The aim of this thesis is to detect the phylogeny and the population dynamics of the European termites of the genera Reticulitermes and Kalotermes, by the use of different mitochondrial (16S, COI/tRNA/COII, CR) and nuclear (microsatellites and Inter-SINE) molecular markers. In the phylogenetic analyses, the obtained results have well defined the cladogenetic events that generated the nowadays species biodiversity of the genus Reticulitermes, while the analysis of the Kalotermes flavicollis taxon showed the presence of at least four genetic clades, defined on the basis of the geographical distance. The second part of the thesis is centred on the population dynamics of two species: Reticulitermes urbis and Kalotermes flavicollis. The first species, native of the Balkans, is known to be present in some cities of Italy and France. I’ve analyzed the colony genetic structure of the introduced population of Bagnacavallo (RA, Italy), using nine microsatellite loci. The obtained results are in accordance with those obtained from another population in France: this species in fact confirms its invasive and infestation capacities. The analysis of the natural population of K. flavicollis, performed with a combination of mitochondrial (control region) and nuclear (microsatellites and I-SINE) markers, clearly evidenced the presence of two genetic lineages that coexist in the same area. Moreover, results clearly indicate that the cross-breeding is allowed. Finally, the whole results are discussed in a comparative view to better understand the differences in ecology, evolutionary dynamics and colony social structure between these two genera.

Eukaryotic ribosomal DNA constitutes a multi gene family organized in a cluster called nucleolar organizer region (NOR); this region is composed usually by hundreds to thousands of tandemly repeated units. Ribosomal genes, being repeated sequences, evolve following the typical pattern of concerted evolution. The autonomous retroelement R2 inserts in the ribosomal gene 28S, leading to defective 28S rDNA genes. R2 element, being a retrotransposon, performs its activity in the genome multiplying its copy number through a “copy and paste” mechanism called target primed reverse transcription. It consists in the retrotranscription of the element’s mRNA into DNA, then the DNA is integrated in the target site. Since the retrotranscription can be interrupted, but the integration will be carried out anyway, truncated copies of the element will also be present in the genome. The study of these truncated variants is a tool to examine the activity of the element. R2 phylogeny appears, in general, not consistent with that of its hosts, except some cases (e.g. Drosophila spp. and Reticulitermes spp.); moreover R2 is absent in some species (Fugu rubripes, human, mouse, etc.), while other species have more R2 lineages in their genome (the turtle Mauremys reevesii, the Japanese beetle Popilia japonica, etc). R2 elements here presented are isolated in 4 species of notostracan branchiopods and in two species of stick insects, whose reproductive strategies range from strict gonochorism to unisexuality. From sequencing data emerges that in Triops cancriformis (Spanish gonochoric population), in Lepidurus arcticus (two putatively unisexual populations from Iceland) and in Bacillus rossius (gonochoric population from Capalbio) the R2 elements are complete and encode functional proteins, reflecting the general features of this family of transposable elements. On the other hand, R2 from Italian and Austrian populations of T. cancriformis (respectively unisexual and hermaphroditic), Lepidurus lubbocki (two elements within the same Italian population, gonochoric but with unfunctional males) and Bacillus grandii grandii (gonochoric population from Ponte Manghisi) have sequences that encode incomplete or non-functional proteins in which it is possible to recognize only part of the characteristic domains. In Lepidurus couesii (Italian gonochoric populations) different elements were found as in L. lubbocki, and the sequencing is still in progress. Two hypothesis are given to explain the inconsistency of R2/host phylogeny: vertical inheritance of the element followed by extinction/diversification or horizontal transmission. My data support previous study that state the vertical transmission as the most likely explanation; nevertheless horizontal transfer events can’t be excluded. I also studied the element’s activity in Spanish populations of T. cancriformis, in L. lubbocki, in L. arcticus and in gonochoric and parthenogenetic populations of B. rossius. In gonochoric populations of T. cancriformis and B. rossius I found that each individual has its own private set of truncated variants. The situation is the opposite for the remaining hermaphroditic/parthenogenetic species and populations, all individuals sharing – in the so far analyzed samples - the majority of variants. This situation is very interesting, because it isn’t concordant with the Muller’s ratchet theory that hypothesizes the parthenogenetic populations being either devoided of transposable elements or TEs overloaded. My data suggest a possible epigenetic mechanism that can block the retrotransposon activity, and in this way deleterious mutations don’t accumulate.

