Noninvasive molecular-imaging technologies are playing a keyrole in drug discovery, development and delivery. Positron Emission Tomography (PET) is such a molecular imaging technology and a powerful tool for the observation of various diseases. However, it is limited by the availability of agents with high selectivity to the target and a physical half-life of the used positron emitting nuclide which matches the biological half-life of the observed process. For the long lasting enrichment of antibodies in tumor tissue few suitable isotopes for PET imaging are currently available. The element arsenic provides a range of isotopes, which could be used for diagnosis and also for endoradiotherapy. This work describes the development of radiochemical separation procedures to separate arsenic isotopes in no-carrier-added (nca) purity from reactor or cyclotron irradiated targets, the development and evaluation of a labeling chemistry to attach these separated arsenic isotopes to monoclonal antibodies, the in vitro and in vivo evaluation of antibodies labeled with radioactive arsenic isotopes and the molecular imaging using small animal PET.

The aim of this work presented here is the characterization of structure and dynamics of different types of supramolecular systems by advanced NMR spectroscopy. One of the characteristic features of NMR spectroscopy is based on its high selectivity. Thus, it is desirable to exploit this technique for studying structure and dynamics of large supramolecular systems without isotopic enrichment. The observed resonance frequencies are not only isotope specific but also influenced by local fields, in particular by the distribution of electron density around the investigated nucleus. Barbituric acid are well known for forming strongly hydrogen-bonded complexes with variety of adenine derivatives. The prototropic tautomerism of this material facilitates an adjustment to complementary bases containing a DDA(A = hydrogen bond acceptor site, D = hydrogen bond donor site) or ADA sequences, thereby yielding strongly hydrogen-bonded complexes. In this contribution solid-state structures of the enolizable chromophor "1-n-butyl-5-(4-nitrophenyl)-barbituric acid" that features adjustable hydrogen-bonding properties and the molecular assemblies with three different strength of bases (Proton sponge, adenine mimetic 2,6-diaminopyridine (DAP) and 2,6-diacetamidopyridine (DAC)) are studied. Diffusion NMR spectroscopy gives information over such interactions and has become the method of choice for measuring the diffusion coefficient, thereby reflecting the effective size and shape of a molecular species. In this work the investigation of supramolecular aggregates in solution state by means of DOSY NMR techniques are performed. The underlying principles of DOSY NMR experiment are discussed briefly and more importantly two applications demonstrating the potential of this method are focused on. Calix[n]arenes have gained a rather prominent position, both as host materials and as platforms to design specific receptors. In this respect, several different capsular contents of tetra urea calix[4]arenes (benzene, benzene-d6, 1-fluorobenzene, 1-fluorobenzene-d5, 1,4-difluorobenzene, and cobaltocenium) are studied by solid state NMR spectroscopy. In the solid state, the study of the interaction between tetra urea calix[4]arenes and guest is simplified by the fact that the guests molecule remains complexed and positioned within the cavity, thus allowing a more direct investigation of the host-guest interactions.

Positron Emission Tomography (PET) is such a molecular imaging technology and a powerful tool for the observation of various deceases in vivo. However, it is limited by the availability of vectors with high selectivity to the target and radionuclides with a physical half-life which matches the biological half-life of the observed process. The 68Ge/68Ga radionuclide generator makes the PET-nuclide anywhere available without an on-site cyclotron. Besides the perfect availability 68Ga shows well suited nuclide properties for PET, but it has to be co-ordinated by a chelator to introduce it in a radiopharmaceuticals.rnHowever, the physical half-life of 68Ga (67.7 min) might limit the spectrum of clinical applications of 68Ga-labelled radiodiagnostics. Furthermore, 68Ga-labelled analogues of endoradiotherapeuticals of longer biological half-live such as 90Y- or 177Lu-labeled peptides and proteins cannot be used to determine individual radiation dosimetry directly. rnThus, radionuclide generator systems providing positron emitting daughters of extended physical half-life are of renewed interest. In this context, generator-derived positron emitters with longer physical half-life are needed, such as 72As (T½ = 26 h) from the 72Se/72As generator, or 44Sc (T½ = 3.97 h) from the 44Ti/44Sc generator.rnIn this thesis the implementation of radioactive gallium-68 and scandium-44 for molecular imaging and nuclear medical diagnosis, beginning with chemical separation and purification of 44Ti as a radionuclide mother, investigation of pilot generators with different elution mode, building a prototype generator, development and investigation of post-processing of the generator eluate, its concentration and further purification, the labeling chemistry under different conditions, in vitro and in vivo studies of labeled compounds and, finally, in vivo imaging experiments are described.

