Docking simulations have been used to assess protein complexes with some success. Small angle X-ray scattering (SAXS) is a well-established technique to investigate protein spatial configuration. This work describes the integration of geometric docking with SAXS to investigate the quaternary structure of recombinant human purine nucleoside phosphorylase (PNP). This enzyme catalyzes the reversible phosphorolysis of N-ribosidic bonds of purine nucleosides and deoxynucleosides. A genetic deficiency due to mutations in the gene encoding for PNP causes gradual decrease in T-cell immunity. Inappropriate activation of T-cells has been implicated in several clinically relevant human conditions such as transplant rejection, rheumatoid arthritis, lupus, and T-cell lymphomas. PNP is therefore a target for inhibitor development aiming at T-cell immune response modulation and has been submitted to extensive structure-based drug design. The present analysis confirms the trimeric structure observed in the crystal. The potential application of the present procedure to other systems is discussed. (C) 2003 Elsevier B.V. Studies of post-dispersal seed removal in the Neotropics have rarely examined the magnitude of seed removal by different types of granivores. The relative impact of invertebrates, small rodents, and birds on seed removal was investigated in a 2,178 ha Atlantic forest fragment in southeastern Brazil. We used popcorn kernels (Zea mays-Poaceae) to investigate seed removal in a series of selective exclosure treatments in a replicated, paired design experiment that included forest understory, gaps, and forest edge sites. We recorded the vegetation around the experimental seed stations in detail in order to evaluate the influence of microhabitat traits on seed removal. Vertebrate granivores (rodents and birds) were surveyed to determine whether granivore abundance was correlated with seed removal levels. Seed removal varied spatially and in unpredictable ways at the study site. Seed encounter and seed use varied with treatments, but not with habitat type. However, seed removal by invertebrates was negatively correlated with gap-related traits, which suggested an avoidance of large gaps by granivorous ants. The abundance of small mammals was remarkably low, but granivorous birds (tinamous and doves) were abundant at the study site. Birds were the main seed consumers in open treatments, but there was no correlation between local granivorous bird abundance and seed removal. These results emphasize the stochastic spatial pattern of seed removal, and, contrary to previous studies, highlight the importance of birds as seed predators in forest habitats. In the present study, a single procedure was established to investigate the effect of the spatial distribution of immatures in patchy resources, on the outcome of larval competition for food. in experimental populations of Chrysomya megacephala. A theoretical model of intraspecific competition was extended and applied to experimental data on survival to adulthood for 20 larval densities, to obtain the theoretical mean number of individuals that will survive, considering a hypothetical previous random adult oviposition in a system of homogeneous patches. The survival curve obtained suggests that the larval competition for food in C. megacephala is of the scramble/exploitative type, which corroborates results from previous studies, although the latter did not consider the correlation between local and global abundances. The present model allows that experimental data could be perfectly applicable, and it incorporates fundamental assumptions about the spatial context of competition for patchy resources in blowflies, and may be applied to the optimization of mass rearing techniques and to the maintenance of insect colonies under experimental conditions.

In recent years, studies based on isoenzymatic patterns of geographic variation have revealed that what is usually called the Africanized honey bee does not constitute a single population. Instead, several local populations exist with various degrees of admixture with European honey bees. In this paper, we evaluated new data on morphometric patterns of Africanized honey bees collected at 42 localities in Brazil, using univariate and multivariate (canonical) trend surface and spatial autocorrelation analyses. The clinal patterns of variation found for genetically independent characters (wing size characters and some wing venation angles) are concordant with previous studies of malate dehydrogenase (MDH) allelic frequencies and support the hypothesis that larger honey bees in southern and southeastern Brazil originated by racial admixture in the initial phases of African honey bee colonization. Geographic variation patterns of Africanized honey bee populations reflect a demic diffusion process in which European genes were gradually lost because of the higher fitness of the African gene pool in Neotropical environmental conditions.

Two studies were carried out to analyze whether learning technical drawing improves a person's ability for spatial visualization. Visualization and inductive reasoning tests were applied at the beginning and end of a course in technical drawing in samples of first year engineering students. In both studies it was observed that a moderate percentage of students improved their Visualization test execution. The improvement was similar in men and women. There was no improvement on the inductive reasoning test. The results support the conclusion that the spatial visualization ability can be improved with training.

Several studies suggest that, on a large scale, relief conditions influence the Atlantic Forest cover. The aim of this work was to explore these relationships on a local scale, in Caucaia do Alto, on the Ibiúna Plateau. Within an area of about 78 km2, the distribution of forest cover, divided into two successional stages, was associated with relief attribute data (slope, slope orientation and altitude). The mapping of the vegetation was based on the interpretation of stereoscopic pairs of aerial photographs, from April 2000, on a scale of 1:10,000, while the relief attributes were obtained by geoprocessing from digitalized topographic maps on a scale of 1:10,000. Statistical analyses, based on qui-square tests, revealed that there was a more extensive forest cover, irrespective of the successional stage, in steeper areas (>10 degrees) located at higher altitudes (>923 m), but no influence of the slope orientation. There was no sign of direct influence of relief on the forest cover through environmental gradients that might have contributed to the forest regeneration. Likewise, there was no evidence that these results could have been influenced by the distance from roads or urban areas or with respect to permanent preservation areas. Relief seems to influence the forest cover indirectly, since agricultural land use is preferably made in flatter and lower areas. These results suggest a general distribution pattern of the forest remnants, independent of the scale of study, on which relief indirectly has a strong influence, since it determines human occupation.

Few studies have examined the effects of temperature on spatial and temporal trends in soil CO2-C emissions in Antarctica. In this work, we present in situ measurements of CO2-C emissions and assess their relation with soil temperature, using dynamic chambers. We found an exponential relation between CO2 emissions and soil temperature, with the value of Q10 being close to 2.1. Mean emission rates were as low as 0.026 and 0.072 g of CO2-C m-2 h-1 for bare soil and soil covered with moss, respectively, and as high as 0.162 g of CO2-C m-2 h-1 for soil covered with grass, Deschampsia antarctica Desv. (Poaceae). A spatial variability analysis conducted using a 60-point grid, for an area with mosses (Sannionia uncianata) and D. antarctica, yielded a spherical semivariogram model for CO2-C emissions with a range of 1 m. The results suggest that soil temperature is a controlling factor on temporal variations in soil CO2-C emissions, although spatial variations appear to be more strongly related to the distribution of vegetation types. © 2010 Elsevier B.V. and NIPR.

How individual-level movement decisions in response to habitat edges influence population-level patterns of persistence and spread of a species is a major challenge in spatial ecology and conservation biology. Here, we integrate novel insights into edge behavior, based on habitat preference and movement rates, into spatially explicit growth-dispersal models. We demonstrate how crucial ecological quantities (e.g., minimal patch size, spread rate) depend critically on these individual-level decisions. In particular, we find that including edge behavior properly in these models gives qualitatively different and intuitively more reasonable results than those of some previous studies that did not consider this level of detail. Our results highlight the importance of new empirical work on individual movement response to habitat edges. © 2013 by The University of Chicago.
