Ausgangslage und Fragestellung In der Forschung zum Selbstgespräch im Sport dominiert die Frage nach dessen Auswir-kungen auf die sprechende Person selber. Gemäss Diaz (1992) besitzt das offene Selbstgespräch aber neben der individuell-regulatorischen auch eine sozial-kommunikative Funktion. Diese sozial-kommunikative Funktion und der damit verbunde-ne Einfluss des Selbstgesprächs auf Beobachter wurden in der Sportwissenschaft bisher nur marginal untersucht (z.B. Gould & Weiss, 1981; Van Raalte, Brewer, Cornelius & Pe-titpas, 2006). Im angestrebten Forschungsvorhaben sollen zwei Fragen geklärt werden: Steht das of-fene, während dem Wettkampf geäusserte Selbstgespräch eines Athleten in Zusammen-hang mit dessen Selbstdarstellung? Beeinflusst das offene Selbstgespräch eines Athle-ten den Dyadenpartner und wie sie sich gegenseitig wahrnehmen? Methode Es werden Doppelpartien im Badminton per Videokamera aufgezeichnet. Mittels einer Korrelationsanalyse soll überprüft werden, ob die so erhobenen Selbstgesprächsaussa-gen mit dem Persönlichkeitsmerkmal „Selbstdarstellung“ der Probanden zusammenhän-gen. Nach Spielschluss werden die Probanden entsprechend eines video-stimulierten Fremdkonfrontationsinterviews mit Spielausschnitten konfrontiert, die offenes Selbstge-spräch ihres Dyadenpartners enthalten. Sie werden dabei nach ihren Kognitionen sowie Emotionen gefragt, die sie während der entsprechenden Spielsituation erlebten. Aktuelle Fragen Welcher Fragebogen zur Erhebung des Persönlichkeitsmerkmals „Selbstdarstellung“ soll Verwendung finden? Zur Auswahl steht ein noch zu übersetzender, validierter, engli-scher und sportspezifischer Fragebogen und die Verwendung eines deutschen Frage-bogens, der habituelle Selbstdarstellungstechniken im Allgemeinen erhebt. Wie soll beim video-stimulierten Fremdkonfrontationsinterview vorgegangen werden? Werden den Probanden z.B. das ganze Spiel oder nur ausgewählte Ausschnitte gezeigt, in denen offenes Selbstgespräch vorkommt? Wie zeitnah muss das Interview durchge-führt werden? Literatur Diaz, R. OBJECT: The aim of this study was to develop and characterize a new orthotopic, syngeneic, transplantable mouse brain tumor model by using the cell lines Tu-9648 and Tu-2449, which were previously isolated from tumors that arose spontaneously in glial fibrillary acidic protein (GFAP)-v-src transgenic mice. METHODS: Striatal implantation of a 1-microl suspension of 5000 to 10,000 cells from either clone into syngeneic B6C3F1 mice resulted in tumors that were histologically identified as malignant gliomas. Prior subcutaneous inoculations with irradiated autologous cells inhibited the otherwise robust development of a microscopically infiltrating malignant glioma. Untreated mice with implanted tumor cells were killed 12 days later, when the resultant gliomas were several millimeters in diameter. Immunohistochemically, the gliomas displayed both the astroglial marker GFAP and the oncogenic form of signal transducer and activator of transcription-3 (Stat3). This form is called tyrosine-705 phosphorylated Stat3, and is found in many malignant entities, including human gliomas. Phosphorylated Stat3 was particularly prominent, not only in the nucleus but also in the plasma membrane of peripherally infiltrating glioma cells, reflecting persistent overactivation of the Janus kinase/Stat3 signal transduction pathway. The Tu-2449 cells exhibited three non-random structural chromosomal aberrations, including a deletion of the long arm of chromosome 2 and an apparently balanced translocation between chromosomes 1 and 3. The GFAP-v-src transgene was mapped to the pericentromeric region of chromosome 18. CONCLUSIONS: The high rate of engraftment, the similarity to the high-grade malignant glioma of origin, and the rapid, locally invasive growth of these tumors should make this murine model useful in testing novel therapies for human malignant gliomas.

