The biomechanical roles of both tendons and ligaments are fulfilled by extracellular matrix of these tissues. In particular, tension is mainly transmitted and resisted by fibrous proteins (collagen, elastin), whereas compressive load is absorbed by water-soluble glycosaminoglycans (GAGs). GAGs spanning the interfibrillar spaces and interacting with fibrils also seem to play a part in transmitting and resisting tensile stresses. Apart from different functional roles and collagen array, tendons and ligaments share the same basic structure showing periodic undulations of collagen fibers or crimps. Each crimp is composed of many knots of each single fibril or fibrillar crimps. Fibrillar and fiber crimps act as shock absorbers during the initial elongation of both tendons and ligaments and assist the elastic recoil of fibrils and fibers when the tensile stress is removed. The aim of this thesis was to evaluate whether GAGs directly affect the 3D microstructural integrity of fibrillar crimp and fiber crimps in both tendons and ligaments. Achilles tendons and medial collateral ligaments of the knee from eight female Sprague-Dawley rats (90 days old) were digested with chondroitinase ABC to remove GAGs and observed under a scanning electron microscope (SEM). In addition, isolated fibrils from these tissues obtained by mechanical homogenization were analyzed by a transmission electron microscope (TEM). Both samples digested with chondroitinase ABC or mechanically disrupted still showed crimps and fibrillar crimps comparable to tissues with a normal GAGs content. All fibrils in the fibrillar crimp region always twisted leftwards, thus changing their running plane, and then sharply bent, changing their course on a new plane. These data suggest that GAGs do not affect structural integrity or fibrillar crimps functions that seem mainly related to the local fibril leftward twisting and the alternating handedness of collagen from a molecular to a supramolecular level.

Il presente lavoro affronta la questione del rapporto tra l’azione cartolare e l’azione causale nell’ambito dei titoli cambiari; nonché il problema della relazione, di diritto sostanziale, che intercorre tra i menzionati titoli ed i rapporti causali che soggiacciono alla loro emissione e circolazione; con costante riferimento sia al diritto italiano che al diritto paraguaiano. Vengono così analizzati i concetti tradizionali della causa della cambiale e della convenzione esecutiva, per poi passare alla analisi della funzione economica tipica dei titoli cambiari e, segnatamente, la previsione del loro rilascio o negoziazione nell’economia del rapporto fondamentale. Si affrontano, così, i noti problemi della novazione e della dazione in pagamento attuate mediante questi titoli, tenendo in particolare considerazione anche la struttura delegatoria che presentano alcuni di essi. Particolare attenzione viene dedicata al fatto che, nella maggior parte dei casi, le parti non prevedono specificamente l’incidenza del rilascio o girata del titolo cambiario sul rapporto sottostante. Per questa ragione, si mette in luce che nella maggior parte dei casi la fattispecie configura una modificazione della relazione causale intercorrente tra le parti, proprio per l’assenza di pattuizione espressa al riguardo. Strettamente connesso con quest’aspetto risulta l’esame dell’emissione dei titoli cambiari solvendi causa, problematica nell’ambito della quale non si è mancato di tener conto della disciplina antiriciclaggio e dei moderni contributi dottrinari sull’oggetto dell’obbligazione pecuniaria. Viene, infine, vagliata anche l’incidenza dell’ulteriore circolazione del titolo sul rapporto causale che giustifica il suo possesso da parte del girante; per poi passare, come conclusione del lavoro, a stabilire l’interpretazione non solo processuale ma anche sostanziale che deve darsi, alla luce dei risultati raggiunti, alla disciplina degli artt. 66 della legge cambiaria e 58 della legge assegni.

