Few studies dealing with effort intensity during swimming exercise in rats have been reported in the literature. Recently, with the use of the lactate minimum test (LMT), our group estimated the minimum blood lactate (MBL) of rats during swimming exercises. This information allowed accurate evaluation of the effort intensity developed by rats during swimming exercise. The present study was designed to evaluate the effects of swimming exercise sessions in below, equivalent and above intensities to MBL, on protein metabolism of rats. Adult (90 days) sedentary male Wistar rats were used in the present study. Mean values of MBL, in the present study, were obtained at blood concentration of 6.7 +/- 0.4 mmol/L with a load of 5% bw. The animals were sacrificed at rest (R) or immediately after a single swimming session (30 min) supporting loads below (3.5% bw), equivalent (5.0% bw) and high load (6.5% bw) to AT. Blood samples were collected each 5 min of exercise for lactate determination. Soleus muscle protein synthesis (amount of L-[C-14] fenil alanyn incorporation to protein) and breakdown (tyrosin release) rates were evaluated. Blood lactate concentrations (mmol/L) stabilized with the below (5.4 +/- 0.01) and equivalent (6.4 +/- 0.006) to MBL but increased, progressively, with the high load. There were no differences in protein synthesis (pmol/mg.h) among rest values (65.2 +/- 3.4) and after-exercise supporting the loads below (61.5 +/- 1.3) and the equivalent (60.7+/-1.7) to MBL but there was a decrease with the high load (36.6+/-2.0). Protein breakdown rates (pmol/g.h) increase after exercise supporting the loads below (227.0 +/- 6.1), equivalent (227.9 +/- 6.0) and high (363.6 +/- 7.1) to MBL in relation to the rest (214.3 +/- 6.0). The results indicate the viability of the application of LMT in studies with rats since it detected alterations imposed by exercise.

In vitro rates of overall proteolysis and the activities of four different proteolytic pathways (lysosomal, Ca2+ dependent, ATP dependent, and ATP independent), as well as rates of protein synthesis, were measured in soleus and extensor digitorum longus (EDL) muscles from streptozotocin- diabetic rats. In the acute phase (1-3 days) of diabetes, there was an increase in overall proteolysis that coincided with an increased activity of the Ca2+-dependent pathway in both soleus and EDL and of the ATP-dependent pathway in EDL. After longer periods (5-10 days) of diabetes, the overall rate of protein degradation decreased and reached values similar to or even lower than those of controls as a result of a reduction in the activities of Ca2+-dependent and ATP-dependent pathways. No change was detected at any time interval in the activity of the intralysosomal proteolytic system in muscles from diabetic animals. Rates of protein synthesis were already reduced 24 h after diabetes induction and decreased further thereafter. Insulin treatment restored to normal the activities of the proteolytic pathways and rates of protein synthesis.

