Recognition by the T-cell receptor (TCR) of immunogenic peptides (p) presented by Class I major histocompatibility complexes (MHC) is the key event in the immune response against virus-infected cells or tumor cells. A study of the 2C TCR/SIYR/H-2K(b) system using a computational alanine scanning and a much faster binding free energy decomposition based on the Molecular Mechanics-Generalized Born Surface Area (MM-GBSA) method is presented. The results show that the TCR-p-MHC binding free energy decomposition using this approach and including entropic terms provides a detailed and reliable description of the interactions between the molecules at an atomistic level. Comparison of the decomposition results with experimentally determined activity differences for alanine mutants yields a correlation of 0.67 when the entropy is neglected and 0.72 when the entropy is taken into account. Similarly, comparison of experimental activities with variations in binding free energies determined by computational alanine scanning yields correlations of 0.72 and 0.74 when the entropy is neglected or taken into account, respectively. Some key interactions for the TCR-p-MHC binding are analyzed and some possible side chains replacements are proposed in the context of TCR protein engineering. In addition, a comparison of the two theoretical approaches for estimating the role of each side chain in the complexation is given, and a new ad hoc approach to decompose the vibrational entropy term into atomic contributions, the linear decomposition of the vibrational entropy (LDVE), is introduced. The latter allows the rapid calculation of the entropic contribution of interesting side chains to the binding. This new method is based on the idea that the most important contributions to the vibrational entropy of a molecule originate from residues that contribute most to the vibrational amplitude of the normal modes. The LDVE approach is shown to provide results very similar to those of the exact but highly computationally demanding method.

Prevention of tuberculosis (TB) through vaccination would substantially reduce the global TB burden. Mtb72F/AS02 is a candidate TB vaccine shown to be immunogenic and well tolerated in PPD-negative adults. We evaluated the safety and immunogenicity of Mtb72F/AS02 in Mycobacterium-primed adults (BCG-vaccinated, or infected adults who had received post-exposure chemoprophylaxis or treatment for pulmonary TB disease). In this observer-blind controlled trial, 20 BCG-vaccinated adults and 18 adults previously infected with Mycobacterium tuberculosis (Mtb), were randomized 3:1 to receive three doses of Mtb72F/AS02 or AS02 at one-month intervals, and followed for 6 months post third vaccination. Mtb72F/AS02 was well tolerated in BCG-vaccinated adults, and tended to be more reactogenic in Mtb-infected adults. Adverse events were mainly self-limiting, resolving without sequelae. No serious adverse events were reported. The adverse events in Mtb72F/AS02 vaccinees were not clearly associated with vaccine-induced responses (as assessed by proinflammatory cytokines, total IgE and C-reactive protein levels). No Th2 T-cell responses, or vaccine-induced T-cell responses to Mtb antigens (CFP-10/PPD/ESAT-6) were detected by ICS. In both cohorts, Mtb72F/AS02 induced persistent polyfunctional Mtb72F-specific CD4(+) T-cell responses and anti-Mtb72F humoral responses. IFN-γ was detectable in serum one day post each vaccination. Further evaluation of the candidate vaccine, Mtb72F/AS02, is warranted. Trial registration: ClinicalTrials.gov identifier: NCT00146744.

Some of the anti-neoplastic effects of anthracyclines in mice originate from the induction of innate and T cell-mediated anticancer immune responses. Here we demonstrate that anthracyclines stimulate the rapid production of type I interferons (IFNs) by malignant cells after activation of the endosomal pattern recognition receptor Toll-like receptor 3 (TLR3). By binding to IFN-α and IFN-β receptors (IFNARs) on neoplastic cells, type I IFNs trigger autocrine and paracrine circuitries that result in the release of chemokine (C-X-C motif) ligand 10 (CXCL10). Tumors lacking Tlr3 or Ifnar failed to respond to chemotherapy unless type I IFN or Cxcl10, respectively, was artificially supplied. Moreover, a type I IFN-related signature predicted clinical responses to anthracycline-based chemotherapy in several independent cohorts of patients with breast carcinoma characterized by poor prognosis. Our data suggest that anthracycline-mediated immune responses mimic those induced by viral pathogens. We surmise that such 'viral mimicry' constitutes a hallmark of successful chemotherapy.