La Muay Thai, comunemente detta “Boxe Thailandese” è un'arte marziale che rientra nella classificazione delle attività intermittenti con entrambi i sistemi energetici reclutati, aerobico e anaerobico, è inoltre caratterizzata dal fatto che il combattimento alla distanza si alterna alla lotta, denominata “clinch”. Nonostante la popolarità della Muay Thai, in ambito mondiale, stia progressivamente aumentando così come è in aumento il numero di atleti che la praticano, le ricerche incentrate su questa arte marziale e gli studi relativi agli aggiustamenti cardiometabolici nonché alle modalità temporali con cui gli specifici gesti atletici si possono succedere nel tempo durante un match, sono ancora estremamente esigui. L’oggetto del nostro studio è stato l’analisi della struttura temporale del combattimento, tramite la Match Analysis off line (analisi visiva del combattimento), con comparazione dei dati ottenuti tra il vincitore (winner) e il perdente (loser) e la valutazione dell’andamento di alcuni importanti parametri metabolici attraverso la misurazione del lattato e della HR, durante un incontro reale di Boxe Thailandese. La sperimentazione è stata condotta su un gruppo di dieci soggetti di sesso maschile, praticanti la disciplina ad un alto livello nazionale, la cui media ± deviazione standard (DS), di età, peso e altezza è di 24,6 ±4,01 anni, 69,4 ±7 kg e 174,1 ±4,3 cm. Gli atleti sono stati sottoposti, in due diverse giornate separate da almeno tre giorni, a due test; durante una prima seduta sperimentale preliminare abbiamo determinato il massimo consumo di ossigeno (VO2max) nel corso di un test sul nastro trasportare, con concomitante stima della Soglia anaerobica (SA) e misura della massima frequenza cardiaca (HR max). In una seconda seduta sperimentale abbiamo effettuato i test di combattimento in palestra e infine abbiamo analizzato i video degli incontri attraverso la Match Analysis. Dai risultati della Match - Analysis è scaturito che i vincitori hanno eseguito un numero più elevato di azioni efficaci (p < 0,05) rispetto ai non-vincitori, grazie ad un numero maggiore di combinazioni (C ) e di attacchi singoli (A) e un numero minore di difese (D) e di tecniche inefficaci. È così emerso come il livello delle realizzazioni sia quasi esclusivamente dovuto all’efficacia della tecnica e alla tattica delle azioni. Abbiamo quindi focalizzato la nostra attenzione sul clinch e sulle azioni di attacco perché si ipotizzava che potessero essere attività dispendiose e probabilmente responsabili dell’incremento di lattato durante il combattimento, dall’ analisi dei dati però non è stata riscontrata nessuna significativa correlazione tra l’andamento dei dati metabolici e le fasi di attacco e di lotta. Dai nostri risultati emerge in maniera interessante come durante le fasi attive del combattimento si siano raggiunti alti valori di lattato ematico e di frequenza cardiaca, rispettivamente di 12,55 mmol/L e di 182,68 b/min, ben oltre la SA rilevata nel test incrementale dove la HR si posizionava a 168,2 b/min. In conclusione si evidenzia come la Boxe Thailandese sia una disciplina caratterizzata da un considerevole impegno energetico-metabolico, sia aerobico che anaerobico. La predominanza del metabolismo lattacido è dimostrata dagli elevati valori di lattato osservati nel presente studio e dalla frequenza degli attacchi (8,6 ± 3,5 sec.). Questo studio potrà essere utilizzato dagli allenatori per la predisposizione di allenamenti specifici che inducano gli adattamenti propri della Muay Thai.

