Uterine leiomyomas are extremely common, benign, smooth muscle tumors that represent a significant public health problem. Although there have been few molecular studies of uterine leiomyomas, most of them have reported a very low frequency of loss of heterozygosity (LOH) in different regions of the genome. The detection of LOH has been used to identify genomic regions that harbor tumor suppressor genes and to characterize different tumor types. We have used a set of 15 microsatellite polymorphism markers to examine the frequency of allele loss in a panel of 64 human uterine leiomyomas matched to normal DNAs. The markers were chosen from regions involved in losses identified by comparative genomic hybridization in a subset of uterine leiomyomas described in a previous report. DNA from tumors and normal tissue was amplified by the polymerase chain reaction and subsequently analyzed using an ABI Prism 377 DNA automated sequencer. The frequency of LOH observed was low, except for the markers D15S87 (15q26.3), D7S493 (7p15.3), and D7S517 (7p22.2). No changes in microsatellite size were detected in our samples. These results provide useful clues for identifying putative tumor suppressor genes associated with a subset of uterine leiomyomas. (C) 2004 Wiley-Liss, Inc.

Objective: To report a rare case of self-inflicted eye injury secondary to obsessive-compulsive disorder (OCD).Method: Case report.Results: A 41 year-old patient had significant bilateral visual loss and blunt trauma through touching his ocular cavities due to recurrent doubts regarding their shape and urges to check the format of bones and cartilages. Differential diagnosis with tic disorders is discussed, and the patient's treatment approach is described.Conclusion: Although rare, severe clinical complications can occur in OCD. Aims: To determine the occurrence of isolated and recurrent episodes of conductive hearing loss (CHL) during the first two years of life in very low birth weight (VLBW) infants with and without bronchopulmonary dysplasia (BPD).Study design, subjects and outcome measures: In a longitudinal clinical study. 187 children were evaluated at 6, 9, 12,15 18 and 24 months of age by visual reinforcement audiometry, tympanometry and auditory brain response system.Results: of the children with BPD, 54.5% presented with episodes of CHL, as opposed to 34.7% of the children without BPD. This difference was found to be statistically significant. The recurrent or persistent episodes were more frequent among children with BPD (25.7%) than among those without BPD (8.3%). The independent variables that contributed to this finding were small for gestational age and a 5 min Apgar score.Conclusions: Recurrent CHL episodes are more frequent among VLBW infants with BPD than among VLBW infants without BPD. The tissue microarray (TMA) technique allows multiple tissue samples in a single block. Commercial adhesive tape is used to avoid the loss of tissue samples during the immunostaining process. Few reports exist in the literature comparing the use of these adhesive tapes to other adhesive techniques. The objective of this study was to compare loss of sections adhered to slides using commercial adhesive tapes versus using silanized only slides. TMA was constructed with varying tissues using a fixed-base device (Beecher Instruments), placing 108 cylinders of 1 mm diameter in duplicate, spaced 1.2 mm apart. Section of 4 mu m were cut from the TMA block and adhered to 30 silanized slides and 30 commercial glass slides using adhesive tape, according to manufacturer's recommendations. Vimentin immunoexpression was evaluated by immunohistochemistry. Antigenic recovery was realized in citrate buffer using a microwave oven. Cylinder loss in the immunohistochemical process was quantified and expressed as: total (>80%), almost complete (75-79%), or partial (50-74%). The commercial adhesive tape group presented lesser total loss (1.1 versus 6.4%), almost complete loss (2.2 versus 3.5%), and partial loss (2.1 versus 3.8%) than the silanized slide group (ANOVA, P < 0.05). The sum of total and almost complete losses in the silanized slide group was 9.9%, greater than the losses in slides using commercial adhesive tapes (3.3%) and less than reported and considered acceptable in the literature (10-30%). In conclusion, the use of silanized only slides presents very satisfactory results, requires less training, and reduces costs significantly, thus justifying their use in research.

