The purpose of this study was to evaluate methods for predicting availability of Zn in soils. Surface soil samples representing Podzolized soils from Lins and Marilia var. Marilia and var. Lins, medium texture Dark-Red Latosol, Dusky-Red Latosol and Terra Roxa Estruturada, were gathered from fields in Jaboticabal, SP, Brazil. Com was used as indicator plant in a greenhouse experiment. Availability of Zn was determined with: 0.5 M CaCl2, 2.0 N MgCl2, N CH3COONH4 (pH 6.0 and 4.8), 0.05 and 0.1 N HCl, 0.05 N H2SO4, 0.005 M DTPA (pH 7.3 and 6.0), 1.0% Na2-EDTA, 0.01 M EDTA + 1.0 M (NH4)2CO3 (pH 8.6) and 0.02 M EDTA + 0.5 M CH3COOH + 0.5 M CH3COONH4. By using the simple correlation coefficients (Zn extracted from soils x absorbed by plants it can be concluded that the complexant agents and acid solutions showed to be efficient. The salt solutions showed low extraction capacity, low correlation coefficients and less accuracy. By considering aspects as accuracy and easy management DTPA pH 7,3 and Na2-EDTA can be indicated as the most apropriates (r values, respectively, 0.873** and 0.868** and V.C. 7.52% and 7.98%).

A label-free electrochemical detection method for DNA hybridization based on electrostatic modulation of the ion-exchange kinetics of a polypyrrole film deposited at microelectrodes is reported. Synthetic single-stranded 27-mer oligonucleotides (probe) have been immobilized at 2,5-bis(2-thienyl)-N-(3-phosphorylpropyl)pyrrole film formed by electropolymerization on the previously formed polypyrrole layer. The 27- or 18-mer target oligonucleotides were monitored via the electrochemically driven anion exchange of the inner polypyrrole film. The performance of the miniaturized DNA biosensor system was studied in respect to selectivity, sensitivity, reproducibility, and regeneration of the sensor. Control experiments were performed with a noncomplementary target of 27-mer DNA and 12 base-pair mismatched 18-mer sequences, respectively, and did not show any unspecific binding. Under optimized experimental conditions, the label-free electrochemical biosensor enabled the detection limits of 0.16 and 3.5 fmol for the 18- and 2 7-mer DNA strand, respectively. Furthermore, we demonstrate reusability of the electrochemical DNA biosensor after successful recovery of up to 100% of the original signal by regenerating the DNA label-free electrode with 50 mM HCl at room temperature.

Tin(II) complexes with 8-hydroxyquinolinate in solid state have been obtained by adding aqueous ammonium to a solution containing stannous chloride and 8-hydroxiquinoline in medium of HCl and acetone up to pH 5 and 9, respectively. The products obtained show the same composition, Sn(C9H6ON)(2); however there are some differences regarding both the thermal behaviour in an oxidant atmosphere and morphology. These products were characterised by elemental and complexometric analysis, TG and DTA curves, infrared and X-ray diffractometry. TG curves show, above 448 K, the partial oxidation on air atmosphere of Sn(II) complexes to Sn(IV) complexes, SnO(C9H6ON)(2). This behaviour does not depend only on pH in which the compounds were obtained but also on the heating rate in TG curves. Sn(II) complexes volatilise almost completely on nitrogen atmosphere and partially on air atmosphere depending on the oxidation degree of the compound.

A simple, rapid inexpensive voltammetric method have been developed for the quantitative determination of albendazole (ABZ) as the pure assay, by direct dissolution of commercial tablets in HCl solutions. Studies with linear sweep (LSV), square-wave (SWV) and differential pulse voltammetry (DPV) were carried out ABZ in aqueous medium at a glassy carbon electrode. A well defined irreversible oxidation peak current was obtained at 1,00V vs. SCE. The method permits a precise quantitative determination of ABZ using the standard addition method. The detection limits for the three voltammetric techniques were found to be 3.0 x 10(-5) M (LSV), 6.2 x 10(-5) M (SWV) and 4.0 x 10(-5) M (DPV).

