The seed lectin from Lotus tetragonolobus (LTA) has been crystallized. The best crystals grew over several days and were obtained using the vapour-diffusion method at a constant temperature of 293 K. A complete structural data set was collected at 2.00 angstrom resolution using a synchrotron-radiation source. LTA crystals were found to be monoclinic, belonging to space group P2(1), with unit-cell parameters a = 68.89, b = 65.83, c = 102.53 angstrom, alpha = gamma = 90, beta = 92 degrees. Molecular replacement yielded a solution with a correlation coefficient and R factor of 34.4 and 51.6%, respectively. Preliminary analysis of the molecular-replacement solution indicates a new quaternary association in the LTA structure. Crystallographic refinement is under way.

Lotus (Nelumbo nucifera Gaertn.) is a perennial herbaceous aquatic ornamental plant with potential to be used as a new cut flower for the Brazilian ornamental market. It shows exotic and attractive flowers and has a strong market appeal, once it is known as a symbol of purity, holiness and immortality. However, flowers have a short-vase life. Lotus flower stem exudes a large quantity of sticky milky sap from the cut surface, which is produced in laticifers, spatially associated with both xylem and phloem. It has been reported that latex coagulates on the cut surface preventing or reducing water absorption and reducing flowers' vaselife, requiring treatments to stop the flow of latex. The objective of this study was to report observations of lotus postharvest characteristics and evaluate treatments to overcome latex flow. The experiment was conducted as a complete randomized design with three replications of four stems in each vase and eight treatments; a control (distilled water), pretreatment of cut stem-ends with hot water (40° C/1 minute), boiling water (3 seconds), isopropyl alcohol 90% (10 minutes) or citric acid (pH = 2.8/1h) and, maintenance of stems in a holding solution of Tween® 20 (0.01%), citric acid (200 mg L-1) or Tween® 20 (0.01%) plus citric acid (200 mg L-1). Treatments had no significant effect on flowers vaselife which was only about three days, although isopropyl alcohol, hot and boiling water completely stopped latex flow. Cut stem-ends pretreated with citric acid (pH = 2.8/1 h) showed a significantly higher relative water content of petals compared to others treatments. The senescence symptom of lotus cut flowers was mainly characterized by abscission of turgid petals and dehiscence of stamens without any visual change of petal color and brightness.

Lotus tetragonolobus lectin (LTA) is a fucose-specific legume lectin. Although several studies report a diverse combination of biological activities for LTA, little is known about the mechanisms involved in L-fucosyl oligosaccharide recognition. The crystal structure of LTA at 2.0 angstrom resolution reveals a different legume lectin tetramer. Its structure consists of a homotetramer composed of two back-to-back GS4-like dimers arranged in a new mode, resulting in a novel tetramer. The LTA N-linked carbohydrate at Asn4 and the unusual LTA dimer-dimer interaction are related to its particular mode of tetramerization. In addition, we used small angle X-ray scattering to investigate the quaternary structure of LTA in solution and to compare it to the crystalline structure. Although the crystal structure of LTA has revealed a conserved metal-binding site, its L-fucose-binding site presents some punctual differences. Our investigation of the new tetramer of LTA and its fucose-binding site is essential for further studies related to cross-linking between LTA and complex divalent L-fucosyl carbohydrates. Based on material from several collections, summarized distributions and new records are presented for selected Neotropical species of Gerromorpha of the families Gerridae (Brachymetra albinervis albinervis, B. furva, B. lata, B. shawi, Cylindrostethus erythropus, C. linearis, C. palmaris, C. regulus, Halobatopsis platensis, Limnogonus aduncus aduncus, L. hyalinus, L. ignotus, L. profugus, L. recurvus, Neogerris lubricus, N. magnus, N. lotus, N. visendus, Ovatametra gualeguay, Rheumatobates crassifemur crassifemur, R. c. esakii, Tachygerris adamsoni, T. celocis and T. surinamensis), Hydrometridae (Hydrometra guianana and H. sztolcmani) and Mesoveliidae (Mesovelia amoena, M. mulsanti and Mesoveloidea williamsi).
