedly formed in the same areas but they do not repeat the exact pattern from year to year. This leads to the conclusion that they are cracks formed in the seasonal ice layer. Some of models of seasonal surface processes rely on the existence of a transparent form of CO2 ice, so-called slab ice. For the creation of the observed cracks the ice is required to be a continuous media, not an agglomeration of relatively separate particles like a firn. The best explanation for our observations is a slab ice with relatively high transparency in the visible wavelength range. This transparency allows a solid state green-house effect to act underneath the ice sheet raising the pressure by sublimation from below. The trapped gas creates overpressure and the ice sheet breaks at some point creating the observed cracks. We show that the times when the cracks appear are in agreement with the model calculation, providing one more piece of evidence that CO2 slab ice covers polar areas in spring.
