The objective of this paper was to study the aqueous extract of Bauhinia forficata L. (pata-de-vaca) effects on streptozotocin-induced diabete pregnant rats. We used Wistar pregnant rats, dividided in 3 experimental groups: control (C, n=7), non-treated diabetics (DNT, n=7) and streptozotocin induced-diabetics treated with an aqueous extract of pata-de-vaca leaves, 200 mg/kg dose (DT, n=7). The animals received the extract through a gastric tube (gavage). The blood glucose level were verified on day 0, 5, 14 and 20 of pregnancy. During pregnancy, the daily mean water intake, food intake and average maternal weight gains of rats were measured. The results demonstrated that plant extract reduced the postimplantation loss porcentage, increasing the number of live fetuses likely to the control group. We found increased food and water intake of the DT and DNT pregnant rats compared to control due to hyperglycemic state. We also observed average maternal weight gains was likely to the DT and control groups on different pregnant periods, suggesting treatment with the plant contributed for the rat weight gains. The blood glucose level of dams did not present significative differences between DT and DNT groups. Thus, the B. forficata aqueous extract, 200 mg/kg dose, did not present hypoglycemic effect on streptozotocin-induced diabete pregnant rats. Nevertheless, the results suggest that DT pregnant rats were kept safe the for B. forficata aqueous extract, allowing at term pregnant occurence.

Cobb male broiler chicks (1,000) on new litter were used to evaluate effects of dietary electrolyte balance [DEB; Na+K-Cl, milliequivalents (mEq) per kilogram] under tropical summer conditions. Corn-soybean meal-based mash diets had salt (NaCl) alone or in combination with one or more supplements: sodium bicarbonate (NaHCO3), ammonium chloride (NH4Cl), or potassium bicarbonate (KHCO3). A completely randomized design, with five starter and grower feed treatments (control: 145, then 130 mEq/kg; or 0, 120, 240, or 360 mEq/kg throughout) and four replicate pens (1.5 x 3.2 m) per treatment (50 chicks per pen), was used. Diets were analyzed for Na, K, and Cl for confirmation. There were no significant (P < 0.05) effects of treatments on mortality or processing parameters. Water intake increased linearly with increasing DEB, giving higher litter moistures and lower rectal temperatures. Blood HCO3 and pH increased with the highest DEB (360 mEq/kg) causing respiratory alkalosis. The DEB of 240 mEg/kg gave best weight gain and feed conversion ratio, and ideal DEB predicted by regression analyses were 186 and 197 mEq/kg from 0 to 21 d of age and 236 and 207 mEq/kg of feed from 0 to 42 d, respectively. These DEB corresponded to estimated (interpolated) values in predicted optimal 186 to 197 mEq/kg starter of Na 0.38 to 0.40% and Cl 0.405 to 0.39% (K = 0.52%), in 207 to 236 mEq/kg starter, Na 0.409 to 0.445% and Cl 0.326 to 0.372% Cl (K = 0.52%), and in grower Na 0.41 to 0.445%, Cl 0.315 to 0.267% (K = 0.47%).

Individually caged male Cobb broilers (24), 44 d of age, were used to evaluate effects of heat stress (1 d of data collection) and dietary electrolyte balance (DEB; Na + K - Cl, mEq/kg from 1 d of age). During summer rearing, mortality was variable, but DEB 240 improved growth, feed conversion ratio, water intake, and waterrfeed ratio vs. DEB 0. The temperature sequence for heat stress was 24 to 32°C in 30 min, 32 to 36°C in 30 min, 36 to 37°C in 15 min, and 37 to 41°C in 45 min. Maximum temperature was held for 15, 60, 90, or 360 min for data collection (relative humidity averaged 42 ± 7%). Results from the same room before and after heat stress were analyzed by DEB (1-factor ANOVA) and before vs. after heat stress compared across DEB (2-sample t-test). Heat stress decreased blood Na, K, and pCO2, and lymphocytes but increased heterophils. Blood HCO3 rose, Cl declined, and hematocrit gave a concave pattern (lowest at DEB 120) as DEB increased. After heat stress, DEB O decreased blood Na and K, and DEB O and 120 levels decreased blood HCO3. After heat stress blood pCO2 and hemoglobin decreased with DEB 240, but it had highest pCO2, a key factor. The DEB 120 gave longest times to panting and prostration with DEB O and 240 results lower but similar statistically. In heat stress, DEB 360 was excessive, DEB 120 and 240 were favorable, and DEB 0 was intermediate based on hematology, panting, and prostration responses.