Blends possessing the elastomeric properties of natural rubber (NR) and the conducting properties of conducting polymer (polyaniline, PANI) were obtained, which are promising for further application in deformation sensors. Blends containing 20% (v/v) of PANI in 80% of NR latex were fabricated by casting in the form of free-standing films and treated either with HCl or with corona discharge, which lead PANI to its conducting state (doping process). Characterization was carried out by Raman spectroscopy, d.c. conductivity and thermogravimetric analysis. Evidence for chemical interaction between PANI and NR was observed, which allowed the conclusion that the NR latex itself is able partially to induce both the primary doping of PANI (by protonation) and the secondary doping of PANI (by changing the chain conformation). Further improvement in the primary doping could be obtained for the blends either by corona discharge or by exposing them to HCl the electrical conductivity reached in the blends was dependent on the doping conditions used, as observed by Raman scattering. Tendon composition changes according to differentiation, mechanical load, and aging. In this study, we attempted to identify, localize, and quantify type VI collagen in bovine tendons. Type VI collagen was identified by the electrophoretic behavior of the alpha chains and Western blotting, and by rotary shadowing. Type VI collagen was extracted from powdered tendon with three sequential 24-h extractions with 4 M guanidine-HCl. The amount of type VI collagen was determined by enzyme-linked immunosorbent assay for purely tensional areas and for the compressive fibrocartilage regions of the deep flexor tendon of the digits, for the corresponding fetal and calf tendons, and for the extensor digital tendon. The distal fibrocartilaginous region of the adult tendon was richer in type VI collagen than the tensional area, reaching as much as 3.3 mg/g (0.33%) of the wet weight. Calf tendons showed an accumulation of type VI at the fibrocartilage site. Immunocytochemistry demonstrated that type VI collagen was evenly distributed in the tensional areas of tendons but was highly concentrated around the fibrochondrocytes in the fibrocartilages. The results demonstrate that tendons are variable with regard to the presence and distribution of type VI collagen. The early accumulation of type VI collagen in the region of calf tendon that will become fibrocartilage in the adult suggests that it is a good marker of fibrocartilage differentiation. Furthermore, the distribution of type VI collagen in tendon fibrocartilage indicates that it organizes the pericellular environment and may represent a survival factor for these cells.

This trial was carried out under laboratory conditions, with three lots of Iguacu soybean seeds, to determine the respiration rate by the titulation method. A randomized complete block design was used, with four repetitions, each one of them with 50 seeds and a control without seeds for each lot. The seeds were placed in gerbox with 40 mi of KOH 0.1N, on distilled water wet blotting paper for fixing the CO2 produced by seeds respiration. The material was placed in germinator at constant 25 degrees C for 16, 24 and 48 hours. After these periods, the titulation of the fixative solution was performed with HCl 0.1N to check the respiration rate. The most deteriorated seeds were those that presented the highest respiration rates, which were measured in mg of CO2/g of dry matter, when compared with the most vigorous lots. The titulation method was suitable for the evaluation of soybean seed respiration rate because the final results separated the lots in three different quality levels that were compatible with the results obtained from the accelerated aging and emergence.

The kinetics of ultrasound-stimulated and HCl-catalyzed hydrolysis of solventless TEOS-water mixtures was studied as a function of temperature ranging from 10 degrees C up to 65 degrees C by means of flux calorimetry measurements. A specially designed device was utilized for this purpose. The exothermic peak arising few minutes after sonication began has been attributed mainly to the hydrolysis reaction. The overall hydrolysis process, which was measured through the irradiation time up to the hydrolysis peak, was found to be thermally activated, with an apparent activation energy Delta E = 36.4 kJ/mol. The alcohol produced at the early hydrolysis due to sonication seems to further enhance the reaction, via a parallel autocatalytic path, which is controlled by a faster pseudo second order rate constant (k'). Our modeling yielded k' = 6.3 x 10(-2) M(-1) min(-1) at 20 degrees C, which is in a reasonable agreement with the literature, and an activation energy Delta E = 40.4 kJ/mol for the specific process of hydrolysis in presence of alcohol.