In this study we have analysed the genetic variability in ca. 700 samples belonging to six species of genus Lepus using maternal and biparental molecular markers (mitochondrial DNA, microsatellites, Single Nucleotide Polimorphisms). We aimed to reconstruct the phylogenetic relationships of species of hares living in Europe, and assess the occurrence of hybridization between the European hare Lepus europaeus and the Italian hare Lepus corsicanus. Results showed a deep genetic differentiation and absence of hybridization between L. corsicanus and L. europaeus, confirming that they are distinct and distantly related biological species. In contrast, we showed small genetic distances and a close phylogenetic relationship between the Italian hare and Cantabrian hare L. castroviejoi, which suggest a deeper evaluation of their taxonomic status. Populations of L. corsicanus are geographically differentiated. In particular, the peninsular and Sicilian populations of Italian hares are sharply genetically distinct, which calls for avoiding any translocation between Italy and Sicily. Information on genetic variability and population structure is being used to implement the Italian Action Plan for L. corsicanus.

Il rapido progresso della tecnologia, lo sviluppo di prodotti altamente sofisticati, la forte competizione globale e l’aumento delle aspettative dei clienti hanno messo nuove pressioni sui produttori per garantire la commercializzazione di beni caratterizzati da una qualità sempre crescente. Sono gli stessi clienti che da anni si aspettano di trovare sul mercato prodotti contraddistinti da un livello estremo di affidabilità e sicurezza. Tutti siamo consapevoli della necessità per un prodotto di essere quanto più sicuro ed affidabile possibile; ma, nonostante siano passati oramai 30 anni di studi e ricerche, quando cerchiamo di quantificare ingegneristicamente queste caratteristiche riconducibili genericamente al termine qualità, oppure quando vogliamo provare a calcolare i benefici concreti che l’attenzione a questi fattori quali affidabilità e sicurezza producono su un business, allora le discordanze restano forti. E le discordanze restano evidenti anche quando si tratta di definire quali siano gli “strumenti più idonei” da utilizzare per migliorare l’affidabilità e la sicurezza di un prodotto o processo. Sebbene lo stato dell’arte internazionale proponga un numero significativo di metodologie per il miglioramento della qualità, tutte in continuo perfezionamento, tuttavia molti di questi strumenti della “Total Quality” non sono concretamente applicabili nella maggior parte delle realtà industriale da noi incontrate. La non applicabilità di queste tecniche non riguarda solo la dimensione più limitata delle aziende italiane rispetto a quelle americane e giapponesi dove sono nati e stati sviluppati questi strumenti, oppure alla poca possibilità di effettuare investimenti massicci in R&D, ma è collegata anche alla difficoltà che una azienda italiana avrebbe di sfruttare opportunamente i risultati sui propri territori e propri mercati. Questo lavoro si propone di sviluppare una metodologia semplice e organica per stimare i livelli di affidabilità e di sicurezza raggiunti dai sistemi produttivi e dai prodotti industriali. Si pone inoltre di andare al di là del semplice sviluppo di una metodologia teorica, per quanto rigorosa e completa, ma di applicare in forma integrata alcuni dei suoi strumenti a casi concreti di elevata valenza industriale. Questa metodologia come anche, più in generale, tutti gli strumenti di miglioramento di affidabilità qui presentati, interessano potenzialmente una vasta gamma di campi produttivi, ma si prestano con particolare efficacia in quei settori dove coesistono elevate produzioni e fortissime esigenze qualitative dei prodotti. Di conseguenza, per la validazione ed applicazione ci si è rivolti al settore dell’automotive, che da sempre risulta particolarmente sensibile ai problemi di miglioramento di affidabilità e sicurezza. Questa scelta ha portato a conclusioni la cui validità va al di là di valori puramente tecnici, per toccare aspetti non secondari di “spendibilità” sul mercato dei risultati ed ha investito aziende di primissimo piano sul panorama industriale italiano.

