HER2 gene amplification is observed in about 15% of breast cancers. The subgroup of HER2-positive breast cancers appears to be heterogeneous and presents complex patterns of gene amplification at the locus on chromosome 17q12-21. The molecular variations within the chromosome 17q amplicon and their clinical implications remain largely unknown. Besides the well-known TOP2A gene encoding Topoisomerase IIA, other genes might also be amplified and could play functional roles in breast cancer development and progression. This review will focus on the current knowledge concerning the HER2 amplicon heterogeneity, its clinical and biological impact and the pitfalls associated with the evaluation of gene amplifications at this locus, with particular attention to TOP2A and the link between TOP2A and anthracycline benefit. In addition it will discuss the clinical and biological implications of the amplification of ten other genes at this locus (MED1, STARD3, GRB7, THRA, RARA, IGFPB4, CCR7, KRT20, KRT19 and GAST) in breast cancer.

There are at least six psychotherapeutic treatments of personality disorders having received empirical and clinical validation in terms of their efficacy. These treatments are based on different theoretical models, namely the cognitive-behavioural, psychodynamic and interpersonal models. This article briefly presents these treatments, focusing on the process of therapeutic change. It is assumed that the process of emotional activation is one of the most interesting theoretical psychotherapy ingredient in treatments of these patients. The treatments are discussed regarding this hypothesis and its clinical implications.

BACKGROUND: This study was designed to determine whether the pain pattern in patients with an internal mammary artery (IMA) harvest differs from that in other cardiac operations and whether these patients present specific characteristics with clinical implications. METHODS: One hundred patients with left IMA grafting (IMA group) were compared prospectively with 100 patients who had a heart operation without IMA harvest (non-IMA group). Pain assessment was performed on postoperative days (POD) 1, 2, 3, and 7, and included pain intensity (10-point scale) and pain localization. RESULTS: In the IMA group, pain intensity was higher on POD 2 (4.2 +/- 2.4 versus 3.2 +/- 2.3, p < 0.01), and there were more patients without pain on POD 7 (32 versus 19, p = 0.03). In the IMA group, more patients had left basal thoracic pain throughout the entire study period and had sternal pain on POD 7, whereas more patients in the non-IMA group complained about back pain during the early postoperative period. CONCLUSIONS: The impact of IMA harvest on pain intensity is moderate, but the pain localization pattern of each group exhibits specific features that could help to better target pain management.

Primary objectives: Awake surgeries of slow-growing tumours invading the brain and guided by direct electrical stimulation induce major brain reorganizations accompanied with slight impairments post-operatively. In most cases, these deficits are so slight after a few days that they are often not detectable on classical neuropsychological evaluations. Consequently, this study investigated whether simple visuo-manual reaction time paradigms would sign some level of functional asymmetries between both hemispheres. Importantly, the visual stimulus was located in the saggital plane in order to limit attentional biases and to focus mainly on the inter-hemispheric asymmetry. Methods and procedures: Three patients (aged 41, 59 and 59 years) after resections in parietal regions and a control group (age¼44, SD¼6.9) were compared during simple uni- and bimanual reaction times (RTs). Main outcomes and results: Longer RTs were observed for the contralesional compared to the ipsilesional hand in the unimanual condition. This asymmetry was reversed for the bimanual condition despite longer RTs. Conclusion and clinical implications: Reaction time paradigms are useful in these patients to monitor more precisely their functional deficits, especially their level of functional asymmetry, and to understand brain (re)organization following slowgrowing lesions.

Context: Ovarian tumors (OT) typing is a competency expected from pathologists, with significant clinical implications. OT however come in numerous different types, some rather rare, with the consequence of few opportunities for practice in some departments. Aim: Our aim was to design a tool for pathologists to train in less common OT typing. Method and Results: Representative slides of 20 less common OT were scanned (Nano Zoomer Digital Hamamatsu®) and the diagnostic algorithm proposed by Young and Scully applied to each case (Young RH and Scully RE, Seminars in Diagnostic Pathology 2001, 18: 161-235) to include: recognition of morphological pattern(s); shortlisting of differential diagnosis; proposition of relevant immunohistochemical markers. The next steps of this project will be: evaluation of the tool in several post-graduate training centers in Europe and Québec; improvement of its design based on evaluation results; diffusion to a larger public. Discussion: In clinical medicine, solving many cases is recognized as of utmost importance for a novice to become an expert. This project relies on the virtual slides technology to provide pathologists with a learning tool aimed at increasing their skills in OT typing. After due evaluation, this model might be extended to other uncommon tumors.

