Effective processes to fractionate the main compounds in biomass, such as wood, are a prerequisite for an effective biorefinery. Water is environmentally friendly and widely used in industry, which makes it a potential solvent also for forest biomass. At elevated temperatures over 100 °C, water can readily hydrolyse and dissolve hemicelluloses from biomass. In this work, birch sawdust was extracted using pressurized hot water (PHWE) flow-through systems. The hypothesis of the work was that it is possible to obtain polymeric, water-soluble hemicelluloses from birch sawdust using flow-through PHW extractions at both laboratory and large scale. Different extraction temperatures in the range 140–200 °C were evaluated to see the effect of temperature to the xylan yield. The yields and extracted hemicelluloses were analysed to obtain sugar ratios, the amount of acetyl groups, furfurals and the xylan yields. Higher extraction temperatures increased the xylan yield, but decreased the molar mass of the dissolved xylan. As the extraction temperature increased, more acetic acid was released from the hemicelluloses, thus further decreasing the pH of the extract. There were only trace amounts of furfurals present after the extractions, indicating that the treatment was mild enough not to degrade the sugars further. The sawdust extraction density was increased by packing more sawdust in the laboratory scale extraction vessel. The aim was to obtain extracts with higher concentration than in typical extraction densities. The extraction times and water flow rates were kept constant during these extractions. The higher sawdust packing degree decreased the water use in the extractions and the extracts had higher hemicellulose concentrations than extractions with lower sawdust degrees of packing. The molar masses of the hemicelluloses were similar in higher packing degrees and in the degrees of packing that were used in typical PHWE flow-through extractions. The structure of extracted sawdust was investigated using small angle-(SAXS) and wide angle (WAXS) x-ray scattering. The cell wall topography of birch sawdust and extracted sawdust was compared using x-ray tomography. The results showed that the structure of the cell walls of extracted birch sawdust was preserved but the cell walls were thinner after the extractions. Larger pores were opened inside the fibres and cellulose microfibrils were more tightly packed after the extraction. Acetate buffers were used to control the pH of the extracts during the extractions. The pH control prevented excessive xylan hydrolysis and increased the molar masses of the extracted xylans. The yields of buffered extractions were lower than for plain water extractions at 160–170 °C, but at 180 °C yields were similar to those from plain water and pH buffers. The pH can thus be controlled during extraction with acetate buffer to obtain xylan with higher molar mass than those obtainable using plain water. Birch sawdust was extracted both in the laboratory and pilot scale. The performance of the PHWE flow-through system was evaluated in the laboratory and the pilot scale using vessels with the same shape but different volumes, with the same relative water flow through the sawdust bed, and in the same extraction temperature. Pre-steaming improved the extraction efficiency and the water flow through the sawdust bed. The extracted birch sawdust and the extracted xylan were similar in both laboratory and pilot scale. The PHWE system was successfully scaled up by a factor of 6000 from the laboratory to pilot scale and extractions performed equally well in both scales. The results show that a flow-through system can be further scaled up and used to extract water-soluble xylans from birch sawdust. Extracted xylans can be concentrated, purified, and then used in e.g. films and barriers, or as building blocks for novel material applications.

Scanning optics create different types of phenomena and limitation to cladding process compared to cladding with static optics. This work concentrates on identifying and explaining the special features of laser cladding with scanning optics. Scanner optics changes cladding process energy input mechanics. Laser energy is introduced into the process through a relatively small laser spot which moves rapidly back and forth, distributing the energy to a relatively large area. The moving laser spot was noticed to cause dynamic movement in the melt pool. Due to different energy input mechanism scanner optic can make cladding process unstable if parameter selection is not done carefully. Especially laser beam intensity and scanning frequency have significant role in the process stability. The laser beam scanning frequency determines how long the laser beam affects with specific place local specific energy input. It was determined that if the scanning frequency in too low, under 40 Hz, scanned beam can start to vaporize material. The intensity in turn determines on how large package this energy is brought and if the intensity of the laser beam was too high, over 191 kW/cm2, laser beam started to vaporize material. If there was vapor formation noticed in the melt pool, the process starts to resample more laser alloying due to deep penetration of laser beam in to the substrate. Scanner optics enables more flexibility to the process than static optics. The numerical adjustment of scanning amplitude enables clad bead width adjustment. In turn scanner power modulation (where laser power is adjusted according to where the scanner is pointing) enables modification of clad bead cross-section geometry when laser power can be adjusted locally and thus affect how much laser beam melts material in each sector. Power modulation is also an important factor in terms of process stability. When a linear scanner is used, oscillating the scanning mirror causes a dwell time in scanning amplitude border area, where the scanning mirror changes the direction of movement. This can cause excessive energy input to this area which in turn can cause vaporization and process instability. This process instability can be avoided by decreasing energy in this region by power modulation. Powder feeding parameters have a significant role in terms of process stability. It was determined that with certain powder feeding parameter combinations powder cloud behavior became unstable, due to the vaporizing powder material in powder cloud. Mainly this was noticed, when either or both the scanning frequency or powder feeding gas flow was low or steep powder feeding angle was used. When powder material vaporization occurred, it created vapor flow, which prevented powder material to reach the melt pool and thus dilution increased. Also powder material vaporization was noticed to produce emission of light at wavelength range of visible light. This emission intensity was noticed to be correlated with the amount of vaporization in the powder cloud.