The goal of adoptive T cell therapy in cancer is to provide effective antitumor immunity by transfer of selected populations of tumor Ag-specific T cells. Transfer of T cells with high TCR avidity is critical for in vivo efficacy. In this study, we demonstrate that fluorescent peptide/MHC class I multimeric complexes incorporating mutations in the alpha3 domain (D227K/T228A) that abrogate binding to the CD8 coreceptor can be used to selectively isolate tumor Ag-specific T cells of high functional avidity from both in vitro expanded and ex vivo T cell populations. Sorting, cloning, and expansion of alpha3 domain mutant multimer-positive CD8 T cells enabled rapid selection of high avidity tumor-reactive T cell clones. Our results are relevant for ex vivo identification and isolation of T cells with potent antitumor activity for adoptive T cell therapy.

MHC-peptide multimers containing biotinylated MHC-peptide complexes bound to phycoerythrin (PE) streptavidin (SA) are widely used for analyzing and sorting antigen-specific T cells. Here we describe alternative T cell-staining reagents that are superior to conventional reagents. They are built on reversible chelate complexes of Ni(2+)-nitrilotriacetic acid (NTA) with oligohistidines. We synthesized biotinylated linear mono-, di-, and tetra-NTA compounds using conventional solid phase peptide chemistry and studied their interaction with HLA-A*0201-peptide complexes containing a His(6), His(12), or 2×His(6) tag by surface plasmon resonance on SA-coated sensor chips and equilibrium dialysis. The binding avidity increased in the order His(6) < His(12) < 2×His(6) and NTA(1) < NTA(2) < NTA(4), respectively, depending on the configuration of the NTA moieties and increased to picomolar K(D) for the combination of a 2×His(6) tag and a 2×Ni(2+)-NTA(2). We demonstrate that HLA-A2-2×His(6)-peptide multimers containing either Ni(2+)-NTA(4)-biotin and PE-SA- or PE-NTA(4)-stained influenza and Melan A-specific CD8+ T cells equal or better than conventional multimers. Although these complexes were highly stable, they very rapidly dissociated in the presence of imidazole, which allowed sorting of bona fide antigen-specific CD8+ T cells without inducing T cell death as well as assessment of HLA-A2-peptide monomer dissociation kinetics on CD8+ T cells.

Purpose: SIOPEN scoring of 123I mIBG imaging has been shown to predict response to induction chemotherapy and outcome at diagnosis in children with HRN.Method: Patterns of skeletal 123I mIBG uptake were assigned numerical scores (Mscore) ranging from 0 (no metastasis) to 72 (diffuse metastases) within 12 body areas as described previously. 271 anonymised, paired image data sets acquired at diagnosis and on completion of Rapid COJEC induction chemotherapy were reviewed, constituting a representative sample of 1602 children treated prospectively within the HR-NBL1/SIOPEN trial. Pre-and post-treatment Mscores were compared with bone marrow cytology (BM) and 3 year event free survival (EFS).Results: Results 224/271 patients showed skeletal MIBG-uptake at diagnosis and were evaluable forMIBG-response. Complete response (CR) on MIBG to Rapid COJEC induction was achieved by 66%, 34% and 15% of patients who had pre-treatment Mscores of <18 (n¼65, 29%), 18-44 (n¼95,42%) and Y ´ 45 (n¼64, 28.5%) respectively (chi squared test p&lt;.0001). Mscore at diagnosis and on completion of Rapid COJEC correlated strongly with BM involvement (p&lt;0.0001). The correlation of pre score with post scores and response was highly significant (p&lt;0.001). Most importantly, the 3 year EFS in 47 children with Mscore 0 at diagnosis was 0.68 (A ` 0.07), by comparison with 0.42 (A` 0.06), 0.35 (A` 0.05) and 0.25 (A` 0.06) for patients in pre-treatment score groups <18, 18-44 and Y ´ 45, respectively (p&lt;0.001). AnMscore threshold ofY ´ 45 at diagnosis was associated with significantly worse outcome by comparison with all other Mscore groups (¼0.029). The 3 year EFS of 0.53 (A` 0.07) of patients in metastatic CR (mIBG and BM) after Rapid Cojec (33%) is clearly superior to patients not achieving metastatic CR (0.24 (A ` 0.04), ¼0.005).Conclusion: SIOPEN scoring of 123I mIBG imaging has been shown to predict response to induction chemotherapy and outcome at diagnosis in children with HRN.