OBJECTIVES: The pharmacokinetic (PK) parameters of artesunate, recently discovered to possess promising trematocidal activity, and its main metabolite dihydroartemisinin (DHA) were determined in rats infected with hepatic and biliary stages of Fasciola hepatica and compared with uninfected rats after single intragastric and intravenous (iv) doses. METHODS: Rats infected with F. hepatica for 25 and 83 days and uninfected rats were cannulated in the right jugular vein and blood samples were withdrawn at selected timepoints following 10 mg/kg of iv and a single 100 mg/kg oral dose of artesunate. Plasma was analysed for artesunate and DHA by liquid chromatography coupled to tandem mass spectrometry. RESULTS: Rats harbouring juvenile and adult F. hepatica infections revealed considerable changes in PK parameters of artesunate and DHA. Following oral administration, maximum plasma concentrations (C(max)) of artesunate and DHA were 1.8-2.3-fold higher in infected rats [artesunate: 1334 +/- 1404 ng/mL (no infection) versus 2454 +/- 1494 ng/mL (acute infection) and 2768 +/- 538 ng/mL (chronic infection); DHA: 3802 +/- 2149 ng/mL (no infection) versus 6507 +/- 3283 ng/mL (acute infection) and 9093 +/- 884 ng/mL (chronic infection)]. The AUCs of artesunate and DHA were 2.1-4.4-fold greater in infected rats. An opposite trend was observed after iv injection. C(max) and AUC of artesunate and DHA following iv dosing were 5784 +/- 3718 and 140 938 +/- 128 783 ng.min/mL and 3849 +/- 3060 and 86 107 +/- 41 863 ng.min/mL, respectively, in uninfected rats versus 2623 +/- 1554 and 21 617 +/- 12 230 ng.min/mL and 2835 +/- 980 and 64 290 +/- 29 057 ng.min/mL, respectively, in rats harbouring a chronic infection. The elimination half-lives (t(1/2)) of artesunate and DHA were considerably altered in infected rats following oral and iv administration of artesunate. CONCLUSIONS: F. hepatica infections strongly influence the disposition kinetics of artesunate and its metabolite in rats. The clinical implications of this finding need to be carefully studied.

Child sexual abuse is associated with problems in children's emotional development, particularly increased insecurity of attachment. However, few studies have examined its effect on the organization of attachment representations in preschoolers, and the findings of those that have been conducted have not been entirely consistent. Therefore, this study aims to analyze the effect of child sexual abuse on attachment representation quality in a sample of children 3 to 7 years old in Chile. The results indicate child sexual abuse does affect children's attachment representation quality. The attachment narratives of child sexual abuse victims scored significantly higher than nonvictims on the hyperactivity and disorganization dimensions of attachment. These results are discussed in terms of attachment theory, clinical findings on child sexual abuse, and clinical implications.

Vascular calcification is a hallmark of advanced atherosclerosis. Here we show that deletion of the nuclear receptor PPARγ in vascular smooth muscle cells of low density lipoprotein receptor (LDLr)-deficient mice fed an atherogenic diet high in cholesterol, accelerates vascular calcification with chondrogenic metaplasia within the lesions. Vascular calcification in the absence of PPARγ requires expression of the transmembrane receptor LDLr-related protein-1 in vascular smooth muscle cells. LDLr-related protein-1 promotes a previously unknown Wnt5a-dependent prochondrogenic pathway. We show that PPARγ protects against vascular calcification by inducing the expression of secreted frizzled-related protein-2, which functions as a Wnt5a antagonist. Targeting this signalling pathway may have clinical implications in the context of common complications of atherosclerosis, including coronary artery calcification and valvular sclerosis.

We investigated the moderating effect of family relationships on the links between maternal postpartum depression and child symptoms in a low-risk community sample of families with 3-month-old infants (n = 57). The level of maternal depression was assessed by the Montgomery-Asberg Depression Rating Scale from a clinical interview, child symptoms by the Symptom Check List completed by both parents, and family relationships by direct observation of father-mother-baby interactions (Lausanne Trilogue Play). Families were categorized as high coordination or low coordination from their overall coordination level throughout the play. Results showed no significant links between maternal depression level and child symptoms reported by both parents. Mothers with a high depressive level in high coordination families tended to report more symptoms in their child than did mothers with lower depressive scores, whereas this link was not found in low coordination families. Prevention perspectives and clinical implications of these results are discussed.

