The research aimed to estimate body contents of protein and energy and net requirements of energy for maintenance of buffaloes, slaughtered at different stages of maturity. There were used 14 Mediterranean intact males with initial average body weight of 352.2 +/- 24.3 kg and average age of 24 months. The animais were randomly divided into four experimental groups. One group was designed to slaughter at the beginning of the experimental period (IS). The animals of another group were restricting fed, receiving, individually, levels of protein and energy 15% above maintenance (RF). The animals of the two remaining groups were individually fed ad libitum (SW450 and SW500) to reach weights corresponding to 100 and 110 percent of the mature weight of the buffalo cows (respectively 450 and 550 kg). The ration contained ground-corn cobs, soybean meal, urea, minerals, and signal-grass (Brachiaria decumbens) hay, with a concentrate: roughage ratio of 50: 50 and 13% of crude protein on a dry matter basis. To estimate changes in body composition inside the range of weights included in the trial, linear regression equations of log protein (kg), fat (kg) and energy (Mcal) as a function of log empty-body-weight (EBW), in kg, were fitted. Energy requirements for maintenance were obtained as estimated heat production at zero level of energy intake. Buffaloes submitted to fattening in feedlot presented early body fat deposition, and had with the same live weight lower protein content and higher fat content and energy per unit weight than european-zebu crossbred cattle.

Background. About 130 million people are infected with the hepatitis C virus (HCV) worldwide, but effective treatment options are not yet available. One of the most promising targets for antiviral therapy is nonstructural protein 3 (NS3). To identify possible changes in the structure of NS3 associated with virological sustained response or non-response of patients, a model was constructed for each helicase NS3 protein coding sequence. From this, the goal was to verify the interaction between helicases variants and their ligands. Findings. Evidence was found that the NS3 helicase portion of non-responder patients contained substitutions in its ATP and RNA binding sites. K210E substitution can cause an imbalance in the distribution of loads, leading to a decrease in the number of ligations between the essential amino acids required for the hydrolysis of ATP. W501R substitution causes an imbalance in the distribution of loads, leading and forcing the RNA to interact with the amino acid Thr269, but not preventing binding of ribavirin inhibitor. Conclusions. Useful information is provided on the genetic profiling of the HCV genotype 3, specifically the coding region of the NS3 protein, improving our understanding of the viral genome and the regions of its protein catalytic site. © 2010 Rahal et al; licensee BioMed Central Ltd.

O objetivo deste estudo foi identificar um marcador bioquímico de folículos bovinos com diâmetro maior do que 10 mm, o qual poderia ser utilizado para identificar folículos dominantes nesta espécie. Líquido folicular era aspirado de folículos com diâmetros menores do que 5 mm, entre 5 e 10 mm e maiores do que 10 mm, provenientes de ovários de 37 fêmeas bovinas abatidas. Após aspiração, o líquido folicular (LF) era acondicionado em ependorfs etiquetados e congelados. Os padrões eletroforéticos em SDS-PAGE das proteínas do LF foram determinados e verificou-se que os folículos com diâmetro maior do que 10 mm apresentaram um polipeptídeo com PM entre 39 e 43 KDa identificado como a proteína ligante de IGF-3 (IGFBP-3), o qual não foi observado em folículos com diâmetro menor do que 5 mm e entre 5 e 10 mm. Este estudo sugere que este polipeptídeo possa ser utilizado como marcador bioquímico de folículos maiores do que 10 mm.

Um experimento foi conduzido para comparar diversos níveis e fontes protéicas utilizados em rações sobre o desempenho, a morfometria intestinal e a relação peso de pâncreas/peso de carcaça de leitões de 36 a 70 dias de idade. Foram utilizados 96 leitões desmamados distribuídos em delineamento de blocos ao acaso com seis tratamentos e quatro repetições de quatro animais. Avaliaram-se seis fontes protéicas (tratamentos): leite em pó desnatado (8,80 e 12,00%); isolado protéico de soja (3,20 e 4,50%); farinha de peixe (5,00%); e levedura seca (10,00%). As dietas, isoenergéticas e isoprotéicas, não afetaram o ganho de peso e a conversão alimentar dos animais, contudo, os animais que receberam a dieta contendo leite em pó desnatado apresentaram maior consumo no período de 56 a 63 dias de idade. Não houve efeito significativo das fontes protéicas sobre a altura de vilos, a profundidade de cripta e a relação peso do pâncreas/peso corporal. As fontes protéicas estudadas e os níveis utilizados nas dietas não influenciaram o desempenho, a morfologia intestinal e a relação peso de pâncreas/peso de carcaça em leitões de 36 a 70 dias de idade

