The microstructure and mechanical response of three extruded magnesium alloys, Mg-3Al-1Zn (AZ31), Mg-1.5Mn (Ml) and Mg-lMn-0.4RE (ME10) are examined. The tensile yield strength of ME10 was nearly half that of AZ31 and Ml. The tensile elongations were 6%, 11% and 19% for Ml, AZ31 and ME10, respectively. This range of properties is large and is attributed to the unique extrusion texture produced in ME10, and the high density of fine particles in Ml.

We tested the hypothesis that there are sex differences in the inhibitory effect of cortisol on pulsatile LH secretion and pituitary responsiveness to GnRH in gonadectomized sheep. In experiment 1, pulsatile LH secretion was examined in gonadectomized ewes and rams infused with either saline, a low (250 µg/kg·h) or a high (500 µg/kg·h) dose of cortisol for 30 h. In experiment 2, direct pituitary actions of cortisol were assessed by monitoring LH pulse amplitude in response to exogenous GnRH in hypothalamo-pituitary disconnected ewes and rams infused with the low dose of cortisol. In experiment 1, the mean (±SEM) plasma LH concentration was (P < 0.05) reduced significantly during cortisol infusion in both sexes, but the effect was greater in rams. In ewes, LH pulse amplitude and frequency were reduced (P < 0.05) at the high, but not the low, cortisol dose, whereas total LH output (LH pulse amplitude multiplied by frequency) was reduced (P < 0.05) at both doses. In rams, LH pulse frequency and amplitude and total LH output were (P < 0.05) reduced significantly at both cortisol doses. In experiment 2, plasma LH concentration and pulse amplitude in response to exogenous GnRH were not affected by infusion of cortisol in either sex. We conclude that gonadectomized rams are more sensitive than gonadectomized ewes to the effects of cortisol to inhibit LH secretion and that sex differences exist in the specific actions of cortisol on LH pulses. The results of experiment 2 suggest that intact hypothalamic input to the pituitary is necessary for cortisol to inhibit pituitary responsiveness to GnRH.

The maintenance of skeletal muscle mass is a critical component of health in both chronic wasting diseases and aging. A considerable amount of progress has been made in the understanding of the signalling pathways that mediate skeletal muscle hypertrophy and atrophy. Akt is seen as a key molecular protein involved in the maintenance of skeletal muscle mass as it has the dual ability to positively influence protein syntheses and negatively regulate protein degradation in its active state (Glass, 2003). Potential mechanisms which may assist with maintaining skeletal muscle mass are the estrogen hormones. Estrogens increase the proliferation of mouse and rat myoblasts and can also attenuate immobilization-induced skeletal muscle atrophy in rats in vivo (Kahlert et al., 1997). No studies have investigated the effect of estrogens on the activation of skeletal muscle hypertrophy and atrophy signalling pathways. Estrogens may contribute to maintaining skeletal muscle mass via their activation of the Akt signalling pathways. Therefore, the aims of the present study were to determine if treatment of C2C12 myotubes with either 17β-estrodiol or estrone increases the activity of Akt and its downstream anabolic signalling proteins, GSK, p70s6k and 4E-BP1 and decreases its catabolic stimulating targets, FOXO, atrogin-1 and MuRF-1. A secondary aim was to determine if this was associated with an increased rate of protein synthesis.

C2C12 myotubes were incubated at 37°C in serum free DMEM without phenol red containing 10 000 units/ml penicillin, 10 000 μg/ml streptomycin, and 250μg/ml amphotericin B for 24h. Myotubes were then stimulated with 17-β estradiol (10nM) for 24h. Phosphorylated and total proteins for Akt, p70S6k, GSK3β, 4E-BP1, FOXO and atrogin-1 were measured using western blotting techniques. Atrogin-1 and MuRF1 mRNA levels were measured using real time-PCR. Protein synthesis rates were measured by incorporation of [3H]-tyrosine into the myotubes during the last hour of treatment.

Compared to control myotubes, treatment with 17β-estradiol increased the ratio of phosphorylated to total protein contents for Akt, GSK-3β and P70s6k by, 1.62, 1.53 and 2.2 fold, respectively (n=6 per group; p < 0.05). There was, however, no difference in the ratios of phosphorylated to total 4E-BP1 or Foxo3a or Atrogin-1 and MuRF1 mRNA. Protein synthesis rates remained unchanged.

