One of the main objectives of the Spanish and Portuguese-Speaking Group of the International Society for Forensic Genetics (GHEP-ISFG) is to promote and contribute to the development and dissemination of scientific knowledge in the area of forensic genetics. Due to this fact, GHEP-ISFG holds different working commissions that are set up to develop activities in scientific aspects of general interest. One of them, the Mixture Commission of GHEP-ISFG, has organized annually, since 2009, a collaborative exercise on analysis and interpretation of autosomal short tandem repeat (STR) mixture profiles. Until now, three exercises have been organized (GHEP-MIX01, GHEP-MIX02 and GHEP-MIX03), with 32, 24 and 17 participant laboratories respectively. The exercise aims to give a general vision by addressing, through the proposal of mock cases, aspects related to the edition of mixture profiles and the statistical treatment. The main conclusions obtained from these exercises may be summarized as follows. Firstly, the data show an increased tendency of the laboratories toward validation of DNA mixture profiles analysis following international recommendations (ISO/IEC 17025: 2005). Secondly, the majority of discrepancies are mainly encountered in stutters positions (53.4%, 96.0% and 74.9%, respectively for the three editions). On the other hand, the results submitted reveal the importance of performing duplicate analysis by using different kits in order to reduce errors as much as possible. Regarding the statistical aspect (GHEP-MIX02 and 03), all participants employed the likelihood ratio (LR) parameter to evaluate the statistical compatibility and the formulas employed were quite similar. When the hypotheses to evaluate the LR value were locked by the coordinators (GHEP-MIX02) the results revealed a minor number of discrepancies that were mainly due to clerical reasons. However, the GHEP-MIX03 exercise allowed the participants to freely come up with their own hypotheses to calculate the LR value. In this situation the laboratories reported several options to explain the mock cases proposed and therefore significant differences between the final LR values were obtained. Complete information concerning the background of the criminal case is a critical aspect in order to select the adequate hypotheses to calculate the LR value. Although this should be a task for the judicial court to decide, it is important for the expert to account for the different possibilities and scenarios, and also offer this expertise to the judge. In addition, continuing education in the analysis and interpretation of mixture DNA profiles may also be a priority for the vast majority of forensic laboratories. (C) 2014 Elsevier Ireland Ltd. In this study were carried out toxicity tests and the evaluation of sub lethal effects of the insecticides malathion and fipronil, over the test system of Girardia tigrina, using for these analyses a great variety of methods. For evaluation of the lethal concentrations of the products, adult planarians were exposed to several different concentrations. All the tests were carried out in duplicate, maintained on the shelter of the light, on a temperature 24-26ºC and they had the duration of 96h, with observations at each 24h. The results were statically valued by the tests U of Mann- Whitney, Spearman-Karber, ANOVA and Pearson correlation. The effects over the system-test, in the different tested concentrations, varied in accordance with the time of exposition. The EC50 given by the test of Spearman-Karber was of 12,5mg/L for malathion and 13,4mg/L for fipronil. The evaluation of sub lethal effects was composed by analyses of the potential of regeneration; histological, for tissue damage evaluation; d; and the “comet assay”, for DNA damage detection. The specimens split up and exposed to the insecticides demonstrated, between other effects, retardation in development of the regenerative blastema. The histological evaluation has shown that exposed planarians to lower concentrations of the insecticides cause more significant damage against the tissues and some specific cell types. Through the comet assay, significant damages were noticed in the DNA in all the treatments, when compared with the control group. Such damages may result in reproduction and immunity alterations, which reduce the competitive efficiency and, consequently, the survival chances of these organisms.

