Candida albicans is present in the oral cavity and in the whole digestive tract of humans and other animals, being frequently related to endodontic treatment failure. The present study determined the incidence of C. albicans in the oral cavity and the susceptibility of isolates to different pH values and saturated calcium hydroxide aqueous solution at pH 12.5. Sixty-five patients attending the Endodontic Clinic at the Sagrado Coração University participated in the study. The collected samples were cultivated in selective media for C. albicans and the isolates were tested in terms of resistance to both alkaline pH and saturated aqueous solution of calcium hydroxide. In relation to time variables, yeast viability was assessed by the Sabouraud's agar culture and fluorescein diacetate and ethidium bromide fluorescent staining method. Results from the different pHs and experimental times, including those from different techniques measuring fungal viability, were compared using the chi-square and Fisher's exact tests (α=0.05). The yeasts became completely inviable after 48 h of contact with the calcium hydroxide solution. On the other hand, when exposed to the alkaline culture broth, the yeasts were found to be viable at pHs 9.5 and 10.5 for up to 7 days. In conclusion, C. albicans can only be completely inhibited by direct contact with saturated calcium hydroxide aqueous solution after 48 h of exposure.

The climates of the central and southern regions of São Paulo state in Brazil favor pathogens such as Puccinia psidii Winter, which causes a common and severe disease in Eucalyptus plantations under 2 years old. We studied genetic parameters including genotype by environment interaction (G × E) of resistance to P. psidii rust in Eucalyptus grandis at nine sites in São Paulo State. Open-pollinated progeny from ten 'provenances' were established in a randomized complete block design; at individual sites there were from 134 to 160 progenies, from four to eight blocks, and five to six trees per plot. Significant provenance and progeny(provenance) differences were detected, as was G × E involving progeny(provenance). However, the G × E involved little if any rank changes, indicating that selection can be done efficiently at a single site, if the disease level is sufficient. The estimated coefficient of genetic variation among the progeny within provenances CVg was high and variable among the sites (ranging from 11 % to 36. 7 %), demonstrating different expression of genetic variability among the sites. The estimated heritability at the individual-tree level h2 and within a plot hw 2 ranged from low to intermediate (ranging from 0. 04 to 0. 46) and was high at the progeny-mean level hf 2 (ranging from 0. 30 to 0. 86). Our study shows good prospects of controlling this disease by selection among and within progenies in a single site. © 2012 Springer-Verlag Berlin Heidelberg.

We compared different cultivars and hybrids of crucifers in relation to development and life-history of diamondback moth (Plutella xylostella) to classify the plants according to their resistance to the pest. The plants used were Manteiga da Geórgia kale, Bola de Neve cauliflower, Ramoso Piracicaba Precoce broccoli, Chato-de-quintal cabbage, and the hybrid cabbages Midori, TPC668, TPC308, and TPC681. We evaluated performance daily until the pupal stage. Pupae were assessed individually to determine the pupal weight, performance, and pupal period. We determined the sex ratio, fecundity, fertility, and longevity of the emerged adults and calculated their reproductive potential. Cabbage hybrids TPC668, TPC308, and TPC681 do not support the development and reproduction of the diamondback moth. These hybrids show a level of resistance that is similar to that found the commercially available hybrid Midori and cultivar Chato de Quintal, which are known to be resistant to the diamondback moth. This finding implies that the capitata (cabbage) cultivars are the most suitable for planting because they are more resistant to pest than the cultivar's moth, acephala (kale). © 2013 Copyright Taylor and Francis Group, LLC.

