Este trabalho teve por objetivo avaliar, comparativamente, a capacidade extratora de P da soja (Glycine max), milho (Zea mays), braquiária brizantha (Brachiaria brizantha) e milheto (Pennisetum glaucum), submetidos a diferentes doses do fertilizante fosfatado natural fosforita Alvorada, em condições controladas. Utilizou-se um Argissolo Vermelho distroférrico de textura média, corrigido e adubado com N, K e micronutrientes. As espécies foram cultivadas em vasos de 18 dm³ por 50 dias em casa de vegetação, com aplicação de 0, 100, 200 e 400 kg ha-1 de P2O5. As quantidades de fosfato natural foram calculadas com base na teor total de P2O5. O milho, ao contrário da soja, respondeu positivamente ao aumento da dose de P2O5 via fosforita Alvorada. A Brachiaria brizantha cv. Marandu, apesar da menor produção de matéria seca em relação ao milheto, apresentou alta eficiência na absorção de P, mesmo com o fornecido deste nutriente por meio de fonte pouco solúvel. O milheto apresentou-se como importante espécie de cobertura do solo, graças ao alto potencial para produção de fitomassa e reciclagem de P num intervalo de tempo relativamente curto (50 dias).

Avaliaram-se a composição química da madeira e da casca de sete espécies (E. saligna E. grandis, E. urophylla, E. camaldulensis, E. citriodora, E. paniculata e E. pellita) e três clones de eucalipto (híbridos de E. grandis x E. urophylla), antes e durante o cultivo das linhagens LE-95/01 e LE-96/18 de shiitake (Lentinula edodes), em toras. Cada linhagem de shiitake foi inoculada em nove toras de cada tipo de eucalipto com 1 m de comprimento e 9 a 14 cm de diâmetro. Assim, o delineamento experimental foi inteiramente casualizado, com 20 tratamentos e nove repetições, sendo cada repetição correspondente a uma tora. As toras foram mantidas em estufa climatizada, com temperatura de 25 ºC ± 5 e umidade relativa do ar entre 60-80%, durante 12 meses. Para a determinação da composição química da madeira, analisaram-se cunhas de discos e cascas de eucalipto recém-cortadas (sem inoculação das linhagens de L. edodes) e cunhas de discos e cascas retirados de toras já inoculadas com as linhagens de L. edodes após oito meses de incubação. Os resultados mostraram diferenças nos teores de holocelulose, lignina e extrativos totais na madeira e casca após o corte e depois de oito meses de incubação nas espécies e clones de eucalipto; o maior índice de decomposição da holocelulose na madeira, ao longo do tempo, ocorreu no E. saligna (5,5%), indicando, assim, ser o mais favorável para o desenvolvimento micelial do L. edodes. Já na casca aconteceu no clone 24 (22,2%). O E. camaldulensis apresentou o maior índice de decomposição da lignina na madeira (6,8%), ao longo do tempo. Já na casca, entre os eucaliptos testados, o E. grandis sofreu a maior decomposição de lignina (21,9%); o L. edodes degradou muito mais a holocelulose e lignina da casca que da madeira, tornando evidente a importância da casca; a casca da maioria dos tipos de eucaliptos apresentou menor teor de holocelulose, maior teor de extrativos totais e teores de lignina semelhantes ou superiores quando comparados com a madeira. O fator tipo de eucalipto (espécies e clones) teve maior efeito que o fator linhagem de L. edodes na degradação da holocelulose e lignina.

A pimenta vermelha é rica em vitamina C e outros fitoquímicos e pode ser consumida como produto desidratado. A avaliação das melhores condições de secagem pode garantir melhor qualidade do produto. Assim, o objetivo deste trabalho foi estudar o efeito da temperatura do ar de secagem (55, 65, 75 ºC) sobre a cinética de secagem, conteúdos de vitamina C e fenólicos totais e cor do produto desidratado, comparando-os à pimenta in natura. A desidratação foi feita por convecção forçada em estufa. A cinética de secagem foi determinada por pesagens periódicas até peso constante. A umidade da pimenta in natura foi de aproximadamente 86%. As curvas de secagem foram ajustadas por três modelos diferentes, avaliados na literatura. O modelo de Page apresentou o melhor ajuste para este processo. A análise de variância mostrou que a temperatura de secagem influenciou significativamente (p < 0,05) os parâmetros de qualidade (conteúdo de vitamina C, conteúdo de fenólicos totais, cor) da pimenta desidratada quando comparados aos da pimenta in natura. Após a secagem, a retenção de vitamina C aumentou com a redução da temperatura de secagem. de maneira geral, a qualidade do produto foi favorecida na secagem com menor temperatura, devido à redução nas perdas de compostos bioativos.