The thesis describes the molluscan biodiversity of the infralittoral off-shore reefs in the "Secche di Tor Paterno" marine protected area lying in the Central Tyrrhenian Sea off the coasts of Lazio south of Roma. Data originate from underwater sampling activities carried out by SCUBA diving in four biocoenoses: Posidonia oceanica leaves and rhizomes, coralligenous concretions and detritic pools. The representativeness of molluscs as descriptors of biocoenoses is evaluated by preliminary comparisons with data about Polychaeta, Pleocyemata (Crustacea) and Brachiopoda obtained in the same survey. The malacocoenoses of the four biocoenoses are treated in detail. Then data are compared with other data sets to assess differences and similarities with other communities. The agreement between death and living assemblages in the reefs is evaluated for the Posidonia oceanica and the coralligenous biocoenosis and was carried out by a set of standard metrics and some benthic ecology methods. Molluscs perform very well as descriptors of biocoenoses, better than the other phyla. The molluscan assemblages of the reefs are very rich in species despite richness is mainly concentrated in the coralligenous and in the rhizomes of Posidonia oceanica. The leaves of Posidonia oceanica host a rather poor assemblage. Detritic pools host a poor but peculiar species assemblage. The dead-live agreement showed that death assemblages are highly representative of sediments of nearby biocoenoses as a result of low bottom transport. Fidelity metrics suggest a good agreement between the living and death assemblages when species richness and taxonomic composition are considered. The study suggests that fidelity is lower when considering the species dominance. These differences could be associated to the trophism of species and possibly to the species life span.

Il lavoro svolto durante il dottorato di ricerca ha permesso lo sviluppo e la verifica della attendibilità di marcatori molecolari neutrali (loci microsatelliti) specifici per Aristeus antennatus. Tali marcatori sono stati poi utilizzati per studiare la struttura genetica di popolazione della specie del Mediterraneo occidentale e i risultati ottenuti sono stati confrontati con quelli di un progetto di ricerca parallelo su Aristeaomorpha foliacea, analizzando differenze ed analogie fra le due specie. I risultati delle analisi su Aristeus antennatus hanno evidenziato una completa assenza di struttura di popolazione e come i due sessi contribuiscano in modo diverso al flusso genico. La specie infatti presenta un sex-ratio a favore dei maschi oltre gli 800m, mentre tale rappoorto è a favore delle femmine in strati più superficiali, dove sono probabilmente soggette a condizioni oceanografiche più dispersive. Tramite test genetici appropriati è stato possibile valutare indirettamente il grado di dospersione dei sessi dimostrando che nell'area analizzati i maschi erano rappresentati maggiormente da individui stanziali, mentre gli individui di sesso femminile erano migranti. Le femmine appaiono pertanto giocare un ruolo preminente rispetto ai maschi nel determinare l'entità del flusso genico. Il confronto dei risultati ottenuti in Aristeus antennatus con quelli di Aristaeomorpha foliacea ha evidenziato la relazione fra alta capacità dispersiva, sia allo stato larvale che adulto, e completo rimescolamento genetico nei gamberi aristeidi nel Mediterraneo occidentale anche se in quest'ultima specie non ci sono evidenze di dispersione genetica mediata dal sesso. E' pertanto di forte interesse (dato anche il valore economico di questi organismi) come una struttura di popolazione qualitativamente e quantitavamente comporabile venga raggiunta con dinamiche di popolazione molto diverse.