La Piastinopenia Immune (PTI) è una patologia autoimmune ad eziologia ignota caratterizzata da piastrinopenia. I linfociti T regolatori (Tregs) sono coinvolti nei meccanismi di tolleranza immunologica e agiscono regolando l’attività delle cellule T autoreattive e delle cellule dendritiche (DCs). Viceversa, le DCs, che esprimono Indoleamine 2,3-dioxygenase (IDO), partecipano al mantenimento della tolleranza agli auto-antigeni attraverso l’espansione dei Tregs. Inoltre, recenti studi hanno dimostrato che i linfociti T helper 17 (Th17) sono coinvolti nell’autoimmunità e che l’espressione di Interleuchina (IL)-17 è associata a numerose patologie autoimmuni. Il ruolo dell’interazione fra DCs e Tregs ed il ruolo dei Th17 nella patogenesi della PTI non sono mai stati studiati in maniera approfondita. Gli obiettivi principali di questa tesi sono stati: a) caratterizzare fenotipicamente e funzionalmente i linfociti Tregs e DCs; b) valutare il ruolo patogenetico dell’interazione tra Tregs e DCs; c) quantificare i Th17 circolanti. I risultati dimostrano che: 1) il numero dei Tregs circolanti dei pazienti, identificati tramite i marcatori Foxp3 e CD127, è significativamente ridotto rispetto alla controparte normale; 2) la conversione in vitro delle cellule CD4+CD25- in linfociti Tregs (CD4+CD25+Foxp3+) dopo stimolazione con DCs mature è significativamente ridotta nei pazienti rispetto ai controlli; 3) sia l’espressione dell’enzima IDO nelle DCs mature (mRNA) che i livelli di chinurenine prodotte (indice di attività enzimatica) sono risultati significativamente ridotti nei pazienti rispetto alla controparte normale. Questi risultati suggeriscono quindi che il ridotto numero dei Tregs circolanti nei pazienti con PTI può essere, almeno in parte, attribuito alla scarsa capacità di conversione delle DCs, in quanto tali cellule esprimono meno IDO. I risultati dimostrano inoltre che: 1) i Tregs dei pazienti con PTI hanno una capacità soppressoria che è significativamente inferiore rispetto ai soggetti normali; tale dato è stato confermato dal dosaggio di IFN- nel surnatante della coltura; 2) i Tregs di pazienti con PTI non sono in grado di inibire la maturazione delle DCs, a differenza di quanto avviene nei soggetti sani: infatti, l’espressione delle molecole costimolatorie CD80 e CD86 sulle DCs è risultata invariata in seguito a cocoltura con DCs immature; 3) il dosaggio delle citochine nel surnatante delle cocolture dimostra che la concentrazione di IL-10 e IL-6 è significativamente ridotta nei pazienti rispetto ai controlli. La scarsa abilità dei Tregs di inibire la maturazione delle DCs e l’alterato pattern di secrezione di citochine potrebbero quindi contribuire all’ insorgenza del fenotipo più maturo delle DCs nei pazienti con PTI. I linfociti Th17 circolanti di pazienti e controlli sono stati identificati in citofluorimetria come cellule CD4+CD161+CD196+. Da tale analisi è emerso che la frequenza dei Th17 circolanti non è significativamente diversa nei due gruppi. Questi dati dimostrano quindi che nella PTI l’interazione bidirezionale tra Tregs e DCs risulta alterata e svolge un ruolo patogenetico, in quanto, da un lato, ci sono Tregs con un deficit numerico e funzionale e, dall’altro, DCs con maggiore capacità immunogenica. Il numero dei Th17 non risulta invece alterato.

The "sustainability" concept relates to the prolonging of human economic systems with as little detrimental impact on ecological systems as possible. Construction that exhibits good environmental stewardship and practices that conserve resources in a manner that allow growth and development to be sustained for the long-term without degrading the environment are indispensable in a developed society. Past, current and future advancements in asphalt as an environmentally sustainable paving material are especially important because the quantities of asphalt used annually in Europe as well as in the U.S. are large. The asphalt industry is still developing technological improvements that will reduce the environmental impact without affecting the final mechanical performance. Warm mix asphalt (WMA) is a type of asphalt mix requiring lower production temperatures compared to hot mix asphalt (HMA), while aiming to maintain the desired post construction properties of traditional HMA. Lowering the production temperature reduce the fuel usage and the production of emissions therefore and that improve conditions for workers and supports the sustainable development. Even the crumb-rubber modifier (CRM), with shredded automobile tires and used in the United States since the mid 1980s, has proven to be an environmentally friendly alternative to conventional asphalt pavement. Furthermore, the use of waste tires is not only relevant in an environmental aspect but also for the engineering properties of asphalt [Pennisi E., 1992]. This research project is aimed to demonstrate the dual value of these Asphalt Mixes in regards to the environmental and mechanical performance and to suggest a low environmental impact design procedure. In fact, the use of eco-friendly materials is the first phase towards an eco-compatible design but it cannot be the only step. The eco-compatible approach should be extended also to the design method and material characterization because only with these phases is it possible to exploit the maximum potential properties of the used materials. Appropriate asphalt concrete characterization is essential and vital for realistic performance prediction of asphalt concrete pavements. Volumetric (Mix design) and mechanical (Permanent deformation and Fatigue performance) properties are important factors to consider. Moreover, an advanced and efficient design method is necessary in order to correctly use the material. A design method such as a Mechanistic-Empirical approach, consisting of a structural model capable of predicting the state of stresses and strains within the pavement structure under the different traffic and