Apocynin, a methoxy-catechol originally extracted from the root of Picrorhiza kurroa, has been used as an inhibitor of the NADPH oxidase complex in phagocytic and nonphagocytic cells. Its mechanism of inhibition is linked to their prior activation through the action of peroxidases leading to oxidation of the dimeric product, diapocynin. In this study, dipocinina was synthesized and investigated its effect as an inhibitor of activation NADPH oxidase in neutrophils (PMN) and peripheral blood mononuclear cells (PBMC). The synthesis of diapocinina was performed by oxidation of apocinina by potassium persulphate in the midst of water for 5 minutes at room temperature. The precipitate was filtered and washed with water and methanol. Diapocinina was characterized by mass spectrometry. PMN and PBMC were obtained from peripheral blood of healthy donors and purified for gelatin sedimentation, or centrifugation with Histopaque ®, the red cells were lysed with ice water or ammonium chloride. Diapocinina or apocinina were incubated with opsonized zymosan, activation of PMNs and release of superoxide anion, these monitored by chemiluminescent assay dependent lucigenina. We found that diapocinina inhibitor was no better than the apocinina in PMN. However, diapocinina was more efficient than apocinina as an inhibitor of NADPH oxidase in PBMC. In conclusion, whereas PBMC are relatively poor compared with peroxidases PMN, our results are consistent with the need for oxidation apocinina for its effect as an inhibitor of NADPH oxidase

O veneno bruto de abelha Apis mellifera é composto por uma série de peptídeos, entre eles a melitina, adolapina, procamina A e B, apamina, tertiapina, secapina, peptídeos desgranuladores de mastócitos, entre outros. Estas substâncias apresentam diversos efeitos já estudadas, dentre elas, efeitos antitumorais, atividades hiperalgésicas ou analgésicas, inflamatórias ou anti-inflamatórias. O peptídeo secapina foi descrito há 35 anos atrás e, posteriormente nada mais foi relatado na literatura. Portanto, este é o primeiro trabalho que visa caracterizar o possível efeito hiperalgésico e/ou analgésico, inflamatório e/ou antiinflamatório, bem como analisar alguns dos mecanismos envolvidos nestes fenômenos. Para tanto, para caracterizar os possíveis efeitos hiperalgésicos/analgésicos da secapina, em diversas doses (0,005, 0,35, 1, 2, 10 e 30 μg / 50 μL), foi utilizado o teste de avaliação da sensibilidade dolorosa denominado teste do von Frey eletrônico. Para a caracterização do efeito inflamatório/anti-inflamatório da secapina foi utilizado o paquímetro digital determinando o volume da pata dos animais. Para investigar os possíveis mecanismos envolvidos nestes efeitos, foram realizados tratamentos farmacológicos utilizando indometacina, zileuton e zafirlikaste (inibidores da síntese da via da cicloxigenase, lipoxigenase e antagonista de receptor de leucotrieno, respectivamente). Os resultados mostraram que a secapina, em todas as doses testadas, induziu efeito hiperalgésico a partir dos primeiros 15 minutos após a administração deste peptídeo. Este efeito hiperalgésico persistiu por até 240 minutos nas doses de 0,005 e 0,35 μg/ 50μL e 480 minutos nas doses de 1 e 2 μg/ 50μL. O efeito edematogênico da secapina na dose de 0,35 μg/50 μL foi iniciado nos primeiros 15 minutos após a administração da secapina e persistiu por até 120 minutos, sendo... O remodelamento da matriz extracelular (MEC), um importante componente dos organismos multicelulares, relaciona-se diretamente com o desenvolvimento embrionário, a angiogênese, a morfogênese de órgãos e a formação de cartilagens. Especificamente nos ovários, este remodelamento permite a ocorrência dos diversos eventos observados no ciclo ovariano, a citar: o crescimento folicular, a ovulação e a formação e regressão do corpo lúteo. A reorganização tecidual advinda destes eventos é regulada, em parte, pela ação de enzimas proteolíticas conhecidas como metaloproteinases de matriz (MMPs) e de seus inibidores (TIMPs e RECK) (revisado por Curry et al., 2003). Atualmente, o remodelamento da MEC pelo sistema MMPs/TIMPs é fortemente correlacionado com a expressão de alguns genes, tais como o gene Basigin (BSG), que induz a expressão de MMPs durante o ciclo ovariano em ratos (Smedts et al., 2005), ou o gene SPARC, que modula a expressão do TGF-β, fator de crescimento este associado ao aumento da agressividade tumoral (Podhajcer et al., 2008). A expressão do gene RECK foi descrita em diversos processos de remodelamento tecidual fisiológicos. Porém, uma diminuição em sua expressão, juntamente com o aumento na expressão das MMPs, tem sido associada com tumores mais agressivos e metastáticos (Meng et al., 2008). No entanto, apesar de RECK e SPARC estarem amplamente associados com remodelamento tecidual em diversas patologias, ainda não foi descrita a associação destes com remodelamento tecidual promovido pelo sistema MMPs/TIMPs que ocorre naturalmente durante a dinâmica ovariana. Considerando este quadro, o trabalho aqui proposto tem por objetivo analisar a expressão gênica de algumas MMPs- (-2, -9, -13, -14 e -19), e alguns de seus inibidores (TIMPs -1, -2, e -3 e RECK) e correlacioná-la com a expressão de SPARC e BSG durante a foliculogênese ovariana de ratas... Aortic regurgitation (AR) leads to a left ventricle dilation and hypertrophy in response to a chronic volume overload. It is still very frequent in developing countries, for instance Brazil, and often as secondary to rheumatic fever. Usually, chronic AR is generally well tolerated for many years, when with the heart dilated the patient searches for treatment. Bidirectional association with depression and cardiovascular disease has been described. Selective serotonin reuptake inhibitors (SSRI) are widely prescribed to treat several affective disorders, especially for cardiovascular patients since they decrease arrhythmia probability. These SSRI improves cardiac function in rats submitted to stress protocols. Preliminary study from our laboratory showed that following 4 weeks of treatment with one SSRI (paroxetine) in subchronic AR rats there was a decreased in daily sodium intake and an improvement in systolic function. An increase in the central oxytocinergic transmission may be involved in this peripheral improvement to the heart. The investigations about the mechanisms underlying this improvement are necessary. Therefore the aims of this project is investigate the effects of 4 weeks of treatment of paroxetine, a SSRI, in rats with a subchronic AR over the central central gene expression of oxytocin and vasopressin using a reverse transcription polymerase chain reaction (RT-PCR)