Purpose: To date, the genotype/phenotype correlation of p.G56R-linked autosomal dominant retinitis pigmentosa (ADRP) is limited to less than 10 kindred. The purpose of this study is to report an unusual appearance of fundus autofluorescence (AF) with NR2E3 p.G56R-linked ADRP in a single kindred.Methods: Patients were enrolled among three generations in a previously unreported family. Molecular diagnosis was performed on all exons of NR2E3 and a p.G56R mutation was identified in affected family members only. Examinations included fundus photography, visual fields, optical coherence tomography, AF, near-infrared AF and ISCEV-standard electrophysiology (ERG).Results: Among 10 examined family members, 5 were affected. The youngest and oldest patients were 16 and 65 years old, respectively. Fundus examination revealed a range of retinal disorder from normal to optic nerve pallor, attenuated arterial caliber and bone spicule-like pigment deposits. In all patients, AF showed a double hyperfluorescent ring; an inner paramacular ring which extension was comparable among patients and an outer ring along the vascular arcades which extended towards periphery in older patients and became hypofluorescent. Maximal scotopic ERGs when recordable showed an increased a/b wave ratio.Conclusions: A double hyperfluorescent ring on AF is an uncommon observation and might be a specific clinical finding in NR2E3 p.G56R-linked ADRP. The consistency of that finding in all affected members of our 3-generation family confirms a previous study. Further analysis is required to determine whether AF changes are associated with particular retinal layer abnormalities.

OBJECTIVES: In this population-based study, reference values were generated for renal length, and the heritability and factors associated with kidney length were assessed. METHODS: Anthropometric parameters and renal ultrasound measurements were assessed in randomly selected nuclear families of European ancestry (Switzerland). The adjusted narrow sense heritability of kidney size parameters was estimated by maximum likelihood assuming multivariate normality after power transformation. Gender-specific reference centiles were generated for renal length according to body height in the subset of non-diabetic non-obese participants with normal renal function. RESULTS: We included 374 men and 419 women (mean ± SD, age 47 ± 18 and 48 ± 17 years, BMI 26.2 ± 4 and 24.5 ± 5 kg/m(2), respectively) from 205 families. Renal length was 11.4 ± 0.8 cm in men and 10.7 ± 0.8 cm in women; there was no difference between right and left renal length. Body height, weight and estimated glomerular filtration rate (eGFR) were positively associated with renal length, kidney function negatively, age quadratically, whereas gender and hypertension were not. The adjusted heritability estimates of renal length and volume were 47.3 ± 8.5 % and 45.5 ± 8.8 %, respectively (P&#8201;< 0.001). CONCLUSION: The significant heritability of renal length and volume highlights the familial aggregation of this trait, independently of age and body size. Population-based references for renal length provide a useful guide for clinicians. KEY POINTS: • Renal length and volume are heritable traits, independent of age and size. • Based on a European population, gender-specific reference values/percentiles are provided for renal length. • Renal length correlates positively with body length and weight. • There was no difference between right and left renal lengths in this study. • This negates general teaching that the left kidney is larger and longer.

Introduction. Selective embolization of the left-gastric artery (LGA) reduces levels of ghrelin and achieves significant short-term weight loss. However, embolization of the LGA would prevent the performance of bariatric procedures because the high-risk leakage area (gastroesophageal junction [GEJ]) would be devascularized. Aim. To assess an alternative vascular approach to the modulation of ghrelin levels and generate a blood flow manipulation, consequently increasing the vascular supply to the GEJ. Materials and methods. A total of 6 pigs underwent a laparoscopic clipping of the left gastroepiploic artery. Preoperative and postoperative CT angiographies were performed. Ghrelin levels were assessed perioperatively and then once per week for 3 weeks. Reactive oxygen species (ROS; expressed as ROS/mg of dry weight [DW]), mitochondria respiratory rate, and capillary lactates were assessed before and 1 hour after clipping (T0 and T1) and after 3 weeks of survival (T2), on seromuscular biopsies. A celiac trunk angiography was performed at 3 weeks. Results. Mean (±standard deviation) ghrelin levels were significantly reduced 1 hour after clipping (1902 ± 307.8 pg/mL vs 1084 ± 680.0; P = .04) and at 3 weeks (954.5 ± 473.2 pg/mL; P = .01). Mean ROS levels were statistically significantly decreased at the cardia at T2 when compared with T0 (0.018 ± 0.006 mg/DW vs 0.02957 ± 0.0096 mg/DW; P = .01) and T1 (0.0376 ± 0.008mg/DW; P = .007). Capillary lactates were significantly decreased after 3 weeks, and the mitochondria respiratory rate remained constant over time at the cardia and pylorus, showing significant regional differences. Conclusions. Manipulation of the gastric flow targeting the gastroepiploic arcade induces ghrelin reduction. An endovascular approach is currently under evaluation.

