Control Centre operators are essential to assure a good performance of Power Systems. Operators’ actions are critical in dealing with incidents, especially severe faults, like blackouts. In this paper we present an Intelligent Tutoring approach for training Portuguese Control Centre operators in incident analysis and diagnosis, and service restoration of Power Systems, offering context awareness and an easy integration in the working environment.

Introduction: Paper and thin layer chromatography methods are frequently used in Classic Nuclear Medicine for the determination of radiochemical purity (RCP) on radiopharmaceutical preparations. An aliquot of the radiopharmaceutical to be tested is spotted at the origin of a chromatographic strip (stationary phase), which in turn is placed in a chromatographic chamber in order to separate and quantify radiochemical species present in the radiopharmaceutical preparation. There are several methods for the RCP measurement, based on the use of equipment as dose calibrators, well scintillation counters, radiochromatografic scanners and gamma cameras. The purpose of this study was to compare these quantification methods for the determination of RCP. Material and Methods: 99mTc-Tetrofosmin and 99mTc-HDP are the radiopharmaceuticals chosen to serve as the basis for this study. For the determination of RCP of 99mTc-Tetrofosmin we used ITLC-SG (2.5 x 10 cm) and 2-butanone (99mTc-tetrofosmin Rf = 0.55, 99mTcO4- Rf = 1.0, other labeled impurities 99mTc-RH RF = 0.0). For the determination of RCP of 99mTc-HDP, Whatman 31ET and acetone was used (99mTc-HDP Rf = 0.0, 99mTcO4- Rf = 1.0, other labeled impurities RF = 0.0). After the development of the solvent front, the strips were allowed to dry and then imaged on the gamma camera (256x256 matrix; zoom 2; LEHR parallel-hole collimator; 5-minute image) and on the radiochromatogram scanner. Then, strips were cut in Rf 0.8 in the case of 99mTc-tetrofosmin and Rf 0.5 in the case of 99mTc-HDP. The resultant pieces were smashed in an assay tube (to minimize the effect of counting geometry) and counted in the dose calibrator and in the well scintillation counter (during 1 minute). The RCP was calculated using the formula: % 99mTc-Complex = [(99mTc-Complex) / (Total amount of 99mTc-labeled species)] x 100. Statistical analysis was done using the test of hypotheses for the difference between means in independent samples. Results:The gamma camera based method demonstrated higher operator-dependency (especially concerning the drawing of the ROIs) and the measures obtained using the dose calibrator are very sensitive to the amount of activity spotted in the chromatographic strip, so the use of a minimum of 3.7 MBq activity is essential to minimize quantification errors. Radiochromatographic scanner and well scintillation counter showed concordant results and demonstrated the higher level of precision. Conclusions: Radiochromatographic scanners and well scintillation counters based methods demonstrate to be the most accurate and less operator-dependant methods.

Em Portugal, como em outros países, podem ser encontrados milhares de trabalhadores com doenças e outros problemas resultantes da exposição aos compostos orgânicos voláteis (COV’s). No entanto são poucos os estudos aplicados à actividade industrial. Neste trabalho validou-se/estimou-se a exposição ocupacional a COV´s dos trabalhadores que executam a tarefa de ‘aplicação de revestimento do mobiliário’, mais conhecida por ‘acabamento de móveis’ na secção de acabamentos do sector de mobiliário de madeira, que se situa nos concelhos de Paços de Ferreira e Paredes, concelhos do Norte de Portugal. A amostra foi constituída por 17 empresas e foram avaliados 34 tarefas que corresponde ao mesmo número de trabalhadores, uma vez que eles executam essa tarefa durante as oito horas diárias de trabalho. Para avaliar a exposição dos trabalhadores foram utilizadas duas abordagens da higiene do trabalho: a pragmática, através da aplicação do método Toolkit e a tradicional com recurso a amostragens de ar e análises laboratoriais. Os resultados obtidos sugerem que o Toolkit é uma boa ferramenta para ser utilizada pelas Pequenas e Médias Empresas (PME’s), que trabalhem com substâncias em pó ou líquidas. É um método expedito e que não acarreta grandes esforços financeiros. Verificou-se ainda que a maioria dos trabalhadores estão em risco de exposição a COV’s. Sendo necessário tomar medidas de controlo.

