Magnetic images of the disintegration process of tablets in the human stomach by ac biosusceptometry

Oral administration of solid dosage forms is usually preferred in drug therapy. Conventional imaging methods are essential tools to investigate the in vivo performance of these formulations. The non-invasive technique of ac biosusceptometry has been introduced as an alternative in studies focusing on gastrointestinal motility and, more recently, to evaluate the behaviour of magnetic tablets in vivo. The aim of this work was to employ a multisensor ac biosusceptometer system to obtain magnetic images of disintegration of tablets in vitro and in the human stomach. The results showed that the transition between the magnetic marker and the magnetic tracer characterized the onset of disintegration (t(50)) and occurred in a short time interval (1.1 +/- 0.4 min). The multisensor ac biosusceptometer was reliable to monitor and analyse the in vivo performance of magnetic tablets showing accuracy to quantify disintegration through the magnetic images and to characterize the profile of this process.

The mechanical nature of gastric contraction activity (GCA) plays an important role in gastrointestinal motility. The aim of this study was to detect GCA in anaesthetized dogs, using simultaneously the techniques of AC biosusceptometry (ACB) and manometry, analysing the characteristics of frequency and amplitude (motility index) of GCA, modified by drugs such as prostigmine and N-butyl-scopolamine. The ACB method is based on a differential transformer of magnetic flux and the magnetic tracer works as a changeable external nucleus. This magnetic tracer causes a modification in the magnetic flux, which is detected by the coils. The results obtained from the ACB showed a performance comparable to the manometry in measuring the modifications in the frequency and amplitude of the GCA. We concluded that this ACB technique, non-invasive and free of ionizing radiation, is an option for evaluating GCA and can be employed in future clinical studies.

Conventionally, pharmaceutical substances are administered orally because the gastrointestinal tract possesses the appropriate features for drug absorption. Nevertheless, the gastrointestinal tract physiology is complex and influenced by many factors. These factors must be completely understood for the optimization of oral drug delivery systems. Although in vitro tests provide information about release and drug absorption profiles, in vivo studies are essential, due to the biological variability. Several techniques have been employed in an attempt to conveniently characterize the behavior of solid dosage forms in vivo. The noninvasive biomagnetic technique of alternate current biosusceptometry (ACB) has been used in studies focusing on gastrointestinal motility and, more recently, to evaluate the performance of magnetic dosage forms. This article will discuss the main characteristics of AC biosusceptometry and its applicability for determination of the relationship between the human gastrointestinal tract and orally administered pharmaceutical dosage forms. Irritable bowel syndrome is a high prevalent functional disorder. Its diagnosis is based in symptoms and the Rome Criteria. The treatment takes in account psychosocial and biologic factors. The therapeutic measures aim to correct the predominant symptoms: constipation, diarrhea and abdominal pain.

Objective - To investigate the effects of buprenorphine on cardiopulmonary variables and on abdominal auscultation scores in horses. Animals - 6 healthy adult horses. Procedures - Horses were restrained in stocks and allocated to 2 treatments in a randomized crossover design, with 1-week intervals between each treatment. Saline (0.9% NaCl) solution was administered IV as a control, whereas buprenorphine (10 μg/kg, IV) was administered to the experimental group. Cardiopulmonary data were collected for 120 minutes after buprenorphine or saline solution administration. Abdominal auscultation scores were monitored for 2 and 12 hours after drug administration in the control and experimental groups, respectively. Results - Following control treatment, horses remained calm while restrained in the stocks and no significant changes in cardiopulmonary variables were observed throughout the study. Buprenorphine administration caused excitatory phenomena (restlessness and head shaking). Heart rate, cardiac index, and arterial blood pressure were significantly increased after buprenorphine administration until the end of the observational period (120 minutes). Minimal changes were found in arterial blood gas tensions. Abdominal auscultation scores decreased significantly from baseline for 4 hours after buprenorphine administration. Conclusions and Clinical Relevance - Buprenorphine induced excitement and hemodynamic stimulation with minimal changes in arterial blood gas tensions. These effects may impact the clinical use of buprenorphine in horses. Further studies are indicated to investigate the effects of buprenorphine on gastrointestinal motility and fecal output.

Several hormonal and physiological changes that occur during pregnancy directly affect gastrointestinal motility (GI), however, very little is known about the relationship between the electrical and mechanical activities of the stomach and its implications. The aim of this study was to compare the female sex hormones, with profiles of gastrointestinal motility using AC biosusceptometry (BAC) in pregnant rats. Seven female Wistar rats (weighing 250-350 g) were used and all procedures were approved by the local Ethics Committee of animal. Laparotomy was performed on anesthetized rats and a magnetic marker (3.5 mm in diameter and 3.0 mm height) was implanted in the distal stomach. After 10 days of recuperation, the mice were put to mate until confirmation of pregnancy. Fed animals were anesthetized (mg pentobarbital 30 / kg) and kept supine for recording the signal with a sensor 30 min BAC placed on the abdominal surface on days 0, 7, 14 and 20 of gestation. Statistical analysis was performed by Student's t test statistical Newman-Keuls and a P value of .05 was considered statistically significant. The frequency of contraction of the stomach was 74 mHz ± 3, 67 ± 7 mHz, 59 mHz ± 6 (p <0.05 vs day -1) and 71 ± 5 mHz on days -0, -7, -14, and -20, respectively. The values of 44 ± 15 pg / mL and 31 ± 9 pg / ml 25 ± 6 pg / ml, 22 ± 9 pg / ml on days -0, -7, -14 and -20, respectively, obtained by 17ß- estradiol were not statistically significant. The values of progesterone were 13 ± 6 pg / ml at 54 ± 15 pg / ml, 127 ± 42 pg / ml (p <0.05 vs day-1), 22 ± 13 pg / mL on day -0, - 7, -14 and -20, respectively. A linear correlation (Pearson's test) between the lower frequency gastric and increased levels of progesterone was 80% (p <0.0000001).