Kaposi's sarcoma-associated herpesvirus (KSHV/human herpesvirus 8 [HHV8]) and Epstein-Barr virus (EBV/HHV4) are distantly related gammaherpesviruses causing tumors in humans. KSHV latency-associated nuclear antigen 1 (LANA1) is functionally similar to the EBV nuclear antigen-1 (EBNA1) protein expressed during viral latency, although they have no amino acid similarities. EBNA1 escapes cytotoxic lymphocyte (CTL) antigen processing by inhibiting its own proteosomal degradation and retarding its own synthesis to reduce defective ribosomal product processing. We show here that the LANA1 QED-rich central repeat (CR) region, particularly the CR2CR3 subdomain, also retards LANA1 synthesis and markedly enhances LANA1 stability in vitro and in vivo. LANA1 isoforms have half-lives greater than 24 h, and fusion of the LANA1 CR2CR3 domain to a destabilized heterologous protein markedly decreases protein turnover. Unlike EBNA1, the LANA1 CR2CR3 subdomain retards translation regardless of whether it is fused to the 5′ or 3′ end of a heterologous gene construct. Manipulation of sequence order, orientation, and composition of the CR2 and CR3 subdomains suggests that specific peptide sequences rather than RNA structures are responsible for synthesis retardation. Although mechanistic differences exist between LANA1 and EBNA1, the primary structures of both proteins have evolved to minimize provoking CTL immune responses. Simple strategies to eliminate these viral inhibitory regions may markedly improve vaccine effectiveness by maximizing CTL responses. Copyright © 2007, American Society for Microbiology. The nucleolar material of Chariesterus armatus was analyzed during spermiogenesis in cell preparations impregnated with silver nitrate. Nucleolar corpuscles were observed in spermatids at the beginning of the process, showing that this organoid is also maintained after meiosis. In addition, nucleoli were seen in the round spermatids connected to the X-chromosome (bearer of the nucleolar organizer in C. armatus), indicating de novo synthesis of nucleolar material. This differs from the reorganization of ribosomal granules, transported from meiotic spermatocytes to round spermatids, where they would support protein synthesis, which is reported for other species. We also observed connections of nucleolar corpuscles to the nuclear membrane regions where the tail and the acrosome will be formed, suggesting close involvement of the nucleolar material in the formation of these structures. In addition to the nucleolar bodies, we detected silver-positive structures, which will require new approaches to clarify their role. One of these structures, observed in the cytoplasm, appears to correspond to the chromatoid body, which has been found in several organisms, but is still poorly understood; another is a complex structure to which the tail appears to be connected. We conclude that C. armatus is an appropriate model for understanding not only the synthesis of rRNA in the spermiogenesis, but also the functional meaning of the close relationship of nucleolar material with other structures during this process.

We evaluated associations between the concentrations of heat shock proteins (hsp60 and hsp70) and their respective antibodies, alterations in maternal reproductive performance, and fetal malformations in pregnant rats with hyperglycemia. Mild diabetes (MD) or severe diabetes (SD) was induced in Sprague-Dawley rats prior to mating; non-treated non-diabetic rats (ND) served as controls. On day 21 of pregnancy, maternal blood was analyzed for hsp60 and hsp70 and their antibodies; and fetuses were weighed and analyzed for congenital malformations. Hsp and anti-hsp levels were correlated with blood glucose levels during gestation. There was a positive correlation between hsp60 and hsp70 levels and the total number of malformations (R∈=∈0.5908, P∈=∈0.0024; R∈=∈0.4877, P∈=∈0.0134, respectively) and the number of malformations per fetus (R∈=∈0.6103, P∈=∈0.0015; R∈=∈0.4875, P∈=∈0.0134, respectively). The anti-hsp60 IgG concentration was correlated with the number of malformations per fetus (R∈=∈0.3887, P∈=∈0.0451) and the anti-hsp70 IgG level correlated with the total number of malformations (R∈=∈0.3999, P∈=∈0.0387). Moreover, both hsp and anti-hsp antibodies showed negative correlations with fetal weight. The results suggest that there is a relationship between hsp60 and hsp70 levels and their respective antibodies and alterations in maternal reproductive performance and impaired fetal development and growth in pregnancies associated with diabetes. © 2012 Cell Stress Society International.

Six hundred and forty one-day-old Cobb male broilers were used to evaluate ornithine decarboxylase (ODC) expression in the mucosa of the small intestine. Birds were submitted to early feed restriction from 7 to 14 days of age. The provided feed was supplemented with glutamine. A completely randomized design with a 2 x 2 factorial arrangement was used (with or without glutamine, with or without feed restriction). Restricted-fed birds were fed at 30% the amount of the ad libitum fed group from 7 to 14 days of age. Glutamine was added at the level of 1% in the diet supplied from 1 to 28 days of age. Protein concentration in the small intestine mucosa was determined, and ODC expression at 7, 14, 21, and 28 days of age was evaluated by dot blotting. ODC was present in the mucosa of broilers, and the presence of glutamine in the diet increased ODC activation. Glutamine prevented mucosa atrophy by stimulating protein synthesis, and was effective against the effects of feed restriction. Dot blotting can be used to quantify ODC expression in the intestinal mucosa of broilers.

