Objective: the aim of this study was to evaluate the bone healing in spontaneously hypertensive rats (SHR) and compare the results with normotensive rats, evaluating male and female animals.Methods: A bone drill defect was created in the left femur of 24 SHR (12 males and 12 females) and 24 normotensive rats (12 males and 12 females). The animals were divided into two groups and sacrificed 7 and 21 days after the surgical procedure. After the routine laboratory processing, histological and histometric analysis were carried out and data were submitted to ANOVA and Tukey's test (5%).Results: Males and females from the same group had similar histological characteristics. After seven days, all animals presented irregular bone trabeculae. The periosteal osteoblasts were flattened in SHR, and presented a cuboid shape in normotensive animals. After 21 days, the bone defects of all specimens showed a linear closure in all the superficial extension. In addition, SHR presented flattened osteoblasts surrounding the bone trabeculae, while normotensive ones showed cuboidal cells. Statistical analysis of the histometric data indicated similar means between the male and female groups, except for normotensive rats on day 7. In addition, a larger amount of new bone formation was observed in hypertensive when compared to normotensive rats on day 27, in males as well as females.Conclusion: We conclude that bone healing in SHR was more significant than in normotensive ones, as shown by the histological and histometric evaluation 21 days after surgery.

The objective of this experiment was to determine the normal values of Bone Radiographic Density (BRD) by using the optical densitometry in radiographic images and the Bone Breaking Strength (BBS) of broiler femurs at different ages (8, 22 and 42 d of age). A total of 60 Cobb male broilers were distributed in three age groups of 20 birds. The BRD and the BBS (maxim force and rigidity) values increased (p<0.01) over the course of ages, presenting greater values at 42 d of age when comparing to 8 and 22 d of age, evidencing a biomechanical adaptation of femur to growth. This experiment offers results that can be used in other experiments of broilers fed with different nutritional levels and they can also be related to pathological values, allowing the diagnosis of diseases that affect the integrity of the poultry leg. © Asian Network for Scientific Information, 2011.

Aim. To investigate the biomechanical effects of zoledronic acid (ZA) on femurs of female osteoporotic rats after follow-up periods of 9 and 12 months. Methods. Eighty female Wistar rats were prospectively assessed. At 60 days of age, the animals were randomly divided into two groups: bilateral oophorectomy (O) (n=40) and sham surgery (S) (n=40). At 90 days of age, groups O and S were randomly subdivided into four groups, according to whether 0.1 mg/kg of ZA or distilled water (DW) was intraperitoneally administered: OZA (n=20), ODW (n=20), SZA (n=20), and SDW (n=20). The animals were sacrificed at 9 and 12 months after the administration of the substances, and then their right femurs were removed and analyzed biomechanically. Axial compression tests that focused on determining the maximum load (N), yield point (N), and stiffness coefficient (N/mm) of the proximal femur were performed in the biomechanical study. Results. ZA significantly increased the maximum load and yield point, reducing the stiffness coefficient concerning the oophorectomy status and follow-up period. Conclusion. Zoledronic acid, at a dose of 0.1 mg/kg, significantly increased the maximum loads and yield points and reduced the stiffness coefficients in the femurs of female rats with osteoporosis caused by bilateral oophorectomy. © 2012 Evandro Pereira Palacio et al.

The aim of study was to investigate if different intake levels of concentrate (supplementation levels) would have an influence on chemical composition, biometric aspects and bone mineral density (BMD) of femurs from Moxoto growing goats in the Brazilian semiarid region. Thirty-two castrated kids (15.69 +/- 0.78 kg initial BW) were used allocated randomly to one of the four levels of supplementation (SL) (treatments groups: 0: 5; 10 and 15 g/kg BW). When the animals of treatment group 15 g/kg BW reached 25 kg BW, the animals in the others treatments groups were also slaughtered. The increase SL resulted in linear increase (P<0.001) in the performance of animals as well as in fresh and dry weight of the femur and decrease of relative weight of the femur. The dry matter contents (g/kg) of femur increased linearly, however, the amounts of MM, ether extract, Ca, P. Mg, Na and K were not affected by the treatments. The values found in this study ranged from 134.84 to 155.90g of Ca, 48.14 to 55.95g of P. 1.69 to 1.87g of Mg, 2.87 to 3.24g of Na and 1.42 to 1.61 g of K/kg of bone in natura for animals with smaller and higher slaughter weight, respectively. The biometric and densitometric variables increased linearly with increasing treatments, except for the thickness of the sponge layer of the proximal and distal epiphysis and BMD measured in the distal epiphysis. The supplementation with concentrate did not affect the chemical composition of the femur, although it has positively influenced the structure and the bone mineral density. A BMD estimated via radiographic bone aluminum equivalent did not have correspondence with bone mineral content measured chemically. (C) 2011 Elsevier BM. All rights reserved.

