In neuroscience, the extracellular actions potentials of neurons are the most important signals, which are called spikes. However, a single extracellular electrode can capture spikes from more than one neuron. Spike sorting is an important task to diagnose various neural activities. The more we can understand neurons the more we can cure more neural diseases. The process of sorting these spikes is typically made in some steps which are detection, feature extraction and clustering. In this paper we propose to use the Mel-frequency cepstral coefficients (MFCC) to extract spike features associated with Hidden Markov model (HMM) in the clustering step. Our results show that using MFCC features can differentiate between spikes more clearly than the other feature extraction methods, and also using HMM as a clustering algorithm also yields a better sorting accuracy.

Zn and DHA have putative neuroprotective effects and these two essential nutrients are known to interact biochemically. We aimed to identify novel protein candidates that are differentially expressed in human neuronal cell line M17 in response to Zn and DHA that would explain the molecular basis of this interaction. Two-dimensional gel electrophoresis and MS were applied to identify major protein expression changes in the protein lysates of human Ml7 neuronal cells that had been grown in the presence and absence of Zn and DHA. Proteomic findings were further investigated using Western immunoblot and real-time PCR analyses. Four protein spots, which had significant differential expression, were identified and selected for in-gel trypsin digestion followed by matrix-assisted laser desorption ionisation MS analysis. The resultant peptide mass fingerprint for each spot allowed their respective identities to be deduced. Two human histone variants H3 and H4 were identified. Both H3 and H4 were downregulated by Zn in the absence of DHA (Zn effect) and upregulated by DHA (DHA effect) in the presence of Zn (physiological condition). These proteomic findings were further supported by Western immunoblot and real-time PCR analyses using H3- and H4-specific monoclonal antibodies and oligonucleotide primers, respectively. We propose that dietary Zn and DHA cause a global effect on gene expression, which is mediated by histones. Such novel information provides possible clues to the molecular basis of neuroprotection by Zn and DHA that may contribute to the future treatment, prevention and management of neurodegenerative diseases such as Alzheimer's disease.

Docosahexaenoic acid (DHA) is the major polyunsaturated fatty acid in neuronal cell membranes. We hypothesize that DHA induces a decrease in neuronal cell death through reduced ZnT3 expression and zinc uptake. Exposure of M17 cells to DHA-deficient medium increased the levels of active caspase-3, relative to levels in DHA-replete cells, confirming the adverse effects of DHA deficiency in promoting neuronal cell death. In DHA-treated M17 cells, zinc uptake was 65% less and ZnT3 mRNA and protein levels were reduced in comparison with DHA-depleted cells. We propose that the neuroprotective function of DHA is exerted through a reduction in cellular zinc levels that in turn inhibits apoptosis.

Large overbank flood events play an important role in maintaining largescale ecological processes and connectivity along and across the floodplains and between the rivers and their floodplains in the southern Murray-Darling Basin. However, the regulation of rivers means that extensive overbank flooding can only occur in the rare circumstance of extreme flood events. Recent environmental water allocations have focussed on the largest floodplain blocks (‘icon’ sites) and a small set of specific values (e.g. colonial nesting waterbirds), as well as on trialling fine-scale manipulation of infrastructure (e.g. pumping) to water relatively small areas. There has been no comprehensive systematic assessment of the entire floodplain and its wider set of flood-dependent natural assets (such as ecosystems and species; herein referred to as ‘natural values’) to maximise the effectiveness of environmental water use and to catalogue values likely to be lost. This paper describes an assessment of some 220 000 ha found to support flood-dependent natural values in Victoria. We mapped the geographic distribution and estimated components of the flooding requirements (natural flooding frequency, and maximum period without flooding and minimum duration of each flooding event before significant deterioration) for each natural value. Using an example of one stretch of the River Murray, we show how the resultant spatial data can be used with floodplain inundation modelling to compare the outcomes of real or planned environmental watering events; potentially providing tools for management agencies to conserve a wider range of floodplain values than is currently the case. That is, water managers and the public can see what ecosystems and threatened species are intended to be maintained by environmental watering and what values are intended to be abandoned across the whole floodplain, rather than just seeing the small subset of values and ‘icon’ sites that are intended to be maintained. Examples are provided to illustrate how information about the location, water requirements and extent covered by potential floods for specific values can be used to build adaptive watering strategies for areas as large as the whole floodplain.

Overbank flooding of rivers is a key process in the maintenance of vegetation types and the species that rely on the floodplain forests and woodlands of northern Victoria. Yet the flooding requirements of species and vegetation types are poorly known. Here we present initial estimates of the water requirements for flood dependent Ecological Vegetation Classes (EVCs) and rare and threatened flora and fauna species associated with the floodplain of the Murray River and its tributaries. Some 110 EVCs were found to be at least partly flood-dependent on the Murray River floodplains. The total current extent of these EVCs in the study area is 224 247 ha, of which 162 266 ha are on public land. One hundred and twenty-four rare or threatened plant taxa and 62 threatened vertebrate fauna taxa (excluding fish) were classified as at least partly flood-dependent. These initial estimates provide important information for land and water managers and researchers alike.