The aim of this study was to examine the role of nifedipine and Nitric Oxide (NO) on salivary flow and compounds (salivary amylase, saliva total proteins, saliva calcium, sodium and potassium). Male Holtzman rats weighting 200-250 g were anesthetized with zoletil 50 mg kg -1 (tiletamine chloridrate 125.0 mg and zolazepan chloridrate 125.0 mg) into quadriceps muscle and stainless steel cannulas were implanted into their lateral ventricle of the brain (LV). Animals in divided group were injected with nifedipine (50 μg μL -1) alone and in combination with 7-nitroindazol (7-NIT) (40 μg μL -1), neuronal NO Sinthase Inhibitor (nNOSI) and Sodium Nitroprussate (SNP) (30 μg μL -1) NO donor agent. As a secretory stimuli, pilocarpine dissolved in isotonic was administered intraperitoneally (ip) at a dosage of 10 mg kg -1 body weight. Saliva was collected for 7 min with four cotton balls weighing approximately 20 mg each, two of which were placed on either side of the oral cavity, with the other two placed under the tongue. Nifedipine treatment induced a reduction in saliva secretion rate and concentration of amylase, total protein and calcium without changes in sodium and potassium concentration in comparison with controls. Co-treatment of animals with nifedipine and SNP retained flow rate and concentration of amylase, total protein and calcium in normal levels. Co-treatment of animals with nifedipine and 7-NIT potentiated the effect of nifedipine on the reduction of saliva secretion and concentrations of amylase, total protein and calcium. Nifedipine (dihydroperidine) calcium-channel blocker widely in use is associated with salivary dysfunction acting in the central nervous system structures. NO might be the mechanism for protective effect against the nifedipine-induce salivary dysfunction, acting in the CNS. © 2006 Asian Network for Scientific Information.

Polymnia sonchifolia is a medicinal plant used as hypoglycemic agent and other ends. The aim was to evaluate the yacon effect on diabetes in Wistar female rats, which was divided in 4 groups: non-diabetic (n=24) given (gavage) 32.4 mg kg -1 of an aqueous extract of plant (treated group) or vehicle (non-treated group), and one diabetic group (n=24) with similar treatment compared to non-diabetic rats. Diabetes was induced using streptozotocin (i.v. route, 40 mg kg -1). Glycemia was monitored after rodent adaptation period, 15 days after diabetes induction, and each 10 days after yacon/vehicle in early treatment. During experiment period, body weights, food and water intake were daily measured. The rats (non-diabetic or diabetic) given yacon extract treatment presented no significant difference related with glycemia, body weight gain, and food and water intake. Thus, further studies such increase of rat number in agreement with different glycemic levels and increase of the plant extract dose are necessary.

To address daily fluctuations in electricity demands, the quantities of water passing through the turbines of hydropower plants can vary significantly (up to fourfold) during a 24-h cycle. This study evaluates the effects of hourly variations in water discharges on the limnological conditions observed in two below-dam river stretches. The study reservoirs, Capivara and Taquaruçu, are the 9th and 10th reservoirs in a cascade of dams in the Paranapanema River in south-east Brazil. The reservoirs exhibit different trophic conditions, water retention times, thermal regimes and spillway positions. Capivara Reservoir is deeper, meso-eutrophic, with a high water retention time and hypolimnetic discharges (32 m) varying between 500 and 1400 m3 s-1. In contrast, Taquaruçu Reservoir is relatively shallow, oligo-mesotrophic, and has a low retention time, with water discharges varying between 500 and 2000 m3 s-1. Its turbine water intake zone also is more superficial (7 m). For two periods of the year, winter and summer, profiles of limnological measurements were developed in the lacustrine (above-dam site) zones of the reservoirs, as well as in the downstream river stretches (below-dam site). In both cases, the sampling was carried out at 4-h intervals over a complete nictemeral cycle. The results demonstrated that the reservoir operating regime (water discharge variations) promoted significant differences in the conditions of the river below the dams, especially for water velocity, turbidity, and nutrient and suspended solids concentrations. The reservoir physical characteristics, including depth, thermal stratification and outlet structure, are also key factors influencing the limnology and water quality at the below-dam sampling sites. In the case of Capivara Reservoir, for example, the low dissolved oxygen concentration (<5.0 mg L-1) in its bottom water layer was transferred to the downstream river stretches during the summer. These study results demonstrated that it is important to continue such investigations as a means of verifying whether or not these high-amplitude/low-frequency variations could negatively affect the downstream river biota. © 2009 The Authors. The effects of posthatch fasting on villi height and number, crypt depth and number of globet cells in duodenum, jejunum and ileum of broiler chicks from heavy and light eggs were compared. The 2×3×3 factorial design (egg weight: light and heavy eggs; treatments: with water and feed, with water, without water and feed; treatment duration: 24, 48 and 72 h) was used. The villi presented higher size in chicks from heavy than from light eggs. The fasting resulted in lower villi in duodenum (at 48 h), jejunum and ileum (at 72 h). The villi number increased in duodenum and jejunum of chicks from light eggs and only in jejunum of chicks from heavy eggs, but the increase was more accented in chicks from light ones. The fasting reduced the globet cells number in jejunum. Water intake avoided the fasting effects on villi height but had no effect on villi number. Chicks from heavy eggs fed with water and ration presented deeper crypts in all regions of the small intestine. The duodenum and ileum crypt depth of the chicks from heavy eggs reduced when they were submitted to fasting and when they were fed only with water. The results showed that chicks from light eggs were more affected than chicks from heavy eggs. The water intake partially avoided the fasting effects. © Asian Network for Scientific Information, 2011.