Introdução: Apesar dos muitos estudos sobre a temática da sensação retardada de desconforto muscular, atualmente, ainda se discute a explicação dos mecanismos subjacentes a esta condição clínica, bem como, a sua prevenção e tratamento. A literatura sugere a massagem como uma das formas de terapia, contudo, os estudos têm mostrado resultados controversos. Objetivo: Verificar se a massagem aplicada 2 horas após um protocolo de exercício excêntrico tem influência na sensação retardada de desconforto muscular, bem como, se o seu efeito varia dependendo do tempo de aplicação. Métodos: 21 participantes (23,62±1,32 anos; 76,95±12,17 kg; 174,71±4,78 cm; 25,25±4,26 Kg/m 2) foram divididos em três grupos. Foi avaliada a dor, força muscular e a perimetria antes, e 2h, 24h, 48h, e 72h após um protocolo de exercício constituído por três séries de dez repetições de contrações excêntricas dos isquiotibiais do membro dominante, com 80% da força máxima, a uma velocidade constante de 60º/s, numa amplitude entre 0º e 80º, utilizando o dinamómetro isocinético Biodex System 4. A massagem foi efetuada 2 horas após o exercício em dois grupos experimentais com durações diferentes, sendo o terceiro grupo de controlo. Para identificar diferenças entre os grupos no momento inicial e na variável diferença entre o momento inicial e os restantes momentos, recorreu-se ao teste de Kruskal-Wallis, seguido de uma análise Post-Hoc através do teste de Dunn com um nível de significância de 0,05. Resultados: Verificou-se que a massagem teve efeito na redução da dor e na perimetria. Relativamente à força não foram encontradas alterações significativas. Conclusão: Os resultados mostraram que a massagem aplicada 2 horas após o exercício excêntrico, independentemente da duração utilizada, teve efeito na redução da dor, mas não na força muscular. Na perimetria apesar de haver alterações, estas não foram consideradas relevantes.

Grande parte dos triples-stores são open source e desenvolvidos em Java, disponibilizando interfaces standards e privadas de acesso. A grande maioria destes sistemas não dispõe de mecanismos de controlo de acessos nativos, o que dificulta ou impossibilita a sua adopção em ambientes em que a segurança dos factos é importante (e.g. ambiente empresarial). Complementarmente observa-se que o modelo de controlo de acesso a triplos e em particular a triplos descritos por ontologias não está standardizado nem sequer estabilizado, havendo diversos modelos de descrição e algoritmos de avaliação de permissões de acesso. O trabalho desenvolvido nesta tese/dissertação propõe um modelo e interface de controlo de acesso que permite e facilite a sua adopção por diferentes triple-stores já existentes e a integração dos triples-stores com outros sistemas já existentes na organização. Complementarmente, a plataforma de controlo de acesso não impõe qualquer modelo ou algoritmo de avaliação de permissões, mas pelo contrário permite a adopção de modelos e algoritmos distintos em função das necessidades ou desejos. Finalmente demonstra-se a aplicabilidade e validade do modelo e interface propostos, através da sua implementação e adopção ao triple-store SwiftOWLIM já existente, que não dispõe de mecanismo de controlo de acessos nativo.

Devido à grande quantidade de dados disponíveis na Internet, um dos maiores desafios no mundo virtual é recomendar informação aos seus utilizadores. Por outro lado, esta grande quantidade de dados pode ser útil para melhorar recomendações se for anotada e interligada por dados de proveniência. Neste trabalho é abordada a temática de recomendação de (alteração de) permissões acesso sobre recursos ao seu proprietário, ao invés da recomendação do próprio recurso a um potencial consumidor/leitor. Para permitir a recomendação de acessos a um determinado recurso, independentemente do domínio onde o mesmo se encontra alojado, é essencial a utilização de sistemas de controlo de acessos distribuídos, mecanismos de rastreamento de recursos e recomendação independentes do domínio. Assim sendo, o principal objectivo desta tese é utilizar informação de rastreamento de acções realizadas sobre recursos (i.e. informação que relaciona recursos e utilizadores através da Web independentemente do domínio de rede) e utiliza-la para permitir a recomendação de privilégios de acesso a esses recursos por outros utilizadores. Ao longo do desenvolvimento da tese resultaram as seguintes contribuições: A análise do estado da arte de recomendação e de sistemas de recomendação potencialmente utilizáveis na recomendação de privilégios (secção 2.3); A análise do estado da arte de mecanismos de rastreamento e proveniência de informação (secção 2.2); A proposta de um sistema de recomendação de privilégios de acesso independente do domínio e a sua integração no sistema de controlo de acessos proposto anteriormente (secção 3.1); Levantamento, análise e especificação da informação relativa a privilégios de acesso, para ser utilizada no sistema de recomendação (secção 2.1); A especificação da informação resultante do rastreamento de acções para ser utilizada na recomendação de privilégios de acesso (secção 4.1.1); A especificação da informação de feedback resultante do sistema de recomendação de acessos e sua reutilização no sistema de recomendação(secção 4.1.3); A especificação, implementação e integração do sistema de recomendação de privilégios de acesso na plataforma já existente (secção 4.2 e secção 4.3); Realização de experiências de avaliação ao sistema de recomendação de privilégios, bem como a análise dos resultados obtidos (secção 5).

