Esta dissertação de mestrado é baseada numa investigação – ação, realizada numa turma de 3º e 4º anos do 1º ciclo. Partindo da análise de uma situação problemática de ensino a crianças oriundas de meios de imigração foi realizada uma investigação que visava intervir na aprendizagem da escrita. Foi aplicado um pré-teste, escrita de uma narrativa tendo como estímulo um conjunto de imagens. Após a sua análise, delineámos uma intervenção de modo a melhorar os aspetos em que as crianças apresentavam dificuldades. A intervenção pedagógica focalizou o ensino da escrita enquanto processo e, dado do que o português não é a língua materna das crianças, demos uma atenção especial ao input textual, recorrendo à leitura obras literárias para crianças. Após a intervenção, foi realizado o pós-teste, sob as mesmas condições que o pré-teste, e observaram-se melhorias acentuadas na competência de escrita das crianças.

O cancro colo-rectal é o terceiro tipo de cancro mais comum em ambos os sexos. Os factores ambientais, o sedentarismo, a obesidade, o tabagismo, o álcool, uma dieta rica em carne vermelha e pobre em fibras, desempenham um forte papel na etiologia do cancro do recto. Nas últimas duas décadas a adopção generalizada da excisão mesoretal total (TME) e uso da quimio-radioterapia (QRT) pré-operatória, aumentou as taxas de controlo local, sobrevida global e livre de doença. Quando comparada com a QRT pós-operatória, a QRT pré-operatória demonstra ser mais eficaz, uma vez que possibilita um maior controlo local, como também uma menor toxicidade, rnomeadamente do intestino delgado. Contudo, verifica-se efeitos colaterais adversos, que influenciam negativamente a qualidade de vida (QoL) dos doentes, sendo os mais comuns a incontinência urinária e fecal, e a disfunção sexual. Com este estudo pretende-se avaliar a QoL dos doentes com cancro do recto localmente avançado (T3-T4), em 3 momentos de avaliação, isto é, antes, durante e no final do tratamento. Pretende-se ainda determinar se a idade influencia a QoL dos doentes.

Objective: To evaluate the influence of Everolimus (RAD001) on chemically induced urothelial lesions in mice and its influence on in vitro human bladder cancer cell lines. Methods: ICR male mice were given N-butyl-N-(4-hydroxybutyl) nitrosamine in drinking water for a period of 12 weeks. Subsequently, RAD001 was administered via oral gavage, for 6 weeks. At the end of the experiment, all the animals were sacrificed and tumor development was determined by means of histopathologic evaluation; mammalian target of rapamycin (mTOR) expressivity was evaluated by immunohistochemistry. Three human bladder cancer cell lines (T24, HT1376, and 5637) were treated using a range of RAD001 concentrations. MTT assay, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL), and flow cytometry were used to assess cell proliferation, apoptosis index, and cell cycle analysis, respectively. Immunoblotting analysis of 3 cell line extracts using mTOR and Akt antibodies was performed in order to study the expression of Akt and mTOR proteins and their phosphorylated forms. Results: The incidence of urothelial lesions in animals treated with RAD001 was similar to those animals not treated. RAD001 did not block T24 and HT1376 cell proliferation or induce apoptosis. A reduction in cell proliferation rate and therefore G0/G1 phase arrest, as well as a statistically significant induction of apoptosis (P 0.001), was only observed in the 5637 cell line. Conclusion: RAD001 seems not to have a significant effect on chemically induced murine bladder tumors. The effect of RAD001 on tumor proliferation and apoptosis was achieved only in superficial derived bladder cancer cell line, no effect was observed in invasive cell lines.