The main goal of this study was to identify and analyse environmental problems related to human occupation at the south part of the city of Paranagua PR, Brazil, aiming the diagnosis and conceptual evaluation of landscape use. For landscape use assessments and degradation diagnosis purposes, the environment was classified considering mainly geological and geomorphologic characteristics, such as declivity and sediment origin. The identification of potential environmental impacts was made by aerial photographs, Landsat-TM images, photographic records, surface sediment samples, water-column samples, and mainly observation visits to each classified unit. Surface sediment samples were taken with a Petit-Ponar sampler, in 13 station located in Paranagua Bay. In order to characterize the contamination level on sediments, the concentration of the following elements were analysed: arsenic (As), copper (Cu), cadmium (Cd), nickel (Ni), lead (Ph) e zinc (Zn). For the extraction of these elements, samples were treated with HCl 0.1N for 12 hours. This method has the advantage of determining contaminants which are weakly bind to the sediment, thus more available to the system. Analysis were performed by atomic absorption spectroscopy. According to the adopted criteria, sectors were classified as hills, mangroves, continental sediments and beach ridges urban and port industrial. Water quality and concentrations of metallic elements were determined at three sectors one control and two with the influence of urban and port/industrial activities, respectively the major upland impacts found were: deforestation due to disordered occupation, and solid waste emission through domestic and industrial sewage. The diagnosis of the actual situation was used as a basis for a management plan, once it allows the identification of impacts, and consequent environmental irregularities. Based on this considerations, suggestions for land use and management were presented, which offers a better approach for recuperation and protection of the studied ecosystem.

Siloxane-polyoxypropylene (PPO) hybrids obtained by the sol-gel process and containing short polymer chain have been doped with different sodium salts NaX (X = ClO4, BF4 or I). The effect of the counter-ion (X) on the chemical environment of the sodium ions and on the ionic conductivity of these hybrids was investigated by Na-23 NMR, small angle X-ray scattering (SAXS), complex impedance, Raman spectroscopy and differential scanning calorimetry (DSC). Results reveal that the different sodium salts have essentially the same effect on the nanoscopic structure of the hybrids. The formation of immobile Na+ cations involved in NaCl-like species could be minimized by using a low amount of HCl as hydrolytic catalyst. The differences in the ionic conductivity of hybrids doped with different sodium salts were correlated with the proportion of Na ions solvated by ether-type oxygen of the polymeric chains and by the carboxyl oxygen located in the urea groups of the PPO chain extremities. Labial salivary glands are found in the majority of insects. They are relatively large, extend back into the thorax, and in Rhodnius, they are cherry red in color due to a pigment derived from traces of hemoglobin absorbed form the gut. In most insects they are acinous shaped, with long excretion channels that present differentiated regions which from salivary reservoirs. The glands may be relatively simple or complexly branched and convoluted. In Rhodnius they are described as being unilobed with no traces of division. The main duct leaves the gland at its anterior extremity. The acini have different kinds of cells but all of them are seen as sources of secretion. Our material has a different shape due to the fact that the animals spent 20 days under starvation conditions. New data are also obtained through treatment with collagenase and HCl. The importance of the study of these glands lies in the fact that it will further understanding of the transmission of Chagas' disease.

