The obtainment of transgenic edible plants carrying recombinant antigens is a desired issue in search for economic alternatives viewing vaccine production. Here we report a strategy for genetic transformation of lettuce plants (Lactuca sativa L.) using the surface antigen HBsAg of hepatitis B virus. Transgenic lettuce seedlings were obtained through the application of a regulated balance of plant growth regulators. Genetic transformation process was acquired by cocultivation of cotyledons with Agrobacterium tumefaciens harboring the recombinant plasmid. It is the first description of a lettuce Brazilian variety Vitória de Verão genetically modified.

The strategy for breeding F-1 hybrid squash is to develop parental lines through self-pollination. However, it increases plant mean homozygosis, which is not the natural genetic state of a cross-pollinated species, and can cause inbreeding depression. The objective of this work was to evaluate this depression with sucessive generations of self-pollination (without selection) in Cucurbita moschata, cv. Piramoita. Populations were obtained from lines with one to four generations of self-pollination (obtained by the SSD method), from the original cv. Piramoita (population SO). Randomized blocks were used with five treatments (different generations of self-pollination -S-0 to S-4), six replicates and five plants per plot. Regression analysis was made by the Wright inbreeding coefficient (F) to measure the homozygosis level effect on vigor loss. There was a linear reduction of mean weight and fruit length, seed production (number and weight) per fruit with the increase of the homozygosis level; however inbreeding did not affect seed quality (weight of 100 seeds and germination).

An Arabidopsis thaliana cDNA clone encoding a plant uncoupling mitochondrial protein (AtPUMP1) was overexpressed in transgenic tobacco plants. Analysis of the AtPUMP1 mRNA content in the transgenic lines, determined by Northern blot, revealed variable levels of transgene expression. Antibody probing of Western blots of mitochondrial proteins from three independent transgenic lines showed significant accumulation of AtPUMP1 in this organelle. Overproduction of AtPUMP1 in transgenic tobacco plants led to a significant increase in tolerance to oxidative stress promoted by exogenous hydrogen peroxide as compared to wild-type control plants. These results provide the first biological evidence for a role of PUMP in protection of plant cells against oxidative stress damage.

O rendimento e a qualidade de sementes de abóbora podem ser aprimorados por uma densidade populacional de plantas adequada, além da quantidade de pólen, que influi na qualidade da polinização e fertilização. Para este estudo, aplicaram-se nove tratamentos, que resultaram da combinação fatorial de três espaçamentos entre plantas (0,8 x 0,3; 0,8 x 0,6 e 0,8 x 0,9 m) com duas quantidades de pólen (50% de uma antera e uma antera inteira) e a polinização natural por insetos. Foram avaliados parâmetros de produção de frutos e sementes, além da qualidade física e fisiológica das sementes. O delineamento experimental adotado foi em blocos ao acaso com cinco repetições e dez plantas úteis por amostragem. O aumento do espaçamento entre plantas elevou o número médio de frutos maduros e a produção de sementes por planta. A produtividade de sementes foi diretamente proporcional à quantidade de pólen usada durante a polinização. Já o fator espaçamento entre plantas não afetou a produção de sementes por área. A maior quantidade de pólen resultou em uma maior produção de sementes por planta e por área. A polinização manual, com a utilização de uma antera inteira, não diferiu da polinização natural com insetos, tanto para rendimento como para a qualidade de sementes.

The objective was to determine the effect of a mouse metallothionein/bovine growth hormone transgene on resting metabolic rate (RMR), cold-induced thermogenesis, and beta-agonist stimulated nonshivering thermogenesis in mice. Non-transgenic littermates were used as controls. Open-circuit indirect calorimetry was used to assess RMR and cold-induced thermogenesis in 64 mice. Air temperature in the chamber was set at 31 degrees C for RMR and was decreased to 28, 25, 21, or 17 degrees C to determine cold-induced thermogenesis. Response to the beta-agonist isoproterenol was evaluated by monitoring changes in colonic temperature of 34 mice upon injection of the drug or saline. Despite the fact that RMR tended to be lower in transgenics than in nontransgenics, at 31 degrees C transgenic mice were able to regulate colonic temperature at the same level as nontransgenics, but colonic temperature decreased in transgenics relative to nontransgenics as air temperature was reduced. For each degree decrease in air temperature between 31 and 17 degrees C, nontransgenic mice increased heat production by 1.03 +/- .10 watt/kg, whereas transgenic mice increased it by only .56 +/- .08 watt/kg, indicating that the thermogenic response of transgenics to cold was inferior. The magnitude of the maximal increase in colonic temperature after isoproterenol injection was similar for both groups, but the response was slower in transgenics. We suggest that lean body mass and substrate availability for shivering thermogenesis are reduced in transgenics relative to total body weight, and that they allow colonic temperature to decrease to conserve energy.
