Copper oxide nanoparticles (CuO-NPs) are frequently used for many technical applications, but are also known for their cell toxic potential. In order to investigate a potential use of CuO-NPs as a therapeutic drug for glioma treatment, we have investigated the consequences of an application of CuO-NPs on the cellular copper content and cell viability of C6 glioma cells. CuO-NPs were synthesized by a wet-chemical method and were coated with dimercaptosuccinic acid and bovine serum albumin to improve colloidal stability in physiological media. Application of these protein-coated nanoparticles (pCuO-NPs) to C6 cells caused a strong time-, concentration- and temperature-dependent copper accumulation and severe cell death. The observed loss in cellular MTT-reduction capacity, the loss in cellular LDH activity and the increase in the number of propidium iodide-positive cells correlated well with the specific cellular copper content. C6 glioma cells were less vulnerable to pCuO-NPs compared to primary astrocytes and toxicity of pCuO-NPs to C6 cells was only observed for incubation conditions that increased specific cellular copper contents above 20 nmol copper per mg protein. Both cellular copper accumulation as well as the pCuO-NP-induced toxicity in C6 cells were prevented by application of copper chelators, but not by endocytosis inhibitors, suggesting that liberation of copper ions from the pCuO-NPs is the first step leading to the observed toxicity of pCuO-NP-treated glioma cells.

A high degradation extent of benzo[a]pyrene (BaP) should not be considered as the sole desirable criterion for the bioremediation of BaP-contaminated soils because some of its accumulated metabolites still have severe health risks to human. Two main metabolites of BaP, benzo[a]pyrene-1,6-quinone (BP1,6-quinone) and 3-hydroxybenzo[a]pyrene (3-OHBP) were identified by high performance liquid chromatography (HPLC) with standards. This study was the first time that degradation of both BaP and the two metabolites was carried out by chemical oxidation and biodegradation. Three main phases during the whole degradation process were proposed.

Hydrogen peroxide–zinc (H2O2–Zn), the fungus – Aspergillus niger and the bacteria – Zoogloea sp. played an important role in the different phases. The degradation parameters of the system were also optimized, and the results showed that the effect of degradation was the best when fungus–bacteria combined with H2O2–Zn, the concentration range of BaP in the cultures was 30–120 mg/l, the initial pH of the cultures was 6.0. However, as co-metabolites, phenanthrene significant inhibited the degradation of BaP. This combined degradation system compared with the conventional method of degradation by domestic fungus only, enhanced the degradation extent of BaP by more than 20% on the 12 d. The highest accumulation of BP1,6-quinone and 3-OHBP were reduced by nearly 10% in the degradation experiments, which further proved that the combined degradation system was more effective as far as joint toxicity of BaP and its metabolites are concerned.

Supercritical fluid extracts of New Zealand green-lipped mussels (NZGLM) have been suggested to have therapeutic properties related to their oil components. The large number of minor FA in NZGLM extract was characterized by a GC-CIMS/MS method that excels at identification of double-bond positions in FAME. The extract contained five major lipid classes: sterol esters, TAG, FFA, sterols, and polar lipids. The total FA content of the lipid extract was 0.664 g/mL. Fifty-three unsaturated FA (UFA) were fully identified, of which 37 were PUFA, and a further 21 UFA were detected for which concentrations were too low for assignment of double-bond positions. There were 17 saturated FA, with 14∶0, 16∶0, and 18∶0 present in the greatest concentration. The 10 n−3 PUFA detected included 20∶5n−3 and 22∶6n−3, the two main n−3 FA; n−3 PUFA at low concentrations were 18∶3, 18∶4, 20∶3, 20∶4, 21∶5, 22∶5, 24∶6, and 28∶8. There were 43 UFA from the n−4, n−5, n−6, n−7, n−8, n−9, n−10, n−11 families, with 16∶2n−4, 16∶1n−5, 18∶1n−5, 18∶2n−6, 20∶4n−6, 16∶1n−7, 20∶1n−7, 16∶1n−9, 18∶1n−9, and 20∶1n−9 being the most abundant. In general, we estimated that FAME concentrations greater than 0.05% (w/w) were sufficient to assign double-bond positions. In total, 91 FA were detected in an extract of the NZGLM, whereas previous studies of fresh flesh from the NZGLM had reported identification of 42 FA. These data demonstrate a remarkable diversity of NZGLM FA.