environmental conditions, was the application of choice. In particular this study focus on the CalME and its Incremental-Recursive (I-R) procedure, based on damage models for fatigue and permanent shear strain related to the surface cracking and to the rutting respectively. It works in increments of time and, using the output from one increment, recursively, as input to the next increment, predicts the pavement conditions in terms of layer moduli, fatigue cracking, rutting and roughness. This software procedure was adopted in order to verify the mechanical properties of the study mixes and the reciprocal relationship between surface layer and pavement structure in terms of fatigue and permanent deformation with defined traffic and environmental conditions. The asphalt mixes studied were used in a pavement structure as surface layer of 60 mm thickness. The performance of the pavement was compared to the performance of the same pavement structure where different kinds of asphalt concrete were used as surface layer. In comparison to a conventional asphalt concrete, three eco-friendly materials, two warm mix asphalt and a rubberized asphalt concrete, were analyzed. The First Two Chapters summarize the necessary steps aimed to satisfy the sustainable pavement design procedure. In Chapter I the problem of asphalt pavement eco-compatible design was introduced. The low environmental impact materials such as the Warm Mix Asphalt and the Rubberized Asphalt Concrete were described in detail. In addition the value of a rational asphalt pavement design method was discussed. Chapter II underlines the importance of a deep laboratory characterization based on appropriate materials selection and performance evaluation. In Chapter III, CalME is introduced trough a specific explanation of the different equipped design approaches and specifically explaining the I-R procedure. In Chapter IV, the experimental program is presented with a explanation of test laboratory devices adopted. The Fatigue and Rutting performances of the study mixes are shown respectively in Chapter V and VI. Through these laboratory test data the CalME I-R models parameters for Master Curve, fatigue damage and permanent shear strain were evaluated. Lastly, in Chapter VII, the results of the asphalt pavement structures simulations with different surface layers were reported. For each pavement structure, the total surface cracking, the total rutting, the fatigue damage and the rutting depth in each bound layer were analyzed.

Iodine is an essential microelement for human health because it is a constituent of the thyroid hormones that regulate growth and development of the organism. Iodine Deficiency Disorders (IDDs) are believed to be one of the commonest preventable human health problems in the world today, according to the World Health Organization: that diseases include endemic goiter, cretinism and fetal abnormalities, among others, and they are caused by lack of iodine in the diet, that is the main source of iodine. Since iodine intake from food is not enough respect to human needs, this can be remedied through dietary diversification, mineral supplementation, food fortification, or increasing the concentration and/or bioavailability of mineral elements in the edible portions of crops through agricultural intervention or genetic selection (biofortification). The introduction of iodized salt is a strategy widely used and accepted to eradicate iodine deficiency, because it is an inexpensive source of stable iodine. Since the intake of salt, though iodized, must still be limited according to the risk of cardiovascular disease, so the increase of iodine content in plants for the production of functional foods is representing a field of study of particular interest and a potential market. In Italy potatoes enriched with iodine are produced by a patented procedure of agronomic biofortification for the fresh market since several years, furthermore they are recently accepted and recommended by Italian Thyroid Association, as an alternative source of iodine. Researches performed during the PhD course intended to characterize this innovative vegetables products, focusing the attention on different aspects, such as chemistry, agriculture, and quality of fresh and fried potatoes. For this purpose, lipid fraction of raw material was firstly investigated, in order to assess whether the presence of iodine in plant metabolism can affect fatty acid or sterol biosynthesis, according to the hypothesis that iodine can be bounded to polyunsaturated fatty acids of cell membranes, protecting them from peroxydation; phytosterols of plant sterol are also studied because their importance in reducing serum cholesterol, especially in potato plant sterols are also involved in synthesis of glycoalkaloid, a family of steroidal toxic secondary metabolites present in plants of the Solanaceae family. To achieve this goal chromatographic analytical techniques were employed to identify and quantify fatty acids and sterols profile of common and iodine enriched row potatoes. Another aim of the project was to evaluate the effects of frying on the quality of iodine-enriched and common potatoes. Since iodine-enriched potatoes are nowadays produced only for the fresh market, preliminary trials of cultivation under controlled environment were carried out to verify if potato varieties suitable for processing were able to absorb and accumulate iodine in the tuber. In a successive phase, these varieties were grown in the field, to evaluate their potential productivity and quality at harvest and after storage. The best potato variety to be destined for processing purposes, was finally subjected to repeated frying cycles; the effects of lipid oxidation on the composition and quality of both potatoes and frying oil bath were evaluated by chromatographic and spectrophotometric analytical techniques. Special attention were paid on volatile compounds of fried potatoes.