I. Facteurs associés avec l'infection tuberculose latent chez les requérants d'asile entrant dans le canton de Vaud : Une étude transversale dans le canton de Vaud. Objectifs : Les objectifs de cette étude étaient l'identification des facteurs associés à l'infection tuberculeuse latente (ITBL) chez les requérants d'asile récemment arrivés au Canton de Vaud et leur utilisation pondérée pour l'élaboration d'un score prédictif qui pourrait permettre la meilleure sélection des individus à dépister avec les Interferon Gamma Release Assays (IGRA). Méthode : Le protocole de l'étude prévoyait l'inclusion des requérants d'asile de plus de 16 ans, récemment arrivés dans deux centre de requérant du canton de Vaud ceux de Sainte-Croix et de Crissier. De septembre 2009 à juillet 2010 les requérants d'asile ont bénéficié lors des visites au centre de soins infirmier (CSI) d'informations sur l'ITBL et le protocole et les enjeux de l'étude. Les requérants d'asile ont d'emblée été informées que leur participation à l'étude n'aurait pas d'impact sur le débouché de leur dossier d'asile et qu'il n'y aurait pas de compensation financière à leur participation. Après avoir signé le consentement éclairé les requérants d'asile bénéficiaient d'une entrevue avec l'infirmière du centre où un questionnaire démographique et médical était remplit. 10cc de sang étaient prélevés à la fin de l'entrevue pour l'examen IGRA. Les patients présentant des symptômes évocateurs de tuberculose active ou un anamnèse de traitement pour une tuberculose active étaient exclus de l'étude et adressés au médecin référant du centre pour une visite médicale. Selon les résultats du test T-SPOT.TB (IGRA), les requérants étaient classés en deux groupes : positifs et négatifs. Le groupe IGRA positif était adressé au médecin référant. L'analyse statistique des données de l'étude a été réalisée par le logiciel STATA 11.2. Les coefficients de l'analyse multivariée ont été combinées pour la création d'un score pronostic dont la puissance de discrimination a été évaluée par une courbe ROC. Le protocole de l'étude avait reçu l'aval de la commission d'éthique de l'Université de Lausanne. Résultats : Durant la ©riode de l'étude, 788 requérants d'asile ont été hébergés dans les deux centres de l'étude. 639 avaient plus de 16 ans et 393 d'entre eux ont partici© à l'étude (61.50%). 295 (75.06%) avaient un IGRA négatif et 98 (24.93%) étaient positifs. A noter que parmi les 98 positifs, 5 avaient une tuberculose active non détecté précédemment. Les analyses univarié et multivarié ont permis d'identifier 6 facteurs associées à l'ITBL : Région d'origine, moyen de transport, état civil, âge, toux et antécédent d'exposition à la tuberculose. Le score élaboré en combinant ces 6 facteurs présente un AUC de 81% avec une sensibilité de 80%, une s©cificité de 70% et des valeurs prédictive positive et négative respectivement de 45% et 92% quant un seuil de 13 est utilisé. Conclusion : Les requérants d'asile qui immigrent en Suisse proviennent de pays où l'incidence de la tuberculose est su©rieure à celle des pays de l'Europe occidentale et présentent un risque élevé pour l'infection tuberculose latente (ITBL). L'origine comme seul facteur n'est pas suffisant pour stratifier le risque d'ITBL et ne peut pas justifier la prescription d'un traitement préventif d'ITBL. L'introduction des tests de détection, hautement s©cifiques de l'infection au M. tuberculosis tel que les IGRA ainsi que le taux élevé de réussite des traitements préventifs de l'infection latente ont ouvert la voie à un dépistage précoce de l'ITBL qui compléterait le dépistage de la tuberculose active actuellement effectué à la frontière. Afin de mieux cibler le dépistage par ces tests une meilleure sélection des individus à dépister est im©rative. Elle pourrait se faire en évaluant le score individuel de risque ITBL par requérant. -- II. Taux élevé d'adhérence au traitement préventif de l'infection tuberculeuse latente prescrit à un collectif de requérants d'asile dans un canton suisse. Objectifs: L'efficacité du traitement préventif de l'infection tuberculeuse latente dépend de l'adherence du sujet au traitement. Un traitement bien conduit pour une duré prévue est en mesure de prévenir l'activation des cas d'infection tuberculeuse latente (ITBL). Le plus grand enjeu dans un programme préventif pour la tuberculose est, outre de cibler la détection des individus les plus à risque pour l'ITBL, de pouvoir traiter efficacement le collectif dépisté positif. Cette étude évaluait la faisabilité d'un traitement préventif court parmi un collectif de requérants d'asile porteurs d'une ITBL dans le canton de Vaud. Méthode: Nous avons effectué une étude prospective de cohorte parmi des requérants d'asile récemment attribués dans le canton de Vaud, âgés de plus de 16 ans et qui avaient été dépistés positifs par IGRA. L'ensemble du collectif selon le protocole de l'étude était adressé au médecin référant afin d'exclure une tuberculose active et pour discuter du traitement préventif si le diagnostic d'ITBL était confirmé. Lors de la première visite médicale, outre l'examen clinique, un bilan radiologique avec une radiographie du thorax et un bilan de la biologie hépatique ainsi qu'un test de dépistage HIV était proposé à l'ensemble du collectif. En cas de suspicion clinique ou d'image radiologique suspecte de tuberculose active le sujet était adressé pour des examens complémentaires. Les sujets porteurs d'ITBL se voyaient proposés, en l'absence de contre indications, un traitement de rifampicine de quatre mois. En acceptant de participer à l'étude ils s'engageaient de se présenter à leur contrôle médical mensuel où était évaluée l'adhérence au traitement et l'apparition d'effets indésirable ou de complications. Si l'adhérence était jugée correcte l'ordonnance du traitement était renouvelée d'un mois et le requérant recevait son prochain rendez-vous de contrôle. L'adhérence était considéré satisfaisante si le patient était adhérent à son schéma de visites médicales et demandait le renouvellement de son ordonnance. Si le requérant d'asile ne se présentait pas à deux contrôles il était considéré comme non adhérent et son traitement est suspendu. Résultats : Notre collectif comptait 98 sujet présument atteint de ITBL sur la base du test T-SPOT.TB ce qui représentait 24.9% du collectif initial. L'âge moyen était de 26.7 ans, 74% était des hommes. La majorité étaient des africains: 66 %, 17% étaient asiatiques et les populations balkaniques et de l'exunion soviétique étaient représentés à part égale d'huit pourcent. Parmi notre collectif nous n'avions pas de sujet immunodéficient notamment HIV positif. Des 98 sujets, 11 ne se sont pas présenté à leur visite médicale initiale. La visite médicale initiale a permis la détection de 8 patients porteurs d'une tuberculose active, dont cinq ont reçu un traitement antituberculeux, ou d'une autre affection pulmonaire non tuberculeuse. Chez deux patients il y avait une contre-indication au traitement préventif et deux avaient un anamnèse positif de traitement antituberculeux non précédemment déclaré. Le traitement préventif a été prescrit à 74 requérants d'asile. Durant le suivi mensuel trois requérants ne se sont pas présentés lors de la première visite de suivi, trois lors de la seconde et sept lors de la troisième pour un total de 13 sujets. Chez deux sujets le traitement préventif a du être suspendu à cause d'une adhérence problématique secondaire à des abus de substances illégales. Durant le suivi, nous n'avons pas eu de sérieuses complications ni d'effets indésirables au traitement qui auraient nécessité son arrêt. En final 60/75 des sujets ont achevé leur traitement soit 80% du collectif. Conclusion: Malgré la vulnérabilité et la volatilité inhérente à cette population qui est d'ailleurs la plus à risqué de réactivation d'une ITBL, cette étude montre que il est possible d'obtenir de taux d'adhérence très élevés au traitement préventif. Nous considérons que les conditions qui ont permis ces résultats sont la prescription d'un schéma de traitement préventif court, un suivi médico-soignant régulier et l'hébergement contrôlée et stable où résidait notre collectif.

