No Brasil, 90% de pacientes com insuficiência renal crônica ou aguda dependem dos procedimentos de hemodiálise para remover produtos de degradação metabólica, excesso de água e de sais minerais do organismo, a fim de restaurar o equilíbrio ácido-base e eletrolítico. Entretanto, o processo de osmose reversa, que visa remover todos os íons da água, também remove o cloro, que exerce efeito bacteriostático sob diversas bactérias autóctones, que passam a se multiplicar na água. Entre os principais grupos dessas bactérias, estão os bastonetes Gram negativos não fermentadores (BGN) e os mais frequentemente associados a bacteremias, em pacientes de hemodiálise são Pseudomonas aeruginosa, Stenotrophomonas maltophilia e Burkholderia cepacia. Essas bactérias podem produzir biofilmes, o que torna sua eliminação do encanamento quase impossível. Assim, o objetivo do trabalho foi a análise de água e do dialisato, da Unidade de Hemodiálise de um Hospital Universitário, na pesquisa dos três BGNs citados. Também foram utilizadas 42 cepas previamente isoladas do mesmo local. Entre as 67 amostras de água, foram isoladas 8 cepas de Pseudomonas aeruginosa e as 50 cepas foram submetidas à pesquisa do gene rpoS, um dos responsáveis pela produção de biofilme. Esse gene foi observado em 20 (40%) cepas, sendo 16 (80%) P. aeruginosa, seguida por 3 (15%) cepas de S. maltophilia e 1 (5%) de B. cepacia. Essas cepas foram submetidas à produção de biofilme a 35ºC (temperatura ótima de crescimento) e a 20ºC (temperatura média da água no encanamento da Unidade de Hemodiálise), em aço inoxidável (material de muitos instrumentos cirúrgicos), PVC (matéria prima da tubulação) e plástico de poliestireno (capacidade de produção de biofilmes em diferentes materiais). Aim: The role of saliva on Candida adhesion to biomaterials has not been clearly defined. The present study investigates whether different periods of preconditioning with saliva would influence the adhesion of Candida albicans to a denture base resin. Methods: Ninety samples of acrylic resin with smooth surfaces were made and then divided into five groups: one control without saliva, and four experimental groups conditioned in saliva for periods of 30 min, 1, 3, or 12 h. Candida adhesion was evaluated by crystal violet staining and 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-([phenylamino] carbonyl)-2H-tetrazolium-hydroxide assay. Results: The one-way analysis of variance revealed that there were no significant differences among the mean number of adherent cells or among the mean absorbance for all groups. No significant correlation was found between the two methods used for assessing Candida albicans adhesion. Conclusion: The different periods of preconditioning with saliva had no significant influence on the adhesion of Candida albicans to the denture base acrylic resin.

Composite resins are materials that can present color changing when exposed to pigments. Objective: The aim of this study was to evaluate, in vitro, the color changing of composites after immersion in different substances for different periods. Material and methods: Two microhybrid composite resins: Charisma (Heraeus – Kulzer) and Opallis (FGM) were used. Red wine and acai pulp were also used as immersion medium. For this study, 32 specimens with 10 mm of diameter and 2 mm of thickness were used, divided into 4 groups: Group 1 – Opallis composite immersed in red wine solution; Group 2 – Opallis composite immersed in acai berry pulp solution; Group 3 – Charisma composite immersed in red wine solution; Group 4 – Charisma composite immersed in acai berry pulp solution. The specimens were evaluated in the following time periods: T0 – baseline, T1 – 24 hours, T2 – 48 hours, T3 – 72 hours and T4 – 96 hours. For the assessment of staining, a spectrophotometer for colorimetry was used (Color Guide 45 / 0, PCB 6807 BYK-Gardner Gerestsried GmBH, Germany), and the values obtained were transferred to a computer and recorded according to CIELAB system. Results: The data were evaluated using Kruskal- Wallis non-parametric tests with the following mean values for the immersion periods of 24, 48, 72 and 96 hours, respectively: G1 – 7.35, 7.84, 9.04,10.48; G2 – 2.92, 4.15, 4.30, 4.64; G3 – 3.14, 7.35, 8.13, 8.43, G4 – 4.49, 5.99, 6.92, 6.76. Conclusion: Red wine showed a higher tendency toward altering the composite color than acai berry pulp. In addition, no significant difference was found concerning to the behavior of the two composite resins. Concerning to the immersion time periods, significant differences were only observed among the groups in the 24 hour time period.

The authors have studied the effect of four staining agents on the optical properties of esthetic restorative materials through translucency tests. Two commercial brands of composite resins were used: Point 4 (Kerr) and Charisma (Heraeus-Kulzer). The liquids tested were: wine, cola, chlorhexidine solution and nicotine solution. The translucency was measured at different periods of time: P0 – before immersion (baseline), P1 - 1 h after immersion, P2 - 2 h after immersion and successively, P3 – 24 h after immersion up to a period of 7 weeks. ANOVA statistical analysis was applied to the data (p < 0.05). The results lead to the following conclusions: (1) Composite resins submitted to the tested immersion mediums were stained, (2) the lower percentage of translucency was observed for nicotine containing solution, (3) The percentage of translucency decreased with the period of immersion.