Personalized medicine will revolutionize our capabilities to combat disease. Working toward this goal, a fundamental task is the deciphering of geneticvariants that are predictive of complex diseases. Modern studies, in the formof genome-wide association studies (GWAS) have afforded researchers with the opportunity to reveal new genotype-phenotype relationships through the extensive scanning of genetic variants. These studies typically contain over half a million genetic features for thousands of individuals. Examining this with methods other than univariate statistics is a challenging task requiring advanced algorithms that are scalable to the genome-wide level. In the future, next-generation sequencing studies (NGS) will contain an even larger number of common and rare variants. Machine learning-based feature selection algorithms have been shown to have the ability to effectively create predictive models for various genotype-phenotype relationships. This work explores the problem of selecting genetic variant subsets that are the most predictive of complex disease phenotypes through various feature selection methodologies, including filter, wrapper and embedded algorithms. The examined machine learning algorithms were demonstrated to not only be effective at predicting the disease phenotypes, but also doing so efficiently through the use of computational shortcuts. While much of the work was able to be run on high-end desktops, some work was further extended so that it could be implemented on parallel computers helping to assure that they will also scale to the NGS data sets. Further, these studies analyzed the relationships between various feature selection methods and demonstrated the need for careful testing when selecting an algorithm. It was shown that there is no universally optimal algorithm for variant selection in GWAS, but rather methodologies need to be selected based on the desired outcome, such as the number of features to be included in the prediction model. It was also demonstrated that without proper model validation, for example using nested cross-validation, the models can result in overly-optimistic prediction accuracies and decreased generalization ability. It is through the implementation and application of machine learning methods that one can extract predictive genotype–phenotype relationships and biological insights from genetic data sets.

Diplomityön aiheena oli selvittää onko Suomessa GSM-tukiasemissa käytössä vaiheohjattuja antenniryhmiäja olisiko tällaisten antennien käyttöön kiinnostusta tai mitään es-teitä. Lähtökohtana tälle työlle oli ajatus GSM-tukiasema-antennista, jota voitaisiin kääntää tarpeen mukaan haluttuun suuntaan. Vaiheohjatut antenniryhmät mahdollistavat juuri tällaisen antennin keilan kääntämisen ja muokkaamisen elektronisesti, ilman kuluvia osia. Keilan muitakin ominaisuuksia voidaan säätää, kuten muotoa ja keilojen määrää. Nämä ominaisuudet mahdollistaisivat esimerkiksi ruuhkaisilla alueilla keilojen lisäämisen, jolloin alueen puhelujen välityskapasiteetti kasvaisi.Tarvittaessa voitaisiin myös keilan muotoa muuttaa. Esimerkiksi hätätilanteessasaadaan haluttu keila suunnattua tarkasti tietylle alueelle tai toiselle tukiasemalle ja näin varmistettua kuuluvuus. Myös huoltotoimenpiteet joissakin tapauksissa helpottuisivat. Etenkin vaikeakulkuisissa paikoissa sijaitsevien tukiasemien ensiapu, esimerkiksi antennin fyysisesti kääntyessä, voitaisiin hoitaa etänä kääntämällä pelkkää keilaa ja tässä tapauksessa kääntää antenni tukiaseman normaalin huollon yhteydessä. Suurimpia ongelmakohtia kyseisen tekniikan käyttöönotossa on ollut hinta, mutta muun muassa uusien valmistustekniikoiden avulla vaiheohjattujen antenniryhmien hintoja ollaan saatu pudotettua.