Résumé pour large public Unité de Biochimie et Psychopharmacologie Clinique, Centre de neurosciences Psychiatrique, Département de Psychiatrie Adulte, Faculté de Biologie et de Médecine, Université de Lausanne Lors de la prise d'un médicament, celui-ci va passer par différentes étapes que sont l'absorption, la distribution, le métabolisme et enfin l'élimination. Ces quatre étapes sont regroupées sous le nom de pharmacocinétique. A noter que ces quatre paramètres sont dynamiques et en constante évolution. Durant cette thèse, nous avons investigué différents aspects de la pharmacocinétique, tout d'abord par une revue de la littérature sur la glycoprotéine-P (Pgp). Récemment découverte, cette protéine de membrane est située aux endroits stratégiques de l'organisme comme la barrière hématoencéphalée, le placenta ou les intestins où elle influencera l'entrée de différentes substances, en particulier les médicaments. La Pgp serait impliquée dans les phénomènes de résistances aux agents thérapeutiques en oncologie. La Pgp influence donc l'absorption des médicaments, et son impact en clinique, en termes d'efficacité de traitement et de toxicité prend chaque jour plus d'importance. Ensuite nous avons mis au point une méthode d'analyse quantitative d'un antidépresseur d'une nouvelle génération : la mirtazapine (Remeron®). La nouveauté réside dans la façon dont la mirtazapine interagit avec les neurotransmetteurs impliqués dans la dépression que sont la sérotonine et la noradrénaline. Cette méthode utilise la chromatographie liquide pour séparer la mirtazapine de ses principaux métabolites dans le sang. La spectrométrie de masse est utilisée pour les détecter et les quantifier. Les métabolites sont des substances issues de réactions chimiques entre la substance mère, la mirtazapine, et généralement des enzymes hépatiques, dans le but de rendre cette substance plus soluble en vue de son élimination. Cette méthode permet de quantifier la mirtazapine et ses métabolites dans le sang de patients traités et de déterminer la variation des taux plasmatiques chez ces patients. Puis nous avons étudié le métabolisme d'un autre antidépresseur, le citalopram, qui a un métabolisme complexe. Le citalopram est un racémate, c'est-à-dire qu'il existe sous forme de deux entités chimiques (R-(-) et S-(+) citalopram) qui ont le même nombre d'éléments mais arrangés différemment dans l'espace. La voie métabolique cérébrale du citalopram est sous le contrôle d'une enzyme, la monoamine oxydase (MAO), conduisant à une forme acide du citalopram (l'acide propionique du citalopram). La MAO existe sous deux formes : MAO-A et MAO-B. Nous avons utilisé des souris déficientes d'un gène, celui de la MAO-A, pour mieux en comprendre le métabolisme en les comparants à des souris sauvages (sans déficience de ce gène). Nous avons utilisé le citalopram et deux de ses métabolites (le déméthylcitaloprarn et le didéméthyícitalopram) comme substrats pour tester la formation in vitro de l'acide propionique du citalopram. Nos résultats montrent que la MAO-A favorise la formation de l'entité R-(-) et présente une plus grande affinité pour le citalopram, tandis que la MAO-B métabolise préférentiellement l'entité S-(+) et a une plus grande affinité pour les deux métabolites déméthylés. De plus, la déficience en MAO-A est partiellement compensée parla MAO-B chez les souris déficientes du gène de la MAO-A. Enfin, nous avons étudié une deuxième voie métabolique du citalopram qui s'est avérée toxique chez le chien Beagle. Celle-ci est catalysée par une autre famille d'enzymes, les cytochromes P-450, et mène aux métabolites déméthylés et didéméthylés du citalopram. Nous avons utilisé des tissus hépatiques de chiens Beagle. Plusieurs cytochromes P-450 sont impliqués dans le métabolisme du citalopram menant à sa forme déméthylée, ceci tant chez l'homme que chez le chien. Par contre, dans le métabolisme de la forme déméthylée menant à 1a forme didéméthylée, un seul cytochrome P-450 serait impliqué chez l'Homme, tandis qu'ils seraient plusieurs chez le chien. L'activité enzymatique produisant la forme didéméthylée est beaucoup plus importante chez le chien comparé à l'homme. Cette observation soutien