The size of reinforced particles notably affects the electro-discharge machining (EDM) of metal matrix composites (MMCs). This paper explores the mechanism of wire EDM of MMCs with different sizes of reinforced particles as well as the corresponding unreinforced matrix material. The mechanisms of material removal, surface generation, and taper kerf formation were investigated. This study shows that the particles’ ability to protect matrix materials from the intense heat of electric arc controls the material removal rate, surface generation, and taper of kerf. The low melting point matrix material is removed very easily, but the heat resistance reinforced particles delay the removal of material and facilitate the transfer of the workpiece material to wire electrode and vice versa. Thus, the material stays longer in touch with intense heat and affects the surface generation, wire electrode wear, and width of the kerf.

To examine whether genes associated with cellular defense against oxidative stress are associated with insulin sensitivity, patients with type 2 diabetes (n = 7) and age-matched (n = 5) and young (n = 9) control subjects underwent a euglycemic-hyperinsulinemic clamp for 120 min. Muscle samples were obtained before and after the clamp and analyzed for heat shock protein (HSP)72 and heme oxygenase (HO)-1 mRNA, intramuscular triglyceride content, and the maximal activities of β-hyroxyacyl-CoA dehydrogenase (β-HAD) and citrate synthase (CS). Basal expression of both HSP72 and HO-1 mRNA were lower (P < 0.05) by 33 and 55%, respectively, when comparing diabetic patients with age-matched and young control subjects, with no differences between the latter groups. Both basal HSP72 (r = 0.75, P < 0.001) and HO-1 (r = 0.50, P < 0.05) mRNA expression correlated with the glucose infusion rate during the clamp. Significant correlations were also observed between HSP72 mRNA and both β-HAD (r = 0.61, P < 0.01) and CS (r = 0.65, P < 0.01). HSP72 mRNA was induced (P < 0.05) by the clamp in all groups. Although HO-1 mRNA was unaffected by the clamp in both the young and age-matched control subjects, it was increased (P < 0.05) ∼70-fold in the diabetic patients after the clamp. These data demonstrate that genes involved in providing cellular protection against oxidative stress are defective in patients with type 2 diabetes and correlate with insulin-stimulated glucose disposal and markers of muscle oxidative capacity. The data provide new evidence that the pathogenesis of type 2 diabetes involves perturbations to the antioxidant defense mechanism within skeletal muscle.