Elderly patients generally use several types of medication, some of which may cause oral side effects. Aim: To investigate the oral side effects caused by medication in an elderly sample. Methods: Three hundred patients were interviewed about their use of medication and were divided in two groups: institutionalized (n=150) and community-dwelling (n=150) elderly. Results: The most used drugs were antihypertensives (53%) for community-dwelling elders and antiulceratives (76%) for the institutionalized ones. The more prevalent side effects were taste alterations that occurred in 19%, dry mouth in 17% and teeth staining in 2%. Conclusions: A high prevalence of oral side effects from medications used by the elderly was found in this study. The health professionals should be aware of the possible side effects caused by prescribed medications.

Background: Solobacterium moorei is a volatile sulfide compound (VSC)-producing Gram-positive anaerobic bacterium that has been associated with halitosis. The aim of this study was to investigate the effects of green tea extract and its major constituent epigallocatechin-3-gallate (EGCG) on growth and severalhalitosis-related properties of S. moorei.Methods: A microplate dilution assay was used to determine the antibacterial activity of green tea extract and EGCG against S. moorei. Their effects on bacterial cell membrane integrity were investigated by transmission electron microscopy and a fluorescence-based permeability assay. Biofilm formation was quantified by crystal violet staining. Adhesion of FITC-labeled S. moorei to oral epithelial cells was monitored by fluorometry. The modulation of beta-galactosidase gene expression in S. moorei was evaluated by quantitative RT-PCR.Results: The green tea extract as well as EGCG inhibited the growth of S. moorei, with MIC values of 500 and 250 mu g/ml, respectively. Transmission electron microscopy analysis and a permeabilization assay brought evidence that the bacterial cell membrane was the target of green tea polyphenols. Regarding the effects of green tea polyphenols on the S. moorei colonization properties, it was found that biofilm formation on EGCG-treated surfaces was significantly affected, and that green tea extract and EGCG can cause the eradication of pre-formed S. moorei biofilms. Moreover, both the green tea extract and EGCG were found to reduce the adherence of S. moorei to oral epithelial cells. The beta-galactosidase activity of S. moorei, which plays a key role in VSC production, was dose-dependently inhibited by green tea polyphenols. In addition, EGCG at 1/2 MIC significantly decreased the beta-galactosidase gene expression.Conclusion: Our study brought evidence to support that green tea polyphenols possess a number of properties that may contribute to reduce S. moorei-related halitosis. Therefore, these natural compounds may be of interest to be used to supplement oral healthcare products.

Bacterial quorum sensing (QS) is a density dependent communication system that regulates the expression of certain genes including production of virulence factors in many pathogens. Bioactive plant extract/compounds inhibiting QS regulated gene expression may be a potential candidate as antipathogenic drug. In this study anti-QS activity of peppermint (Menthe piperita) oil was first tested using the Chromobacterium violaceum CVO26 biosensor. Further, the findings of the present investigation revealed that peppermint oil (PMO) at sub-Minimum Inhibitory Concentrations (sub-MICs) strongly interfered with acyl homoserine lactone (AHL) regulated virulence factors and biofilm formation in Pseudomonas aeruginosa and Aeromonas hydrophila. The result of molecular docking analysis attributed the QS inhibitory activity exhibited by PMO to menthol. Assessment of ability of menthol to interfere with QS systems of various Gram-negative pathogens comprising diverse AHL molecules revealed that it reduced the AHL dependent production of violacein, virulence factors, and biofilm formation indicating broad-spectrum anti-QS activity. Using two Escherichia colt biosensors, MG4/pKDT17 and pEAL08-2, we also confirmed that menthol inhibited both the las and pqs QS systems. Further, findings of the in vivo studies with menthol on nematode model Caenorhabditis elegans showed significantly enhanced survival of the nematode. Our data identified menthol as a novel broad spectrum QS inhibitor.

Color stability of restorative materials is essential for longevity of esthetic composite restoration over time. The aim of this investigation was assess the effect of prior water immersion on the color stability of a composite resin to red wine staining. Seventy disccshaped specimens (6mm x 1.5mm) were carried out and randomized in 7 groups (n = 10), according to distilled water immersion time at 0 (control), 24, 48, 72,120,192, and 240 h. Baseline color was measured according to the ciel*a*b* system using a reflection spectrophotometer(uvc2450, shimadzu).After that, the specimens were storage in red wine for 7 days. Color difference (∆e) after aging was calculated based on the color coordinates before(baseline) and after storage period.Data were subjected to onecway anova(alpha=0.05).The different times of immersion in.Water before to the red wine storage showed similar behavior on the color stability, without statistical difference compared to control group, immersed directly in the wine(p=0.7057).The previous water uptake of composite resin evaluated did not decrease the susceptibility to red wine staining.

The purpose of this study was to evaluate, in vitro, the chromatic behavior of esthetic brackets stored in potentially staining solutions. The sample were divided into four groups according to the commercial brand and stored in four different solutions (distilled water, cola soda, coffee and mouthrinse) at 37°C for 14 days. Possible color changes measured according to the CIE L*a*b* color system with a spectrophotometer at five intervals of time after storage. The statistical analysis was carried out using ANOVA to 1%, Tukey's tests and decomposition of interactions with a significance level of 5%.The color changes were dependent on the solution, storage time and the brand of brackets. The largest color changes were observed in the G3, followed by G2, G1/G4. The esthetic brackets do not present satisfactory and stable chromatic behavior.