Foods that provide medical and health benefits or have a role in disease risk prevention are termed functional foods. The functionality of functional foods is derived from bioactive compounds that are extranutritional constituents present in small quantities in food. Bioactive components include a range of chemical compounds with varying structures such as carotenoids, flavonoids, plant sterols, omega-3 fatty acids (n-3), allyl and diallyl sulfides, indoles (benzopyrroles), and phenolic acids. The increasing consumer interest in natural bioactive compounds has brought about a rise in demand for these kinds of compounds and, in parallel, an increasing number of scientific studies have this type of substance as main topic. The principal aim of this PhD research project was the study of different bioactive and toxic compounds in several natural matrices. To achieve this goal, chromatographic, spectroscopic and sensorial analysis were performed. This manuscript reports the main results obtained in the six activities briefly summarized as follows: • SECTION I: the influence of conventional packaging on lipid oxidation of pasta was evaluated in egg spaghetti. • SECTION II: the effect of the storage at different temperatures of virgin olive oil was monitored by peroxide value, fatty acid activity, OSI test and sensory analysis. • SECTION III: the glucosinolate and phenolic content of 37 rocket salad accessions were evaluated, comparing Eruca sativa and Diplotaxis tenuifolia species. Sensory analysis and the influence of the phenolic and glucosinolate composition on sensory attributes of rocket salads has been also studied. • SECTION IV: ten buckwheat honeys were characterised on the basis of their pollen, physicochemical, phenolic and volatile composition. • SECTION V: the polyphenolic fraction, anthocyanins and other polar compounds, the antioxidant capacity and the anty-hyperlipemic action of the aqueous extract of Hibiscus sabdariffa were achieved. • SECTION VI: the optimization of a normal phase high pressure liquid chromatography–fluorescence detection method for the quantitation of flavanols and procyanidins in cocoa powder and chocolate samples was performed.