l'hypothèse que des taux élevés de la forme didéméthylée participent à la toxicité spécifique du citalopram chez le chien. Nous pouvons conclure que plusieurs famille d'enzymes sont impliquées tant au niveau cérébral qu'hépatique dans la métabolisation de médicaments psychotropes. Sachant que les enzymes peuvent être stimulées ou inhibées, il importe de pouvoir suivre au plus prés les taux plasmatiques des différents psychotropes et de leurs métabolites. Les substances issues de cette métabolisation ne sont pas toujours dotées d'une activité pharmacologique. Il s'est avéré par conséquent indispensable de suivre les taux plasmatiques d'une substance et de ses métabolites et d'établir ou non l'existence d'une relation avec l'effet clinique observé. Ce concept nommé « therapeutic drag monitoring » (TDM) est particulièrement utile en psychiatrie ou un manque de compliance des patients est fréquemment observé. Les médicaments psychotropes ont un métabolisme principalement hépatique (cytochromes P-450) et parfois cérébral (monoamines oxydases), comme pour le citalopram par exemple. Une méthode stéréosélective de chromatographie liquide couplée à la spectrométrie de masse a été développée pour analyser les énantiomères R-(-) et S-(+) d'un antidépresseur agissant sur les récepteurs noradrénergiques et sérotoninergiques, la mirtazapine et de ses métabolites déméthylmirtazapine et 8-hydroxymirtazapine. Les données préliminaires obtenues dans les plasmas dosés suggèrent que les concentrations de R-(-)-mirtazapine sont plus élevées que celles de S-(+)-mirtazapine, à l'exception des patients qui auraient comme co-médication des inhibiteurs du CYP2D6, telle que la fluoxétine ou la thioridazine. Il y a une enantiosélectivité du métabolisme de la mirtazapine. En particulier pour la 8-hydroxymirtazapine qui est glucuroconjuguée et pour laquelle le ratio S/R varie considérablement. Cette méthode analytique présente l'avantage d'être utilisable pour le dosage stéréosélectif de la mirtazapine et de ses métabolites dans le plasma de patients ayant d'autres substances en co-médication. La glycoprotéine P fonctionne comme une pompe transmembranaire transportant les xénobiotiques depuis le milieu intracellulaire vers le milieu extracellulaire. Son induction et son inhibition, bien que moins étudiées que pour les cytochromes P-450, ont des implications cliniques importantes en termes d'efficacité de traitement et de toxicité. Cette glycoprotéine P a fait l'objet d'une recherche bibliographique. Nous avons étudié le métabolisme du citalopram, un antidépresseur de la classe des inhibiteurs spécifiques de la recapture de la sérotonine chez la souris et chez le chien. Cette substance subit un métabolisme complexe. La voie de métabolisation conduisant à la formation de l'acide propionique du citalopram, catalysée par les monoamines oxydases, a été étudiée in vitro dans les mitochondries cérébrales chez la souris déficiente du gène de la MAO-A (Tg8). La monoamine oxydase A catalyse la formation de l'énantiomère R-(-) et présente une plus grande affinité pour les amines tertiaires, tandis que la monoamine oxydase B favorise la formation de la forme S-(+) et a une affinité plus marquée pour les amines secondaires et primaires. L'étude du citalopram chez la souris Tg8 adulte a montré que la monoamine oxydase B compense la déficience de la monoamine oxydase A chez ces souris génétiquement modifiées. Une autre voie de métabolisation du citalopram conduisant à la formation de didéméthylcitalopram, catalysée par les cytochromes P-450, a été étudiée in vitro dans des microsomes hépatiques de chiens Beagle. Nos études ont montré que les cinétiques de N-déméthylation du citalopram sont biphasiques chez le chien. Les orthologues canins impliqués dans la première N-déméthylation semblent être identiques aux cytochromes P-450 humains. Par contre, dans la deuxième Ndéméthylation, un seul cytochrome P-450 semble être impliqué chez l'homme (CYP2D6), tandis qu'on retrouve jusqu'à cinq orthologues chez le chien. Le CYP2D15, orthologue canin du CYP2D6, est majoritairement impliqué. De plus, l'activité enzymatique, reflétée par les clairances intrinsèques, dans la première N-déméthylation