Some wild species of the genus Arachis have demonstrated potential for improvement of peanuts. This work was performed to evaluate the occurrence and symptoms of Enneothrips flavens and Stegasta bosquella and its effects on agronomic traits of wild Arachis accessions. Nine accessions of wild Arachis species and a commercial A. hypogaea variety were studied in a split plot statistical scheme with a completely randomized block design and four replications. The main plots consisted of plants sprayed or not sprayed for insect control, while the subplots comprised the peanut accessions. Accessions GKP10017 (A. cardenasii) and V7639 (A. kuhlmannii) showed the lowest percentages of leaflets with E. flavens and S. bosquella. Accessions V9912, V7639 and V8979 (all three A. kuhlmannii) and V13250 (A. kempff-mercadoi) showed the lowest responses to insecticide application among the various plant traits evaluated. These accessions are of interest for further studies to identify mechanisms of resistance, to be used in breeding programs for resistance to these insects.

This study analyzed the histopathology of rabbit skin, previously immunized with SGE2, SGE4, and SGE6 gland extracts prepared from salivary glands of Rhipicephalus sanguineus female with 2, 4, and 6 days of feeding, at the region of the R. sanguineus female feeding lesion 2, 4, and 6 days after tick attachment. In this work, infestation-naïve New Zealand White rabbits were inoculated either with the extracts (test group (TG)) or with phosphate buffer and complete Freund's adjuvant mixture (control group 2 (CG2)). Each extract-inoculated- (TG and CG2) and non-inoculated (CG1) rabbit was subsequently infested with R. sanguineus. Skin biopsies were collected from the rabbit at the tick feeding lesion at 2, 4, and 6 days of feeding. Results revealed that rabbit immunization with gland extracts induced acquisition of resistance against this species. It should be stated that the SGE4 extract was the most effective in developing an immune-inflammatory response against ectoparasites, being this process characterized by the presence of an early and intense inflammatory cell infiltrate. On the other hand, SGE6 extract caused a later appearance of resistance with less infiltrate occurrence and intense edema at the feeding lesion site. As to the inflammatory process deriving from SGE2 extract inoculation, it was the less intense. It was concluded that immunization with different extracts from R. sanguineus female salivary glands did not change microscope features of the inflammatory process, although an earlier or more intense and later response, which was also dependent on the inoculate extract, was noticed. © 2012 Springer-Verlag Berlin Heidelberg.

Rhipicephalus sanguineus, commonly known as the brown dog tick, is one of the most widely distributed species of tick. In dogs, it can cause anemia and provide the transmission of pathogenic microorganisms such as Babesia canis, Ehrlichia canis, Hepatozoon canis, Anaplasma platys, and Mycoplasma haemocanis. To man, it can transmit the intracellular parasites Rickettsia rickettsii and Rickettsia conorii, the causative agents of the Rocky Mountain spotted fever in the Americas and Mediterranean and spotted fever in Europe and North Africa. Its control is performed by applying synthetic formulations composed of pyrethroids; however, continued use of these products results in environmental damage and acquisition of resistance. Alternatively, studies with botanical insecticides have been increasingly recurrent. Therefore, this study aimed to test the efficacy of essential oil of Tagetes patula, a ruderal species widely described in the literature for its insecticidal properties, in engorged females of R. sanguineus by the adults immersion test (AIT) and impregnated paper disk test (IPDT). The essential oil used, through gas chromatography coupled to mass spectrometry, revealed the presence of 55 compounds, being the 4-vinyl guaiacol and gamma terpinene the majority ones. The AIT compared to the IPDT was more efficient in inhibiting oviposition of tick; however, the eggs laid by the females submitted to saturated atmosphere with essential oil, from IPDT, not hatched, interrupted their development cycle. Besides being a pioneer work, the results presented here contributes to new researches, aiming the incorporation of essential oil in an acaricide for use in the environment. © 2013 Springer-Verlag Berlin Heidelberg.