The aim of this study was to evaluate the antimicrobial activity of alcohol-free mouthwashes on Candida albicans. Twenty clinical isolates of C. albicans and one reference strain (ATCC 18804) were evaluated after exposure to two 0.12% chlorhexidine-based and alcohol-free (“Ca” and “Or”) in comparison to gluconate chlorhexidine with ethanol (positive control). The maximum inhibitory dilution (MID) and maximum fugal dilution (MFD) were determined by the microdilution method. Twelve serial dilutions (from 50 to 0.02%) were prepared in duplicate. Then, 100 µL of C. albicans suspension (106 cells.mL–1) were added to the wells. After incubation (37 °C/24 hours), MID was determined by reading the optical density. For MFD determination, the content of the wells were plated on Saouraud agar. For MID, there were no differences between groups Or and control, but Ca group showed a MID statistically higher (Kruskal-Wallis, p = 0.0012). For MFD, there were no differences between Ca and control (Mann-Whitney test, p = 0.1631). It can be concluded that Ca group showed a fungicid activity against C. albicans similar to the control, but lower fungistatic activity when compared to the control. Group Or showed only a fungistatic action similar to control.

This work examined the histological effects, on the rat palatal mucosa, of a denture base acrylic resin, submitted or not to a post-polymerization heat-treatment. Methods: Fifteen adult female Wistar rats, with sixty days old, weighting 150 g – 250 g were divided in G1: animals being maintained under the same conditions as the experimental groups following described, but without the use acrylic palatal plates (control group); G2: use of heat-polymerized acrylic resin palatal plates made of Lucitone 550; G3: use of palatal plates identical to G2, but subjected to a post-polymerization treatment in a water bath at 55°C for 60 min. The plates covered all the palate and were fixed in the molar region with light-cured resin, thus being kept there for 14 days. After the sacrifice, the palate was removed, fixed in formaldehyde 10% and decalcified with EDTA. Sections were stained using haematoxylin and eosin. Images in duplicate were made from the central region of the cuts, to measure the thickness (μm) of the keratin layers (TKC), epithelium total (TET) and connective tissue (TCC). Statistical analyses were carried out by one-way ANOVA and Tukey post-tests (α=0.05). Results: According to the results there was significant difference in the thickness of keratin between G2 and G3, with G1 having the intermediate value and similar to the other groups. To date little is known about the reliability of peak oxygen consumption (VO2pEAK) in incremental metronome paced step tests (1ST) and the reliability of on-kinetics VO2 has never been studied. We aimed to study the reliability of both tests. Eleven healthy subjects performed two ISTs until exhaustion. On two different days two duplicate 4 min constant metronome paced step tests (CST) were performed. VO2PEAK, mean response time (MRT) and phase II time constant (tau) were tested for reproducibility using the paired t-tests, in addition to the limits of agreement (LOA) and within subject coefficient of variation (COV). With a 95% LOA of 0.38 to 0.26 L min(-1), -8.7 to 9.1 s and -9.9 to 10.5 s they exhibit a COV of 3%, 4.5% and 6.9% for VO2PEAK, MRT and tau respectively. ST are sufficiently reliable for maximal and submaximal aerobic power assessments in healthy subjects and new studies of oxygen uptake kinetics in selected patient groups are warranted. A multiseries integrable model (MSIM) is defined as a family of compatible flows on an infinite-dimensional Lie group of N-tuples of formal series around N given poles on the Riemann sphere. Broad classes of solutions to a MSIM are characterized through modules over rings of rational functions, called asymptotic modules. Possible ways for constructing asymptotic modules are Riemann-Hilbert and ∂̄ problems. When MSIM's are written in terms of the group coordinates, some of them can be contracted into standard integrable models involving a small number of scalar functions only. Simple contractible MSIM's corresponding to one pole, yield the Ablowitz-Kaup-Newell-Segur (AKNS) hierarchy. Two-pole contractible MSIM's are exhibited, which lead to a hierarchy of solvable systems of nonlinear differential equations consisting of (2 + 1) -dimensional evolution equations and of quite strong differential constraints. © 1989 American Institute of Physics.

During the meiotic division of Dermatobia hominis spermatogenesis, the centrioles duplicate only in prophase I, giving rise to short cilia which are exposed on the cellular surface. In metaphase I they are internalized and distributed to the daughter cells. Consequently, the secondary spermatocytes have two centrioles which repeat the cycle of cilia externalization followed by internalization. The spermatids receive only one centriole, which changes into a basal body and originates a flagellum. This centriole behaviour seems to be a general feature in insect male germ cell meiosis.