The goal of treatment of chronic hepatitis C is to achieve a sustained virological response, which is defined as exhibiting undetectable hepatitis C virus (HCV) RNA levels in serum following therapy for at least six months. However, the current treatment is only effective in 50% of patients infected with HCV genotype 1, the most prevalent genotype in Brazil. Inhibitors of the serine protease non-structural protein 3 (NS3) have therefore been developed to improve the responses of HCV-infected patients. However, the emergence of drug-resistant variants has been the major obstacle to therapeutic success. The goal of this study was to evaluate the presence of resistance mutations and genetic polymorphisms in the NS3 genomic region of HCV from 37 patients infected with HCV genotype 1 had not been treated with protease inhibitors. Plasma viral RNA was used to amplify and sequence the HCV NS3 gene. The results indicate that the catalytic triad is conserved. A large number of substitutions were observed in codons 153, 40 and 91; the resistant variants T54A, T54S, V55A, R155K and A156T were also detected. This study shows that resistance mutations and genetic polymorphisms are present in the NS3 region of HCV in patients who have not been treated with protease inhibitors, data that are important in determining the efficiency of this new class of drugs in Brazil.

The objective of this study was to evaluate the chemical composition and dry matter in vitro digestibility of stem, leaf, straw, cob and kernel fractions of eleven corn (Zea mays) cultivars, harvested at two cutting heights. The experiment was designed as randomized blocks, with three replicates, in a 2 × 11 factorial arrangement (eleven cultivars and two cutting heights). The corn cultivars evaluated were D 766, D 657, D 1000, P 3021, P 3041, C 805, C 333, AG 5011, FOR 01, CO 9621 and BR 205, harvested at a low cutting height (5 cm above ground) and a high cutting height (5 cm below the first ear insertion). Cutting height influenced the dry matter content of the stem fraction, which was lower (23.95%) in plants harvested at the low, than in plants harvested at the high cutting height (26.28%). The kernel fraction had the highest dry matter in vitro digestibility (85.13%), while cultivars did not differ between each other. Cob and straw were the fractions with the highest level of neutral detergent fiber (80.74 and 79.77%, respectively) and the lowest level of crude protein (3.84% and 3.69%, respectively). The leaf fraction had the highest crude protein content, both for plants of low and high cuttings (15.55% and 16.20%, respectively). The increase in the plant cutting height enhanced the dry matter content and dry matter in vitro digestibility of stem fraction, but did not affect the DM content of the leaf fraction.

To investigate the alterations of glucose homeostasis and variables of the insulin-like growth factor-I (IGF- 1) growth system in sedentary and trained diabetic (TD) rats, Wistar rats were divided into sedentary control (SC), trained control (TC), sedentary diabetic (SD), and TD groups. Diabetes was induced by Alloxan (35 mg kg(-1) b.w.). Training program consisted of swimming 5 days week(-1), 1 h day(-1), during 8 weeks. Rats were sacrificed and blood was collected for determinations of serum glucose, insulin, growth hormone (GH), IGF-1, and IGF binding protein-3(IGFBP-3). Muscle and liver were removed to evaluate glycogen content. Cerebellum was extracted to determinate IGF-1 content. Diabetes decreased serum GH, IGF-1, IGFBP-3, liver glycogen, and cerebellum IGF-1 peptide content in baseline condition. Physical training recovered liver glycogen and increased serum and cerebellum IGF-1 peptide in diabetic rats. Physical training induces important metabolic and hormonal alterations that are associated with an improvement in glucose homeostasis and serum and cerebellum IGF-1 concentrations. O objetivo deste trabalho foi avaliar a produção e composição do leite de vacas de corte das raças Charolês (C), Nelore (N), mestiças CN filhas de touros C e mestiças NC filhas de touros N, mantidas em pastagem nativa e submetidas a diferentes manejos no pré-desmame: suplementadas com farelo de arroz integral (0,7% do peso vivo) e que desmamaram aos 42 ou 63 dias pós-parto, ou não suplementadas e que desmamaram aos 63 dias. A idade das vacas variou de 3 a 12 anos, sendo agrupadas em quatro classes: primíparas, jovens, adultas e velhas. Os dados foram submetidos à análise de variância, cujo modelo estatístico incluiu os efeitos de manejo, grupo genético e idade da vaca, período e as interações entre esses fatores. Os teores de lactose e gordura não foram influenciados pelos fatores estudados. Houve interação significativa entre idade da vaca e período para a produção de leite e entre manejo e idade da vaca para o teor de proteína. Vacas suplementadas apresentaram maior produção de leite (3,85 contra 3,25 L/dia), teor de extrato seco total - EST (12,18 contra 11,83%) e teor de extrato seco desengordurado - ESD (8,75 contra 8,57%). A produção de leite decresceu com o avanço do período de lactação, sendo a queda mais acentuada nas vacas não suplementadas. Vacas CN produziram mais leite (4,17 L/dia) do que as puras, não diferindo das NC (3,76 L/dia). Vacas N apresentaram produção de leite similar (3,76 L/dia) às C (3,11 L/dia). No entanto, as primeiras apresentaram leite com melhor qualidade, incluindo proteína (3,16 contra 2,86%), EST (12,52 contra 11,46%) e ESD (8,87 contra 8,49%). Vacas adultas apresentaram maior teor de ESD, seguidas das vacas jovens, das primíparas e das velhas, com valores de 8,86; 8,62; 8,62; e 8,54%, respectivamente.

