P>Reasons for performing study:Carbonic anhydrase (CA) catalyses the hydration/dehydration reaction of CO(2) and increases the rate of Cl- and HCO(3)- exchange between the erythrocytes and plasma. Therefore, chronic inhibition of CA has a potential to attenuate CO(2) output and induce greater metabolic and respiratory acidosis in exercising horses.Objectives:To determine the effects of Carbonic anhydrase inhibition on CO(2) output and ionic exchange between erythrocytes and plasma and their influence on acid-base balance in the pulmonary circulation (across the lung) in exercising horses with and without CA inhibition.Methods:Six horses were exercised to exhaustion on a treadmill without (Con) and with CA inhibition (AczTr). CA inhibition was achieved with administration of acetazolamide (10 mg/kg bwt t.i.d. for 3 days and 30 mg/kg bwt before exercise). Arterial, mixed venous blood and CO(2) output were sampled at rest and during exercise. An integrated physicochemical systems approach was used to describe acid base changes.Results:AczTr decreased the duration of exercise by 45% (P < 0.0001). During the transition from rest to exercise CO(2) output was lower in AczTr (P < 0.0001). Arterial PCO(2) (P < 0.0001; mean +/- s.e. 71 +/- 2 mmHg AczTr, 46 +/- 2 mmHg Con) was higher, whereas hydrogen ion (P = 0.01; 12.8 +/- 0.6 nEq/l AczTr, 15.5 +/- 0.6 nEq/l Con) and bicarbonate (P = 0.007; 5.5 +/- 0.7 mEq/l AczTr, 10.1 +/- 1.3 mEq/l Con) differences across the lung were lower in AczTr compared to Con. No difference was observed in weak electrolytes across the lung. Strong ion difference across the lung was lower in AczTr (P = 0.0003; 4.9 +/- 0.8 mEq AczTr, 7.5 +/- 1.2 mEq Con), which was affected by strong ion changes across the lung with exception of lactate.Conclusions:CO(2) and chloride changes in erythrocytes across the lung seem to be the major contributors to acid-base and ions balance in pulmonary circulation in exercising horses.

Background: Leishmaniasis is one of the most important vector-borne diseases of humans. This parasitic disease can be caused by many species of Leishmania. In humans, different species of the parasite are associated with different forms of the disease, cutaneous and visceral. Among domesticated animals, dogs are the most important species in the epidemiology of this disease. Leishmania chagasi, an important zoonosis, is well established as the agent of visceral leishmaniasis in Brazil. The disease is endemic in north, northeast, midwest and southeast, and is transmitted to mammals by hematophagous insects such as the Lutzomyia longipalpis. In 2008, our research group has diagnosed a case of canine leishmaniasis in the municipality of Uruguaiana and subsequently there were several cases in the city and the neighbor municipality of Sao Borja. Most Brazilian states are endemic for leishmaniasis, with the exception of Rio Grande do Sul. In southern Brazil, the reports of humans and dogs infected by Leishmania spp. are the source of endemic area in the country. Therefore, the aim of this study is register the first clinical case of canine visceral leishmaniasis in the municipality of Santa Maria, RS.Case: In october 2010, a veterinary clinic of Santa Maria received a canine, female, Doberman, with two years of age. The animal had severe skin lesions on the head and limbs, pale mucous membranes, and enlarged lymph nodes. According to the owner, the animal showed progressive weight loss and anorexia for more than five days. During the clinical examination the blood was collected for hemogram and cytology of lymph nodes was performed by puncture aspiration with a fine needle. In the erythrogram, it was observed a decrease in the total number of erythrocytes (2.8 x 10(6)/mu L), hematocrit (21%), hemoglobin (6.8 g/dL) and platelets (98 x 10(3)/mu L). In the leucogram, any alteration was observed. The cytology of lymph nodes showed amastigotes forms, suggestive of the Leishmania spp. Based on this finding; we performed the blood collection for PCR, to confirm parasitism and to determine the species of Leishmania. At the molecular test was used PCR-specific for L. chagasi, and the result was positive.Discussion: This is the first autochthonous clinical case in the central region of the RS, non-endemic area for leishmaniasis. In serological studies of visceral leishmaniasis it was diagnosed in five asymptomatic dogs in the municipalities of Santa Maria, Julio de Castilhos and Itaara, however not confirmed by molecular analysis. In the municipalities of Cruz Alta and Uruguaiana cases of L. chagasi have been reported in dogs which previously resided in Leishmania sp. endemic areas. The municipality of Sao Borja had the first record of L. longipalpis in the RS during the leishmaniasis outbreak in 2008-2009. In the central region of the RS vector has not been found, but because in this first autochthonous case dog in Santa Maria believe that the parasite is present and/or doing other insect transmission of leishmaniasis. Clinical signs associated with hematologic and coagulation disorders observed in the canine are commonly described in symptomatic dogs in endemic regions. This case of autochthonous leishmaniasis reinforces the idea of the vector presence in Santa Maria, center of the RS. We believe that canine leishmaniasis is an emerging disease in the southern region of Brazil.

Os efeitos de diferentes níveis de vitamina C e ferro no desempenho produtivo e parâmetros fisiológicos da tilápia do Nilo (Oreochromis niloticus) foram avaliados por um período de 73 dias. O delineamento foi inteiramente casualizado, com tratamentos dispostos em esquema fatorial com três níveis de vitamina C (125; 375 e 1115 mg/kg) e três níveis de ferro (30, 90 e 270 mg/kg), mais um tratamento adicional (0 mg/kg de suplementação de vitamina C e ferro), com quatro repetições cada tratamento, totalizando 10 grupos experimentais. Utilizaram-se 240 alevinos revertidos com peso médio inicial de 7,46 ± 0,49 g, distribuídos aleatoriamente em 40 aquários de 250 L, numa lotação de 6 peixes/aquário. Confeccionou-se dieta purificada com 32,0% de proteína bruta e 3300 kcal/kg de energia digestível. A ausência de vitamina C e ferro nas dietas propiciou o aparecimento de anemia microcítica e hipocrômica aos alevinos. A presença dessa vitamina em dosagens elevadas estimulou a liberação de eritrócitos imaturos na corrente sangüínea. Determinou-se, também, que níveis desses acima das exigências nutricionais descritas para a espécie não determinaram efeito detrimental no desempenho produtivo, na produção de proteínas plasmáticas ou nos aspectos morfológicos do fígado.