Electroanalytical methods based on square-wave adsorptive-stripping voltammetry (SWAdSV) and flow-injection analysis with square-wave adsorptive-stripping voltammetric detection (FIA-SWAdSV) were developed for the determination of fluoxetine (FXT). The methods were based on the reduction of FXT at a mercury drop electrode at -1.2 V versus Ag/AgCl, in a phosphate buffer of pH 12.0, and on the possibility of accumulating the compound at the electrode surface. The SWAdSV method was successfully applied in the quantification of FXT in pharmaceutical products, human serum samples, and in drug dissolution studies. Because the presence of dissolved oxygen did not interfere significantly with the analysis, it was possible to quantify FXT in several pharmaceutical products using FIA-SWAdSV. This method enables analysis of up to 120 samples per hour at reduced costs.

Celiac disease (CD) is an autoimmune enteropathy, characterized by an inappropriate T-cell-mediated immune response to the ingestion of certain dietary cereal proteins in genetically susceptible individuals. This disorder presents environmental, genetic, and immunological components. CD presents a prevalence of up to 1% in populations of European ancestry, yet a high percentage of cases remain underdiagnosed. The diagnosis and treatment should be made early since untreated disease causes growth retardation and atypical symptoms, like infertility or neurological disorders. The diagnostic criteria for CD, which requires endoscopy with small bowel biopsy, have been changing over the last few decades, especially due to the advent of serological tests with higher sensitivity and specificity. The use of serological markers can be very useful to rule out clinical suspicious cases and also to help monitor the patients, after adherence to a gluten-free diet. Since the current treatment consists of a life-long glutenfree diet, which leads to significant clinical and histological improvement, the standardization of an assay to assess in an unequivocal way gluten in gluten-free foodstuff is of major importance.

The purpose of the present work is to determine the antioxidant capacity (AC) of 27 commercial beers. The AC indicates the degree of protection of a certain organism against oxidative damage provoked by reactive oxygen and nitrogen species. Assays were carried out by the following methods: (i) total radical trapping antioxidant parameter (TRAP); (ii) trolox equivalent antioxidant capacity (TEAC); (iii) trolox equivalent antioxidant capacity (DPPH); (iv) ferric-ion reducing antioxidant parameter (FRAP); (v) cupric reducing antioxidant capacity (CUPRAC); (vi) oxygen radical absorbance capacity (ORAC). Ascorbic acid (AA), gallic acid (GA) and trolox (TR) were used as standards. All beers showed antioxidant power, but a wide range of ACs was observed. The effect of several factors upon these differences was studied. Statistical differences were found between ACs of beers of different colours. ORAC method provided always higher experimental ACs, of significant statistical differences to other assays.

The state of the art of voltammetric and amperometric methods used in the study and determination of pesticides in crops, food, phytopharmaceutical products, and environmental samples is reviewed. The main structural groups of pesticides, i.e., triazines, organophosphates, organochlorides, nitrocompounds, carbamates, thiocarbamates, sulfonylureas, and bipyridinium compounds are considered with some degradation products. The advantages, drawbacks, and trends in the development of voltammetric and amperometric methods for study and determination of pesticides in these samples are discussed.

This study investigated the influence of wearing unstable shoe construction (WUS) on compensatory postural adjustments (CPA) associated with external perturbations. Thirty-two subjects stood on a force platform resisting an anterior-posterior horizontal force applied to a pelvic belt via a cable, which was suddenly released. They stood under two conditions: barefoot and WUS. The electromyographic (EMG) activity of gastrocnemius medialis, tibialis anterior, rectus femoris, biceps femoris, rectus abdominis, and erector spinae muscles and the center of pressure (CoP) displacement were acquired to study CPA. The EMG signal was used to assess individual muscle activity and latency, antagonist co-activation and reciprocal activation at joint and muscle group levels. Compared to barefoot, WUS led to: (1) increased gastrocnemius medialis activity, (2) increased total agonist activity, (3) decreased antagonist co-activation at the ankle joint and muscle group levels, (4) increased reciprocal activation at the ankle joint and muscle group levels, and (5) decrease in all muscle latencies. No differences were observed in CoP displacement between conditions. These findings demonstrate that WUS led to a reorganization of the postural control system associated to improved performance of some components of postural control responses.