est jusqu'à 45 fois plus élevée chez le chien comparé à l'homme. Ces différentes observations soutiennent l'hypothèse que des taux élevés de didéméthylcitalopram sont responsables de la toxicité du citalopram chez le chien. Nous pouvons conclure que plusieurs famille d'enzymes sont impliquées tant au niveau cérébral qu'hépatique dans la métabolisation de médicaments psychotropes. Sachant -que les enzymes peuvent être induits ou inhibés, il importe de pouvoir suivre au plus près les taux plasmatiques des différents psychotropes et de leurs métabolites. Summary Most of the drugs are metabolized in the organism. Substances issued from this metabolic activity do not always show a pharmacological activity. Therefore, it is necessary to monitor plasmatic levels of drugs and their metabolites, and establish the relationship with the clinical effect. This concept named therapeutic drug monitoring is very useful in psychiatry where lack of compliance is commonly observed. Antidepressants are mainly metabolized in the liver (cytochrome P-450) and sometimes in the brain (monoamine oxidase) like the citalopram, for exemple. A LC-MS method was developed, which allows the simultaneous analysis of R-(-) and S-(+) enantiomers of mirtazapine, an antidepressant acting specifically on noradrenergic and serotonergic receptors, and its metabolites demethylmirtazapine and 8-hydroxymirtazapine in plasma of mirtazapine treated patients. Preliminary data obtained suggested that R-(-) mirtazapine concentrations were higher than those of S-(+) mirtazapine, except in patients comedicated with CYP2D6 inhibitors such as fluoxetine or thioridazine. There is an enantioselectivity in the metabolism of mirtazapine. In particular for the 8-hydroxymirtazapine, which is glucuroconjugated and S/R ratio varies considerably. Therefore this method seems to be suitable for the stereoselective assay of mirtazapine and its metabolites in plasma of patients comedicated with mirtazapine and other drugs for routine and research purposes. P-glycoprotein is working as an efflux transporter of xenobiotics from intracellular to extracellular environment. Its induction or inhibition, although less studied than cytochrome P-450, has huge clinical implications in terms of treatment efficacy and toxicity. An extensive literature search on P-glycoprotein was performed as part of this thesis. The study of citalopram metabolism, an antidepressant belonging to the class of selective serotonin reuptake inhibitors. This substance undergoes a complex metabolism. First metabolization route leading to citalopram propionic acid, catalyzed by monoamine oxidase was studied in vitro in mice brain mitochondria. Monoamine oxidase A catalyzed the formation of R-(-) enantiomer and showed greater affinity for tertiary amines, whereas monoamine oxidase B triggered the formation of S-(+) enantiomer and demonstrated higher affinity for primary and secondary amines. citalopram evaluation in adult Tg8 mice showed that monoamine oxidase B compensated monoamine oxidase A deficiency in those genetically transformed mice. The second metabolization route of citalopram leading to didemethylcitalopram and catalyzed by cytochrome P-450 was studied in vitro in Beagle dog's livers. Our results showed that citalopram N-demethylation kinetics are biphasic in dogs. Canine orthologs involved in the first N-demethylation seemed to be identical to human cytochromes P-450. However, in the second N-demethylation only one cytochrome P-450 seemed to be involved in human (CYP2D6), whereas up to five canine orthologs were found in dogs. CYP2D15 canine ortholog of CYP2D6 was mainly involved. In addition, enzymatic activity reflected by intrinsic clearance in the first N-demethylation was up to 45 fold higher in dogs compared to humans. Those observations support the assumption that elevated rates of didemethylcitalopram are responsible for citalopram toxicity in dogs. We can conclude that several enzymes groups are involved in the brain, as well as in the liver, in antidepressant metabolization. Knowing that enzymes may be induced or inhibited, it makes sense to closely monitor plasmatic levels of antidepressants and their metabolites.