Apocynin has been used as an efficient inhibitor of the multi-enzymatic complex NADPH oxidase in many experimental models involving phagocytic and nonphagocytic cells. The mechanism of inhibition has been linked with the previous activation of apocynin through the action of cellular peroxidases leading to the formation of a dimeric oxidation product, diapocynin. In this study we compared apocynin with pure diapocynin regarding their effects as scavenger of hydrogen peroxide and hypochlorous generated by glucose/glucose oxidase and myeloperoxidase respectively, and as inhibitors of the production of hydrogen peroxide and hypochlorous acid by activated neutrophils. The production of hydrogen peroxide was measured by the oxidation of the fluorescent substance Amplex Red and the production of hypochlorous acid by was measured as taurine-chloramine derivative using the chromogenic substrate 3,3’,5,5’- tetramethylbenzidine (TMB). Neutrophils (1 x106 cells/mL) were pre-incubated in PBS buffer supplemented with 1 mM calcium chloride, 0.5 mM magnesium chloride, 1 mg/mL glucose and 5 mM taurine in the presence or absence of inhibitors. The reactions were triggered by adding the soluble stimulus Forbol Miristate Acetate PMA or zymosan and incubated by additional 30 minutes. We found that pure diapocynin was not better than apocynin regarding its scavenger and inhibitory properties. These results suggest that the formation of diapocynin is not essential for the action of apocynin as inhibitor of NADPH oxidase activation

Aortic regurgitation (AR) is still common in developing countries as secondary of rheumatic fever, and its incidence have increased in senile degenerative form. The AR develops severe myocardial hypertrophy. A common comorbidity associated with cardiovascular disease is depression. Among the most prescribed antidepressants in the world are the serotonin selective reuptake inhibitors (SSRIs). Central serotonergic pathways are involved with the inhibition of sodium intake and can be modify the excretion of this íon. Therefore, we investigated whether treatment with an antidepressants SSRI, the paroxetine, for four weeks can modfy the behavior of water and NaCl 0,3M intake, excretion of sodium and morphofunctional parameters of rats with AR induce. Wistar rats (280 - 300g) underwent surgery for AR (n=15) or control surgery (n=14). The AR was induced by retrograde puncture of valve leaflets. The animals were divided into 4 groups: AR + paroxetine (n=8), AR + control (n=7), control + paroxetine (n=7), control + control (n=7). From the 4th to the 8th week after inductuin of AR was administered paroxetine (10mg/kg pc) daily and subcutaneously. In the 4th and 8th week after induction of AR echocardiograms were performed to collect data morphofunctional. During the 4 weeks of treatment were analyzed intake of water and saline daily and once a week urine samples were collected for analysis by flame photometer of excretion of sodium and potassium. In the 10th week the animals were submitted to a challenge protocol hidromineral by combining furosemide (10 mg / kg bw) associated with the low dose of Captopril (5 mg / kg bw). During the challenged urine samples were collected for analysis by flame photometer of excretion of sodium and potassium at the time zero and after 2 hours of treatment.