Purpose: This study investigated the influence of long-term wearing of unstable shoes (WUS) on compensatory postural adjustments (CPA) to an external perturbation. Methods: Participants were divided into two groups: one wore unstable shoes while the other wore conventional shoes for 8 weeks. The ground reaction force signal was used to calculate the anterior– posterior (AP) displacement of the centre of pressure (CoP) and the electromyographic signal of gastrocnemius medialis (GM), tibialis anterior (TA), rectus femoris (RF) and biceps femoris (BF) muscles was used to assess individual muscle activity, antagonist co-activation and reciprocal activation at the joint (TA/GM and RF/(BF + GM) pairs) and muscle group levels (ventral (TA + RF)/dorsal (GM + BF) pair) within time intervals typical for CPA. The electromyographic signal was also used to assess muscle latency. The variables described were evaluated before and after the 8-week period while wearing the unstable shoes and barefoot. Results: Long-term WUS led to: an increase of BF activity in both conditions (barefoot and wearing the unstable shoes); a decrease of GM activity; an increase of antagonist co-activation and a decrease of reciprocal activation level at the TA/GM and ventral/dorsal pairs in the unstable shoe condition. Additionally, WUS led to a decrease in CoP displacement. However, no differences were observed in muscle onset and offset. Conclusion: Results suggest that the prolonged use of unstable shoes leads to increased ankle and muscle groups’ antagonist co-activation levels and higher performance by the postural control system.

We study a fractional model for malaria transmission under control strategies.Weconsider the integer order model proposed by Chiyaka et al. (2008) in [15] and modify it to become a fractional order model. We study numerically the model for variation of the values of the fractional derivative and of the parameter that models personal protection, b. From observation of the figures we conclude that as b is increased from 0 to 1 there is a corresponding decrease in the number of infectious humans and infectious mosquitoes, for all values of α. This means that this result is invariant for variation of fractional derivative, in the values tested. These results are in agreement with those obtained in Chiyaka et al.(2008) [15] for α = 1.0 and suggest that our fractional model is epidemiologically wellposed.

Bone is constantly being molded and shaped by the action of osteoclasts and osteoblasts. A proper equilibrium between both cell types metabolic activities is required to ensure an adequate skeletal tissue structure, and it involves resorption of old bone and formation of new bone tissue. It is reported that treatment with antiepileptic drugs (AEDs) can elicit alterations in skeletal structure, in particular in bone mineral density. Nevertheless, the knowledge regarding the effects of AEDs on bone cells are still scarce. In this context, the aim of this study was to investigate the effects of five different AEDs on human osteoclastic, osteoblastic and co-cultured cells. Osteoclastic cell cultures were established from precursor cells isolated from human peripheral blood and were characterized for tartrate-resistant acid phosphatase (TRAP) activity, number of TRAP+ multinucleated cells, presence of cells with actin rings and expressing vitronectin and calcitonin receptors and apoptosis rate. Also, the involvement of several signaling pathways on the cellular response was addressed. Osteoblastic cell cultures were obtained from femur heads of patients (25-45 years old) undergoing orthopaedic surgery procedures and were then studied for cellular proliferation/viability, ALP activity, histochemical staining of ALP and apoptosis rate. Also the expression of osteoblast-related genes and the involvement of some osteoblastogenesis-related signalling pathways on cellular response were addressed. For co-cultured cells, osteoblastic cells were firstly seeded and cultured. After that, PBMC were added to the osteoblastic cells and co-cultures were evaluated using the same osteoclast and osteoblast parameters mentioned above for the corresponding isolated cell. Cell-cultures were maintained in the absence (control) or in the presence of different AEDs (carbamazepine, gabapentin, lamotrigine, topiramate and valproic acid). All the tested drugs were able to affect osteoclastic and osteoblastic cells development, although with different profiles on their osteoclastogenic and osteoblastogenic modulation properties. Globally, the tendency was to inhibit the process. Furthermore, the signaling pathways involved in the process also seemed to be differently affected by the AEDs, suggesting that the different drugs may affect osteoclastogenesis and/or osteoblastogenesis through different mechanisms. In conclusion, the present study showed that the different AEDs had the ability to directly and indirectly modulate bone cells differentiation, shedding new light towards a better understanding of how these drugs can affect bone tissue.