Lipids are important components that contribute very significantly to nutritional and technological quality of foods because they are the least stable macro-components in foods, due to high susceptibility to oxidation. When rancidity take place, it makes food unhealthy and unacceptable for consumers. Thus, the presence of antioxidants, naturally present of added to foods, is required to enhance shelf life of foods. Moreover, antioxidant like phenolic compounds play an important role in human health enhancing the functionality of foods. The aim of this PhD project was the study of lipid quality and lipid oxidation in different vegetable foods focusing on analytical and technological aspects in order to figure out the effects of lipid composition and bioactive compounds (phenolic compounds, omega-3 fatty acids and dietary fiber) addition on their shelf life. In addition, bioavailability and antioxidant effects of phenolic compounds in human and animals, respectively, were evaluated after consumption of vegetable foods. The first section of the work was focused on the evaluation of lipid quality impact on technological behaviour of vegetable foods. Because of that, cocoa butter with different melting point were evaluated by chromatographic techniques (GC, TLC) and the sample with the higher melting point showed the presence of fatty acids, triglycerides, 2-monoglycerides and FT-IR profile different from genuine cocoa butter, meaning an adding of foreign fat (lauric-fat) not allowed by the law. Looking at lipid quality of other vegetable foods, an accelerated shelf life test (OXITEST®), was used to evaluate of lipid stability to oxidation in tarallini snacks made up using different lipid matrices (sunflower oil, extravirgin olive oil and a blend of extravirgin olive oil and lard). The results showed a good ability of OXITEST® to discriminate between lipid unsaturation and different cooking times, without any samples fat extraction. In the second section, the role of bioactive compounds on cereal based food shelf life was studied in different bakeries by GC, spectrophotometric methods and capillary electrophoresis. It was examined the relationships between phenolic compounds, added with flour, and lipid oxidation of tarallini and frollini. Both products showed an increase in lipid oxidation during storage and antioxidant effects on lipid oxidation were not as expected. Furthermore, the influence of enrichment in polyunsaturated fatty acids on lipid oxidation of pasta was evaluated. The results proved that LC n-3 PUFA were not significantly implicated in the onset of oxidation in spaghetti stored under daylight and accelerated oxidation in a laboratory heater. The importance of phenolic compounds as antioxidant in humans and rats was also studied, by HPLC/MS in the latter section. For this purpose, apigenin and apigenin glycosides excretion was investigated in six women’s urine in a 24 hours study. After a single dose of steamed artichokes, both aglicone and glucuronide metabolites were recovered in 24 h urine. Moreover, the effect of whole grain durum wheat bread and whole grain Kamut® khorasan bread in rats were evaluated. Both cereals were good sources of antioxidants but Kamut® bread fed animals had a better response to stress than wheat durum fed, especially when a sourdough bread was supplied.

Il trapianto delle cellule staminali emopoietiche umane CD34+ in combinazione con le cellule T regolatorie CD4+/CD25+ FoxP3+ (Tregs) potrebbe prevenire l'alloreattività verso le cellule staminali emopoietiche e ridurre il rischio di rigetto in trapianti allogenici HLA non correlati. Per dimostrare questa ipotesi abbiamo messo in coltura le cellule CD34+ e le cellule CD4+/CD25+ isolate con metodica immunomagnetica (Miltnyi Biotec)da sangue periferico non manipolato,sangue periferico mobilizzato con G-CSF o da Cordone ombelicale. Gli esperimenti svolti in vitro, hanno evidenziato che le cellule Tregs arricchite, ottenute dalla stessa fonte delle cellule CD34+( autologhe), mostravano un effetto inibitorio maggiore sulle celulle T alloreattive, rispetto alle cellule Tregs ottenute da un donatore terzo(allogenico).Inoltre l'attività immunosoppressoria delle Tregs era mantenuta dopo stimolazione con cellule CD34+ allogeniche e i Tregs non modificavano l'attività clonogenica delle cellule staminali CD34+. Avendo ottenuto questi dati in vitro abbiamo trapiantato in topi NOD/SCID le cellule Tregs e le cellule CD34+ in rapporto 1:1 o 1:2 ed è stato osservato un normale attecchimento delle cellule staminali. Incubando queste cellule con dosi fisiologiche di timoglobulina derivata da coniglio (nota molecola immunosopressoria) non veniva modificato il numero dei Tregs dopo 6 giorni di coltura. Dopo l'esposizione alla Timoglobulina, inoltre, i Tregs mantenevano la loro attività soppressoria, aumentava l'espressione del recettore chemochinico CCR7, e venivano rilasciate diverse citochine principalmente l'interleuchina 10(IL-10). Tali dati dimostrano come sia le cellule tregs autologhe che quelle allogeniche potrebbero essere trapiantate insieme alle cellule staminali CD34+ dopo un regime preparatorio di terapia che include la timoglobulina. A tale scopo sono state eseguite selezioni di Tregs da aferesi di donatori sani mobilizzati con G-CSF su scala clinica utilizzando biglie immunomagnetiche Clinical grade (Miltenyi Biotec) e sono state confrontate 2 modalità di selezione con due o tre passaggi con o senza la deplezione dei monociti CD14+.Si è dimostrato così che è possibile selezionare un numero uguale di CD34+ e Tregs ,che con la metodica che prevede la deplezione dei monociti si ottiene una popolazione di cellule Tregs più pura ( > 80%) e infine le applicazioni possibili di questi risultati includo: trapianto di cellule CD34+ del donatore insieme a cellule Tregs in trapianti aploidentici ; infusione of cellule tregs isolate da PBSC mobilizzati con G-nei casi di pazienti con GVHD steroide-resistente; infusione di G-Tregs del donatore nei casi di rigetto di organo trapiantato da of donatore ancora vivente.