Aldosterone acting on the brain stimulates sodium appetite and sympathetic activity by mechanisms that are still not completely clear. In the present study, we investigated the effects of chronic infusion of aldosterone and acute injection of the mineralocorticoid receptor (MR) antagonist RU 28318 into the fourth ventricle (4th V) on sodium appetite. Male Wistar rats (280-350 g) with a stainless-steel cannula in either the 4th V or lateral ventricle (LV) were used. Daily intake of 0.3 M NaCl increased to 46 ± 15 and 130 ± 6 ml/24 h after 6 days of infusion of 10 and 100 ng/h of aldosterone into the 4th V (intake with vehicle infusion: 2 ± 1 ml/24 h). Water intake fell slightly and not consistently, and food intake was not affected by aldosterone. Sodium appetite induced by diuretic (furosemide) combined with 24 h of a low-sodium diet fell from 12 ± 1.7 ml/2 h to 5.6 ± 0.8 ml/2 h after injection of the MR antagonist RU 28318 (100 ng/2 μl) into the 4th V. RU 28318 also reduced the intake of 0.3 M NaCl induced by 9 days of a low-sodium diet from 9.5 ± 2.6 ml/2 h to 1.2 ± 0.6 ml/2 h. Infusion of 100 or 500 ng/h of aldosterone into the LV did not affect daily intake of 0.3 M NaCl. The results are functional evidence that aldosterone acting on MR in the hindbrain activates a powerful mechanism involved in the control of sodium appetite. © 2013 the American Physiological Society.

The objective of this present experiment was to determine intake and apparent digestibility of dry matter and nutrients, nitrogen (NB) and water balances (WB) of sheep fed with buffel grass silages. The buffel grass cultivars used were Tanzania, Buchuma and Biloela. 21 male, castrated, crossbred Santa Ines x Non defined genotype sheep, with initial body weight of 31.8±3.16kg were used, kept in metabolic cages. The experimental design was completely randomized with seven replicates. No differences were observed in dry matter intake in g/day (919.2) and % of body weight (2.9). In general, digestibility coefficients of dry matter varied from 37.7% to 60.0%. The NB and WB were positive, with higher NB values in sheep fed with Tanzania (5.1g/day) and Biloela (3.9g/day) and higher WB in animals fed with Buchuma (1.38kg/animal/day) and Biloela (1.42kg/animal/day). All cultivars of buffel grass evaluated (Tanzania, Buchuma and Biloela) presented adequate chemical composition, where the silage of buffel grass used corresponded to 60% of the total daily water intake by sheep.

The nucleus of the solitary tract (NTS) is the primary site of visceral afferents to the central nervous system. In the present study, we investigated the effects of lesions in the commissural portion of the NTS (commNTS) on the activity of vasopressinergic neurons in the hypothalamic paraventricular (PVN) and supraoptic (SON) nuclei, plasma vasopressin, arterial pressure, water intake, and sodium excretion in rats with plasma hyperosmolality produced by intragastric 2 M NaCl (2 ml/rat). Male Holtzman rats with 15-20 days of sham or electrolytic lesion (1 mA; 10 s) of the commNTS were used. CommNTS lesions enhanced a 2 M NaCl intragastrically induced increase in the number of vasopressinergic neurons expressing c-Fos in the PVN (28 ± 1, vs. sham: 22 ± 2 c-Fos/AVP cells) and SON (26 ± 4, vs. sham: 11 ± 1 c-Fos/AVP cells), plasma vasopressin levels (21 ± 8, vs. sham: 6.6 ± 1.3 pg/ml), pressor responses (25 ± 7 mmHg, vs. sham: 7 ± 2 mmHg), water intake (17.5 ± 0.8, vs. sham: 11.2 ± 1.8 ml/2 h), and natriuresis (4.9 ± 0.8, vs. sham: 1.4 ± 0.3 meq/1 h). The pretreatment with vasopressin antagonist abolished the pressor response to intragastric 2 M NaCl in commNTS-lesioned rats (8 ± 2.4 mmHg at 10 min), suggesting that this response is dependent on vasopressin secretion. The results suggest that inhibitory mechanisms dependent on commNTS act to limit or counterbalance behavioral, hormonal, cardiovascular, and renal responses to an acute increase in plasma osmolality. © 2013 the American Physiological Society.