Four new triphenyltin(IV) complexes of composition Ph3SnLH (where LH = 2-/4-[(E)-2-(aryl)-1-diazenyl]benzoate) (1–4) were synthesized and characterized by spectroscopic (1H, 13C and 119Sn NMR, IR, 119Sn Mössbauer) techniques in combination with elemental analysis. The 119Sn NMR spectroscopic data indicate a tetrahedral coordination geometry in non-coordinating solvents. The crystal structures of three complexes, Ph3SnL1H (1), Ph3SnL3H (3), Ph3SnL4H (4), were determined. All display an essentially tetrahedral geometry with angles ranging from 93.50(8) to 124.5(2)°; 119Sn Mössbauer spectral data support this assignment. The cytotoxicity studies were performed with complexes 1–4, along with a previously reported complex (5) in vitro across a panel of human tumor cell lines viz., A498, EVSA-T, H226, IGROV, M19 MEL, MCF-7 and WIDR. The screening results were compared with the results from other related triphenyltin(IV) complexes (6–7) and tributyltin(IV) complexes (8–11) having 2-/4-[(E)-2-(aryl)-1-diazenyl]benzoates framework. In general, the complexes exhibit stronger cytotoxic activity. The results obtained for 1–3 are also comparable to those of its o-analogs i.e. 4–7, except 5, but the advantage is the former set of complexes demonstrated two folds more cytotoxic activity for the cell line MCF-7 with ID50 values in the range 41–53 ng/ml. Undoubtedly, the cytotoxic results of complexes 1–3 are far superior to CDDP, 5-FU and ETO, and related tributyltin(IV) complexes 8–11. The quantitative structure-activity relationship (QSAR) studies for the cytotoxicity of triphenyltin(IV) complexes 1–7 and tributyltin(IV) complexes 8–11 is also discussed against a panel of human tumor cell lines.

Omega-3 (ω-3) fatty acids are one of the two main families of long chain polyunsaturated fatty acids (PUFA). The main omega-3 fatty acids in the mammalian body are α-linolenic acid (ALA), docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). Central nervous tissues of vertebrates are characterized by a high concentration of omega-3 fatty acids. Moreover, in the human brain, DHA is considered as the main structural omega-3 fatty acid, which comprises about 40% of the PUFAs in total. DHA deficiency may be the cause of many disorders such as depression, inability to concentrate, excessive mood swings, anxiety, cardiovascular disease, type 2 diabetes, dry skin and so on. On the other hand, zinc is the most abundant trace metal in the human brain. There are many scientific studies linking zinc, especially excess amounts of free zinc, to cellular death. Neurodegenerative diseases, such as Alzheimer's disease, are characterized by altered zinc metabolism. Both animal model studies and human cell culture studies have shown a possible link between omega-3 fatty acids, zinc transporter levels and free zinc availability at cellular levels. Many other studies have also suggested a possible omega-3 and zinc effect on neurodegeneration and cellular death. Therefore, in this review, we will examine the effect of omega-3 fatty acids on zinc transporters and the importance of free zinc for human neuronal cells. Moreover, we will evaluate the collective understanding of mechanism(s) for the interaction of these elements in neuronal research and their significance for the diagnosis and treatment of neurodegeneration.

Considering that the uncertainty noise produced the decline in the quality of collected neural signal, this paper proposes a signal quality assessment method for neural signal. The method makes an automated measure to detect the noise levels in neural signal. Hidden Markov Models were used to build a classification model that classifies the neural spikes based on the noise level associated with the signal. This neural quality assessment measure will help doctors and researchers to focus on the patterns in the signal that have high signal to noise ratio and carry more information.

 The main focus of this research was to investigate possible link between zinc, DHA, apoptosis and cell survival. Study also analyses the benefits of omega-3 FA’s and the link between free zinc availability to neurodegeneration. Furthermore, this study was focused on developing a suitable cell culture model for neuronal research.

EEG signal is one of the most important signals for diagnosing some diseases. EEG is always recorded with an amount of noise, the more noise is recorded the less quality is the EEG signal. The included noise can represent the quality of the recorded EEG signal, this paper proposes a signal quality assessment method for EEG signal. The method generates an automated measure to detect the noise level of the recorded EEG signal. Mel-Frequency Cepstrum Coefficient is used to represent the signals. Hidden Markov Models were used to build a classification model that classifies the EEG signals based on the noise level associated with the signal. This EEG quality assessment measure will help doctors and researchers to focus on the patterns in the signal that have high signal to noise ratio and carry more information. Moreover, our model was applied on an uncontrolled environment and on controlled environment and a result comparison was applied.