To study the influence of the addition of various antioxidants and their combinations on the artifactual oxidation of cholesterol during analysis, 2 factorial experiments were performed in duplicate. In the first experiment, 2 amounts of the following antioxidants were assayed: ethylenediaminetetraacetic acid (EDTA) disodium salt (0 and 1 mg), pyrogallol (0 and 600 microg), and butylated hydroxytoluene (BHT; 0 and 600 microg); in the second, EDTA disodium salt (0 and 1 mg), ascorbyl palmitate (0 and 600 microg), and BHT (0 and 600 microg). Under low oxidative conditions of dim light, evaporation of solvents at low temperatures, and cold saponification in darkness under nitrogen atmosphere, the addition of antioxidants showed no further protective effect. Furthermore, the presence of ascorbyl palmitate significantly increased the formation of cholesterol-5beta,6beta-epoxide, and 7beta-hydroxycholesterol.

To review the literature searching for a consensus for the choice of orthodontic extrusion as treatment for crown-root fracture. An electronic search was performed in the databases PubMed, Cochrane Central Register of Controlled Trials and Scopus and a manual search of the Journal Dental Traumatology. Forty articles were found in PubMed and 38 in Scopus and after removal of duplicate sample 51 contained articles. Of these, 48 were excluded for not having orthodontic treatment, no follow-up or follow-up less than 6 months, or not report the presence of crown-root fracture. In manual search in Dental Traumatology 20 articles were found, but none of them met the prerequisites established. So, three articles formed the basis of the study. The choice of how to treat orthodontic extrusion of crown-root fracture was effective and stable, without root and periodontal changes. Factors, such as root formation and presence of pulp vitality were decisive for determining the stages of treatment, however, there is no consensus based on scientific evidence about these protocols.

Mouthwashes are used as an adjunct to tooth brushing for improving breath and preventing oral diseases. The aim of this study was to compare the in vitro Maximum Inhibitory Dilution (MID) of 3 mouthwashes with different active ingredients against mutans streptococci (MS). The products analyzed were PeriogardR, CepacolR and PlaxR Fresh Mint. Their antibacterial activity was assessed in duplicate in 96-well microtiter plates against 36 clinical isolates of MS. Each mouthwash was submitted to a serial two-fold dilution (1/2.5 to 1/5120) using double concentration of Tryptose Soy Broth with 1.0% yeast extract. The final volume in each well was 100 mL plus 5 mL of a bacterial suspension, equivalent to 107 CFU/mL. They were incubated microaerobically at 37oC for 48 hours and the MIDs determined. MID was 1/320 for PeriogardR and CepacolR, and 1/20 for PlaxR. Statistical analysis revealed that the MID of PeriogardR MID did not differ from that of CepacolR (p>0.05), and was higher than that of PlaxR (p<0.05). In conclusion, the antiseptic mouthwashes containing chlorhexidine (PeriogardR) and cetylpyridinium chloride (CepacolR) had higher in vitroantibacterial activity (MID) against MS than the antiseptic mouthwash containing triclosan (PlaxR), according to microbiological method employed.

One of the most primal ways of human work already known is the tessellation and ginning for the production of fabric and clothing - what used to be, back in those days, statement of power and status. The arrival of the Industrial Revolution - in the middle of the XVIII century at Britain - increased the textile industry production, and what used to be manufactured and hard to obtain, starts then to be produced in mechanical ways and large-scale. Despite all the boost given to the economy of an expanding capitalist market, it should be pointed out the consequences of this major industrialization, especially the environmental ones, more and more concerning nowadays. The emissions of waste - that sometimes could be toxic - in effluents can possibly contaminate the aquatic ecosystems, causing a huge damage to its fauna and flora, affecting therefore all the biodiversity, reaching inclusively the humans. To avoid these problems, a few strategies have been taking place in the attempt to eliminate - or at least reduce - the amount of dye found in the effluents, and as the textile industry constantly leaves waste, efficient methods - that present good results in a short period of time - with a low cost are needed. The present study will test the bioremoval capacity of yeast (Saccharomyces cerevisiae) in contact with dyes in a fix concentration, diluted in water with three different pH values. The tests will be done duplicate, and after the concentration analyses - made by spectrophotometry - it will be analyzed which pH shows major efficiency in the dye removal and what is the influence of the biomass in this process