This study demonstrates that in C2C12 mouse myotubes, 17β-estradiol treatment increases the phosphorylation of the hypertrophy signalling protein, Akt, and its downstream hypertrophy signalling targets, GSK-3β and P70s6k; no associated changes in protein synthesis were observed. Future studies should investigate the ability of 17β-estradiol to activate these proteins in a model of myotube catabolism and to determine if protein degradation is attenuated.

Weak photo-induced chemiluminescence (PICL) emission is observed when polymers are exposed to UVA or visible light. The presence of dyes can either increase PICL intensity via Type I photosensitisation which generates polymer free radicals, or reduce it via photo-protection. PICL studies on the eight Blue Wool Standards (BWSs) that are used commercially as lightfastness standards show higher PICL intensity from the least photostable BWSs that use triphenylmethane dyes and lower intensity from more photostable BWSs using UVA and visible wavelengths. The relative PICL intensities do not correlate in a stepwise manner with lightfastness ratings of the BWSs. However dye/polymer combinations that emit high levels of PICL relative to the undyed material are unlikely to have acceptable lightfastness. The xanthene dyes fluorescein and eosin Y are more strongly photosensitising than triphenylmethane dyes on wool and both produce higher PICL emission than undyed wool when irradiated with visible light.

The use of solar collectors with coloured absorbers for water heating is an area of particular interest when considering their integration with buildings. By matching the absorber colour with that of the roof or façade of the building, it is possible to achieve an architecturally and visually pleasing result. Despite the potential for the use of coloured absorbers, very little work has been undertaken in the field.

In this study, the thermal performance of a series of coloured (ranging from white to black), building integrated solar collectors for water heating was examined both theoretically and experimentally. Subsequently, the annual solar fraction for typical water heating systems with coloured absorbers was calculated. The results showed that coloured solar collector absorbers can make noticeable contributions to heating loads. Furthermore, although their thermal efficiency is lower than highly developed selective coating absorbers, they offer the advantage of improved aesthetic integration with buildings.

The time sustained during a graded cycle exercise is ~10% longer in an upright compared with a supine posture. However, during constant-load cycling this effect is unknown. Therefore, we tested the postural effect on the performance of high-intensity constant-load cycling. Twenty-two active subjects (11 men, 11 women) performed two graded tests (one upright, one supine), and of those 22, 10 subjects (5 men, 5 women) performed three high-intensity constant-load tests (one upright, two supine). To test the postural effect on performance at the same absolute intensity, during the upright and one of the supine constant-load tests subjects cycled at 80% of the peak power output achieved during the upright graded test. To test the postural effect on performance at the same relative intensities, during the second supine test subjects cycled at 80% of the peak power output achieved during the supine graded test. Exercise time on the graded and absolute intensity constant-load tests for all subjects was greater (P<0.05) in the upright compared with supine posture (17.9±3.5 vs. 16.1±3.1 min for graded; 13.2±8.7 vs. 5.2±1.9 min for constant-load). This postural effect at the same absolute intensity was larger in men (19.4±8.5 upright vs. 6.6±1.6 supine, P<0.001) than women (7.1±2 upright vs. 3.9±1.4 supine, P>0.05) and it was correlated (P<0.05) with both the difference in VO2 between positions during the first minute of exercise (r=0.67) and the height of the subjects (r=0.72). In conclusion, there is a very large postural effect on performance during constant-load cycling exercise and this effect is significantly larger in men than women.

We have used the hypothalamo-pituitary disconnected (HPD) sheep model to investigate direct pituitary actions of cortisol to suppress LH secretion in response to exogenous GnRH. We previously observed that, during the non-breeding season, treatment with cortisol did not suppress the LH response to GnRH in HPD gonadectomised rams or ewes.1 In the present experiment, we tested the effect of cortisol on the LH response to exogenous GnRH in gonadectomised HPD sheep during the breeding season. Using a cross-over design, HPD gonadectomised Romney Marsh rams (n = 6) and ewes (n = 5) received a saline or cortisol (250 μg/kg/h) infusion for 30 h on each of two days, one week apart. All animals were treated with 125 ng i.v. injections of GnRH every 2 h during a 6 h control period preceding the infusion and during the infusion. Jugular blood samples were taken during the control period and the first 6 h and last 6 h of the infusion (over 3 LH pulses). Mean plasma concentrations of LH and LH pulse amplitudes, driven by programmed GnRH injections, were similar in gonadectomised rams and ewes and there were no significant effects of saline infusion between the control periods or the saline infusion in either sex. The amplitude of LH pulses was significantly (P < 0.05) reduced in rams during the first 6 h of the cortisol infusion compared to the control period, but there were no effects of the cortisol infusion in ewes. These data show that, in the absence of sex steroids, there is a sex difference in the mechanism by which cortisol acts at the pituitary to reduce LH secretion in response to exogenous GnRH in HPD gonadectomized sheep during the breeding season. We conclude that the effect of cortisol to reduce secretion of LH involves an action on the pituitary, at least in gonadectomised rams.