In dieser Arbeit wurde die Methode der resonanten Ionisation von neutralen Atomen mittels Laserstrahlung auf die leichten Aktinide Thorium, Uran, Neptunium und Plutonium angewendet und für die Ultraspurenanalyse optimiert. Der empfindliche Nachweis dieser Aktinide stellt eine Herausforderung für die Beobachtung und Bestimmung von radioaktiven Verunreinigungen aus kerntechnischen Anlagen in der Umwelt dar. In einem für diese Untersuchungen entwickelten Quadrupolmassenspektrometer konnte durch Resonanzionisationsspektroskopie jeweils eine Reihe unbekannter Energiezustände in der Elektronenhülle des neutralen Atoms der oben genannten Aktinide identifiziert, sowie effiziente Anregungsschemata für die resonante Ionisation entwickelt und charakterisiert werden. Durch die verwendete in-source-Ionisation, die aufgrund der guten Überlagerung von Laserstrahlung und Atomstrahl eine hohe Nachweiseffizienz gewährleistet, konnten diese Untersuchungen bereits mit einem, für Radionuklide notwendigen, geringen Probeneintrag erfolgen. Die resonante Ionisation erlaubt durch die selektiven resonanten Prozesse eine Unterdrückung unerwünschter Kontaminationen und wurde auf den analytischen Nachweis von Ultraspurengehalten in Umweltproben, sowie die Bestimmung der entsprechenden Isotopenzusammensetzung optimiert. Durch die effiziente in-source-Ionisation mit leistungsstarker gepulster Laserstrahlung, konnten Nachweiseffizienzen im Bereich von bis zu 1% erreicht werden. Dabei wurden für Plutonium in synthetischen Proben, aber auch in ersten Umweltproben, Nachweisgrenzen von 10^4-10^5 Atomen erzielt. Die Verwendung spektral schmalbandiger Dauerstrichlaser und eine Ionisation transversal zum frei propagierenden Atomstrahl ermöglicht durch Auflösung der Isotopieverschiebung eine hohe Selektivität gegenüber dominanten Nachbarisotopen, wohingegen die Ionisationseffizienz deutlich abnimmt. Hiermit konnte für das Ultraspurenisotop U-236 eine Nachweisgrenze bis hinab zu 10^-9 für das Isotopenverhältnis N(U-236)/N(U-238) bestimmt werden.

Membrane proteins play an indispensable role in physiological processes. It is, therefore, not surprising that many diseases are based on the malfunction of membrane proteins. Hence membrane proteins and especially G-protein coupled receptors(GPCRs)- the largest subfamily- have become an important drug target. Due to their high selectivity and sensitivity membrane proteins are also feasible for the detection of small quantities of substances with biosensors. Despite this widespread interest in GPCRs due to their importance as drug targets and biosensors there is still a lack of knowledge of structure, function and endogenous ligands for quiet a few of the previously identified receptors.rnBottlenecks in over-expression, purification, reconstitution and handling of membrane proteins arise due to their hydrophobic nature. Therefore the production of reasonable amounts of functional membrane proteins for structural and functional studies is still challenging. Also the limited stability of lipid based membrane systems hampers their application as platforms forrnscreening applications and biosensors.rnIn recent years the in vitro protein synthesis became a promising alternative to gain better yields for expression of membrane proteins in bio-mimetic membrane systems. These expression systems are based on cell extracts. Therefore cellular effects on protein expression are reduced. The open nature of the cell-free expression systems easily allows for the adjustment of reactionrnconditions for the protein of interest. The cell-free expression in the presence of bio-mimetic membrane systems allows the direct incorporation of the membrane proteins and therefore skips the time-consuming purification and reconstitution processes. Amphiphilic block-copolymers emerged as promising alternative for the less stable lipid-based membrane systems. They, likernlipids, form membraneous structures in aqueous solutions but exhibit increased mechanical and chemical stability.rnThe aim of this work was the generation of a GPCR-functionalised membrane system by combining both promising alternatives: in vitro synthesis and polymeric membrane systems. This novel platform should be feasible for the characterisation of the incorporated GPCR. Immunodetection of Dopamine receptor 1 and 2 expressed in diblock- and triblock-polymersomes demonstrated the successful in vitro expression of GPCRs in polymeric membranes. Antibodyrnbinding studies suggested a favoured orientation of dopamine receptors in triblockpolymersomes.rnA dopamine-replacement assay on DRD2-functionalised immobilised triblockpolymersomes confirmed functionality of the receptor in the polymersomes. The altered binding curve suggests an effect of the altered hydrophobic environment presented by the polymer membrane on protein activity.