Internet-palvelujen määrä kasvaa jatkuvasti. Henkilöllä on yleensä yksi sähköinen identiteetti jokaisessa käyttämässään palvelussa. Autentikointitunnusten turvallinen säilytys käy yhä vaikeammaksi, kun niitä kertyy yhdet jokaisesta uudesta palvelurekisteröitymisestä. Tämä diplomityö tarkastelee ongelmaa ja ratkaisuja sekä palvelulähtöisestä että teknisestä näkökulmasta. Palvelulähtöisen identiteetinhallinnan liiketoimintakonsepti ja toteutustekniikat – kuten single sign-on (SSO) ja Security Assertion Markup Language (SAML) – käydään läpi karkeiden esimerkkien avulla sekä tutustuen Nokia Account -hankkeessa tuotetun ratkaisun konseptiin ja teknisiin yksityiskohtiin. Nokia Account -palvelun ensimmäisen version toteutusta analysoidaan lopuksi identiteetinhallintapalveluiden suunnitteluperiaatteita ja vaatimuksia vasten.

Coating and filler pigments have strong influence to the properties of the paper. Filler content can be even over 30 % and pigment content in coating is about 85-95 weight percent. The physical and chemical properties of the pigments are different and the knowledge of these properties is important for optimising of optical and printing properties of the paper. The size and shape of pigment particles can be measured by different analysers which can be based on sedimentation, laser diffraction, changes in electric field etc. In this master's thesis was researched particle properties especially by scanning electron microscope (SEM) and image analysis programs. Research included nine pigments with different particle size and shape. Pigments were analysed by two image analysis programs (INCA Feature and Poikki), Coulter LS230 (laser diffraction) and SediGraph 5100 (sedimentation). The results were compared to perceive the effect of particle shape to the performance of the analysers. Only image analysis programs gave parameters of the particle shape. One part of research was also the sample preparation for SEM. Individual particles should be separated and distinct in ideal sample. Analysing methods gave different results but results from image analysis programs corresponded even to sedimentation or to laser diffraction depending on the particle shape. Detailed analysis of the particle shape required high magnification in SEM, but measured parameters described very well the shape of the particles. Large particles (ecd~1 µm) could be used also in 3D-modelling which enabled the measurement of the thickness of the particles. Scanning electron microscope and image analysis programs were effective and multifunctional tools for particle analyses. Development and experience will devise the usability of analysing method in routine use.

Protein tyrosine phosphorylation controls a wide array of cellular responses such as growth, migration, proliferation, differentiation, metabolism and cytoskeletal organisation. Tyrosine phosphorylation is a dynamic process involving the competing activities of protein tyrosine kinases and protein tyrosine phosphatases. The protein tyrosine kinases are further divided into non-receptor- and receptor tyrosine kinases. The latter are transmembrane glycoproteins activated by the binding of specific ligands, mostly growth factors, to their extracellular domain, transmitting different signals to the cell. Growth factor receptors such as the epidermal growth factor receptor, vascular endothelial growth factor receptor 2 and platelet-derived growth factor receptor β, belong to the receptor tyrosine kinases, the signalling of which is often disturbed in various diseases, including cancer. This has led to the development of receptor tyrosine kinase antagonists for use as anti-cancer drugs. As the receptor tyrosine kinases, also the protein tyrosine phosphatases can be divided into receptor- and non-receptor types. The protein tyrosine phosphatases have attained much less attention than the receptor tyrosine kinases partly because they were identified later. However, accumulating evidence shows that the protein tyrosine phosphatases have important roles as specific and active regulators of tyrosine phosphorylation in cells and of physiological processes. Consequently, the protein tyrosine phosphatases are receiving arising interest as novel drug targets. The aim of this work was to elucidate the negative regulation of receptor tyrosine kinases by one non-receptor protein tyrosine phosphatase, T-cell protein tyrosine phosphatase TCPTP. The results show that TCPTP activated by cell adhesion receptor integrin α1 functions as a negative regulator of the epidermal growth factor receptor. It was also found that TCPTP affects vascular endothelial growth factor receptor 2 signalling and angiogenesis. Lastly, a High-throughput screen with 64,280 compounds was performed to identify novel TCPTP activators, resulting in identification of one small molecule compound capable of exerting similar effects on TCPTP signalling as integrin α1. This compound is shown to downregulate signalling of epidermal growth factor receptor and platelet-derived growth factor receptor β, as well as to inhibit cell proliferation and angiogenesis. Our results suggest that a suitable small-molecule TCPTP activator could be utilized in the development of novel anti-cancer drugs.

This paper proposes a calibration method which can be utilized for the analysis of SEM images. The field of application of the developed method is a calculation of surface potential distribution of biased silicon edgeless detector. The suggested processing of the data collected by SEM consists of several stages and takes into account different aspects affecting the SEM image. The calibration method doesn’t pretend to be precise but at the same time it gives the basics of potential distribution when the different biasing voltages applied to the detector.