The silverleaf whitefly Bemisia tabaci (Genn.) biotype B (Hemiptera: Aleyrodidae) is an economically important pest of tomatoes Solanum lycopersicum (L.), causing irregular ripening on fruits and transmitting several plant pathogenic geminiviruses. The management of this pest is commonly based on repetitive spraying with synthetic pesticides, causing serious environmental damages and increase of resistance by insect population. In the present study, essential oils from the leaves of Artemisia camphorata Vill., Ageratum conyzoides L., Foeniculum vulgare Mill., Lippia alba (Mill.) N. E. Br., Plectranthus neochilus Schltr., and Tagetes erecta L. were investigated for their possible repellent and oviposition-deterrent effects against B. tabaci biotype B on tomato. In a multi-choice assay, P. neochilus essential oil was the most active repellent and oviposition deterrent. Essential oils of A. conyzoides and T. erecta significantly deterred the female B. tabaci biotype B from laying eggs on treated tomato leaflets compared with the control. (E)-Caryophyllene (30.67 %) and the monoterpenes α-pinene (15.02 %) and α-thujene (11.70 %) were identified as the major constituents of the essential oil of P. neochilus. Our findings demonstrated the potential of essential oil of P. neochilus and other oils in the reduction of settlement and oviposition of B. tabaci biotype B on tomato. © 2012 Springer-Verlag Berlin Heidelberg.

Session ratings of perceived exertion (SRPE) have been considered to provide a quantitative evaluation of the entire exercise session in different types of resistance training. In this study we investigated the ability of SRPE to assess exercise strain in a circuit weight training (CWT) workout and the influence of time lag to report SRPE. Ten healthy male volunteers (22.3±2.8 years, 72.5±6.5kg, and 175±5cm) completed a CWT session involving three circuits of five multiple joint exercises with single sets of 20 repetitions at 30% one repetition maximum (1-RM). Heart rate [63.7-75.0% maximum heart rate (%HRmax)], blood lactate (5.6-7.6mM) as well as overall, chest, and active muscle RPE increased significantly (p<0.05) throughout the CWT, but no significant differences were found between ratings of perceived exertion (RPE) types. Overall, chest and active muscle SRPE were accessed 10 minutes, 20 minutes, and 30 minutes after the workout, with no significant main effects or SRPE type×time interaction being found (p>0.05). Finally, no significant differences (p>0.05) were observed between averaged SRPE and RPE responses (overall: 3.7±0.6 vs. 3.5±0.9; chest: 3.8±0.7 vs. 3.6±0.8; active muscle; 3.7±0.7 vs. 3.5±0.7). These results suggest SRPE, irrespective of the moment at which it is taken, to be a useful tool for assessing global exercise strain in a CWT workout, providing coaches, physicians, and exercisers a practical way for monitoring this type of resistance training. © 2013.

Measures to control the cattle tick, Rhipicephalus (Boophilus) microplus, based only on chemical products are becoming unsustainable, mainly because of the development of resistance. The objective of this study was to test the effect of the aqueous extract of pineapple skin (AEPS) and bromelain extracted from the stem (Sigma-Aldrich®, B4882) on engorged females and larvae of R. (B.) microplus in vitro. These substances were diluted in water and evaluated at eight concentrations. Engorged females were collected and distributed in groups of 10, with three repetitions for each treatment. After immersion in the solutions, the females were placed in an incubator for observation of survival, oviposition and larval hatching. The larval packet method was used, also with three repetitions with about 100 larvae each. The packets were incubated and the readings were performed after 24h. The estimated reproduction and efficacy of the solutions were calculated. The LC50 and LC90 were estimated using the Probit procedure of the SAS program. The eight concentrations were compared within each treatment by the Tukey test. For the experiment with engorged females, the most effective concentrations were 125, 250 and 500mg/mL: 33%, 48% and 59% for the AEPS and 27%, 51% and 55% for the bromelain. The LC50 and LC90 values were, respectively, 276 and 8691mg/mL for AEPS and 373 and 5172mg/mL for bromelain. None of the dilutions tested was effective against the larvae of R. (B.) microplus. This is the first report of the action of pineapple extracts or their constituents on cattle ticks. The results demonstrate that further studies regarding composition of tick cuticle, with evaluation of other solvents and formulations, should be conducted seeking to enhance the effect of pineapple extracts and compounds against this ectoparasite. © 2013 Elsevier Inc.