The CopA copper ATPase of Enterococcus hirae belongs to the family of heavy metal pumping CPx-type ATPases and shares 43% sequence similarity with the human Menkes and Wilson copper ATPases. Due to a lack of suitable protein crystals, only partial three-dimensional structures have so far been obtained for this family of ion pumps. We present a structural model of CopA derived by combining topological information obtained by intramolecular cross-linking with molecular modeling. Purified CopA was cross-linked with different bivalent reagents, followed by tryptic digestion and identification of cross-linked peptides by mass spectrometry. The structural proximity of tryptic fragments provided information about the structural arrangement of the hydrophilic protein domains, which was integrated into a three-dimensional model of CopA. Comparative modeling of CopA was guided by the sequence similarity to the calcium ATPase of the sarcoplasmic reticulum, Serca1, for which detailed structures are available. In addition, known partial structures of CPx-ATPase homologous to CopA were used as modeling templates. A docking approach was used to predict the orientation of the heavy metal binding domain of CopA relative to the core structure, which was verified by distance constraints derived from cross-links. The overall structural model of CopA resembles the Serca1 structure, but reveals distinctive features of CPx-type ATPases. A prominent feature is the positioning of the heavy metal binding domain. It features an orientation of the Cu binding ligands which is appropriate for the interaction with Cu-loaded metallochaperones in solution. Moreover, a novel model of the architecture of the intramembranous Cu binding sites could be derived.

Family change theory suggests three ideal-typical family models characterized by different combinations of emotional and material interdependencies in the family. Its major proposition is that in economically developing countries with a collectivistic background a family model of emotional interdependence emerges from a family model of complete interdependence. The current study aims to identify and compare patterns of family-related value orientations related to family change theory across three cultures and two generations. Overall, N = 919 dyads of mothers and their adolescent children from Germany, Turkey, and India participated in the study. Three clusters were identified representing the family models of independence, interdependence, and emotional interdependence, respectively. Especially the identification of an emotionally interdependent value pattern using a person-oriented approach is an important step in the empirical validation of family change theory. The preference for the three family models differed across as well as within cultures and generations according to theoretical predictions. Dyadic analyses pointed to substantial intergenerational similarities and also to differences in family models, reflecting both cultural continuity as well as change in family-related value orientations.

Purpose: Proper delineation of ocular anatomy in 3D imaging is a big challenge, particularly when developing treatment plans for ocular diseases. Magnetic Resonance Imaging (MRI) is nowadays utilized in clinical practice for the diagnosis confirmation and treatment planning of retinoblastoma in infants, where it serves as a source of information, complementary to the Fundus or Ultrasound imaging. Here we present a framework to fully automatically segment the eye anatomy in the MRI based on 3D Active Shape Models (ASM), we validate the results and present a proof of concept to automatically segment pathological eyes. Material and Methods: Manual and automatic segmentation were performed on 24 images of healthy children eyes (3.29±2.15 years). Imaging was performed using a 3T MRI scanner. The ASM comprises the lens, the vitreous humor, the sclera and the cornea. The model was fitted by first automatically detecting the position of the eye center, the lens and the optic nerve, then aligning the model and fitting it to the patient. We validated our segmentation method using a leave-one-out cross validation. The segmentation results were evaluated by measuring the overlap using the Dice Similarity Coefficient (DSC) and the mean distance error. Results: We obtained a DSC of 94.90±2.12% for the sclera and the cornea, 94.72±1.89% for the vitreous humor and 85.16±4.91% for the lens. The mean distance error was 0.26±0.09mm. The entire process took 14s on average per eye. Conclusion: We provide a reliable and accurate tool that enables clinicians to automatically segment the sclera, the cornea, the vitreous humor and the lens using MRI. We additionally present a proof of concept for fully automatically segmenting pathological eyes. This tool reduces the time needed for eye shape delineation and thus can help clinicians when planning eye treatment and confirming the extent of the tumor.