The carotenoid composition of Brazilian Valencia orange juice was determined by open column chromatography (OCC) and high-performance liquid chromatography. Carotenoid pigments were extracted using acetone and saponified using 10% methanolic potassium hydroxide. Sixteen pigments were isolated by OCC and identified as alpha-carotene, zeta-carotene, beta-carotene, alpha-cryptoxanthin, beta-cryptoxanthin, lutein-5,6-epoxide, violaxanthin, lutein, antheraxanthin, zeaxanthin, luteoxanthin A, luteoxanthin B, mutatoxanthin A, mutatoxanthin B, auroxanthin B and trollichrome B. Thirteen carotenoid pigments were separated using a ternary gradient (acetonitrile-methanol-ethyl acetate) elution on a C-18 reversed-phase column. Among these, violaxanthin, lutein, zeaxanthin, beta-cryptoxanthin, zeta-carotene, alpha-carotene, and beta-carotene were quantified. The total carotenoid content was 12 +/- 6.7 mg/1, and the major carotenoids were lutein (23%), beta-cryptoxanthin (21%), and zeaxanthin (20%). 2005 Elsevier Ltd. All rights reserved.

O objetivo do estudo foi determinar os conteúdos de saponinas, taninos e a solubilidade da proteína (SP) de 28 cultivares de alfafa: Crioula, Monarca, BR 4, Alto Great, MH 4, SW 9210 A, 5929, BR 1, EL Grande, 5715, MH 15, Valley Plus, BR 2, Rio, SW 8210, Maricopa, ICI 990, 5888, P 30, Alfa-200, WL 516, SW 8112 A, BR 3, Florida 77, Araucana, Falcon, Semit 921 e Sutter. O material analisado foi obtido do 10º e 14º corte, respectivamente; em 08/12/97 e 16/04/98, de um experimento desenvolvido na Faculdade de Medicina Veterinária e Zootecnia, UNESP, Botucatu. Amostras de cada cultivar foram colhidas, pesadas e secas em estufa a 52ºC. As saponinas foram extraídas com solvente hidrofílico e lipofílico e o conteúdo foi calculado pela equação y=0,8121x-1,4759, R² = 1,00. A extração dos taninos totais e condensados foi efetuada por meio de ultra-som (12 min), sendo os taninos totais determinados pela equação: y=44,978 + 0,5644 com R²=0,9977 e os condensados, multiplicando-se a absorbância por 78,26 dividido pelo teor de matéria seca. A SP foi determinada pelo método de KOH, de acordo com a seguinte fórmula: SP (%) = proteína solúvel x 100/proteína bruta da amostra. Os teores de saponinas, taninos totais e condensados e a solubilidade da proteína não diferiram (P>0,05) entre as cultivares. Houve efeito (P<0,05) da época de corte apenas sobre o teor de taninos totais. Os teores médios de saponinas de 1,00% aliados à baixa solubilidade média da proteína bruta (34,11%) não se constituem em fatores limitantes para uso dos cultivares de alfafa estudadas.

Purpose: To evaluate the effects of mechanical ventilation (MV) of high-oxygen concentration in pulmonary dysfunction in adult and elderly rats. Methods: Twenty-eight adult (A) and elderly (E), male rats were ventilated for 1 hour (G-AV1 and G-EV1) or for 3 hours (G-AV3 and G-EV3). A and E groups received a tidal volume of 7 mL/kg, a positive end-expiratory pressure of 5 cm H2O, respiratory rate of 70 cycles per minute, and an inspiratory fraction of oxygen of 1. We evaluated total protein content and malondialdehyde in bronchoalveolar lavages (BAL) and performed lung histomorphometrical analyses. Results: In G-EV1 animals, total protein in BAL was higher (33.0 +/- 1.9 mu g/mL) compared with G-AV1 (23.0 +/- 2.0 mu g/mL). Upon 180 minutes of MV, malondialdehyde levels increased in elderly (G-EV3) compared with adult (G-AV3) groups. Malondialdehyde and total proteins in BAL after 3 hours of MV were higher in elderly group than in adults. In G-EV3 group we observed alveolar septa dilatation and significative increase in neutrofiles number in relation to adult group at 60 and 180 minutes on MV. Conclusion: A higher fraction of inspired oxygen in short courses of mechanical ventilation ameliorates the parameters studied in elderly lungs.