Heating effects in polypyrrole-coated polyethyleneterephthalate (PET)-Lycra® fabrics were studied. Chemical synthesis was employed to coat the PET fabrics by polypyrrole using ferric chloride as oxidant and antraquinone- 2-sulfonic acid (AQSA) and naphthalene sulfonic acid (NSA) as dopants. The coated fabrics exhibited reasonable electrical stability, possessed high electrical conductivity, and were effective in heat generation. Surface resistance of polypyrrole-coated fabrics ranged from approximately 150 to 500 /square. Different connections between conductive fabrics and the power source were examined. When subjected to a constant voltage of 24 V, the current transmitted through the fabric decreased about 10% in 72 h. The promise of cancer immunotherapy is that it will not only eradicate primary tumors but will generate systemic antitumor immunity capable of destroying distant metastases. A major problem that must first be surmounted relates to the immune resistance of large tumors. Here we reveal that immune resistance can be overcome by combining immunotherapy with a concerted attack on the tumor vasculature. The functionally related antitumor drugs 5,6-dimethylxanthenone-4-acetic acid (DMXAA) and flavone acetic acid (FAA), which cause tumor vasculature collapse and tumor necrosis, were used to attack the tumor vasculature, whereas the T-cell costimulator B7.1 (CD80), which costimulates T-cell proliferation via the CD28 pathway, was used to stimulate antitumor immunity. The injection of cDNA (60–180 µg) encoding B7.1 into large EL-4 tumors (0.8 cm in diameter) established in C57BL/6 mice, followed 24 h later by i.p. administration of either DMXAA (25 mg/kg) or FAA (300 mg/kg), resulted in complete tumor eradication within 2–6 weeks. In contrast, monotherapies were ineffective. Both vascular attack and B7.1 immunotherapy led to up-regulation of heat shock protein 70 on stressed and dying tumor cells, potentially augmenting immunotherapy. Remarkably, large tumors took on the appearance of a wound that rapidly ameliorated, leaving perfectly healed skin. Combined therapy was mediated by CD8+ T cells and natural killer cells, accompanied by heightened and prolonged antitumor cytolytic activity (P < 0.001), and by a marked increase in tumor cell apoptosis. Cured animals completely rejected a challenge of 1 x 107 parental EL-4 tumor cells but not a challenge of 1 x 104 Lewis lung carcinoma cells, demonstrating that antitumor immunity was tumor specific. Adoptive transfer of 2 x 108 splenocytes from treated mice into recipients bearing established (0.8 cm in diameter) tumors resulted in rapid and complete tumor rejection within 3 weeks. Although DMXAA and B7.1 monotherapies are complicated by a narrow range of effective doses, combined therapy was less dosage dependent. Thus, a broad range of amounts of B7.1 cDNA were effective in combination with 25 mg/kg DMXAA. In contrast, DMXAA, which has a very narrow range of high active doses, was effective at a low dose (18 mg/kg) when administered with a large amount (180 µg) of B7.1 cDNA. Importantly, combinational therapy generated heightened antitumor immunity, such that gene transfer of B7.1 into one tumor, followed by systemic DMXAA treatment, led to the complete rejection of multiple untreated tumor nodules established in the opposing flank. These findings have important implications for the future direction and utility of cancer immunotherapies aimed at harnessing patients’ immune responses to their own tumors.

The effect of the heat treatment on the corrosion behaviour of amorphous Al88Ni6La6 made by melt-spun has been investigated by electrochemical measurements. Heat treatment was carried out at 523 K and 673 K for 4 min and 15 min respectively. The evolution of the crystallization process after annealing was identified by differential scanning calorimeter (DSC) as well as X-ray diffraction. The XRD patterns show that the structure of samples heat-treated at higher temperature changes towards a crystal state. The results obtained from the polarization curves reveal that all Al88Ni6La6 alloys exhibit spontaneously passivated behaviour. Furthermore, it is noted that the partially crystallized alloy has the best corrosion resistance in comparison with as-spun amorphous and fully crystallized alloys, while the fully crystallized sample shows deterioration in the corrosion resistance.

Coral reef fishes are expected to experience rising sea surface temperatures due to climate change. How well tropical reef fishes will respond to these increased temperatures and which genes are important in the response to elevated temperatures is not known. Microarray technology provides a powerful tool for gene discovery studies, but the development of microarrays for individual species can be expensive and time-consuming. In this study, we tested the suitability of a Danio rerio oligonucleotide microarray for application in a species with few genomic resources, the coral reef fish Pomacentrus moluccensis. Results from a comparative genomic hybridization experiment and direct sequence comparisons indicate that for most genes there is considerable sequence similarity between the two species, suggesting that the D. rerio array is useful for genomic studies of P. moluccensis. We employed this heterologous microarray approach to characterize the early transcriptional response to heat stress in P. moluccensis. A total of 111 gene loci, many of which are involved in protein processing, transcription, and cell growth, showed significant changes in transcript abundance following exposure to elevated temperatures. Changes in transcript abundance were validated for a selection of candidate genes using quantitative real-time polymerase chain reaction. This study demonstrates that heterologous microarrays can be successfully employed to study species for which specific microarrays have not yet been developed, and so have the potential to greatly enhance the utility of microarray technology to the field of environmental and functional genomics.

Low-level Nd additions, up to 0.17 wt%, were added to Al-5Mg to explore the impact on the subsequent degree of sensitization. Following heat treatment at 150°C for 1 day and 7 days, nitric acid mass loss (NAMLT) tests revealed that additions of >0.11% Nd were effective at decreasing the amount of subsequent intergranular attack.