This research focuses on reproductive biology and pollination ecology of entomophilous angiosperms, with particular concern to reproductive success in small and isolated populations of species that occur at their distribution limits or are endemic. I considered three perennial herbs as model species: Primula apennina Widmer, Dictamnus albus L. and Convolvulus lineatus L. I carried out field work on natural populations and performed laboratory analyses on specific critical aspects (resource allocation, pollen viability, stigmatic receptivity, physiological self-incompatibility, seed viability), through which I analysed different aspects related to plant fitness, such as production of viable seed, demographic structure of populations, type and efficiency of plant-pollinator system, and limiting factors.

The Neolithic is characterized by the transition from a subsistence economy, based on hunting and gathering, to one based on food producing. This important change was paralleled by one of the most significant demographic increase in the recent history of European populations. The earliest Neolithic sites in Europe are located in Greece. However, the debate regarding the colonization route followed by the Middle-eastern farmers is still open. Based on archaeological, archaeobotanical, craniometric and genetic data, two main hypotheses have been proposed. The first implies the maritime colonization of North-eastern Peloponnesus from Crete, whereas the second points to an island hopping route that finally brought migrants to Central Greece. To test these hypotheses using a genetic approach, 206 samples were collected from the two Greek regions proposed as the arrival point of the two routes (Korinthian district and Euboea). Expectations for each hypothesis were compared with empirical observations based on the analysis of 60 SNPs and 26 microsatellite loci of Y-chromosome and mitochondrial DNA hypervariable region I. The analysis of Y-chromosome haplogroups revealed a strong genetic affinity of Euboea with Anatolian and Middle-eastern populations. The inferences of the time since population expansion suggests an earlier usage of agriculture in Euboea. Moreover, the haplogroup J2a-M410, supposed to be associated with the Neolithic transition, was observed at higher frequency and variance in Euboea showing, for both these parameters, a decreasing gradient moving from this area. The time since expansion estimates for J2a-M410 was found to be compatible with the Neolithic and slightly older in Euboea. The analysis of mtDNA resulted less informative. However, a higher genetic affinity of Euboea with Anatolian and Middle-eastern populations was confirmed. These results taken as a whole suggests that the most probable route followed by Neolithic farmers during the colonization of Greece was the island hopping route.

Il lavoro eseguito comprende 3 differenti studi inerenti l’approfondimento di diversi aspetti della fisiopatologia della prostata, in particolare l’aspetto clinico, biochimico ed ormonale. Per quanto riguarda l’aspetto clinico è stato eseguito uno studio ecografico sull’accrescimento e sviluppo della ghiandola, a partire dai 4 giorni di vita all’anno di età del cane. L’aspetto biochimico dell’organo è stato indagato nel secondo studio, analizzando l’eventuali differenze nel profilo proteico e nel profilo dello zinco, valutato con spettrofotometria ad assorbimento atomico, su campioni di plasma seminale di cani sani o cani affetti da iperplasia prostatica benigna sintomatica od asintomatica. Lo studio è stato eseguito allo scopo di valutare se l’approccio biochimico ai problemi prostatici possa essere d’aiuto nella diagnosi della più comune patologia della prostata, l’iperplasia prostatica benigna. Il terzo studio inerente l’aspetto ormonale della prostata, completa lo studio precedente, analizzando le eventuali differenze ormonali (Testosterone, Estrogeni e Progesterone) riscontrabili fra due gruppi di cani: i cani sani e quelli affetti da iperplasia prostatica benigna.