The aim of the study was to verify whether 8 weeks of resistance training employing maximal isokinetic eccentric (IERT) knee extensor actions would reduce the acute force loss observed after high-intensity treadmill running exercise. It was hypothesized that specific IERT would induce protective effects against muscle fatigue and ultrastructural damages, preventing or reducing the loss in mechanical muscle function after running. Subjects were tested before and after IERT protocol for maximal isometric, concentric and eccentric isokinetic knee extensor strength (60 and 180 s-1). In a second session, subjects performed treadmill running (~35 min) and the previously mentioned measurements were repeated immediately after running. Subsequently, subjects were randomized to training (n = 12) consisting of 24 sessions of maximal IERT knee extensors actions at 180 s-1, or served as controls (n = 8). The effects of acute running-induced fatigue and training on isokinetic and isometric peak torque, and rate of force development (RFD) were investigated. Before IERT, running-induced eccentric torque loss at 180 s-1 was -8 %, and RFD loss was -11 %. Longitudinal IERT led to reduced or absent acute running-induced losses in maximal IERT torque at 180 s-1 (+2 %), being significantly reduced compared to before IERT (p < 0.05), however, RFD loss remained at -11 % (p > 0.05). In conclusion, IERT yields a reduced strength loss after high-intensity running workouts, which may suggest a protective effect against fatigue and/or morphological damages. However, IERT may not avoid reductions in explosive muscle actions. In turn, this may allow more intense training sessions to be performed, facilitating the adaptive response to running training. © 2013 Springer-Verlag Berlin Heidelberg.

The purpose of this study was to investigate the influence of exercise order on one-repetition maximum (1-RM) and ten-repetition maximum (10-RM) strength gains after 6 weeks of resistance training (RT) in trained men. Sixteen men were randomly assigned into two groups based on the order of exercises performed during training sessions: a group that performed large muscle group exercises first and progressed to small muscle group exercises (LG-SM); while a second group performed the opposite sequence and started with small muscle group exercises and progressed to large muscle group exercises (SM-LG). Four sessions of RT were conducted per week; all exercises were performed for three sets of 8-12 repetitions with 1-min rest intervals between sets. Maximal and submaximal strength were assessed at baseline and after 6 weeks of RT with 1-RM and 10-RM testing for the bench press (BP), lat pulldown (LPD), triceps pulley extension (TE) and biceps curl (BC), respectively. Two-way ANOVA for the 1-RM and 10-RM tests indicated a significant group x time interaction. The 1-RM values significantly increased for all exercises in both groups (P<0.05), but were not significantly different between groups. However, effect size (ES) data indicated that the LG-SM group exhibited a greater magnitude of gains (1-RM and 10-RM) for the BP and LPD exercises. Conversely, ES indicated that the SM-LG group exhibited a greater magnitude of gains (1-RM and 10-RM) for the TE and BC exercises. In conclusion, the results suggest that upper body movements should be prioritized and performed according to individual needs to maximize maximal and submaximal strength. © 2013 Scandinavian Society of Clinical Physiology and Nuclear Medicine.

The purpose of this study was to quantify energy expenditure (EE) during multiple sets of leg press (LP) and bench press (BP) exercises in 10 males with at least 1 yr of resistance training (RT). The subjects underwent two sessions to determine 1 repetition maximum (1RM) on the BP and LP and one protocol consisting of a warm up and 4 sets for 10 repetitions at 70% 1RM with a 3-min rest period between sets for each exercise. Energy expenditure was calculated as the sum of oxygen uptake (aerobic component), EPOC, and lactate production (anaerobic component). There were no significant differences in EE between exercises for sets 1 to 4 and the total energy expended. However, statistical analysis revealed a significant difference (P<0.05) between exercises in RT economy (BP, 0.0206 ± 0.0044 kcal·kg-1 vs. LP, 0.0051 ± 0.0015 kcal·kg-1). Within exercise comparison showed set 4 was significantly different from sets 1 and 3 for BP, and for LP a significant difference was found between set 4 and sets 1, 2 and 3. Our results point to an increase in EE during multiple sets at 70% 1RM and show that in spite of the difference in muscle mass involved and total work done during each type of exercise, EE was not different due to greater economy during the LP.

