Milk serum proteins such as alpha-lactalbumin (ALA) and beta-lactoglobulin (BLG) present biochemical polymorphism which is under the control of codominant autosomal alleles. In the present report, we propose modifications of traditional electrophoretic techniques such as increasing the running gel concentration from 5 to 10% and the addition of 5 M urea to the stacking gel, which permitted the detection of two variants (A and B) at the ALA and BLG loci. About 8 mul of milk serum (6 mg/ml protein) and 10 pl of total fresh milk were applied. Bovine serum albumin (BSA) and immunolactoglobulins (ILG) could also be discriminated. Total fresh milk was as useful as the purified serum milk proteins for the discrimination of ALA and BLG serum milk protein polymorphism by alkaline vertical slab polyacrylamide gel electrophoresis. However, BSA and ILG ran with caseins, which prevented their characterization in this system.

The present study was carried out with the objective of evaluating the effects of feeding dairy cows with organic or inorganic sources of zinc (Zn), copper (Cu) and selenium (Se) on blood concentrations of these minerals, blood metabolic profiles, nutrient intake and milk yield and composition. Nineteen Holstein cows were selected and randomly assigned to two groups for receiving organic (n = 9) or inorganic (n = 10) sources of Zn, Cu and Se from 60 days before the expected date of calving to 80 days of lactation. Samples of feed, orts and milk were collected for analysis. Body condition score (BCS) was determined and blood samples were collected for analysis of Zn, Cu and Se concentrations, as well as for metabolic profile. Supplying organic or inorganic sources of Zn, Cu, and Se did not affect dry matter and nutrient intake, blood metabolic profile, milk yield and composition, plasma concentration of these minerals, and BCS or change the BCS in cows from 60 days before the expected date of calving to 80 days of lactation. An effect of time was observed on all feed intake variables, plasma concentrations of Zn and Se, milk yield, milk protein content, BCS and change in BCS.

The objective was to evaluate the effect of beta-lactoglobulin (beta-lg) polymorphism and seasonality on milk composition (fat, lactose, total solids, milk urea nitrogen, total protein, true protein, casein and somatic cell counts) of Holstein and Girolando cows. Milk and blood samples from 278 Holsteins cows and 156 Girolando cows were taken during two dry seasons and two rainy seasons, for milk composition analysis and to determine beta-lg genotypes, respectively. BB genotype was the most frequent for both breeds, followed by AA genotype for Holstein (BB>AA>AB) and by AB for Girolando cows (BB>AB>AA). No differences were found in milk compositional characteristics among genetic variants of beta-lg (AA, AB and BB) either between Holstein or Girolando cows. No association between milk composition and beta-lg genetic polymorphism was observed. During the dry season, independently of the breed considered, higher contents of lactose, true protein, casein and casein :true protein ratio were found.

Data concerning daily milk yield (MY), percentage of milk fat (%F), protein (%P), lactose (%LT), and total solids (%TS), and somatic cell counts (SCC) for a herd of 222 Murrah buffalo reared in the state of São Paulo, Brazil, were collected monthly from 1997 to 2000 in order to study the factors affecting SCC and their relation to milk production and constituents during lactation. SCC decreased in the second month of lactation and increased thereafter, up to the ninth month of lactation. The interaction of month of lactation x order of calving was significant. Mean MY observed during the first month of lactation was 6.87 kg, which increased to 7.65 kg during the second month, and then decreased until the ninth month of lactation (3.83 kg). During the different months of lactation, %F, %P, %LT, and %TS ranged from 6.28 to 8.38%, 4.05 to 4.59%, 4.96 to 5.34%, and 16.94 to 18.55%, respectively. Calving year, calving order, and order of month of lactation significantly affected MY, %F, %P, %LT, and %TS. The regression coefficients of transformed SCC on MY and %LT were negative and significant during all months of lactation, showing that milk and lactose yield decreased with increased transformed SCC, causing losses to buffalo milk producers.

Two hundred and eighteen Bacillus thuringiensis isolates from Brazil were characterized by the presence of crystal protein genes by PCR with primers specific to different cry and cyt genes. Among these isolates, 95 were selected according to their geographic origin for genetic characterization with the 16S rRNA gene, RAPD, and plasmid profile. Isolates containing cryl genes were the most abundant (48%) followed by the cry11 and cyt (7%) and cry8 genes (2%). Finally, 40.3% of the isolates did not produce any PCR product. The plasmid profile and RAPD analysis showed a remarkable diversity among the isolates of B. thuringiensis not observed in the 16S rRNA gene. These results suggest that the genetic diversity of B. thuringiensis species results from the influence of different ecological factors and spatial separation between strains generated by the conquest of different habitats.