BACKGROUND: Pancreaticoduodenectomies (PD) still have a substantial mortality rate. Recently, different scores have been published to predict the mortality risk pre-operatively after PD. This retrospective study was designed to perform an external assessment of an Early Mortality Risk Score (EMRS). METHODS: From 2000 to 2012, all PD cases performed at our institution were documented. Only patients treated for pancreatic head adenocarcinomas were included. Survival time and EMRS (based on age, tumour size, tumour differentiation and comorbidities) were calculated for every patient. Relative risks (RR) of early death 9 and 12 months after PD were then calculated. RESULTS: Of 270 PD for various aetiologies, 120 PD for adenocarcinomas were included. The median follow-up was 37 months, and the overall median survival was 19 months. EMRS of 4 showed a mortality RR of 5.1 at 9 months (P = 0.048) and of 4.5 at 12 months (P = 0.020). CONCLUSIONS: EMRS of 4 is a predictor of tumour-related mortality at 9 and 12 months after PD for adenocarcinoma. The EMRS was externally assessed in our patient cohort and can be implemented in clinical practice. Clinical implications of this score still need to be studied.

Objectifs La chirurgie pancréatique reste associée à une morbidité postopératoire importante. Les efforts sont concentrés la plupart du temps sur la diminution de cette morbidité, mais la détection précoce de patients à risque de complications pourrait être une autre stratégie valable. Un score simple de prédiction des complications après duodénopancréatectomie céphalique a récemment été publié par Braga et al. La présente étude a pour but de valider ce score et de discuter de ses possibles implications cliniques. Méthodes De 2000 à 2015, 245 patients ont bénéficié d'une duodénopancréatectomie céphalique dans notre service. Les complications postopératoires ont été recensées selon la classification de Dindo et Clavien. Le score de Braga se base sur quatre paramètres : le score ASA (American Society of Anesthesiologists), la texture du pancréas, le diamètre du canal de Wirsung (canal pancréatique principal) et les pertes sanguines intra-opératoires. Un score de risque global de 0 à 15 peut être calculé pour chaque patient. La puissance de discrimination du score a été calculée en utilisant une courbe ROC (receiver operating characteristic). Résultats Des complications majeures sont apparues chez 31% des patients, alors que 17% des patients ont eu des complications majeures dans l'article de Braga. La texture du pancréas et les pertes sanguines étaient statistiquement significativement corrélées à une morbidité accrue. Les aires sous la courbe étaient respectivement de 0.95 et 0.99 pour les scores classés en quatre catégories de risques (de 0 à 3, 4 à 7, 8 à 11 et 12 à 15) et pour les scores individuels (de 0 à 15). Conclusions Le score de Braga permet donc une bonne discrimination entre les complications mineures et majeures. Notre étude de validation suggère que ce score peut être utilisé comme un outil pronostique de complications majeures après duodénopancréatectomie céphalique. Les implications cliniques, c'est-à-dire si les stratégies de prise en charge postopératoire doivent être adaptées en fonction du risque individuel du patient, restent cependant à élucider. -- Objectives Pancreatic surgery remains associated with important morbidity. Efforts are most commonly concentrated on decreasing postoperative morbidity, but early detection of patients at risk could be another valuable strategy. A simple prognostic score has recently been published. This study aimed to validate this score and discuss possible clinical implications. Methods From 2000 to 2012, 245 patients underwent pancreaticoduodenectomy. Complications were graded according to the Dindo-Clavien classification. The Braga score is based on American Society of Anesthesiologists score, pancreatic texture, Wirsung duct diameter, and blood loss. An overall risk score (from 0 to 15) can be calculated for each patient. Score discriminant power was calculated using a receiver operating characteristic curve. Results Major complications occurred in 31% of patients compared to 17% in Braga's data. Pancreatic texture and blood loss were independently statistically significant for increased morbidity. The areas under curve were 0.95 and 0.99 for 4-risk categories and for individual scores, respectively. Conclusions The Braga score discriminates well between minor and major complications. Our validation suggests that it can be used as prognostic tool for major complications after pancreaticoduodenectomy. The clinical implications, i.e., whether postoperative treatment strategies should be adapted according to the patient's individual risk, remain to be elucidated.

High-frequency oscillations in the gamma-band reflect rhythmic synchronization of spike timing in active neural networks. The modulation of gamma oscillations is a widely established mechanism in a variety of neurobiological processes, yet its neurochemical basis is not fully understood. Modeling, in-vitro and in-vivo animal studies suggest that gamma oscillation properties depend on GABAergic inhibition. In humans, search for evidence linking total GABA concentration to gamma oscillations has led to promising -but also to partly diverging- observations. Here, we provide the first evidence of a direct relationship between the density of GABAA receptors and gamma oscillatory gamma responses in human primary visual cortex (V1). By combining Flumazenil-PET (to measure resting-levels of GABAA receptor density) and MEG (to measure visually-induced gamma oscillations), we found that GABAA receptor densities correlated positively with the frequency and negatively with amplitude of visually-induced gamma oscillations in V1. Our findings demonstrate that gamma-band response profiles of primary visual cortex across healthy individuals are shaped by GABAA-receptor-mediated inhibitory neurotransmission. These results bridge the gap with in-vitro and animal studies and may have future clinical implications given that altered GABAergic function, including dysregulation of GABAA receptors, has been related to psychiatric disorders including schizophrenia and depression.