The effect of hydrogen content on the compaction of Ti–6Al–4V powder at low temperatures, namely 500 °C, using equal channel angular pressing (ECAP) with back pressure has been investigated. The properties of the compacts before and after a heat treatment and de-hydrogenation cycle have been determined. Compaction of powder by ECAP (500 °C and 260 MPa) has shown maximum levels of relative density of 99.3% and 99.4% when charged with 0.05–0.1 wt.% and 0.61–0.85 wt.% of hydrogen, respectively. After the de-hydrogenation heat treatment the diffusion bonding between individual powder particles was completed and the microstructure was altered, depending on the level of hydrogen content. Two local maxima of 99.2% and 98.1% were observed in the measured density of consolidated compacts for hydrogen contents between 0.05 wt.% and 0.1 wt.% and between 0.61 wt.% and 0.85 wt.%, respectively. However, the mechanical properties of the compacts within these two ranges of hydrogen content were significantly different due to a difference in the observed microstructure. An exceptionally high ductility of 29%, in combination with a relatively high strength of ~560 MPa, was measured in a shear punch test on specimens which had a prior hydrogen level of 0.05 wt.% before the heat treatment. It was shown that material consolidated from powder hydrogenated to low levels of hydrogen before compaction has the potential to offer substantial improvements in mechanical properties after a suitable heat treatment.

Use of high albedo materials reduces the amount of solar radiation absorbed through building envelops and urban structures and thus keeping their surfaces cooler. The cooling energy savings by using high albedo materials have been well documented. Higher surface temperatures add to increasing the ambient temperature as convection intensity is higher. Such temperature increase has significant impacts on the air conditioning energy utilization in hot climates. This study makes use of a parametric approach by varying the temperature of building facades to represent commonly used materials and hence analyzing its effect on the air temperature through a series of CFD (Computational Fluid Dynamics) simulations. A part of the existing CBD (Central Business District) area of Singapore was selected for the study. Series of CFD simulations have been carried out using the software CFX-5.6. Wind tunnel experiments were also conducted for validation. It was found that at low wind speeds, the effect of materials on the air temperature was significant and the temperature at the middle of a narrow canyon increased up to 2.52°C with the façade material having lowest albedo.

Wool fabrics are often treated under conditions of varying pH in dyeing and finishing processes. It is known that in air the dimensions of wool fabrics change with the amount of fiber swelling at different regain. In this work, it has been shown that a similar relationship between fiber swelling and fabric dimensions existed in water at different pH values. The diameters of Merino and Corriedale wool fibers in water at different pH values were measured with an OFDA 2000 fiber diameter analyzer, fitted with a specially constructed accessory liquid cell. The results showed that the mean diameters of swollen wool fibers in water varied with pH. Minimum swelling was obtained in the range pH 5-7. It was found that the dimensions of wool fabric in water were dependent on the pH. The changes in fiber diameter in water could be attributed to changes in ionic interactions between charged acid and basic groups in wool protein with variations in pH.

Nurses' factual knowledge increased and oxygen administration decisions improved following education. Variability in nurses' knowledge and decisions, both before and after education, suggests that factual knowledge is only one component of clinical decisions. If nursing education is to improve clinical practice, then we need to better understand the relationship between factual knowledge and clinical decisions.

Atmospheric pressure plasma with a relatively short exposure time was sufficient to modify the wool surface. The ageing studies showed the hydrophobic recovery of fibre surface took place at the early stages after plasma treatment. While plasma improved wool shrink-resistance, improvement in dyeability depends on the hydrophilic-hydrophobic nature of dyestuff.