There are several factors affecting network performance. Some of these can be controlled whereas the others are more fixed. These factors are studied in this thesis from the wide area network (WAN) perspective and the focus is on corporate networks. Another area of interest is the behavior of application protocols when used through WAN. The aim is to study the performance of commonly used application protocols in corporate networks. After identifying the performance problems in corporate WANs the thesis concentrates on methods for improving WAN performance. WAN acceleration is presented as a possible solution. The different acceleration methods are discussed in order to give the reader a theoretical view on how the accelerators can improve WAN performance. Guidelines on the installation of accelerators into a network are also discussed. After a general overview on accelerators is given, one accelerator vendor currently on market is selected for a further analysis. The work is also a case study where two accelerators are installed into a target company network for testing purposes. The tests are performed with three different application protocols that have been identified as critical applications for the target corporation. The aim of the tests is to serve as a proof of concept for WAN acceleration in the target network.

Ultrasuodatus on tehokas entsyymiliuosten konsentrointi- ja puhdistusmenetelmä. Prosessin ongelmana on kuitenkin kalvojen likaantumisesta johtuva suodatuksen aikainen vuon pienentyminen ja kalvojen hankala puhdistaminen. Oikeilla kalvovalinnoilla ja optimaalisilla pesustrategioilla voidaan suodatus- ja pesuaikoja lyhentää ja nain parantaa suodatusprosessin tehokkuutta ja tuotteen laatua. Työn kirjallisessa osassa on käsitelty entsyymiliuosten ja polymeerikalvojen suodatusprosessiin vaikuttavia ominaisuuksia. Työssä on esitelty myös pesuaineiden ja pesukemikaalien ominaisuuksia sekä pesuprosessiin vaikuttavia tekijöitä. Työn kokeellinen osa koostui kahdesta osasta. Työn ensimmäisessä osassa etsittiin entsyymiliuosten ultrasuodatusprosessiin sopivaa kalvoa vertailemalla kalvojen permeabiliteetteja, hydrofiilisyytta, varausta ja peseytyvyyttä. Suodatuksissa käytettiin kahta Roal Oy:n tuottamaa entsyymiliuosta ja suodatukset tehtiin DSS Labstak M20-laitteistolla. Työn toisen osan tarkoituksena oli etsiä entsyymiliuosten suodatuksessa käytettävien kalvojen likaantumisen syyt sekä tehokas puhdistusaine kalvojen pesuun. Tehokasta pesuainetta etsittiin liotuskokeilla ja laboratoriomittakaavan poikkivirtauslaitteistolla. Analysointeja tehtiin muun muassa FTIR- spektroskoopilla, vuomittauksilla, pyyhkäisyelektronimikroskoopilla ja kontaktikulmamittauksilla. Entsyymiliuosten suodatuksiin sopivan kalvon etsinnässä testattujen kalvojen välillä oli suuria eroja niin permeabiliteeteissa kuin likaantumisessa ja puhdistumisessa, mutta mikään kalvoista ei erottunut ylivoimaisesti parhaaksi. Pesuaineista kaupalliset membraanien puhdistukseen tarkoitetut emäksiset pesuaineet osoittautuivat tehokkaimmiksi lian poistajiksi.

Ceramics are widely used in industrial applications due to their advantageous thermal and mechanical stability. Corrosion of ceramics is a great problem resulting in significant costs. Coating is one method of reducing adversities of corrosion. There are several different thin film deposition processes available such as sol-gel, Physical and Chemical Vapour Deposition (PVD and CVD). One of the CVD processes, called Atomic Layer Deposition (ALD) stands out for its excellent controllability, accuracy and wide process capability. The most commonly mentioned disadvantage of this method is its slowness which is partly compensated by its capability of processing large areas at once. Several factors affect the ALD process. Such factors include temperature, the grade of precursors, pulse-purge times and flux of precursors as well as the substrate used. Wrongly chosen process factors may cause loss of self-limiting growth and thus, non-uniformities in the deposited film. Porous substrates require longer pulse times than flat surfaces. The goal of this thesis was to examine the effects of ALD films on surface properties of a porous ceramic material. The analyses applied were for permeability, bubble point pressure and isoelectric point. In addition, effects of the films on corrosion resistance of the substrate in aqueous environment were investigated. After being exposured to different corrosive media the ceramics and liquid samples collected were analysed both mechanically and chemically. Visual and contentual differences between the exposed and coated ceramics versus the untreated and uncoated ones were analysed by scanning electron microscope. Two ALD film materials, dialuminium trioxide and titanium dioxide were deposited on the ceramic substrate using different pulse times. The results of both film materials indicated that surface properties of the ceramic material can be modified to some extent by the ALD method. The effect of the titanium oxide film on the corrosion resistance of the ceramic samples was observed to be fairly small regardless of the pulse time.