The simultaneous existence of alternative oxidases and uncoupling proteins in plants has raised the question as to why plants need two energy-dissipating systems with apparently similar physiological functions. A probably complete plant uncoupling protein gene family is described and the expression profiles of this family compared with the multigene family of alternative oxidases in Arabidopsis thaliana and sugarcane (Saccharum sp.) employed as dicot and monocot models, respectively. In total, six uncoupling protein genes, AtPUMP1-6, were recognized within the Arabidopsis genome and five (SsPUMP1-5) in a sugarcane EST database. The recombinant AtPUMP5 protein displayed similar biochemical properties as AtPUMP1. Sugarcane possessed four Arabidopsis AOx1-type orthologues (SsAOx1a-1d); no sugarcane orthologue corresponding to Arabidopsis AOx2-type genes was identified. Phylogenetic and expression analyses suggested that AtAOx1d does not belong to the AOx1-type family but forms a new (AOx3-type) family. Tissue-enriched expression profiling revealed that uncoupling protein genes were expressed more ubiquitously than the alternative oxidase genes. Distinct expression patterns among gene family members were observed between monocots and dicots and during chilling stress. These findings suggest that the members of each energy-dissipating system are subject to different cell or tissue/organ transcriptional regulation. As a result, plants may respond more flexibly to adverse biotic and abiotic conditions, in which oxidative stress is involved. © The Author [2006]. Utilizaram-se vacas Holandesas com produção superior a 20 kg de leite/dia, de primeira e segunda lactações, com 19±6 dias em lactação, para avaliar o efeito da suplementação com 8,4 g/dia de metionina protegida (MPDR) ou 8,4 g/dia de metionina não-protegida da degradação ruminal (MNPDR) sobre a produção e composição do leite, comparativamente a vacas controle, durante 90 dias. As vacas foram alimentadas com ração completa constituída por silagem de milho e concentrado. Produção de leite, teor de proteína do leite e produção de proteína não foram afetados pela suplementação com MPDR. As produções médias de leite foram 27,70; 27,09 e 27,61 kg/dia; os teores médios de proteína, 2,83; 2,85 e 2,77%; e as produções de proteína do leite, 0,77; 0,76 e 0,79 kg/dia, respectivamente, para vacas controle, suplementadas com MPDR e MNPDR. O teor de gordura do leite foi de 2,39; 2,12 e 1,89% paras vacas suplementadas com MPDR, MNPDR e controle, respectivamente. A produção diária de gordura foi 0,57; 0,58 e 0,58 kg/dia e a produção diária de leite corrigido para gordura (3,5%), 21,25; 21,19 e 21,35 kg/dia, para os respectivos tratamentos controle, MPDR e MNPDR. A suplementação com MPDR não alterou a produção de leite, porém melhorou a sua composição no início da lactação.

Foram desenvolvidas equações de predição de consumo de pasto de capim-elefante (Pennisetum purpureum, Schumack) por vacas mestiças Holandês x Zebu em lactação, utilizando-se procedimentos de stepwise em regressões múltiplas, aplicados a um banco de dados de experimentos conduzidos ao longo de três anos na Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA) Gado de Leite (Coronel Pacheco, MG). As variáveis independentes disponíveis foram relacionadas a características inerentes às vacas (dias em lactação; teores de proteína, gordura e extrato seco total e produções destes componentes no leite; produção de leite in natura ou corrigida para 4% de gordura; ordem de lactação; peso vivo atual; peso vivo ao parto e grau de sangue Holandês x Zebu); ao manejo (dias de pastejo; disponibilidade de forragem e período de descanso da pastagem); ao ambiente (estação do ano e precipitação pluviométrica) e à alimentação (digestibilidade in vitro e parâmetros da composição química do pasto de capim-elefante e da cana-de-açúcar - Saccharum officinarum (L.) corrigida com 1% de uréia; consumos de suplemento volumoso (cana corrigida com uréia) e concentrado; concentrações fecais de proteína bruta e de fibras em detergente neutro e ácido). Efeitos linear e quadrático e transformações logarítmicas foram adicionalmente incluídos no banco de dados. Foram obtidas equações de predição de consumo de pasto de capim-elefante (expresso em kg/vaca/dia ou % do peso vivo) com coeficientes de determinação de 65,2 a 67,0%. As principais variáveis independentes incluídas nas equações foram o consumo do suplemento volumoso usado na estação seca do ano (cana corrigida com uréia); a digestibilidade in vitro do pasto de capim-elefante; a precipitação pluviométrica; a produção de leite corrigida para 4% de gordura; o peso vivo atual ou, em alternativa a este, o valor da pesagem realizada após o parto da vaca; além do consumo de suplemento concentrado, que evidenciou um efeito de substituição àquele do pasto de capim-elefante.
