Pectate lyase (PL) was produced by the filamentous fungus Penicillium viridicatum RFC3 in solid-state cultures of a mixture of orange bagasse and wheat bran (1 : 1 w/w), or orange bagasse, wheat bran and sugarcane bagasse (1 : 1 : 0.5 w/w), and in a submerged liquid culture with orange bagasse and wheat bran (3%) as the carbon source. PL production was highest (1,500U mL -1 or 300Ug -1 of substrate) in solid-state fermentation (SSF) on wheat bran and orange bagasse at 96 hours. PL production in submerged fermentation (SmF) was influenced by the initial pH of the medium. With the initial pH adjusted to 4.5, 5.0, and 5.5, the peak activity was observed after 72, 48, and 24 hours of fermentation, respectively, when the pH of the medium reached the value 5.0. PL from SSF and SmF were loaded on Sephadex-G75 columns and six activity peaks were obtained from crude enzyme fromSSF and designated PL I, II, III, IV, V, andVI, while five peaks were obtained fromcrude enzyme fromSmF and labeled PL I', II', III', IV', and VII'. Crude enzyme and fraction III from each fermentative process were tested further. The optimum pH for crude PL from either process was 5.5, while that for PL III was 8.0. The maximum activity of enzymes from SSF was observed at 35°C, but crude enzyme was more thermotolerant than PL III, maintaining its maximum activity up to 45°C. Crude enzyme from SmF and PL III' showed thermophilic profiles of activity, with maximum activity at 60 and 55°C, respectively. In the absence of substrate, the crude enzyme from SSF was stable over the pH range 3.0-10.0 and PL III was most stable in the pH range 4.0-7.0. Crude enzyme from SmF retained 70%-80% of its maximum activity in the acid-neutral pH range (4.0-7.0), but PIII showed high stability at alkaline pH (7.5-9.5). PL from SSF was more thermolabile than that from SmF. The latter maintained 60% of its initial activity after 1 h at 55°C. Cyclodextrin glycosyltransferase (CGTase) is an enzyme that produces cyclodextrins from starch by an intramolecular transglycosylation reaction. Cyclodextrins have been shown to have a number of applications in the food, cosmetic, pharmaceutical, and chemical industries. In the current study, the production of CGTase by Paenibacillus campinasensis strain H69-3 was examined in submerged and solid-state fermentations. P. campinasensis strain H69-3 was isolated from the soil, which grows at 45 C, and is a Gram-variable bacterium. Different substrate sources such as wheat bran, soybean bran, soybean extract, cassava solid residue, cassava starch, corn starch, and other combinations were used in the enzyme production. CGTase activity was highest in submerged fermentations with the greatest production observed at 48-72 h. The physical and chemical properties of CGTase were determined from the crude enzyme produced from submerged fermentations. The optimum temperature was found to be 70-75 degrees C, and the activity was stable at 55 degrees C for 1 h. The enzyme displayed two optimum pH values, 5.5 and 9.0 and was found to be stable between a pH of 4.5 and 11.0.

Aim: The present study was developed in a deforested stream located in a region that exhibits marked seasonality with the purpose to investigate whether ecological descriptors of the quantitative structure (i.e., composition, abundance, biomass, species richness, diversity) and feeding of fishes do change between the dry and wet periods. Methods: Sampling was conducted bimonthly from April 2004 to February 2005 by using a standardized effort with electrofishing equipment and environmental variables measurements. Results: We collected 713 fishes belonging to 23 species. The most abundant species were Gymnotus carapo (24.0%) and Poecilia reticulata (23.8%). Species richness, abundance, and biomass showed to be higher in the wet period, but these differences were not significant and did not influence the multivariate pattern of the assemblage (ANOSIM, R = 0.148). Nevertheless, average dissimilarity between community structure in the dry and wet periods was 52.7%, mainly due to the differential contribution of P. reticulata, notably more abundant in the wet season, under quasi-hypoxic water conditions. Examination of 333 gastric contents of 12 species evidenced that food variety was higher in the dry period. of these species, 67% (Astyanax altiparanae, Astyanax fasciatus, Geophagus brasiliensis, Gymnotus carapo, Hypostomus ancistroides, Phalloceros harpagos, Poecilia reticulata, and Rhamdia quelen) kept the diet throughout the year, being classified in the same trophic groups in both periods, and detritus was the most important item for half of them, followed by aquatic insects. Overall, no significant differences in the community's diet between periods were registered (ANOSIM, R = [long dash]0.04). Conclusions: This relative constancy suggests a quite regular availability of resources (mainly shelters in submerged marginal grasses and detritus) along the year.

Lipase production by Trichoderma harzianum was evaluated in submerged fermentation (SF) and solid-state fermentation (SSF) using a variety of agro-industrial residues. Cultures in SF showed the highest activity (1.4 U/mL) in medium containing 0.5 % (w/v) yeast extract, 1 % (v/v) olive oil and 2.5 C:N ratio. This paper is the first to report lipase production by T. harzianum in SSF. A 1:2 mixture of castor oil cake and sugarcane bagasse supplemented with 1 % (v/w) olive oil showed the best results among the cultures in SSF (4 U/g ds). Lipolytic activity was stable in a slightly acidic to neutral pH, maintaining 50 % activity after 30 min at 50 C. Eighty percent of the activity remained after 1 h in 25 % (v/v) methanol, ethanol, isopropanol or acetone. Activity was observed with vegetable oils (olive, soybean, corn and sunflower) and long-chain triacylglycerols (triolein), confirming the presence of a true lipase. The results of this study are promising because they demonstrate an enzyme with interesting properties for application in catalysis produced by fermentation at low cost. © 2012 Springer-Verlag and the University of Milan.

Amylases from Rhizopus oryzae and Rhizopus microsporus var. oligosporus were obtained using agro-industrial wastes as substrates in submerged batch cultures. The enzymatic complex was partially characterised for use in the production of glucose syrup. Type II wheat flour proved better than cassava bagasse as sole carbon source for amylase production. The optimum fermentation condition for both microorganisms was 96 hours at 30°C and the amylase thus produced was used for starch hydrolysis. The product of the enzymatic hydrolysis indicated that the enzyme obtained was glucoamylase, only glucose as final product was attained for both microorganisms. R. oligosporus was of greater interest than R. oryzae for amylase production, taking into account enzyme activity, cultivation time, thermal stability and pH range. Glucose syrup was produced using concentrated enzyme and 100 g L-1 starch in a 4 hours reaction at 50°C. The bioprocess studied can contribute to fungus glucoamylase production and application. © 2013 Institute of Chemistry, Slovak Academy of Sciences.

O presente trabalho teve por objetivo estudar três espaçamentos entre fileiras (30, 40 e 50 cm) e três densidades de semeadura (100, 150 e 200 sementes viáveis/m2) quanto a fenologia da planta, os componentes da produção e a produtividade de grãos do arroz de sequeiro cv. IAC 201. Desta forma, foi instalado um experimento em condições de campo, em um Latossolo Vermelho escuro, epi-eutrófico, textura argilosa, em Selvíria, MS. A redução do espaçamento entre fileiras aumentou o número de colmos e de panículas. O aumento da densidade de semeadura reduziu o perfilhamento por planta. A densidade de 100 sementes/m2 é mais adequada para o cultivar IAC 201. em cultivo de sequeiro sob alta precipitação e boa distribuição pluvial, o espaçamento de 30 cm entre fileiras de plantas, proporcionou maior produtividade de grãos do cultivar IAC 201 de arroz.