Attività di ricerca nell'ambito dell'inidirizzo nefrologia clinica, valutando: la popolazione dei pazienti in trattamento sostitutivo in particolare incidenza e prevalenza dei pazienti in trattamento renale sostitutivo nell'Azienda USL di Cesena dal 2000 al 2009 confronto con i dati del registro italiano (analisi retrospettiva). Analisi di questa popolazione e in particolare referral nefrologico e applicazione di misure atte a ridurre la progressione dell'IRC (prevenzione secondaria, terziaria e riduzione dei costi). Confronto con i dati internazionali.

Researches performed during the PhD course intended to assess innovative applications of near-infrared spectroscopy in reflectance (NIR) in the production chain of beer. The purpose is to measure by NIR the "malting quality" (MQ) parameter of barley, to monitor the malting process and to know if a certain type of barley is suitable for the production of beer and spirits. Moreover, NIR will be applied to monitor the brewing process. First of all, it was possible to check the quality of the raw materials like barley, maize and barley malt using a rapid, non-destructive and reliable method, with a low error of prediction. The more interesting result obtained at this level was that the repeatability of the NIR calibration models developed was comparable with the one of the reference method. Moreover, about malt, new kinds of validation were used in order to estimate the real predictive power of the proposed calibration models and to understand the long-term effects. Furthermore, the precision of all the calibration models developed for malt evaluation was estimated and statistically compared with the reference methods, with good results. Then, new calibration models were developed for monitoring the malting process, measuring the moisture content and other malt quality parameters during germination. Moreover it was possible to obtain by NIR an estimate of the "malting quality" (MQ) of barley and to predict whether if its germination will be rapid and uniform and if a certain type of barley is suitable for the production of beer and spirits. Finally, the NIR technique was applied to monitor the brewing process, using correlations between NIR spectra of beer and analytical parameters, and to assess beer quality. These innovative results are potentially very useful for the actors involved in the beer production chain, especially the calibration models suitable for the control of the malting process and for the assessment of the “malting quality” of barley, which need to be deepened in future studies.

Osmotic Dehydration and Vacuum Impregnation are interesting operations in the food industry with applications in minimal fruit processing and/or freezing, allowing to develop new products with specific innovative characteristics. Osmotic dehydration is widely used for the partial removal of water from cellular tissue by immersion in hypertonic (osmotic) solution. The driving force for the diffusion of water from the tissue is provided by the differences in water chemical potential between the external solution and the internal liquid phase of the cells. Vacuum Impregnation of porous products immersed in a liquid phase consist of reduction of pressure in a solid-liquid system (vacuum step) followed by the restoration of atmospheric pressure (atmospheric step). During the vacuum step the internal gas in the product pores is expanded and partially flows out while during the atmospheric step, there is a compression of residual gas and the external liquid flows into the pores (Fito, 1994). This process is also a very useful unit operation in food engineering as it allows to introduce specific solutes in the tissue which can play different functions (antioxidants, pH regulators, preservatives, cryoprotectants etc.). The present study attempts to enhance our understanding and knowledge of fruit as living organism, interacting dynamically with the environment, and to explore metabolic, structural, physico-chemical changes during fruit processing. The use of innovative approaches and/or technologies such as SAFES (Systematic Approach to Food Engineering System), LF-NMR (Low Frequency Nuclear Magnetic Resonance), GASMAS (Gas in Scattering Media Absorption Spectroscopy) are very promising to deeply study these phenomena. SAFES methodology was applied in order to study irreversibility of the structural changes of kiwifruit during short time of osmotic treatment. The results showed that the deformed tissue can recover its initial state 300 min after osmotic dehydration at 25 °C. The LF-NMR resulted very useful in water status and compartmentalization study, permitting to separate observation of three different water population presented in vacuole, cytoplasm plus extracellular space and cell wall. GASMAS techniques was able to study the pressure equilibration after Vacuum Impregnation showing that after restoration of atmospheric pressure in the solid-liquid system, there was a reminding internal low pressure in the apple tissue that slowly increases until reaching the atmospheric pressure, in a time scale that depends on the vacuum applied during the vacuum step. The physiological response of apple tissue on Vacuum Impregnation process was studied indicating the possibility of vesicular transport within the cells. Finally, the possibility to extend the freezing tolerance of strawberry fruits impregnated with cryoprotectants was proven.

