A new species of the genus Amazophrynella is described from southern Amazonia, Brazil. The new species is characterized by its medium size for the genus (15.2-19.3 mm SVL in males, 20.2-25.7 mm SVL in females), snout rounded in dorsal view, acute in lateral view, presence of subrostral crest not converging anteriorly, finger I shorter than finger II, and by dorsal and ventral coloration. The reproductive behavior of the new species is similar to other Amazophrynella with pigmented and numerous eggs that are probably laid in temporary ponds.

A new genus and species of cricket belonging to the Hapithinae: Neomorphini (Podoscirtidae) is described from the forests of the Iguacu National Park, State of Parana, Brazil. The new genus is thoroughly compared with Neomorpha Desutter, 1987, the only other member of the Neomorphini described so far.

Two new species of brachycephalid frogs are described from Pico Marumbi, municipality of Morretes, and Pico da Igreja, municipality of Guaratuba, Parana State, southern Brazil. The new species share the following attributes: body bufoniform; skin on top of the head, and central part of the back body smooth with no dermal co-ossification; outer metatarsal tubercle distinct; dermal roofing bones of skull unornamented; all paired cranial bones distinct and not fused; quadratojugals, and maxillary odontoids present. The new species from Pico Marumbi is characterized by male SVL = 11.6-12.5 and female SVL = 13.0-14.5 turn; and general color orange with dorsal reddish-brown irregular markings, lateral surfaces with small dark brown spots, and belly with brownish spots and small dots. The new species from Pico da Igreja is characterized by male SVL = 12.6-13.9 and female SVL = 14.6-15.3 mm; and general color orange, lateral surfaces with small dark brown spots, and belly with brownish coalescent spots and small dots. Comparisons with other brachycephalid species and osteological data are provided.

A new species of Aparasphenodon is described from patches of arboreal restinga within the Atlantic Forest Biome, in a region known as Baixo Sul in southern Bahia, northeastern Brazil. Aparasphenodon arapapa sp. nov. is promptly diagnosed from other Aparasphenodon mainly by having small size (male snout-vent length 57.4-58.1 mm), loreal region flattened and wide, and canthus rostralis rounded and poorly elevated. The wide and flattened snout resembles that found in Triprion and Diaglena, and possibly is a parallelism (homoplasy) related to the phragmotic behavior of casque-headed tree frogs to their microhabitat usage. The decision to allocate the new species in the genus Aparasphenodon is discussed in detail, as the single morphological synapomorphy of the genus, the presence of a prenasal bone, is insufficient to morphologically relate the new species to Aparasphenodon, Triprion, or Diaglena.

A new species of Ophiomisidium is described and regards about reproduction are presented. It is the first species in the genus to posses, apparently,. a brooding development. Very few specimens of species of Ophiomisidium have previously been collected, but over 3000 specimens of the new species were collected on the continental shelf and slope (60-810 m depth) off southern Brazil. They were obtained during the REVIZEE Programme, the first extensive surveys to examine Brazilian ophiuroids from depths greater than 200 m.

The fragile X syndrome (FXS), the most common cause of hereditary mental retardation, is caused by expansions of CGG repeats in the FMR1 gene. The gold-standard method to diagnose FXS is the Southern blot (SB). Because SB is laborious and costly, some adaptations in the polymerase chain reaction (PCR) method have been utilized for FXS screening. A previous PCR-based screening method for FXS identification utilizing small amounts of DNA was reported as simple and efficient. The aim of this study was to reproduce the mentioned PCR-based screening method for identification of expanded alleles of the FMR1 gene in Brazilian individuals and to investigate the efficiency of this method in comparison with SB. Utilizing the enzyme Expand Long Template PCR System, 78 individuals were investigated by that PCR-based screening method for FXS identification. Conclusive results were obtained for 75 samples. Considering all the allelic forms of FXS (normal [NL], premutation [PM], and full-mutation [FM]), the comparison of the PCR-based screening method with SB demonstrated 100% of accuracy, sensitivity, and specificity. However, when the PM and the FM were analyzed separately from each other, but together with the NL allele, the accuracy, sensitivity, and specificity decreased (to 42.9%-97.4%). We concluded that the PCR-based screening method was reproducible and capable of identifying all different FXS alleles, but because the differentiation between the PM and the FM alleles was not accurate, SB is still the gold-standard method for the molecular diagnosis of FXS.
