A purified glycoprotein of 43 000 daltons from Paracoccidioides brasiliensis (gp43) was tested as paracoccidioidin in delayed-type hypersensitivity (DTH) tests in both experimental animals (guinea pig and mice) and patients with paracoccidioidomycosis (PCM). The gp43 paracoccidioidin was compared with the traditional Fava Netto antigen (AgFN). In guinea pigs, the intradermal injection of 2 g of gp43 showed a similar response to those obtained with AgFN, showing in histological sections a population of lymphoid cells that participate in DTH. In mice, gp43 at a dose of 3.75g showed positive DTH response. The use of gp43 as paracoccidioidin in humans showed that this molecule can be used to evaluate the DTH response in patients with PCM. Of 25 PCM patients studied, 48% were positive to gp43 while only 28% were positive to AgFN; 12 PCM patients were completely anergic to both antigens. Considering only those 13 PCM patients who were responsive to gp43 and/or to AgFN, 92.3% reacted against gp43 and 53.8% reacted against AgFN (P < 0.05). Gp43 skin test responses (13.67 9.56 mm) were significantly larger than those obtained with AgFN (8.43 3.69 mm). Immunohistochemical study of the human skin showed a perivascular inflammatory response constituted predominantly by T lymphocytes, macrophages and polymorphonuclear leukocytes. 1996 ISHAM.

The purpose of the study was to evaluate the biocompatibility of two current adhesive resins and a calcium hydroxide cement. Fifty-four polyethylene tubes were filled with these dental materials, which were hand-mixed or light-cured according to the manufacturer's directions: group 1-Clearfill Liner Bond 2 (Kuraray); group 2-Single Bond (3M); and group 3-calcium hydroxide cement (Dycal-Dentsply). The materials were implanted into dorsal connective tissue of rats, which were killed 7, 30, and 60 days after the implantation procedure. The implant sites were excised, immersed in buffered Karnovsky's fixative, and processed using routine histological techniques. Sections of 6 m thickness were stained with hematoxylin and eosin and assessed under light microscopy. Both adhesive resins at 7 days elicited a moderate/intense inflammatory reaction that decreased over time. Fibrous capsules surrounding the tubes were observed at 30 days. Half of the samples in groups 1 and 2 showed thin fibrous capsule formation containing macrophages, capillaries, lymphocytes, fibroblasts, and collagen fibers. Connective tissue healing was observed even though many specimens exhibited a persistent inflammatory reaction mediated by macrophages and giant cells at the 60-day evaluation. Dycal allowed complete healing at 30 days with only a thin fibrous capsule. In conclusion, all experimental materials were successfully walled off by the connective tissue of the rat. However the adhesive resins may release particulates that may, in turn, induce a persistent local inflammatory reaction. Consequently, in this specific condition, these materials cannot be regarded as biocompatible. Dycal was less irritating than the adhesive resins and was better tolerated by the connective tissue. Although canine visceral leishmaniasis (CVL) has been extensively studied, muscular damage due to Leishmania (Leishmania) infantum chagasi infection remains to be fully established. The aim of this study was to describe the electromyographic and histological changes, as well as search for the presence of amastigote forms of Leishmania spp, CD3+ T-lymphocytes, macrophages and IgG in skeletal muscles of dogs with visceral leishmaniasis (VL). Four muscles (triceps brachial, extensor carpi radialis, biceps femoris and gastrocnemius) from a total of 17 naturally infected and six healthy dogs were used in this study. Electromyographic alterations such as fibrillation potentials, positive sharp waves and complex repetitive discharges were observed in, at least, three muscles from all infected dogs. Myocyte necrosis and degeneration were the most frequent muscular injury seen, followed by inflammatory reaction, fibrosis and variation in muscle fibers size. Immunohistochemistry in muscle samples revealed amastigote forms in 4/17 (23. 53%), IgG in 12/17 (70. 58%), CD3+ T-lymphocytes in 16/17 (94. 12%) and macrophages in 17/17 (100%) dogs. Statistically positive correlation was observed between: inflammatory infiltrate (p=0. 0305) and CD3+ immunoreaction (p=0. 0307) in relation to the number of amastigote forms; inflammatory infiltrate (p=0. 0101) and macrophage immunoreaction (p=0. 0127) in relation to the amount of CD3+; and inflammatory infiltrate (p=0. 0044) and degeneration/necrosis (p<0. 0001) in relation to the presence of macrophages. Our results suggest that different mechanisms contribute to the development of myocytotoxicity, including celular and humoral immune responses and direct muscular injury by the parasite. Nevertheless, the catabolic nature of the disease can probably interact with other factors, but cannot be incriminated as the only responsible for myositis.