Traditionally, quantum theory has traditionally relied heavily on the use of  mathematics. However, there is a significant cohort of students who are  weak in mathematics, for example, students who are majoring in   biochemistry, biological sciences, etc. This paper reports on the use of  spreadsheets to generate approximate numerical solutions and visual  (graphical) descriptions as a method of avoiding or minimizing symbolic  manipulations, mathematical derivations and numerical computation. A  specific example from quantum theory is provided. Some aspects of  educational pedagogy of spreadsheet usage are discussed.

It is neither comprehensive nor appropriate that the bioremediation of a benzo[a]pyrene (BaP)-contaminated environment be assessed only by its high degradation extent because its metabolites' chemical structures are similar to the parent compound and maybe equally toxic. Therefore, further degradation of BaP metabolites is significant. Three methods, combining the Zoogloea sp. with potassium permanganate, combining the Zoogloea sp. with H2O2, Zoogloea sp. alone, were investigated to degrade cis-BP4,5-dihydrodiol and cis-BP7,8-dihydrodiol, which are the metabolites of BaP formed by bacterium-Zoogloea sp. Optimum parameters of degradation in the best method are that: of the three methods, coupling the Zoogloea sp. and KMnO4 is the best; compared with cis-BP7,8-dihydrodiol, cis-BP4,5-dihydrodiol is the more liable to be accumulated in pure cultures; the degradation effect of the two metabolites is optimal when the initial concentration of KMnO4 in the cultures is 0.05%; initial concentration of cis-BP4,5-dihydrodiol and cis-BP7,8-dihydrodiol is 4 mg L−1, 8 mg L−1, respectively; cometabolic substance is salicylic acid or sodium succinate. The degradation extent of cis-BP4,5-dihydrodiol and cis-BP7,8-dihydrodiol using combining the Zoogloea sp. and KMnO4 reach 76.1% and 85.9% after 12 days of cultivation, respectively, which were more than twice compared with conventional method.

Hydroxyapatite (HAp) is commonly used to coat titanium alloys (Ti–6Al–4V) for orthopedic implants. However, their poor adhesion strength and insufficient long-term stability limit their application. Novel sphene (CaTiSiO5) ceramics possess excellent chemical stability and cytocompatibility. The aim of this study is to use the novel sphene ceramics as coatings for Ti–6Al–4V. The sol–gel method was used to produce the coatings and the thermal properties, phase composition, microstructure, thickness, surface roughness and adhesion strength of sphene coatings were analyzed by differential thermal analysis–thermal gravity (DTA–TG), X-ray diffraction (XRD), scanning electron microscopy (SEM), atom force microscopy (AFM) and scratch test, respectively. DTA analysis confirmed that the temperature of the sphene phase formation is 875 °C and XRD analysis indicated pure sphene coatings were obtained. A uniform structure of the sphene coating was found across the Ti–6Al–4V surface, with a thickness and surface roughness of the coating of about 0.5–1 μm and 0.38 μm, respectively. Sphene-coated Ti–6Al–4V possessed a significantly improved adhesion strength compared to that for HAp coating and their chemical stability was evaluated by testing the profile element distribution and the dissolution kinetics of calcium (Ca) ions after soaking the sphene-coated Ti–6Al–4V in Tris–HCl solution. Sphene coatings had a significantly improved chemical stability compared to the HAp coatings. A layer of apatite formed on the sphene-coated Ti–6Al–4V after they were soaked in simulated body fluids (SBF). Our results indicate that sol–gel coating of novel sphene onto Ti–6Al–4V possessed improved adhesion strength and chemical stability, compared to HAp-coated Ti–6Al–4V prepared under the same conditions, suggesting their potential application as coatings for orthopedic implants.