O experimento foi realizado com objetivo de avaliar os efeitos do estresse calórico e da suplementação de cloreto de potássio (KCl) sobre o desempenho e algumas características fisiológicas de frangos de corte. Quarenta aves, no período de 42 a 49 dias, foram submetidas ao estresse calórico (16 horas a 25 ± 1ºC; duas horas com temperatura crescente; quatro horas a 35 ± 1ºC; e duas horas com temperatura decrescente até a termoneutralidade e com umidade relativa de 63,5 ± 5%), recebendo os seguintes tratamentos: 0,50 e 1,00% KCl na ração; 0,25 e 0,50% KCl na água de bebida. A ração à base de milho e farelo de soja, com 20% de proteína bruta e 3200 kcal EM/kg, foi fornecida ad libitum. A suplementação de KCl na ração ou na água de beber não influiu no ganho de peso, nos consumos de ração e água, na conversão alimentar, na mortalidade, na relação água/ração, no teor de matéria seca das excretas e nas características hematológicas. O estresse calórico aumentou a temperatura retal, o hematócrito, a hemoglobina, o heterófilo e a relação heterófilo/linfócito e reduziu as hemácias, o linfócito, o sódio e o potássio sérico. A suplementação de KCl na água regulou o número de eritrócitos e a hemoglobina em frangos estressados pelo calor. A relação heterófilo:linfócito e a temperatura retal podem ser usadas como índices de estresse.

A EPO é um fator de crescimento glicoprotéico sintetizado pelas células adjacentes aos túbulos proximais renais regulada via mecanismo de feed back envolvendo a tensão de oxigênio tissular. Na baixa tensão de oxigênio arterial, a produção de EPO aumenta causando uma maior produção de eritrócitos na medula óssea. Devido ao pouco conhecimento da concentração de EPO sérica em equinos e a ausência de trabalhos sobre o efeito da idade e sexo sobre a sua concentração o trabalho teve como objetivo comparar a concentração sérica de eritropoietina em equinos da raça Árabe de sexos e idades diferentes. Foram utilizados 31 equinos da raça Árabe, com idades de seis a 12 meses (jovens) e acima de 24 meses (adultos), sendo 13 machos (seis jovens e sete adultos) e 18 fêmeas (oito jovens e 10 adultas), clinicamente sadios. As amostras de sangue foram colhidas por venipunção jugular e o soro armazenado até o momento do processamento. A concentração sérica de eritropoetina foi determinada pelo método de radioimunoensaio (RIA) utilizando kit comercial (EPO Trac TM125I RIA, Diagnostic Systems Laboratories, Webster, Texas, USA). Para análise estatística dos dados utilizou-se o Teste t de Student ao nível de 5% de significância (P<0,05). Não foram observadas diferenças significativas (P<0,05) quando se compararam os animais divididos entre sexos e as idades de seis a 12 meses (jovens) e acima de 24 meses (adultos). Conclui-se que a concentração de eritropoietina em equinos da raça Árabe independe do sexo e idade, e pode ser utilizada como valores de referência, porém ressalta-se a necessidade da obtenção de valores de referência para cada laboratório.

The midgut of adult female Anopheles darlingi is comprised of narrow anterior and dilated posterior regions, with a single layered epithelium composed by cuboidal digestive cells. Densely packed apical microvilli and an intricate basal labyrinth characterize each cell pole. Before blood feeding, apical cytoplasm contains numerous round granules and whorled profiles of rough endoplasmic reticulum. Engorgement causes a great distension of midgut. This provokes the flattening of digestive cells and their nuclei. Simultaneously, apical granules disappear, the whorls of endoplasmic reticulum disassemble and 3 h post bloodmeal (PBM), nucleoli enlarge manyfold. An intense absorptive process takes place during the first 24h PBM, with the formation of large glycogen inclusions, which persist after the end of the digestive process. Endoproteases activities are induced after bloodmeal and attain their maximum values between 10 and 36 h PBM. At least two different aminopeptidases seem to participate in the digestive process, with their maximum activity values at 36 and 48 h PBM, respectively. Coarse electrondense aggregates, possibly debris from digested erythrocytes, begin to appear on the luminal face of the peritrophic membrane from 18 h PBM and persist during all the digestive process, and are excreted at its end. We suggest that these aggregates could contain some kind of insoluble form of haem, in order of neutralize its toxicity. (c) 2005 Elsevier Ltd. All rights reserved.

In this study, micronucleus and nuclear alterations tests were performed on erythrocytes of Oreochromis niloticus (Perciformes, Cichlidae) in order to evaluate the water quality of the Atibaia river, in an area that receives effluents discharge of a petroleum refinery and also to evaluate the effectiveness of the treatments used by the refinery. Water samples were collected in five different sites related with a refinery from São Paulo State, Brazil. For the micronucleus and nuclear alterations tests, O. niloticus specimens were exposed for 72 h to the water samples and in pure ground water (negative control). The results herein obtained indicated that the treatments used by the refinery diminished the cytogenetic damage; however they were not fully effective, since the final mill has induced damages in the genetic material of the test organism.
