The largest chromosome in the river buffalo karyotype, BBU1, is a submetacentric chromosome with reported homology between BBU1q and bovine chromosome 1 and between BBU1p and BTA27. We present the first radiation hybrid map of this chromosome containing 69 cattle derived markers including 48 coding genes, 17 microsatellites and four ESTs distributed in two linkage groups spanning a total length of 1330.1 cR(5000). The RH map was constructed based on analysis of a recently developed river buffalo-hamster whole genome radiation hybrid (BBURH5000) panel. The retention frequency of individual markers across the panel ranged from 17.8 to 52.2%. With few exceptions, the order of markers within linkage groups is identical to the order established for corresponding cattle RH maps. The BBU1 map provides a starting point for comparison of gene order rearrangements between river buffalo chromosome 1 and its bovine homologs. Copyright (C) 2007 S. Karger AG, Basel.

Durante o processo de maturação, as sementes passam por modificações físicas, bioquímicas e fisiológicas até atingir o ponto ideal de colheita, quando apresentam a capacidade máxima de germinação e vigor. O objetivo deste trabalho foi identificar o estágio de colheita e o substrato mais favorável à germinação das sementes e ao crescimento de plântulas de Tabebuia chrysotricha. Os frutos de T. chrysotricha foram colhidos em 16 árvores- matriz na Fazenda Lageado, em Botucatu, SP, em quatro estágios de colheita: fruto fechado menor (ME), fruto fechado maior (MA), fruto em início de abertura (IA) e fruto aberto e em início de dispersão das sementes (D). Para caracterizar os estágios de colheita, foi utilizada a coloração dos frutos e das sementes, mensurados a espessura, a largura e o comprimento dos frutos e determinado o teor de água das sementes. O efeito do estágio de colheita sobre a qualidade das sementes foi verificado mediante o teste de germinação sobre areia e sobre papel a 25 ºC, avaliado após 21 dias da semeadura; e o vigor, pelo teste da primeira contagem (7 dias) e do comprimento de plântulas (21 dias). Os resultados indicaram que as sementes de Tabebuia chrysotricha devem ser colhidas quando o fruto estiver em início de abertura. Este estágio de colheita também pode ser identificado pelo teor de água das sementes de 59,6%. O substrato areia mostrou-se mais favorável ao teste de germinação e ao crescimento das plântulas.

Six cattle that had earlier exposure to Dermatobia hominis were infested experimentally with first-instar larvae of the parasite. Skin biopsies taken at intervals were studied in wax and in plastic sections. The avidin-biotin-peroxidase method was used to detect the presence and localization of host immunoglobulins (Igs) G and M and antigens of first and second instar larvae of Dermatobia hominis. The larvae penetrated actively through the skin and migrated towards the subcutaneous tissues. The great numbers of eosinophils suggest that they are the most important cell in mediating damage to D.hominis larvae. The immunoglobulins bound only to dead or moulting larvae in which access to binding sites may have been altered. This could represent a morphological manifestation of a mechanism that protects larvae from the host immune response. Large amounts of soluble antigens detected along the fistulous tract may be important in the maintenance of this tract by disturbing the normal cicatrization process.

Syagrus schizophylla (Mart.) Glass. belongs to the Arecaccae family. This palm is native of Brazil and presents great potential for use in gardens and cultivation in pot. Palms species, with relatively few exceptions, can only be propagated from seeds; even so, there are no reports in the literature about the germination of this palm seeds. The seed maturity is a factor that interferes in the success of the germination process. For some species, studies showed that palm seeds germinated better when the fruits were completely ripe (showing full color) and for other, when they were with green coloration. Several species of the Arecaccae family presents physical dormancy of seeds in varied degrees, demanding treatments for improve germination. The objective of this work was to study the effects of maturation and of the scarification on seed germination of S. schizophylla. The experimental design used was a factorial 3 x 2 (3 maturation stadiums: green, half-ripe - yellow and completely ripe - red; and mechanical scarification: with and without), entirely casualized, with four replications of 15 seeds per plot. The seeds (with 32,43% of humidity) were placed in plastic boxes with sand (60% of humidity, placing water according to weight in each three days), under controlled conditions of alternated temperature of 25-35 degrees C, photoperiod of 12 hours. The percentage of germination and the speed germination index (SGI) were evaluated until 79 days. It was observed that, so much for germination percentage as for SGI, there was not significant difference for the interaction among the two factors, maturation stadium and scarification, however, there were significant differences among the maturation stadiums and between seeds scarified or not. The seeds from green fruits presented lower germination percentage and slower germination when compared with seeds from yellow or red fruits; the seeds from yellow or red fruits didn't differ statistically to each other. To seeds scarificated, independently of the maturation stadium, presented germination percentage significantly larger and the germination was significantly faster when compared with the seeds without scarification.