The main scope of my PhD is the reconstruction of the large-scale bivalve phylogeny on the basis of four mitochondrial genes, with samples taken from all major groups of the class. To my knowledge, it is the first attempt of such a breadth in Bivalvia. I decided to focus on both ribosomal and protein coding DNA sequences (two ribosomal encoding genes -12s and 16s -, and two protein coding ones - cytochrome c oxidase I and cytochrome b), since either bibliography and my preliminary results confirmed the importance of combined gene signals in improving evolutionary pathways of the group. Moreover, I wanted to propose a methodological pipeline that proved to be useful to obtain robust results in bivalves phylogeny. Actually, best-performing taxon sampling and alignment strategies were tested, and several data partitioning and molecular evolution models were analyzed, thus demonstrating the importance of molding and implementing non-trivial evolutionary models. In the line of a more rigorous approach to data analysis, I also proposed a new method to assess taxon sampling, by developing Clarke and Warwick statistics: taxon sampling is a major concern in phylogenetic studies, and incomplete, biased, or improper taxon assemblies can lead to misleading results in reconstructing evolutionary trees. Theoretical methods are already available to optimize taxon choice in phylogenetic analyses, but most involve some knowledge about genetic relationships of the group of interest, or even a well-established phylogeny itself; these data are not always available in general phylogenetic applications. The method I proposed measures the "phylogenetic representativeness" of a given sample or set of samples and it is based entirely on the pre-existing available taxonomy of the ingroup, which is commonly known to investigators. Moreover, it also accounts for instability and discordance in taxonomies. A Python-based script suite, called PhyRe, has been developed to implement all analyses.

A partire dagli anni '70, si è assistito ad un progressivo riassetto geopolitico a livello mondiale Grazie anche all’evoluzione tecnologica ed alla sua diffusione di massa, il tempo e lo spazio si contraggono nel processo di globalizzazione che ha caratterizzato le società contemporanee ove l'informazione e la comunicazione assumono ormai un ruolo centrale nelle dinamiche di conoscenza. Il presente studio, intende far luce in primis sulla disciplina dell'intelligence, così come enunciata in ambito militare e "civile", in particolare nel contesto USA, NATO ed ONU, al fine quindi di evidenziare le peculiarità di una nuova disciplina di intelligence, cosiddetta Open Source Intelligence, che ha come elemento di innovazione l'utilizio delle informazioni non classificate. Dopo aver affrontato il problema della concettualizzazione ed evoluzione del fenomeno terroristico, sarà posto il focus sull’espressione criminale ad oggi maggiormente preoccupante, il terrorismo internazionale di matrice islamica, in prospettiva multidimensionale, grazie all’adozione di concetti criminologici interdisciplinari. Sotto il profilo della sperimentazione, si è, quindi, deciso di proporre, progettare e sviluppare l’architettura della piattaforma Open Source Intellicence Analysis Platform,un tool operativo di supporto per l’analista Open Source Intelligence, che si pone quale risorsa del’analisi criminologica, in grado di fornire un valido contributo, grazie al merge tra practice e research, nell’applicazione di tale approccio informativo al fenomeno terroristico.

The present issue analyses bluefin tuna (Thynnus thunnus) and swordfish (Xiphias glaudis) diet, caught by professional long-line fishing in the middle Adriatic Sea (Pomo pit). These species represent apex predators in pelagic environment that may play key roles in determining food web structure and ecosystem dynamics. The studies about their feedings habits, based upon stomach contents analysis, are important for the comprehension of biological and ecological interaction. Over the years, many studies have been performed on the diet of tuna and swordfish in the Mediterranean Sea. This research is based on a fairly wide number of analyzed stomach contents, in comparison with the previous ones. In this work, the analysis of 340 stomach contents of bluefin tuna caught by long-line in the central Adriatic sea confirms in general the opportunistic behaviour of this species. Finding support the hypothesis that Adriatic tuna and swordfish chase their food over a wide bathymetric zone and probably near the surface at night. No indication of food preference respect to size of predator or sample season are found. It seems that the two species are able to cohabit because their trophic niche are not overlapped, changing during the time and the vertical and horizontal space.