Zur Entwicklung einer selektiven Tumorimmuntherapie wurden im Rahmen dieser Dissertation potentielle Vakzine dargestellt. Als Leitstruktur für die Impfstoffkandidaten diente das Oberflächenglycoprotein MUC1, das durch fehlregulierte Enzymaktivitäten auf malignen Zellen strukturell verändert überexprimiert wird. Hierbei wurde insbesondere der Einfluss von Fluorsubstituenten in der Glycanseitenkette auf die Immunogenität und die Spezifität der Vakzine untersucht. Dazu wurde ein dreifach fluoriertes Analogon an Tetanus Toxoid (TTox) als Trägerprotein angebunden und in Immunisierungsstudien an transgenen Mäusen konnten hohe Selektivitäten und starke Immunantworten nachgewiesen werden. Die Darstellung des 20 Aminosäuren umfassenden Glycopeptides erfolgte an der festen Phase, wobei unterschiedlich fluorierte Thomsen-Friedenreich-Antigenanaloga anstelle eines Threonins in die Sequenz eingebaut wurden. Diese Bausteine wurden durch Glycosylierungsreaktionen fluorierter Donoren sowie Akzeptoren hergestellt, wobei Letztere durch nachträgliche Fluorierungen der Glycosylaminosäure erhalten wurden. Darüber hinaus wurde die Durchführung der komplexen Glycosylierungsreaktionen in Mikroreaktoren am Beispiel zweier fluorierter Glycosylaminosäuren untersucht. Neben der raschen Optimierung der Reaktionsparameter in Durchflussreaktoren konnte die direkte Umsetzung in einem mikrostrukturierten Reaktor in den präparativen Maßstab demonstriert werden.

In this present work high quality PMMA opals with different sphere sizes, silica opals from large size spheres, multilayer opals, and inverse opals were fabricated. Highly monodisperse PMMA spheres were synthesized by surfactant-free emulsion polymerization (polydispersity ~2%). Large-area and well-ordered PMMA crystalline films with a homogenous thickness were produced by the vertical deposition method using a drawing device. Optical experiments have confirmed the high quality of these PMMA photonic crystals, e.g., well resolved high-energy bands of the transmission and reflectance spectra of the opaline films were observed. For fabrication of high quality opaline photonic crystals from large silica spheres (diameter of 890 nm), self-assembled in patterned Si-substrates a novel technique has been developed, in which the crystallization was performed by using a drawing apparatus in combination with stirring. The achievements comprise a spatial selectivity of opal crystallization without special treatment of the wafer surface, the opal lattice was found to match the pattern precisely in width as well as depth, particularly an absence of cracks within the size of the trenches, and finally a good three-dimensional order of the opal lattice even in trenches with a complex confined geometry. Multilayer opals from opaline films with different sphere sizes or different materials were produced by sequential crystallization procedure. Studies of the transmission in triple-layer hetero-opal revealed that its optical properties cannot only be considered as the linear superposition of two independent photonic bandgaps. The remarkable interface effect is the narrowing of the transmission minima. Large-area, high-quality, and robust photonic opal replicas from silicate-based inorganic-organic hybrid polymers (ORMOCER® s) were prepared by using the template-directed method, in which a high quality PMMA opal template was infiltrated with a neat inorganic-organic ORMOCER® oligomer, which can be photopolymerized within the opaline voids leading to a fully-developed replica structure with a filling factor of nearly 100%. This opal replica is structurally homogeneous, thermally and mechanically stable and the large scale (cm2 size) replica films can be handled easily as free films with a pair of tweezers.