Background

Chemical immobilization of Weddell seals (Leptonychotes weddellii) has previously been, for the most part, problematic and this has been mainly attributed to the type of immobilizing agent used. In addition to individual sensitivity, physiological status may play an important role. We investigated the use of the intravenous administration of a 1:1 mixture of tiletamine and zolazepam (Telazol®) to immobilize adult females at different points during a physiologically demanding 5–6 week lactation period. We also compared performance between IV and IM injection of the same mixture.
Results

The tiletamine:zolazepam mixture administered intravenously was an effective method for immobilization with no fatalities or pronounced apnoeas in 106 procedures; however, there was a 25 % (one animal in four) mortality rate with intramuscular administration. Induction time was slightly longer for females at the end of lactation (54.9 ± 2.3 seconds) than at post-parturition (48.2 ± 2.9 seconds). In addition, the number of previous captures had a positive effect on induction time. There was no evidence for effects due to age, condition (total body lipid), stage of lactation or number of captures on recovery time.
Conclusion

We suggest that intravenous administration of tiletamine and zolazepam is an effective and safe immobilizing agent for female Weddell seals. Although individual traits could not explain variation in recovery time, we suggest careful monitoring of recovery times during longitudinal studies (> 2 captures). We show that physiological pressures do not substantially affect response to chemical immobilization with this mixture; however, consideration must be taken for differences that may exist for immobilization of adult males and juveniles. Nevertheless, we recommend a mass-specific dose of 0.50 – 0.65 mg/kg for future procedures with adult female Weddell seals and a starting dose of 0.50 mg/kg for other age classes and other phocid seals.

 In chemistry education, students not only learn chemical knowledge and skills, but about the culture of chemistry – how scientists think about, and practise, chemistry. Students often learn that science is practised according to the “scientific method”, which is a model of scientific discovery, expounded by science historians and philosophers. The idealised “scientific method” has a number of steps: the collection of information about a phenomenon; the development of a hypothesis to explain those observations; an experiment to test a prediction that arises from the hypothesis, perhaps including more observations and collection of more information; improvement of the hypothesis; and so on.

The problem is that students (and even some science professionals) often do not understand the philosophy behind the scientific method and paradoxically, the scientific method does not seem to apply to most careers in science. The true nature of science is that concepts have been developed though variants of the “scientific method”, and that a process of testing the predictive value of these concepts has lead to advances in that conceptual knowledge. Hence the “scientific method” applies to the development of scientific ideas, not necessarily to the work of all scientists. It is not whether we personally use the scientific method in our day-today work, but how we use, apply, think about and communicate scientific knowledge and skills that makes us chemists.

The mass transfer coefficient is an important kinetic factor to control the thermo-chemical treatment processes of metals and alloys. More importantly, the mass transfer coefficient is different at different surface positions of a metallic part treated, which depends on the dynamic characteristics of the atmosphere close to the treated surface. Understanding the local mass transfer coefficient would be significant to approach the expected physical and mechanical properties of treated surfaces. In this paper, a reverse method was proposed to measure the mass transfer coefficient at component surface and the diffusivity in metal during heat treatment. The methodology of the reverses method and the optimal parame-ters are discussed in some detail. This method was successfully used to determine the car-bon transfer coefficient at the surface of a part in a carburizing furnace and carbon diffusiv-ity from the carbon distribution within the diffusion layer.

Polypyrrole is a material with immensely useful properties suitable for a wide range of electrochemical applications, but its development has been hindered by cumbersome manufacturing processes. Here we show that a simple modification to the standard electrochemical polymerization method produces polypyrrole films of equivalently high conductivity and superior mechanical properties in one-tenth of the polymerization time. Preparing the film as a series of electrodeposited layers with thorough solvent washing between layering was found to produce excellent quality films even when layer deposition was accelerated by high current. The washing step between the sequentially polymerized layers altered the deposition mechanism, eliminating the typical dendritic growth and generating nonporous deposits. Solvent washing was shown to reduce the concentration of oligomeric species in the near-electrode region and hinder the three-dimensional growth mechanism that occurs by deposition of secondary particles from solution. As artificial muscles, the high density sequentially polymerized films produced the highest mechanical work output yet reported for polypyrrole actuators.