BACKGROUND: Zebrafish is a clinically-relevant model of heart regeneration. Unlike mammals, it has a remarkable heart repair capacity after injury, and promises novel translational applications. Amputation and cryoinjury models are key research tools for understanding injury response and regeneration in vivo. An understanding of the transcriptional responses following injury is needed to identify key players of heart tissue repair, as well as potential targets for boosting this property in humans. RESULTS: We investigated amputation and cryoinjury in vivo models of heart damage in the zebrafish through unbiased, integrative analyses of independent molecular datasets. To detect genes with potential biological roles, we derived computational prediction models with microarray data from heart amputation experiments. We focused on a top-ranked set of genes highly activated in the early post-injury stage, whose activity was further verified in independent microarray datasets. Next, we performed independent validations of expression responses with qPCR in a cryoinjury model. Across in vivo models, the top candidates showed highly concordant responses at 1 and 3 days post-injury, which highlights the predictive power of our analysis strategies and the possible biological relevance of these genes. Top candidates are significantly involved in cell fate specification and differentiation, and include heart failure markers such as periostin, as well as potential new targets for heart regeneration. For example, ptgis and ca2 were overexpressed, while usp2a, a regulator of the p53 pathway, was down-regulated in our in vivo models. Interestingly, a high activity of ptgis and ca2 has been previously observed in failing hearts from rats and humans. CONCLUSIONS: We identified genes with potential critical roles in the response to cardiac damage in the zebrafish. Their transcriptional activities are reproducible in different in vivo models of cardiac injury.

