Flucloxacillin sodium (FLU) is a semi-synthetic penicillin active against many gram-positive bacteria such as streptococci and penicilinase-producing staphylococci, including methicillin-susceptible S. aureus. This study describes the development and validation of a microbiological assay, applying the diffusion agar method for the determination of FLU, as well as the evaluation of the ability of the method in determining the stability of FLU in capsules against acidic and basic hydrolysis, photolytic and oxidative degradations, using S. aureus ATCC 25923 as micro-organism test and 3 x 3 parallel line assay design (three doses of the standard and three doses of the sample in each plate), with six plates for each assay, according to the Brazilian Pharmacopoeia. The validation method showed good results including linearity, precision, accuracy, robustness and selectivity. The assay is based on the inhibitory effect of FLU using Staphylococcus aureus ATCC 25923. The results of the assay were treated by analysis of variance (ANOVA) and were found to be linear (r = 0.9997) in the range from 1.5 to 6.0 μg/mL, precise (repeatability: R.S.D. = 1.63 and intermediate precision: R.S.D. = 1.64) and accurate (98.96%). FLU solution (from the capsules) exposed to direct UVC light (254 nm), alkaline and acid hydrolysis and hydrogen peroxide causing oxidation were used to evaluate the specificity of the bioassay. Comparison of bioassay and liquid chromatography by ANOVA showed no difference between methodologies. The results demonstrated the validity of the proposed bioassay, which is a simple and useful alternative methodology for FLU determination in routine quality control.

Introduction: The analysis of body composition through direct and indirect methods allows the study of the various components of the human body, becoming the central hub for assessing nutritional status. Objective: The objective of the study was to develop equations for predicting body fat% from circumferential body arm, waist and calf and propose percentiles to diagnose the nutritional status of school children of both sexes aged 4-10 years. Methods: We selected intentionally (non-probabilistic) 515 children, 261 children and 254 being girls belonging to Program interaction and development of children and adolescents from the State University of Campinas (Sao Paulo, Brazil). Anthropometric variables were evaluated for weight, height, triceps and subscapular skinfolds and body circumferences of arm, waist and calf, and the% fat determined by the equation proposed by Boileau, Lohman and Slaughter (1985). Through regression method 2 were generated equations to predict the percentage of fat from the body circumferences, the equations 1 and 2 were validated by cross validation method. Results: The equations showed high predictive values ranging with a R2 = 64-69%. In cross validation between the criterion and the regression equation proposed no significant difference (p > 0.05) and there was a high level of agreement to a 95% CI. Conclusion: It is concluded that the proposals are validated and shown as an alternative to assess the percentage of fat in school children of both sexes aged 4-10 years in the region of Campinas, SP (Brazil).

A gas chromatographic method to determine caprolactam in multilayer PA-6 films used for meat foodstuffs and cheese was developed and validated. A wide linear range (0.8-400 mu g/ml), RSD <= 4.1% and recovery higher than 90.0% were obtained for the chromatographic system, while precision and accuracy of the method showed RSD <= 3.8%, recovery from 95.5-100.0% and LOQ of 32 mu g/g. Irradiated (3, 7 and 12 kGy) and non-irradiated commercial films were analyzed. Most of them increased caprolactam levels with the increase of irradiation doses. (C) 2008 Elsevier Ltd. Cyclic oligomers were identified in PET bottles used for mineral water and fruit juice using MS and H-1 and C-13 NMR: a first series cyclic trimer, a first series cyclic tetramer, a first series cyclic dimmer and a second series cyclic trimer. An analytical method to determine first series cyclic trimer in these bottles was developed and validated, using HPLC. The first series cyclic trimer levels were 316-462 mg/100 g of PET bottle.
