The synthesis of intracellular glycerol-3-phosphate dehydrogenase (EC 1.1.1.8) in baker's yeast was investigated in submerged culture supplied with glucose or glycerol as sole carbon sources. Inhibitors of the glycolytic pathway, Krebs cycle and respiratory chain did not stimulate glycerol-3-phosphate dehydrogenase synthesis when added in low concentrations in up 7.5 × 10 -5 mol/L. The repression exercised by glucose on the synthesis of glycerol-3-phosphate dehydrogenase in YP-glucose medium was reduced by the addition of fermentation products and of sodium bisulfite. Synthesis of the enzyme was raised 22-110%. However, in YP-glycerol medium, the addition of 0.06% (w/v) sodium bisulfite reduced (29%) the synthesis of the enzyme, while 0.012% (v/v) acetaldehyde stimulated the synthesis of glycerol-3-phosphate dehydrogenase by 12%.

Microwaves have been used in organic synthesis, since 1986, and have proved advantageous in several respects: the possibility of higher yields, greater selectivity and less thermal decomposition. Phthalimide and its derivatives constitute an important class of compounds for use in synthetic organic chemistry; in medicinal chemistry, it is considered an important biophore, acting as a pharmacophoric structural subunit for the synthesis of a number of compounds with different pharmacological uses, such as against sickle-cell disease. The purpose of the work reported here was to develop an alternative method for the synthesis of phthalimide derivatives by exploiting the condensation of phthalic anhydride with amino groups under microwave radiation. The results showed that phthalimide derivatives were obtained in shorter reaction times (5-10 min) and higher yields (60-89%) than by with conventional heating (reflux), demonstrating the potential use of microwaves in the synthesis of this class of molecules.

Anti-inflammatory drugs are known to be the most widely-marketed drugs in the world, despite their serious side effects, mainly on the gastrointestinal tract. Thus, there are constant efforts to discover new prototypes with improved therapeutic activity and safety for the patient. Since the advent of the computational chemistry, the theoretical study of the physiological behavior of a new compound and hence an understanding of its supposed mechanism of action have been made a lot more accessible. Thus, molecule-receptor mathematical modeling was applied to compound I (1-(2,6-dichlorophenyl)indolin-2-one), to predict theoretically its ability to inhibit, selectively, the COX-2 isoform of prostaglandin endoperoxide synthase (PGHS-2), and the best positions to introduce chemical groups and to make molecular modifications.

Schistosomiasis, an important disease in Brazil, is caused by a trematode of the genus Schistosoma, reaching millions of person in one of the most endemic region of this disease in the whole globe. The main goal of this work was to syntetize the 6-formyl- oxamniquine derivative and evaluate its biological activity. The 6-formyl-oxamniquine derivative was obtained by the oxidation of oxamniquine with MnO 2, applying CH 2Cl 2 as solvent at room temperature for 24 hours. The obtaintion of 6-formyl-oxamniquine derivative compound was confirmed by IR spectroscopy and 13C NMR and 1H NMR, presenting similar activity when compared to the commercial oxamniquine (Mansil®).

This work aimed to determine ruminal parameters of dry mater (DM), neutral detergent fiber (NDF) and protein synthesis in the rumen of animals fed diets based on sorghum with different tannin levels. In situ degradation was evaluated by rumen incubation of level 1 (L1) and level 2 (L2) sorghum silages during 0, 6, 24, 48 and 96 hours. A duplicated 4 × 4 Latin Square was used. Potential degradability (PD) of DM was similar among silages, whereas the effective degradability (ED) decreased when the diets were supplemented with concentrate. Microbial nitrogen flux and microbial synthesis efficiency were not affected by roughage source. The synthesis efficiency, expressed in organic matter and crude protein digested in rumen, was higher in L1 tannin diets supplemented with concentrate. There was not relationship between the presence of tannins and the parameters of ruminal degradation.

Given a linear time-invariant plant Gol(s) with one input and q outputs, where q > 1, a method based on the Routh-Hurwitz Stability Criterion is proposed to obtain a constant tandem matrix F ∈ ℝq, such that FGOl(s) is a minimumphase system. From this solution, the system FGol(s) is represented in state space by {A, B, FC} and a constant output feedback matrix K0 ∈ ℝ is obtained such that the feedback system {A - BK0C, B, FC} is Strictly Positive Real (SPR). The proposed procedure offers necessary and sufficient conditions for both problems. Initially, the general case, with a generic q, is analyzed. Following, the particular cases q = 2 and q = 3 are studied.

Sickle cell disease (SCD) is a hereditary hemolytic anemia caused by the inheritance of one S hemoglobin gene from each ancestor. Patients with SCD present increased circulating levels of cytokines, including TNF-alpha (TNF-α). Hydroxyurea (HU) is the available therapeutically strategy for treatment; it acts as a source of nitric oxide and benefits patients by increasing the levels of fetal hemoglobin (HbF). Thus, within one research line that aims at finding new drugs, a series of compounds with TNF-α inhibition and nitric oxide donation properties have been synthesized in order to explore possible synergism of actions beneficial in the treatment of the disease. Six compounds were synthesized: five derivatives of organic nitrates and one of sulfonamide. The compounds, (1,3-dioxo-1,3-dihydro-2Hisoindol-2-yl) methyl nitrate (compound I); (1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) ethyl nitrate (compound II); 3-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) benzyl nitrate (compound III);4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl)-N-hydroxybenzenesulfonamide (compound IV); 4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) benzyl nitrate (compound V) and 2-[4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) phenyl]ethyl nitrate (compound VI), were synthesized using linear synthetic methodology, with excellent overall yields. All compounds showed anti-inflammatory and analgesic effects with a reduction in 43%-65% of ear edema in mice and a reduction of 25%-42% of writhing induced by acetic acid. All compounds showed comparable reductions in the leukocyte infiltration capacity and ability to generate nitric oxide. The aryl compounds (III, IV and V) presented less mutagenic activity compared to compounds I, II and VI according to the salmonella mutagenicity assay (Ames test). Compounds IV and VI showed activity in K562 culture cells, with increases in gamma globin gene expression to levels higher than with hydroxyurea suggesting a potential to increase fetal hemoglobin. This data set characterizes new potentially useful drug candidates for the treatment of symptoms of sickle cell anemia.