The development of resistance to anthelmintics has prompted research into alternative methods of controlling intestinal nematodes in ruminants. This study aimed to assess the activity of Ananas comosus on Haemonchus contortus in Santa Inês sheep. The aqueous extract of pineapple skin (AEPS), bromelain from pineapple stems (B4882) and residue from pineapple processing was evaluated in in vitro and in vivo tests. The enzymatic activity of substances was analyzed by the azocasein method. The egg hatch test (EHT) and larval development test (LDT) were performed using the Embrapa2010 isolate of H. contortus. In the in vivo test, 36 sheep artificially infected with H. contortus were divided into six groups: G1: 2g/kg BW of the aqueous extract administered for three days; G2: 2g/kg BW of the industrial pineapple residue for 60 days; G3: 180mg/animal of bromelain in a single dose; G4: negative control I; G5: positive control (levamisole phosphate); and G6: negative control II. The eggs per gram (EPG) in the feces were counted till 28 days after treatment. LC50 and LC90 were obtained by the probit procedure, while the in vivo test results were analyzed by GLM. The aqueous extract in the in vitro and in vivo test, the bromelain and industrial residue presented 0.102, 0.157, 1.864 and 0.048 enzyme units/mL, respectively. In the egg hatch test, the LC50 and LC90 were respectively 31 and 81mg/mL for the aqueous extract and 0.50 and 2mg/mL for bromelain. In the larval development test, the LC50 and LC90 were respectively 1.7 and 7.3mg/mL for the aqueous extract and 0.019 and 0.086mg/mL for bromelain. In the in vivo test, the general efficacies of the treatments in relation to the negative control were 22.6%, 42.2%, 3.65% and 89% for the aqueous extract, industrial pineapple residue, bromelain and positive control respectively. The transformed EPG values were 3.19±0.59, 3.32±0.25, 2.85±0.66, 3.44±0.50, 2.28±0.93 and 2.75±0.94 for the aqueous extract, industrial residue, bromelain, negative control I, positive control and negative control II respectively. The results for all the treated groups differed significantly (p<0.05) from the positive control, and although the residue presented efficacy of 42.2%, there was no statistical difference (p>0.05) in relation to the negative control. Therefore, both the aqueous extract and bromelain were effective in vitro, but showed reduced anthelmintic efficacy in vivo. For the pineapple residue, the 42.2% in vivo efficacy in reducing the EPG and the possibility of reducing environmental contamination through reuse of industrial residue indicate it can also be useful for control of this parasite. © 2013 Elsevier B.V.

Este trabalho procura identificar selecionadas produções poéticas da antologia 26 Poetas Hoje as faces da resistência indicada por Alfredo Bosi no ensaio “Poesia Resistência”. Do “grito de alarme” pessoal de poetas pertencentes a gerações diferentes, emerge uma poesia espontânea, autêntica e, sobretudo, resistente, como produto da renovação da expressão poética, impregnado de afetividades e de intimismo e que incorporou o coloquial e os recursos da grande poesia brasileira desde 22. This reading presents a sample from this anthology and tries to identify in it the faces of resistance indicated by Alfredo Bosi in his essay: “ Poesia Resistência”. In the several manners the poets of different generations chose for their personal cry of alert we find a poetry wich is spontaneous, authentic and, above all, “resistent”. This is due mainly to the renewal of the poetical expression, impregnated with affective ness and intimism, which included colloquial language and devices used by all great Brazilian poetry since the modernism of 1922.