Anuran amphibians exhibit different patterns of energy substrate utilization that correlate with the intensity of vocal and locomotor activities. Given the remarkable differences among species in breeding and feeding strategies, and the different ways energy is used in the whole animal, the suggested correlations between calling and locomotor behavior and the level of energy substrates in the muscles responsible for such activities are more complex than previously reported. We explored the relationships between calling and locomotor behavior and energy supply to trunk and hindlimb muscles, respectively, within the ecologically diverse tree-frog genus Scinax. Specifically, we measured the relative amount of carbohydrates and lipids in these two groups of muscles, and in the liver of three species of Scinax that differ in vocal and locomotor performance, and compared our results with those of two other species for which comparable data are available. We also compared the contents of lipids and carbohydrates of conspecific males collected at the beginning and after 4 h of calling activity. The stomach content to potential feeding opportunities across species was also assessed in both groups of males. Scinax hiemalis and S. rizibilis exhibit comparatively low and episodic calling during long periods of activity whereas S. crospedospilus calls at higher rates over shorter periods. Male S. hiemalis had highest levels of trunk muscle glycogen followed by those of S. rizilbilis and S. crospedospilus, respectively. There was no correlation between total lipid content in trunk muscle and calling rate among different species, suggesting that other metabolic aspects may be responsible for the energetic support for vocal activity. The levels of lipids and carbohydrates in trunk and hindlimb muscles and liver of males collected at the beginning and 4 h into the calling period were similar across species, so the extent of energetic reserves does not appear to constrain vocal or locomotor activity. Finally, we found exceptionally high levels of carbohydrates and lipids in the liver of S. rizibilis, a trait perhaps related to a long and demanding breeding period.

Trans-dehydrocrotonin, the major diterpene isolated from the bark of Croton cajucara, has good antiulcerogenic activity which, however, is accompanied by toxic effects. on the basis of these results, a semi-synthetic crotonin, named 4SRC, was prepared to determine whether this substance has similar antiulcerogenic activity with lower or no toxicity. The natural crotonin was also isolated from the bark of C. cajucara but was not used due to the small amount obtained. The cytotoxic effect of semi-synthetic crotonin, expressed as cell viability, was assessed in (a) lung fibroblast cell line (V79) derived from Chinese hamsters, a system commonly used for cytotoxicity studies, and (b) rat hepatocytes isolated from male Wistar rats. After treatment, cell viability was determined by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide reduction (MTT reduction), total acid content and neutral red uptake assays. To evaluate V79 cell viability, different concentrations of semi-synthetic crotonin were incubated with the cells. To evaluate the antiulcerogenic effects of semi-synthetic crotonin (50, 100 and 200 mg/kg), we used the models of gastric ulcer induced by ethanol/HCl, stress, indomethacin/bethanechol, and ethanol in male Swiss mice and male Wistar rats. The substance had an IC50 = 500 muM in the neutral red uptake and MTT reduction tests and an IC50 = 200 muM in the nucleic acid content test. With regard to hepatocyte viability after treatment with semi-synthetic crotonin at different concentrations, semi-synthetic crotonin had an IC50 = 10-500 muM in the nucleic acid content and MTT reduction tests and an IC50 = 120 muM in the neutral red uptake test. In another experiment, V79 cells were incubated with the metabolites produced by hepatocytes treated with different concentrations of semi-synthetic crotonin. After a 4-h incubation, semi-synthetic crotonin had an IC50 = 500 muM in the MTT reduction and neutral red uptake tests and an IC50 = 370 muM in nucleic acid content test. The substance had significant antiulcerogenic activity in all models studied, suggesting the presence of a possible antisecretory effect combined with a cytoprotective effect. For this reason, the effect of semi-synthetic crotonin was also evaluated on biochemical parameters of gastric juice and gastric wall mucus, both obtained from pylorus-ligated mice. No significant differences were observed in these parameters between semi-synthetic crotonin-treated and control animals. The results obtained with semi-synthetic crotonin are promising, with a significant preventive effect against gastric ulcer induced by different agents. Our data also show that semi-synthetic crotonin was less toxic than dehydrocrotonin and that the cytotoxic effects decreases with the time that isolated hepatocytes were in culture. Water contaminants have a high potential risk for the health of populations. Protection from toxic effects of environmental water pollutants primarily involves considering the mechanism of low level toxicity and likely biological effects in organisms who live in these polluted waters. The biomarkers assessment of oxidative stress and metabolic alterations to cadmium exposure were evaluated in Nile tilapia, Oreochromis niloticus. The fish were exposed to 0.35, 0.75, 1.5, and 3.0 mg/l concentrations of Cd2+ (CdCl2) in water for 60 days. Fish that survived cadmium exposure showed a metabolic shift and a compensatory development for maintenance of the body weight gain. We observed a decreased glycogen content and decreased glucose uptake in white muscle. Lactate dehydrogenase (LDH) and creatine phosphokinase (CK) activities were also decreased, indicating that the glycolytic capacity was decreased in this tissue. No alterations were observed in total protein content in white muscle due to cadmium exposure suggesting a metabolic shift of carbohydrate metabolism to maintenance of the muscle protein reserve. There was an increase in glucose uptake, CK increased activity, and a clear increase of LDH activity in red muscle of fish with cadmium exposure. Since no alterations were observed in lipoperoxide concentration, while antioxidant enzymes glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) were changed in the liver and the red and white muscle of fish with cadmium exposure, we can conclude that oxygen free radicals are produced as a mediator of cadmium toxicity. Resistance development is related with increased activities of antioxidant enzymes, which were important in the protection against cadmium damage, inhibiting lipoperoxide formation.