Synaptonemal complex (SC) analysis of XY pairing in the goat (Capra hircus; 2n = 60) was investigated by electron microscopy for the first time in this species. Synapsis of the X and Y chromosomes begins during the mid-late zygotene stage as the autosomes complete their pairing. Only a small portion of the total length of the Y is paired with the X chromosome at this time. By the early pachytene, almost 90% of the Y is paired with the X. All the observed stages of the sex bivalent pairing showed the structural difference between the differential and pairing regions. In the pairing region, a synaptonemal complex is formed, while in the differential region the chromosome axes remain free.

This study evaluated the influence of the cementation length of glass fiber-reinforced composite (FRC) on the fatigue resistance of bovine teeth restored with an adhesively cemented FRC. Thirty roots of single-rooted bovine teeth were allocated to 3 groups (n = 10), according to the ratio of crown length/root length (post cementation length): group 1 = 2/3, group 2 = 1/2, and group 3 = 1/1. The roots were prepared, the fiber posts (FRC Postec Plus) were cemented, and the specimens were submitted to 2 million mechanical cycles. After fatigue testing, a score was given based on the number of fatigue cycles until fracture, and data were submitted to statistical analysis. All specimens were resistant to fatigue. Taking into account the methodology and results of this study, the evaluated fiber posts can be cemented based on the ratio of crown/root at 1/1. Further clinical studies must be conducted to verify this ratio.

The 3-isobutyl-1-methylxanthine (IBMX) is able to prevent resumption of meiosis by maintaining elevated cyclic AMP (cAMP) concentrations in the oocyte, and roscovitine, a purine known to specifically inhibit MPF kinase activity, maintains bovine oocytes at the germinal vesicle (GV) stage. The present study was conducted to analyze whether cytoplasmic maturation (examined by the pattern of cortical granule (CG) distribution) of bovine oocytes is improved during meiotic arrest with IBMX and roscovitine. Oocytes were matured in vitro in a 10% Knockout(SR) supplemented TCM-199 medium (Control) with either 0.5 mM IBMX or 25 mu M roscovitine (ROSC). Oocytes were stained with fluorescein isothiocyanate conjugated Lens culinaris agglutinin (FITC-LCA) for CG evaluation and with Hoechst 33342 for nuclear stage assessment. At 16 h of culture, the percentage of oocytes remaining in the GV stage was higher (P < 0.05) in the ROSC group (32.41%) compared with the Control and IBMX groups (8.61% and 9.73%, respectively). At 24h of culture, progression of meiosis to M II stage was retarded (P < 0.05) in the ROSC group (24.05%) compared to the Control (60.20%), whereas the IBMX group (33.88%) showed no significant difference to the other two groups. At 16h of maturation, the proportion of oocytes with CG in clusters (immature cytoplasm) was similar between the groups, as was the percentage of peripheral CG (mature) at 24h of maturation. The results of the present study demonstrated that the meiotic inhibitors IBMX and roscovitine delay the progression of nuclear maturation without affecting cytoplasmic maturation, assessed by the analysis of CG repositioning. Aim To report the diagnosis and successful clinical management of a patient with anatomical variation in maxillary first and second premolars.Summary Maxillary premolars have a highly variable root canal morphology, but it is rare, especially in the second premolar, to find three canals. This article describes the diagnosis and clinical management of first and second premolars with three canals and three separate roots, drawing particular attention to radiographic interpretation and access refinements.

The objective of this research was to study the basic aspects of bullfrog reproduction in Anfigranja system from June 1989 to July 1990. For the 1493 animals involved in this experiment (657 males and 836 females) biometrical data were obtained (weight, length and tympanum diameter) and developmental stages of gonads were determined. Female sizes at first maturation were 10,33 cm and 109,78 g and for males it was around 8,09 cm; all males above 45 g were already in the maturation process. The relationship between tympanum diameter and body length (RTC) was considered a good index for male maturation.