The Ecosystem Approach to Fisheries represents the most recent research line in the international context, showing interest both towards the whole community and toward the identification and protection of all the “critical habitats” in which marine resources complete their life cycles. Using data coming from trawl surveys performed in the Northern and Central Adriatic from 1996 to 2010, this study provides the first attempt to appraise the status of the whole demersal community. It took into account not only fishery target species but also by-catch and discharge species by the use of a suite of biological indicators both at population and multi-specific level, allowing to have a global picture of the status of the demersal system. This study underlined the decline of extremely important species for the Adriatic fishery in recent years; adverse impact on catches is expected for these species in the coming years, since also minimum values of recruits recently were recorded. Both the excessive exploitation and environmental factors affected availability of resources. Moreover both distribution and nursery areas of the most important resources were pinpointed by means of geostatistical methods. The geospatial analysis also confirmed the presence of relevant recruitment areas in the North and Central Adriatic for several commercial species, as reported in the literature. The morphological and oceanographic features, the relevant rivers inflow together with the mosaic pattern of biocenoses with different food availability affected the location of the observed relevant nursery areas.

Two Amerindian populations from the Peruvian Amazon (Yanesha) and from rural lowlands of the Argentinean Gran Chaco (Wichi) were analyzed. They represent two case study of the South American genetic variability. The Yanesha represent a model of population isolated for long-time in the Amazon rainforest, characterized by environmental and altitudinal stratifications. The Wichi represent a model of population living in an area recently colonized by European populations (the Criollos are the population of the admixed descendents), whose aim is to depict the native ancestral gene pool and the degree of admixture, in relation to the very high prevalence of Chagas disease. The methods used for the genotyping are common, concerning the Y chromosome markers (male lineage) and the mitochondrial markers (maternal lineage). The determination of the phylogeographic diagnostic polymorphisms was carried out by the classical techniques of PCR, restriction enzymes, sequencing and specific mini-sequencing. New method for the detection of the protozoa Trypanosoma cruzi was developed by means of the nested PCR. The main results show patterns of genetic stratification in Yanesha forest communities, referable to different migrations at different times, estimated by Bayesian analyses. In particular Yanesha were considered as a population of transition between the Amazon basin and the Andean Cordillera, evaluating the potential migration routes and the separation of clusters of community in relation to different genetic bio-ancestry. As the Wichi, the gene pool analyzed appears clearly differentiated by the admixed sympatric Criollos, due to strict social practices (deeply analyzed with the support of cultural anthropological tools) that have preserved the native identity at a diachronic level. A pattern of distribution of the seropositivity in relation to the different phylogenetic lineages (the adaptation in evolutionary terms) does not appear, neither Amerindian nor European, but in relation to environmental and living conditions of the two distinct subpopulations.

La ricerca consta di quattro capitoli: nella prima parte si analizzano, in prospettiva empirica, i canoni argomentativi utilizzati dalla Corte costituzionale italiana. Tali canoni possono essere generalmente distinti in due grandi famiglie: giuridico-interpretativi per un verso, e ed empirico fattuali per un altro. In un secondo capitolo si è visto come la Corte costituzionale italiana ha frutto uso di questi canoni nella sua giurisprudenza italiana in tema di diritto soggettivo di libertà di espressione e di mezzi di comunicazione del pensiero. Nel primo caso la Corte costituzionale ha fatto ricorso a canoni interpretativi testuali, mentre nel secondo ad argomentazioni empiriche spesso accompagnate da un giudizio di proporzionalità sull'intervento legislativo. Nella terza parte del lavoro si è invece descritta la giurisprudenza della Corte Suprema americana: è stata analizzata l'evoluzione delle tecniche argomentativi in tema di libertà di espressione: partendo dall'ad hoc balancing e dal definitional balancing si è giunti alla descrizione dei diversi livelli di scrutinio e alla progressiva concettualizzazione delle categorie espressive. Infine nel quarto capitolo si sono messe a confronto le due giurisprudenze: si è potuto così notare che la progressiva categorizzazione del diritto di libertà negli Stati Uniti ha dato esiti maggiormente garantisti, mentre più liberisti in relazione alle concrete dinamiche di formazione del discorso pubblico. Viceversa, in Italia i canoni testuali hanno originato una giurisprudenza più restrittiva quanto al diritto soggettivo e più egualitaria quanto al sistema della libertà di espressione. In ogni caso, entrambe le esperienze evidenziano il ruolo di policy-maker delle Corti costituzionali nei moderni ordinamenti liberal-democratici.