Objetivou-se avaliar os efeitos de diferentes formas químicas de compostos nitrogenados (proteicos e não-proteicos) e de carboidratos (amiláceos e fibra solúvel) sobre o consumo, a digestibilidade e a síntese ruminal de proteína microbiana em bovinos sob suplementação durante o período das águas. Foram utilizados cinco novilhos mestiços Holandês × Zebu, com peso vivo (PV) médio inicial de 211 ± 35 kg, fistulados no rúmen e abomaso. Os tratamentos foram: controle (somente pasto); e suplementos formulados à base de milho + farelo de soja; milho + ureia; polpa cítrica + farelo de soja; e polpa cítrica + ureia. Os suplementos foram balanceados para apresentar 30% de proteína bruta (PB), com base na matéria seca (MS), e fornecidos na quantidade de 3 g/kg PV. O experimento foi conduzido segundo delineamento em quadrado latino 5 × 5, em esquema fatorial 2 × 2 + 1, composto de duas fontes de compostos nitrogenados, duas fontes de carboidratos e tratamento controle. O consumo de pasto reduziu com o fornecimento de suplementos, com coeficiente médio de substituição de 2,11 g de MS de pasto/g de MS de suplemento. A suplementação não alterou os coeficientes de digestibilidade total e ruminação da MS nem o teor dietético de nutrientes digestíveis totais (NDT). Os animais sob suplementação apresentaram maiores coeficientes de digestibilidade total e ruminal da proteína bruta. A eficiência de síntese de proteína microbiana (EFSM), média de 123,1 g PB microbiana/kg de NDT, não foi alterada pela suplementação. Contudo, os animais sob suplementação com milho apresentaram maior EFSM em comparação aos animais sob suplementação com polpa cítrica (137,6 e 106,1 g PB microbiana/kg de NDT, respectivamente). A suplementação proteico-energética para bovinos mantidos em pastos tropicais durante o período das águas não causa benefícios nutricionais, o que reflete o alto coeficiente de substituição da forragem pelo suplemento.

Objetivou-se avaliar o efeito da substituição parcial do milho pela casca de soja e pelo farelo de gérmen de milho sobre a produção e eficiência de síntese de proteína microbiana e sobre as taxas de diluição e passagem ruminal. Foram utilizados três novilhos de corte, mestiços, canulados no rúmen e no duodeno, distribuídos em dois delineamentos em quadrado latino 3 x 3. As dietas experimentais, à base de silagem de milho (60%), apresentavam como fonte de proteína farelo de girassol e, como fonte de energia, milho (MI) ou sua substituição parcial pela casca de soja (CS) ou pelo farelo de gérmen de milho (FGM). Para o isolamento da massa microbiana, foram coletadas amostras de conteúdo ruminal às 3, 6, 9 e 12 horas após a alimentação. Utilizaram-se RNA como marcador microbiano e dicromato de sódio e Co-EDTA como indicadores das taxas de passagem e de diluição, respectivamente. A composição dos microrganismos ruminais não foi influenciada pelas dietas experimentais ou pelos horários de coleta. Não houve diferenças significativas no fluxo de matéria orgânica, carboidratos totais, nitrogênio e nitrogênio microbiano para o duodeno e na eficiência de síntese de proteína microbiana. As taxas de diluição foram semelhantes entre as dietas, com média de 13,4%/h. A taxa de passagem da silagem de milho mordentada foi menor, com tendência de menor taxa de passagem também para o farelo de girassol na dieta MI. A casca de soja e o farelo de gérmen de milho podem substituir parcialmente o milho, proporcionando ambiente ruminal adequado ao desenvolvimento da flora microbiana e conseqüente produção de proteína microbiana ruminal em novilhos confinados.