Heat stress has negative effects on pregnancy rates of lactating dairy cattle. There are genetic differences in tolerance to heat stress; Bos taurus indicus (B. t. indicus) cattle and embryos are more thermotolerant than Bos taurus taurus (B. t. taurus). In the present study, the effects of sire and sire breed on conception and embryonic/fetal loss rates of lactating Holstein cows during the Brazilian summer were determined. In Experiment 1, cows (n = 302) were AI after estrus detection or at a fixed-time with semen from one Gyr (B. t. indicus) or one Holstein sire (B. t. taurus). Pregnancy was diagnosed 80 days after AI. In Experiment 2, cows (n = 811) were AI with semen from three Gyr and two Holstein sires. Pregnancy was diagnosed at 30-40 and at 60-80 days after AI. Cows diagnosed pregnant at the first examination but non-pregnant at the second were considered as having lost their embryo or fetus. Data were analyzed by logistic regression. The model considered the effect of sire within breed, sire breed, days postpartum, period of lactation, and AI type (AI after estrus versus fixed-time). There was no effect of the AI type, days postpartum or milk production on conception or embryonic loss rates. The use of Gyr bulls increased pregnancy rate when compared to Holstein bulls [9.1% (60/657) versus 5.0% (23/456), respectively, P = 0.008; data from Experiments 1 and 2 combined]. Additionally, in Experiment 2, cows inseminated using semen from sire #4 (Gyr) had lower embryonic loss (10%) when compared with other B. t. indicus (35.3% and 40%) or B. t. taurus sires (18.2% and 38.5%, P = 0.03). In conclusion, the use of B. t. indicus sires may result in higher conception rates in lactating Holstein cows during summer heat stress. Moreover, sire can affect embryonic loss and selection of bulls according to this criterion may result in higher parturition rates in lactating Holstein cows. (c) 2006 Elsevier B.V. O trabalho foi realizado objetivando-se avaliar o efeito da densidade populacional e da linhagem sobre as características de desempenho, o rendimento de carcaça e a qualidade da carne de peito (perda de peso por cozimento, força de cizalhamento e pH) em frangos de corte. Foram utilizados 2.925 pintos de um dia, sexados, distribuídos em um delineamento experimental inteiramente casualizado, em fatorial 3x3x2, sendo três linhagens (Ross 308, Cobb 500 e Hybro PG), três densidades (10, 13 e 16 aves/m²) e dois sexos, com duas repetições com número variável de aves em função da densidade. A densidade afetou o ganho de peso nas fases inicial e final e no período total, sendo que a densidade de 10 aves/m² apresentou o melhor ganho de peso, porém, as densidades de 13 e 16 aves/m² não diferiram entre si. O aumento da densidade promoveu maior produção de peso vivo/m² de galpão, além de incrementar a renda bruta. No entanto, não afetou o rendimento de carcaça e das partes, bem como as características de qualidade da carne. As linhagens diferiram para ganho de peso em todas as fases e para o consumo de ração na fase final e no período total de criação, porém não diferiram para o rendimento de carcaça, apesar de diferirem para o rendimento de peito, pernas, asas, dorso e gordura abdominal, sendo que para o rendimento de pernas houve diferenças apenas entre os machos e para gordura abdominal apenas entre as fêmeas. As linhagens diferiram para perda de peso da carne de peito por cozimento, mas não para maciez e pH. O sexo das aves influenciou as características de desempenho e rendimento de carcaça, mas não a qualidade da carne do peito.

O trabalho foi conduzido com o objetivo de avaliar o efeito de diferentes linhagens (conformação versus convencionais) sobre o desempenho, rendimento de carcaça e partes, medidas físicas (espessura, largura e comprimento do filé do peito) e qualidade da carne do peito (perda de peso por cozimento, maciez e pH) em frangos de corte com 35, 42 e 49 dias. Foram utilizados 2400 pintos de um dia, distribuídos em um delineamento inteiramente casualizado em fatorial 5x2, com cinco linhagens (conformação A, B e C ; e convencionais D e E) e dois sexos, com quatro repetições de 60 aves cada. O crescimento das aves foi diferente, havendo linhagens com maior crescimento tanto entre as de conformação quanto entre as convencionais. As linhagens apresentaram desempenho diferenciado para ganho de peso, consumo de ração e conversão alimentar. Os animais de algumas linhagens de conformação apresentaram desempenho superior às convencionais, mas o oposto também foi verificado. O rendimento de carcaça diferiu aos 35 dias, com as linhagens de conformação apresentando os maiores rendimentos. O rendimento de pernas e carne de pernas foi semelhante ou melhor nas linhagens convencionais e os maiores rendimentos de peito e carne de peito ocorreram nas linhagens de conformação. A gordura abdominal diferiu aos 42 dias somente entre as linhagens de conformação. A espessura, largura e comprimento dos filés do peito também foram diferentes, com as linhagens de conformação apresentando valores semelhantes entre si e superiores aos das linhagens convencionais. Não houve diferenças nos parâmetros de qualidade da carne. O sexo das aves teve influência tanto no desempenho quanto no rendimento de carcaça e partes, porém não afetou a qualidade da carne do peito.