T. micrantha (L.) Blume (Ulmaceae), a common pioneer tree species in Brazil, is used in the restoration of degraded areas. The fruits are fleshy and indehiscent, with only one water impermeable seed. During the fruiting period, fruits of different colours are found at the same time on the same branch. This research aimed to correlate fruit colour with other physical indicators of seed maturity and to Verify the effect of temperature regime on seed germination. Collected fruits were separated in to green, green-red and red colour and for each of these maturation stages, size, moisture content and dry matter of both fruits and seeds were determined. Seeds were scarified with sulphuric acid and submitted to a germination test conducted at constant (20 degreesC, 30 degreesC and 40 degreesC) and alternating (20-30 degreesC, 30-40 degreesC and 20-40 degreesC) temperatures for 15 weeks. Seed germination percentage and speed were analysed after five weeks and the fmal percentage of germinated and Viable seeds after 15 weeks. Seed maturity is attained when the fruits are green-red. At this stage, moisture content was about 64% for fruits and 10% for seeds. Alternating temperature was required for seed germination and 20-30 degreesC was the best option. Most seeds had germinated after five weeks, providing mature seeds, acid scarification and alternating temperature were used.

Diurea cross-linked bridged silsesquioxanes (BSs) C(10)C(11)C(10) derived from organosilane precursors, including decylene chains as side spacers and alkylene chains with variable length as central spacers (EtO)(3)Si- (CH(2))(10)-Y(CH(2))(n)-Y-(CH(2))(10)-Si(OEt)(3) (n = 7, 9-12; Y = urea group and Et = ethyl), have been synthesized through the combination of self-directed assembly and an acid-catalyzed sol gel route involving the addition of dimethylsulfoxide (DMSO) and a large excess of water. This new family of hybrids has enabled us to conclude that the length of the side spacers plays a unique role in the structuring of alkylene-based BSs, although their morphology remains unaffected. All the samples adopt a lamellar structure. While the alkylene chains are totally disordered in the case of the C(10)C(7)C(10) sample, a variable proportion of all-trans and gauche conformers exists in the materials with longer central spacers. The highest degree of structuring occurs for n = 9. The inclusion of decylene instead of propylene chains as side spacers leads to the formation of a stronger hydrogen-bonded urea-urea array as evidenced by two dimensional correlation Fourier transform infrared spectroscopic analysis. The emission spectra and emission quantum yields of the C(10)C(n)C(10) Cm materials are similar to those reported for diurea cross-linked alkylene-based BSs incorporating propylene chains as side spacers and prepared under different experimental conditions. The emission of the C(10)C(n)C(10) hybrids is ascribed to the overlap of two distinct components that occur within the urea cross-linkages and within the siliceous nanodomains. Time-resolved photoluminescence spectroscopy has provided evidence that the average distance between the siliceous domains and the urea cross-links is similar in the C(10)C(n)C(10) BSs and in oxyethylene-based hybrid analogues incorporating propylene chains as side spacers (diureasils), an indication that the longer side chains in the former materials adopt gauche conformations. It has also allowed us to demonstrate for the first time that the emission features of the urea-related component of the emission of alkylene-based BSs depend critically on the length of the side spacers.

The following sequence of substitution reactions was studied spectrophotometrically in organic solvents: RNH2 + TCNQ →-HCN 7-substituted TCNQ →-HCN +RNH2 7.7-disubstituted TCNQ where R = butyl, octyl, dodecyl, and hexadecyl. The production of 7-(alkylamino)-7,7,8-tricyanoquinodimethanes proceeds via the formation of the anion radical of TCNQ (TCNQ-·) whose rate of appearance was found to be a function of the chain length of R, reaching a maximum for octylamine. The formation of TCNQ-· was sensitive to the solvent polarity and electron-donor power and was associated with a small enthalpy and a highly negative entropy of activation. Above a certain [C8H17NH2] the rate of disappearance of TCNQ-· was independent of the amine concentration, and the reaction had a much higher enthalpy and entropy of activation. The occurrence of tautomerism precluded an investigation of the conversion of 7-(octylamino)-7,8,8-tricyanoquinodimethane into 7,7-bis(octylamino)-8,8-dicyanoquinodimethane. A study of the reaction of octylamine with 7-morpholino-7,8,8-tricyanoquinodimethane (which does not exist in tautomeric forms) showed that the second substitution step proceeds with the same mechanism as the first one. The only difference between the two compounds (TCNQ and its monosubstituted morpholino derivative) is one of reactivity. © 1985 American Chemical Society.
