Further to increase the wood yield, is important goal of forest tree breeding the adaptation of clones through diverse environment, especially for soil and climate. Perennial tree species such as eucalypts, have a long life cycle and the frosts can occur and to be their limiting source of cultivation. The aim of the study was to determine the genetic variability and the genetic correlations between selection ages of clones cultivated in the municipality of Palma So la, SC, Brazil, where frosts are common in the winter. A clonal trial was set up in 2008 in Palma So la, by statistical design of randomized complete blocks considering 29 clones, six replications, six plants per plot, and 3.0m x 3.0 m spacing. The silvicultural characters of total plant height, diameter of breast height (dbh), and wood volume were evaluated through 24, 36, 48, and 60 months old. The deviance analysis and estimates of genetic parameters were based on the REML / BLUP genetic statistical procedure. Significant differences were observed for all traits in the deviance analysis. High correlations and statistically significant between characters were observed, indicating that the early selection will provide significant gains. Part of the clones USP/IPEF 64, USP/IPEF 78, USP/IPEF 52, USP/IPEF 68, and USP/IPEF 74) are potential to be used into the breeding programs and in commercial stands into the studied frost regions.

The Coffee Genome Project made available to the scientific community relevant information that made practical the identification and cloning of important genes, as well as the identification of the major sequences involved on their regulation. The aim of the present study was to amplify, clone and sequence coffee promoters with specific expression patterns. For that, coffee ESTs which known expression profiles were employed. First, the promoter regions of coffee genes showing, respectively, fruitspecific and ubiquitous expression were amplified using the Genome Walking strategy. Amplified sequences were then inserted in the pGEM-Teasy vector (Promega) and sequenced. Once completed the sequencing, an expression cassette was constructed using the binary vector pCAMBIA-1381z (Cambia). These expression cassettes were cloned into Agrobacterium tumefaciens, and transgenic tobacco plants were generated aiming the functional characterization of these promoters

Staphylococcus aureus é um patógeno ubíquo capaz de causar uma variedade de infecções em humanos. A resistência desse microrganismo a antibióticos como oxacilina e meticilina é um problema sério, de crescimento significativo para a terapêutica antimicrobiana clínica em pacientes acometidos por infecções estafilocócicas. A resistência à meticilina em S. aureus é decorrente da alteração do sítio de ação dos antibióticos β-lactâmicos, os quais agem através da inibição de enzimas que catalisam a síntese da parede celular. Essas enzimas são o sítio de ação das penicilinas, e por isso, passaram a ser chamadas de proteínas ligadoras de penicilinas (PBPs). O gene mecA codifica a PBP2a, que substitui a função das outras PBPs neste patógeno e confere resistência a β-lactâmicos. A PBP2a exibe uma afinidade reduzida pelo anel β-lactâmico e permite que a bactéria continue a sintetizar a parede bacteriana. Este gene faz parte de um elemento genético móvel encontrado em isolados de MRSA, designado cassete cromossômico estafilocócico mec (SCCmec), integrado ao cromossomo de S. aureus. O objetivo do estudo foi a caracterização molecular de 139 isolados de S.aureus provenientes de pacientes pediátricos com bacteremia durante o período de 1991 a 2010. Métodos moleculares foram utilizados para a determinação do perfil genético das amostras, incluindo, identificação do tipo de SCCmec, detecção do gene codificador de leucocidina de panton valentine (PVL) e similaridade clonal em gel de eletroforese em campo pulsado (PFGE). O gene mecA foi detectado em 32 (23%) amostras e houve predomínio do SCCmec IV (68,8%) em relação ao SCCmec III (31,2%). A presença de PVL foi encontrada em 18 amostras (12,9%), todas sensíveis à oxacilina. O clone epidêmico brasileiro, relacionado ao SCCmec tipo III, esteve presente na unidade neonatal do hospital... Sucrose esters are biodegradable, non pollutant and safe for health; moreover, they have shown great potential in the pest control. We tested a crude mix of sucrose esters on the survivorship and oviposition of Calacarus heveae Feres (Acari: Eriophyidae) females, an important rubber tree pest mite. The females were collected from rubber leaflets of the GT 1 and RRIM 600 clones. The mites were kept on stock arenas placed in rearing chamber at 28 + 0,1°C, 80 + 10% of relative humidity and 12h of light phase about two weeks before the assays. After this period, the mites were transferred to assay arenas made with leaflets from the same clone of stock arenas. The survivorship of females sprayed with sucrose esters in the concentrations of 1, 2, 3 and 4 g/L and their oviposition effect with 0.5 and 1 g/L were analyzed. The mites sprayed with 4 g/L had about 80% of mortality, while those with 1 g/L around 60%. All mites that died in the treatments with sucrose ester became dark with wrinkled tegument and decreased their body volume, suggesting dehydration. We also observed the efficacy of 1 g/L concentration to decrease the female oviposition in about 50%, in the third day after spraying. No differences were observed in the mortality and oviposition between females kept on both clones.

The lastyears declined the discovery of compounds to use in industrial and naturaldiversity has been the best supplier for novel genes, enzymes and compounds inhigh demand by the biotechnology industry. We know immense diversity of microorganisms,yet most remains unexplored. For these reason we use the metagenômica approach toinvestigate the potential of uncultured microorganisms. With this purpose weused the metagenomic library of from Eucalyptus spp. arboretum (EAA), wedid screening to found positive clone and them was submitted to the process of shotgun,the data obtained was submitted a bioinformatics analyses. Our results showsthe hypothesis of high unexplored microbial diversity of soil are able to foundnovel genes and metagenomic approach is and allowed to isolate novel genes and insilico analyses are essential part to identify a novel Inorganicpyrophosphatase (PPase) prediction indicated the novel gene operate as H+ pumps. Thissuggests that a special feature, our work in situ will be cloning thegene expression vector for subsequent kinetic characterization and crystallization.