The aim of this PhD thesis was to evaluate the effect of a sub-lethal HPH treatment on some probiotic properties and on cell response mechanisms of already-known functional strains, isolated from Argentinean dairy products. The results achieved showed that HPH treatments, performed at a sub-lethal level of 50 MPa, increased some important functional and technological characteristics of the considered non intestinal probiotic strains. In particular, HPH could modify cell hydrophobicity, autoaggregation and resistance to acid gastric conditions (tested in in vitro model), cell viability and cell production of positive aroma compounds, during a refrigerate storage in a simulated dairy product. In addition, HPH process was able to increase also some probiotic properties exerted in vivo and tested for two of the considered strains. In fact, HPH-treated cells were able to enhance the number of IgA+ cells more than other not treated cells, although this capacity was time dependent. On the other hand, HPH treatment was able to modify some important characteristics that are linked to the cell wall and, consequently, could alter the adhesion capacity in vivo and the interaction with the intestinal cells. These modifications, involving cell outermost structures, were highlighted also by Trasmission Electron Microscopy (TEM) analysis. In fact, the micrographs obtained showed a significant effect of the pressure treatment on the cell morphology and particularly on the cell wall. Moreover, the results achieved showed that composition of plasma membranes and their level of unsaturation are involved in response mechanisms adopted by cells exposed to the sub-lethal HPH treatment. Although the response to the treatment varied according to the characteristics of individual strains, time of storage and suspension media employed, the results of present study, could be exploited to enhance the quality of functional products and to improve their organoleptic properties.

This PhD thesis is aimed at studying the possible pathways and the mechanisms that can trigger oxylipins biosynthesis, and particularly that of short chain aldehydes and alcohols, in Lactobacillus helveticus, also in the presence of oxidative stress, using a totally labelled linoleic acid as precursor. In plants and fungi these molecules, involved in defence mechanisms against pathogens and in communication systems, derive from the oxidation of cellular unsaturated fatty acids (UFAs) and their accumulation is associated with stress exposure. Since some oxylipins are produced also by lactobacilli, it is possible to hypothesize that a metabolic pathway from UFAs to oxylipins, similar to what happens in plants and fungi, is present also in lactic acid bacteria. The results obtained pointed out that some volatile molecules are the result of UFAs catabolism, since they appear only when cells are incubated in their presence. Labelled linoleic acid is integrated in the membrane and subsequently transformed into aldehydes and alcohols, whose extent and carbon atoms number depend on stress exposure. The enzymes responsible for this metabolic pathway in plants and fungi (e.g. lipoxygenase, dioxygenase) seem to be absent in Lactobacillus helveticus and in other lactobacilli. Proteomic analyses show the over expression of many proteins, including thioredoxin reductase (part of the bacterial oxidative defence system), mainly in cells grown with linoleic acid without oxidative stress exposure, confirming that linoleic acid itself induces oxidative stress. 6 general oxidoreductases (class including dioxygenases and peroxidase) were found and therefore a deeper investigation on them could be productive in elucidating all steps involved in oxylipins biosynthesis in bacteria. Due to the multiple role of oxylipins (flavouring agents, antimicrobial compounds and interspecific signalling molecules) the identification of genes involved and regulating factors should have an important biotechnological impact, also allowing the overproduction of selected bioactive molecules.