Aims: To evaluate the reliability of fine needle aspirate cell blocks in the assessment of oestrogen receptor (ER), progesterone receptor (PR) and HER-2/neu proteins by immunohistochemistry in comparison with surgical specimens. Materials and methods: This is a retrospective study of 62 cases of breast carcinoma diagnosed by fine needle aspiration cytology (FNAC) and confirmed using the surgical specimen. Immunohistochemical tests were performed to assess the presence of oestrogen receptor (ER), progesterone receptor (PR) and HER-2/neu proteins in cell blocks and the corresponding surgical specimens. The cell block method used alcohol prior to formalin fixation. Cases with 10% or more stained cells were considered positive for ER and PR. Positivity for HER-2/neu was assessed on a scale of 0-3+. The criterion for positivity was a score of 3+. Results: Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV) and accuracy of the cell blocks in the investigation of ER, PR and HER-2/neu protein (3+) were (%): ER, 92.7, 85.7, 92.7, 85.7 and 90.3; PR, 92.7, 94.7, 97.4, 87.0 and 93.5; HER-2/neu, 70.0, 100.0, 100.0, 94.5 and 95.2. Discrepancies were seen in cell blocks in the 1+ and 2+ HER-2/neu staining scores: two of 12 cases scoring 2+ and one case of 26 scoring 1+ on cell blocks scored 3+ on surgical specimens. The correlation index between cell block and corresponding surgical specimen varied from 90% to 94%. Conclusion: Cell blocks provide a useful method of assessing ER, PR and HER-2/neu, mainly for inoperable and recurrent cases, but consideration should be given to carrying out FISH analysis on 1+ as well as 2+ HER-2/neu results. © 2012 Blackwell Publishing Ltd.

Periodontitis is an inflammatory disease caused by pathogenic microorganisms and characterized by the destruction of the periodontium. Obese individuals have an increased risk of periodontitis, and elevated circulating levels of adipokines, such as nicotinamide phosphoribosyltransferase (NAMPT), may be a pathomechanistic link between both diseases. The aim of this in vitro study was to examine the regulation of periodontal ligament (PDL) cells by NAMPT and its production under inflammatory and infectious conditions. NAMPT caused a significant upregulation of 9 genes and downregulation of 3 genes, as analyzed by microarray analysis. Eight of these genes could be confirmed by real-time PCR: NAMPT induced a significant upregulation of EGR1, MMP-1, SYT7, ITPKA, CCL2, NTM, IGF2BP3, and NRP1. NAMPT also increased significantly the MMP-1 and CCL2 protein synthesis. NAMPT was significantly induced by interleukin-1β and the periodontal microorganism P. gingivalis. NAMPT may contribute to periodontitis through upregulation of MMP-1 and CCL2 in PDL cells. Increased NAMPT levels, as found in obesity, may therefore represent a mechanism whereby obesity could confer an increased risk of periodontitis. Furthermore, microbial and inflammatory signals may enhance the NAMPT synthesis in PDL cells and thereby contribute to the increased gingival and serum levels of this adipokine, as found in periodontitis. © 2013 Marjan Nokhbehsaim et al.