The objective of this study was to evaluate the effectiveness of formalin and copper sulphate to control monogeneans in juvenile Hemigrammus sp and its effect on leukocyte count. We used a completely randomized design with five treatments and three replications. All treatments lasted for three days and consisted of: T1, control; T2 and T3 immersion bath for one hour in formalin, an of two concentrations of 0.25 ml formalin and the other 0.1 ml L-1; the T4 bath for 24 hours with 0,025 ml of formalin-L-1; and T5, bath 24 hours with copper sulphate (0.3 mg L-1). After the experimental period was observed that T2, 100% mortality. The T3 and T5 provided 100% efficiency, however, fish presented mortalities and intoxication of the 66 and 80% respectively. T4 showed an efficacy of 77.7% and decrease in parasitological indices: total number of parasites of11, 3 9, mean infection intensity of 3.2 2 and a prevalence of 47.1%. As the leukocytes was observed that at T3, T4 and T5 occurred differences in the proportion of lymphocytes, monocytes and neutrophils as compared to control. The CuSO4 and formalin reduce the parasite load, but presented to be toxic at higher concentrations.

Canine Distemper is a contagious, severe and multisystemic disease caused by a virus from Morbillivrus genus. The virus is distributed worldwide and it presents a high lethality rate, affecting mainly dogs. The diagnosis is based on clinical signs associated with hematological fi ndings. The observation of Lentz bodies in erythrocytes and leukocytes is the defi nitive diagnosis for the disease. The aim of this work was to test the hypothesis that the hematological profi le in dogs positive for canine distemper differs according to blood cell type presenting Lentz bodies. For this purpose, 25 dogs positive for the disease were evaluated at the Veterinary Hospital Lus Quintiliano de Oliveira UNESP, Araatuba city. The diagnosis was based on the observation of Lentz bodies in blood smears. Fromthe total, 64% of dogs presented anemia, 16% leucopenia and 12% leukocytosis. Lymphopenia occurred in 76% of dogs. Viral inclusions were observed solely in neutrophils (32%), lymphocytes (28%) and erythrocytes (12%). Concomitant observation occurred in lymphocytes and erythrocytes (4 %), in lymphocytes and neutrophils (12%), in neutrophils and monocytes (4%) and in neutrophils and erythrocytes (4%). In an isolated case Lentz bodies were observed simultaneously in neuthrophils, monocytes and lymphocytes. In conclusion, hematological profi le is not associated with the presence of viral inclusion in a particular cell type.

The present study aimed to estimate the number of CD8(+) T and natural killer (NK) infiltrating cells and the expression of interleukin-10 (IL-10) and transforming growth factor beta 1 (TGF-beta1) in chemically induced neoplasms in an initiation-promotion bioassay for carcinogenesis. Male Wistar rats were treated with N-nitrosodiethylamine, N-methyl-N-nitrosourea, N-butyl-N-(4-hydroxybutyl) nitrosamine, dihydroxy-di-N-propylnitrosamine, and 1,2-dimethylhydrazine for 4 weeks. Two groups were subsequently exposed through diet to phenobarbital (0.05%) or 2-acetylaminofluorene (0.01%) for 25 weeks. An untreated group was used as a control. Immune cells and cytokines were immunohistochemically evaluated in neoplasms and in surrounding normal tissues at the liver, kidneys, lung, and small and large intestines. When compared to the respective normal tissues, an increased number of NK cells was verified infiltrating the colon, lung, and kidney neoplasms, while the number of CD8+ T cells decreased in the intestine and lung neoplasms. Expression of IL-10 was found mainly in kidney tumors. TGF-beta1 was expressed mainly in the liver and kidneys tumors. The results indicate that the differential occurrence of immune cells between neoplastic and normal tissues could be dependent upon tumor microenvironment.