A general method for the generation of two-dimensional (2D) ordered, large-area, and liftable conducting polymer-nanobowl sheet has been demonstrated via chemical polymerization for the first time. The sheet is made using the monolayer self-assembled from polystyrene (PS) spheres at the aqueous/air interface as template, followed by depositing conducting polymer on the part of PS monolayer submerging in the aqueous phase via chemical polymerization, and core extraction. During the process of polymerization, no substrate is required, which caused the as-prepared patterned conducting polymer sheet can be easily lifted-off and deposited, in full size, on any flat substrate. In order to overcome interfacial incompatibility issues in natural fibre reinforced polymer bio-composites, surface modifications of the natural fibres using complex and environmentally unfriendly chemical methods is necessary. In this paper, we demonstrate that the interfacial properties of cellulose-based bio-composites can be tailored through surface adsorption of polyethylene glycol (PEG) based amphiphilic block copolymers using a greener alternative methodology. Mixtures of water or water/acetone were used to form amphiphilic emulsions or micro-crystal suspensions of PEG based amphiphilic block copolymers, and their deposition from solution onto the cellulosic substrate was carried out by simple dip-coating. The findings of this study evidence that, by tuning the amphiphilicity and the type of building blocks attached to the PEG unit, the flexural and dynamic thermo-mechanical properties of cellulose-based bio-composites comprised of either polylactide (PLA) or high density polyethylene (HDPE) as a matrix, can be remarkably enhanced. The trends, largely driven by interfacial effects, can be ascribed to the combined action of the hydrophilic and hydrophobic components of these amphiphiles. The nature of the interactions formed across the fibre-matrix interface is discussed. The collective outcome from this study provides a technological template to significantly improve the performance of cellulose-based bio-composite materials.

Precise and reliable modelling of polymerization reactor is challenging due to its complex reaction mechanism and non-linear nature. Researchers often make several assumptions when deriving theories and developing models for polymerization reactor. Therefore, traditional available models suffer from high prediction error. In contrast, data-driven modelling techniques provide a powerful framework to describe the dynamic behaviour of polymerization reactor. However, the traditional NN prediction performance is significantly dropped in the presence of polymerization process disturbances. Besides, uncertainty effects caused by disturbances present in reactor operation can be properly quantified through construction of prediction intervals (PIs) for model outputs. In this study, we propose and apply a PI-based neural network (PI-NN) model for the free radical polymerization system. This strategy avoids assumptions made in traditional modelling techniques for polymerization reactor system. Lower upper bound estimation (LUBE) method is used to develop PI-NN model for uncertainty quantification. To further improve the quality of model, a new method is proposed for aggregation of upper and lower bounds of PIs obtained from individual PI-NN models. Simulation results reveal that combined PI-NN performance is superior to those individual PI-NN models in terms of PI quality. Besides, constructed PIs are able to properly quantify effects of uncertainties in reactor operation, where these can be later used as part of the control process. © 2014 Taiwan Institute of Chemical Engineers.

The need to manage otariid populations has necessitated the development of a wide range of capture methods. Chemical restraint by remote drug delivery (i.e., darting) is a highly selective method that can be used to facilitate otariid capture in a range of scenarios, when other methods may be impracticable. However, the risks associated with darting otariids are not widely known and guidelines necessary to promote and refine best practice do not exist. We review the risks associated with darting and in light of our findings, develop darting guidelines to help practitioners assess and minimize risks during capture, anesthesia and recovery. Published studies reveal that mortalities associated with darting predominantly result from complications during anesthetic maintenance (e.g., prolonged respiratory depression, apnea, or hyperthermia), rather than from complications during capture or recovery. In addition to monitoring vital signs and proper intervention, the risk of irreversible complications during anesthesia can be reduced by administering drug doses that are sufficient to enable the capture and masking of animals, after which anesthetic depth can be regulated using gas anesthesia.