The nor-clerodane diterpene trans-crotonin isolated from the bark of Croton cajucara BENTH. was investigated for its ability to prevent the formation of gastric-mucosa ulceration in different experimental models in mice. The results obtained from crotonin were compared with those obtained with another diterpene, DHC (trans-dehydrocrotonin) in the same models. When previously administered (p.o.) at the dose of 100 mg/kg, crotonin, as well as DHC, significantly reduced (p<0.05) gastric injury induced by stress (72, 67%), indomethacin/bethanechol (78, 29%) and pylorus ligature (35, 30%). In the HCl/ethanol-induced gastric ulcer model, at oral doses of 100 and 250 mg/kg, crotonin significantly prevented (p<0.05) the formation of gastric lesions by 51 and 56%, respectively, when compared to the control group. Gastric injury was also of significantly less magnitude in the DHC treatment group (p<0.05). In the pylorus-ligature model, crotonin (p.o.), like cimetidine, increased the volume of gastric juice when compared to the control group (p<0.05). No significant modifications where found in gastric parameters such as pH or total acid content after oral crotonin treatment. However, systemic alterations were observed when crotonin (100 mg/kg) was previously administered intraduodenally to mice. We observed significant changes (p<0.001) in gastric-juice parameters such as an increase in volume and a decrease in gastric acidity. Those pre-treated with crotonin as well as with DHC did not increase free mucus production (p>0.05). The results suggest that crotonin presents a significant anti-ulcer effect when assessed in these ulcer-induced models. As with DHC, the antiulcerogenic effects of crotonin are probably related to anti-secretory or/and gastroprotective properties of this substance. In light of results obtained with DHC and natural trans-crotonin in the present study, we concluded that the A-ring of both diterpenes is not directly involved in the antiulcerogenic activity.

The brown alga Pilayella littoralis was used as a new biosorbent in an on-line metal preconcentration procedure in a flow-injection system. Al, Co, Cu and Fe were determined in lake water samples by inductively coupled plasma optical emission spectrometry (ICP-OES) after preconcentration in a silica-immobilized alga column. Like other algae, P. littoralis exhibited strong affinity for these metals proving to be an effective accumulation medium. Metals were bound at pH 5.5 and were displaced at pH < 2 with diluted HCl. The enrichment factors for Cu-II, Fe-III, Al-III and Co-II were 13, 7, 16 and 11, respectively. Metal sorption efficiency ranged from 86 to 90%. The method accuracy was assessed by using drinking water certified reference material and graphite furnace atomic absorption spectrometry (GFAAS) as a comparison technique. The column procedure allowed a less time consuming, easy regeneration of the biomaterial and rigidity of the alga provided by its immobilization on silica gel. (C) 2003 Elsevier B.V. About similar to 2.1 x 10(-3) Mol SiO2 cm(-3) and similar to 88%-volume liquid-phase silica wet gels were prepared from oxalic-acid-catalyzed tetraethoxysilane (TEOS) sonohydrolysis. Aerogels were obtained by supercritical CO2 extraction. The samples were analyzed by thermogravimetry, small-angle X-ray scattering and nitrogen adsorption. Wet gels can be described as mass fractal structures with fractal dimension D similar to 1.94 and structural characteristic length zeta changing between similar to 3.3 to similar to 3.0 nm in the studied range of the catalyst concentration. A fraction of the porosity is apparently eliminated in the supercritical process. The values of the BET specific surface S-BET, the total pore volume V-p and the mean pore size l(p) of the aerogels were found to change almost randomly around the mean values S-BET = 874 m(2) g(-1), V-p = 0.961 cm(3) g(-1) and l(p) = 4.4 nm with catalyst concentration variation. In this work we investigate the effect of hydrochloric acid (HC) addition on the structure and thermal and magnetic properties of iron-doped siloxane-polyoxyethylene (POE) hybrids prepared by the sol-gel route. X-ray powder diffraction (XRD) and X-ray absorption near edge structure (XANES) results reveal the dominance of ferrihydrite nanoparticles and a mixture of this phase with FeCl4- species in the hybrid prepared without and with HCl, respectively. Thermal analysis reveals the existence of two crystalline polymeric phases in the hybrid prepared with HCl whereas hybrids prepared without HCl are amorphous. The 105 and 60 Angstrom sized ferrihydrite nanoparticles were detected by SAXS analysis of the composite prepared without and with HCl, respectively. The magnetic results suggest that in both samples antiferromagnetic nanoparticles coexist with small clusters/isolated ions. In the sample without HCl addition, larger particles dominate the magnetic behavior, while the opposite occurs for the sample prepared using HCl catalyst. (C) 2004 Elsevier B.V. All rights reserved.