A consecutive series of 353 patients who underwent Lichtenstein mesh repair for inguinal hernia from the 1st of July 1994 to the 30th of July 1995 were studied. We analysed our indication, technique, complications, follow-up and outcome. Special consideration was given to the advantages and acceptance of day-case surgery. Our results suggest that the Lichtenstein repair should be considered as a new standard procedure, especially outside of hernia centres.

Mycorrhizal symbioses--the union of roots and soil fungi--are universal in terrestrial ecosystems and may have been fundamental to land colonization by plants. Boreal, temperate and montane forests all depend on ectomycorrhizae. Identification of the primary factors that regulate symbiotic development and metabolic activity will therefore open the door to understanding the role of ectomycorrhizae in plant development and physiology, allowing the full ecological significance of this symbiosis to be explored. Here we report the genome sequence of the ectomycorrhizal basidiomycete Laccaria bicolor (Fig. 1) and highlight gene sets involved in rhizosphere colonization and symbiosis. This 65-megabase genome assembly contains approximately 20,000 predicted protein-encoding genes and a very large number of transposons and repeated sequences. We detected unexpected genomic features, most notably a battery of effector-type small secreted proteins (SSPs) with unknown function, several of which are only expressed in symbiotic tissues. The most highly expressed SSP accumulates in the proliferating hyphae colonizing the host root. The ectomycorrhizae-specific SSPs probably have a decisive role in the establishment of the symbiosis. The unexpected observation that the genome of L. bicolor lacks carbohydrate-active enzymes involved in degradation of plant cell walls, but maintains the ability to degrade non-plant cell wall polysaccharides, reveals the dual saprotrophic and biotrophic lifestyle of the mycorrhizal fungus that enables it to grow within both soil and living plant roots. The predicted gene inventory of the L. bicolor genome, therefore, points to previously unknown mechanisms of symbiosis operating in biotrophic mycorrhizal fungi. The availability of this genome provides an unparalleled opportunity to develop a deeper understanding of the processes by which symbionts interact with plants within their ecosystem to perform vital functions in the carbon and nitrogen cycles that are fundamental to sustainable plant productivity.

Despite the advantage of avoiding the costs of sexual reproduction, asexual vertebrates are very rare and often considered evolutionarily disadvantaged when compared to sexual species. Asexual species, however, may have advantages when colonizing (new) habitats or competing with sexual counterparts. They are also evolutionary older than expected, leaving the question whether asexual vertebrates are not only rare because of their 'inferior' mode of reproduction but also because of other reasons. A paradigmatic model system is the unisexual Amazon molly, Poecilia formosa, that arose by hybridization of the Atlantic molly, Poecilia mexicana, as the maternal ancestor, and the sailfin molly, Poecilia latipinna, as the paternal ancestor. Our extensive crossing experiments failed to resynthesize asexually reproducing (gynogenetic) hybrids confirming results of previous studies. However, by producing diploid eggs, female F(1) -hybrids showed apparent preadaptation to gynogenesis. In a range-wide analysis of mitochondrial sequences, we examined the origin of P. formosa. Our analyses point to very few or even a single origin(s) of its lineage, which is estimated to be approximately 120,000 years old. A monophyletic origin was supported from nuclear microsatellite data. Furthermore, a considerable degree of genetic variation, apparent by high levels of clonal microsatellite diversity, was found. Our molecular phylogenetic evidence and the failure to resynthesize the gynogenetic P. formosa together with the old age of the species indicate that some unisexual vertebrates might be rare not because they suffer the long-term consequences of clonal reproduction but because they are only very rarely formed as a result of complex genetic preconditions necessary to produce viable and fertile clonal genomes and phenotypes ('rare formation hypothesis').
