No presente estudo procedeu-se ao isolamento e caracterização da fração globulina majoritária (11 S) de grão-de-bico, var. IAC-Marrocos. A globulina majoritária extraÃda foi isolada por cromatografia de filtração em gel e de troca-iônica mostrando apenas uma banda de proteÃna na eletroforese em gel de poliacrilamida. A globulina majoritária, após passagem em coluna de Sephadex, revelou duas bandas protéicas de 55 e 52,5kDa e três bandas menores em gel de poliacrilamida dodecilsulfato de sódio. Na presença de 2-mercaptoetanol 6 polipeptÃdios na faixa de 18 a 42kDa foram revelados na eletroforese. A globulina isolada foi submetida à ação da tripsina e quimotripsina onde a forma nativa mostrou-se resistente à ação enzimática enquanto o aquecimento (96 e 121°C/15min) não foi suficiente para aumentar a susceptibilidade à hidrólise, significativamente. Adição de NaCl 0,3M levou a um aumento da estabilidade estrutural com menor susceptibilidade à digestão proteolÃtica, fato em parte perdido com o aquecimento. As hidrólises foram acompanhadas por eletroforese em gel de poliacrilamida dodecilsulfato de sódio.

A composição quÃmica e a qualidade biológica da torta de colza obtida da semente, variedade CTC-4, foram determinados. A farinha foi preparada me diante prensagem da semente e extração da gordura com solvente, lixiviada durante 2 horas, com água corrente na proporção de 1:5, e agitação constante, a fim de reduzir os nÃveis de glicosinolatos. O efeito tóxico foi observado através do peso dos órgãos e da histopatologia dos órgãos em experimento de 60 dias. A qualidade biológica da torta desintoxicada, ao nÃvel de 5% e 10%, foi semelhante ao da caseÃna (PER controle 3,18 vs 3,08, 5% e 2,98, 10%). O PER da torta crua foi de 2,28. Os ratos que receberam a farinha crua tiveram órgãos maiores. As tireóides dos ratos submetidos as dietas 2 e 3, não demonstraram alterações que suspeitassem de um efeito bociogênico e as alterações ocorridas nas tireóides dos animais que receberam, D-4 foram semelhantes à quelas ocorridas nos grupos 2 e 3. Não houve danos célulares graves em relação ao fÃgado, os rins, o coração, baço e supra-renais para todos os grupos observados. O método de extração dos fatores tóxicos da colza foi eficiente de acordo com os resultados obtidos para o presente estudo.

The chemical and biochemical composition of mango, varies according to the cultivation conditions, variety and maturation state, generally containing a high level of ascorbic acid. In order to establish the correlation between the activity of the ascorbate oxidase [E.C.1.10.3.3], and ascorbic acid level in the ripening process of the Haden mango (MangÃfera Ãndica L.), sample of the fruits related to hard green stage (zero), 2, 4, 6, 8, 10, 12 and 14 days stored at 20 ± 2oC, were tested. The samples were obtained by cutting small cubes of 8 cm3 from pulps of 8 mangoes with texture without significant difference (p£0.05) at Magness-Taylor pressure tester scale. In each sample the activity of ascorbate oxidase was followed, in order to check its participation in possible substrate losses during the ripening fruits. The ascorbic acid level and sensory profile also was determined periodically during the ripening period. The enzymatic activity was spectrophotometrically determined at 245 nm and 30oC. The ascorbic acid was analyzed according modified AOAC methodology, and sensory analysis by descriptive quantitative analysis. Data were analyzed using correlation analysis, analysis of variance (ANOVA), Tukey's test, principal component analysis and stepwise discriminant analysis. During the ripening, the ascorbate oxidase activity increased (from 0 to 5.0 x 10-1 U/ml) and the ascorbic acid level decreased (from 209.3 mg to 110.0 mg per 100g of pulp), showing a significant (p£0.05) inverse linear correlation (r=-0.98). The descriptors terms for mangoes were: characteristic flavor, characteristic aroma, sourness, astringency, yellow coloration of pulp, sweetness and succulence. The sensory profile presented significant improvement during ripening. All sensory attributes increased significantly (p£0.05) except sourness and astringency, wich decreased during the ripening of mangoes.

Farmers are interested in producing popcorn under organic production systems and propane flaming could be a significant component of an integrated weed management program. The objective of this study was to collect baseline information on popcorn tolerance to broadcast flaming as influenced by propane dose and crop growth stage at the time of flaming. Field experiments were conducted at the Haskell Agricultural Laboratory of the University of Nebraska, Concord, NE in 2008 and 2009 using five propane doses (0, 13, 24, 44 and 85 kg ha(-1)) applied at the 2-leaf, 5-leaf and 7-leaf growth stages. Propane was applied using a custom-built research flamer driven at a constant speed of 6.4 km h(-1). Crop response to propane dose was described by log-logistic models on the basis of visual estimates of crop injury, yield components (plants m(-2), ears plant(-1), kernels cob(-1) and 100-kernel weight) and grain yield. Popcorn response to flaming was influenced by the crop growth stage and propane dose. Based on various parameters evaluated, popcorn flamed at the 5-leaf showed the highest tolerance while the 2-leaf was the most susceptible stage. The maximum yield reductions were 45%, 9% and 16% for the 2-leaf, 5-leaf and 7-leaf stages, respectively. In addition, propane doses that resulted in a 5% yield loss were 23 kg ha(-1) for the 2-leaf and 7-leaf and 30 kg ha(-1) for the 5-leaf stage. Flaming has a potential to be used effectively in organic popcorn production if properly used. (C) 2010 Elsevier Ltd. All rights reserved.

The objective of this work was to evaluate the influence of the use of nitrogen and phosphorus in the culture of jambu. The experiment was initiated at the experimental farm of the agency APTA Polo Vale do Ribeira, using the Nazaré cultivar. The experimental design was complete randomized with fourlevelsof nitrogen (0, 37.5, 75 and 112.5 kg ha-1 N) and phosphorus (0, 75, 150 and 300 Kg ha-1P2O5),and four reapplications. After soil preparation, liming and fertilization, seedlings of jambu were transplanted at a spacing of 0.50 x 0.5 m. The experimental plot was 1.0 x 1.0 m. Fresh and dry mass of leaves and flowers and number of flowers were analyzed. The production of fresh and dry leaves and flowers were influenced by fertilization. Thelevelsof nitrogen affected the outcome, as theyprovided a linear increase in all variables, and the phosphorus level of 75 kgha-1 P2O5 caused the highest average production in the number of flowers and in the fresh and dried mass of flowers.