This study was carried out at Faculdade de Medicina Veterinária e Zootecnia, Universidade Estadual Paulista, Botucatu, SP, Brazil, and evaluated bone quality in broiler breeders. Twenty-three families of Ross broiler breeders were housed in 5.0-m² pens. The families were comprised of 13 females and one male at the onset of the experimental period. The mean number of females per family was 9.34 at the end of the trial. The feeding program and management followed strain guidelines (Agroceres Ross, 2003). Bone analyses were performed in the right tibia and femur using optical radiographic densitometry at 4, 8, 12, 15, 20, 24, 30, 35, 42, 47 and 52 weeks of rearing. Trap nests were used to collect eggs from the breeders two weeks before and after the evaluation weeks. At each evaluation day, five birds were sacrificed after radiographs were taken and the tibias and femurs were collected to perform the following analyses: fatfree dry matter, ash percentage, bone resistance and Seedor index. Therefore, it was possible to establish correlations between bone quality and eggshell quality. Characteristics of bone quality were highly correlated to each other; on the other hand, there were no correlations between bone quality and external egg quality. In conclusion, there was no effect of egg production on egg quality, possibly because there was no reabsorption of bone minerals.

An extensible internal device (EID) was developed to preserve growth plate during the treatment of fracture complications or segmental bone loss from tumour resection in children. Since this type of extensible, trans-physeal, internal fixation device has only been used in a few paediatric cases; the aim of this study was to evaluate an in vivo canine study, a surgical application of this device, and its interference with longitudinal growth of the non-fractured distal femur. Ton clinically healthy two- to three-month-old poodles weighing 1.5-2.3 kg were used. Following a medial approach to the right distal femur, one extremity of the EID, similar to a T-plate, was fixed in the femoral condyle with two cortical screws placed below the growth plate. The other extremity, consisting of an adaptable brim with two screw holes and a plate guide, was fixed in the third distal of the femoral diaphysis with two cortical screws. The EID was removed 180 days after application. All of the dogs demonstrated full weight-bearing after surgery. The values of thigh and stifle circumferences, and stifle joint motion range did not show any difference between operated and control hindlimbs. The plate slid in the device according to longitudinal bone growth, in all but one dog. In this dog, a 10.5% shortening of the femoral shaft was observed due to a lack of EID sliding. The other dogs had the some longitudinal lengths in both femurs. The EID permits longitudinal bone growth without blocking the distal femur growth plate if appropriately placed.

Objective: To determine the effects produced by the ingestion of a fermented soy product (soy yoghurt), supplemented with isoflavones and associated with a resistive exercise program, on the bone metabolism of mature ovariectomized (Ovx) and sham-ovariectomized (sham-Ovx) rats.Methods: A total of 56 rats were used. They were divided into 2 sedentary control groups, the Ovx control group (C-Ovx) and the sham-Ovx control group (C-Sovx), each with 7 sedentary animals, and 2 treated groups, Ovx and sham-Ovx, with 21 animals each. These two treated groups were subdivided into three subgroups of seven animals each, which received the following treatments: consuming the soy yoghurt + sedentary, only subjected to resistive exercise, and consuming the soy yoghurt + resistive exercise. Both the program of resistive exercise and the consumption of soy yoghurt (at 3 mL/(kg body weight day)) continued for 12 weeks. The soy yoghurt was supplemented with isoflavones at 50 mg/100 g of product. The animals were sacrificed and their right-side femurs and tibias removed and assessed for bone mineral density (BMD). The alkaline phosphatase activity (AP) was determined in the blood serum.Results: There was a significant increase in both femur and tibia BMD values and in serum alkaline phosphatase activity in all the treated subgroups, compared with the control groups (p < 0.05).Conclusion: the ingestion of the soy yoghurt supplemented with isoflavones was capable of preventing a loss of bone mass in Ovx rats and of increasing bone mass in sham rats, whilst the resistive exercise program was effective in augmenting the bone mass in sham and Ovx rats. We investigated the importance of daily free activity in the cage and body weight gain during the recovering of bone structural and mechanical properties in growing rats after hindlimb unloading. Eight-week-old male Wistar rats were randomly divided into control (CG, n=24) and suspended (SG, n=24) groups. Animals from SG underwent a four-week hindlimb unloading period by tail-suspension. Animals from CG and those from SG after release were kept in collective cages and sacrificed at the age of 12, 16 and 20 weeks. Both femurs were removed and its area, bone mineral density (BMD), resistance to failure and stiffness were determined. Four-week hindlimb unloading decreased (p < 0.05) body weight (CG, 373.00 +/- 9.47 vs. SG, 295.86 +/- 9.19 g), BMD (CG, 0.19 +/- 0.01 vs. SG, 0.15 +/- 0.01 g/cm(2)), bone resistance to failure (CG, 147.75 +/- 5.05 vs. SG, 96.40 +/- 5.95 N) and stiffness (CG, 0.38 +/- 0.01 vs. SG, 0.23 +/- 0.02 N/m). Eight weeks of free activity in cage recovered (p > 0.05) the body weight (CG, 472.75 +/- 14.11 vs. SG, 444.75 +/- 18.91 g), BMD (CG, 0.24 +/- 0.01 vs. SG, 0.22 +/- 0.01 g/cm(2)), bone resistance to failure (CG, 195.73 +/- 10.06 vs. SG, 178.45 +/- 8.48 N) and stiffness (CG, 0.56 +/- 0.02 vs. SG, 0.47 +/- 0.03 N/m) of SG animals. Body weight correlated strongly with bone structural and mechanical properties (p < 0.0001). In conclusion, free activity in the cage associated with body weight gain restored bone structural and mechanical properties in growing rats after hindlimb unloading.