Background: Poor nutritional status and worse health-related quality of life (QoL) have been reported in haemodialysis (HD) patients. The utilization of generic and disease specific QoL questionnaires in the same population may provide a better understanding of the significance of nutrition in QoL dimensions. Objective: To assess nutritional status by easy to use parameters and to evaluate the potential relationship with QoL measured by generic and disease specific questionnaires. Methods: Nutritional status was assessed by subjective global assessment adapted to renal patients (SGA), body mass index (BMI), nutritional intake and appetite. QoL was assessed by the generic EuroQoL and disease specific Kidney Disease Quality of Life-Short Form (KDQoL-SF) questionnaires. Results: The study comprised 130 patients of both genders, mean age 62.7 ± 14.7 years. The prevalence of undernutrition ranged from 3.1% by BMI ≤ 18.5 kg/m2 to 75.4% for patients below energy and protein intake recommendations. With the exception of BMI classification, undernourished patients had worse scores in nearly all QoL dimensions (EuroQoL and KDQoL-SF), a pattern which was dominantly maintained when adjusted for demographics and disease-related variables. Overweight/obese patients (BMI ≥ 25) also had worse scores in some QoL dimensions, but after adjustment the pattern was maintained only in the symptoms and problems dimension of KDQoL-SF (p = 0.011). Conclusion: Our study reveals that even in mildly undernourished HD patients, nutritional status has a significant impact in several QoL dimensions. The questionnaires used provided different, almost complementary perspectives, yet for daily practice EuroQoL is simpler. Assuring a good nutritional status, may positively influence QoL.

Em Portugal, no final da década de 90 do século XX, começaram a ser desenvolvidas estratégias de discriminação positiva visando garantir o cumprimento da escolaridade obrigatória e a luta contra a exclusão escolar e social, designadamente nas periferias das grandes cidades. É neste contexto que surgem as escolas e os Territórios Educativos de Intervenção Prioritária (TEIP) pelo Despacho nº147-B/ME/96, de 1 de Agosto de 1996. Este programa, inicialmente inspirado nas zonas de intervenção prioritárias francesas, visava a intervenção de vários parceiros locais (professores, alunos, pessoal não docente, associações de pais, autarquias locais, associações culturais e associações recreativas) na elaboração do Projeto Educativo. Com o XVII Governo Constitucional o programa foi redefinido passando a incluir novas vertentes: necessidade de existência de um projeto educativo próprio; consultadoria externa; avaliação periódica de resultados em diferentes domínios (taxas de insucesso e abandono escolar, assiduidade, comportamento, participação, inovações organizacionais, parcerias educativas estabelecidas no âmbito do Programa, etc.). Foi também alargado a todo o território nacional, envolvendo atualmente cento e cinco agrupamentos escolares. Com esta apresentação visamos analisar se as novas políticas e diretrizes, no domínio da educação prioritária, contribuíram para a emergência de novas estratégias pedagógicas, organizacionais e de envolvimento comunitário. Os elementos em que basearemos a nossa análise serão os seguintes: (i) relatório nacional do programa TEIP (2010-2011); (ii) relatórios do programa de avaliação externa das escolas; (iii) entrevistas realizadas aos atores locais, designadamente a coordenadores e consultores dos projetos TEIP. As conclusões da nossa comunicação centrar-se-ão no papel dos atores locais no desenvolvimento do Programa TEIP e no impacto deste programa na melhoria dos resultados académicos, na diminuição da indisciplina e violência escolar e na construção de percursos de vida que contrariem as tendências para a exclusão escolar e social.

The population growth of a Staphylococcus aureus culture, an active colloidal system of spherical cells, was followed by rheological measurements, under steady-state and oscillatory shear flows. We observed a rich viscoelastic behavior as a consequence of the bacteria activity, namely, of their multiplication and density-dependent aggregation properties. In the early stages of growth (lag and exponential phases), the viscosity increases by about a factor of 20, presenting several drops and full recoveries. This allows us to evoke the existence of a percolation phenomenon. Remarkably, as the bacteria reach their late phase of development, in which the population stabilizes, the viscosity returns close to its initial value. Most probably, this is caused by a change in the bacteria physiological activity and in particular, by the decrease of their adhesion properties. The viscous and elastic moduli exhibit power-law behaviors compatible with the "soft glassy materials" model, whose exponents are dependent on the bacteria growth stage. Video coding technologies have played a major role in the explosion of large market digital video applications and services. In this context, the very popular MPEG-x and H-26x video coding standards adopted a predictive coding paradigm, where complex encoders exploit the data redundancy and irrelevancy to 'control' much simpler decoders. This codec paradigm fits well applications and services such as digital television and video storage where the decoder complexity is critical, but does not match well the requirements of emerging applications such as visual sensor networks where the encoder complexity is more critical. The Slepian Wolf and Wyner-Ziv theorems brought the possibility to develop the so-called Wyner-Ziv video codecs, following a different coding paradigm where it is the task of the decoder, and not anymore of the encoder, to (fully or partly) exploit the video redundancy. Theoretically, Wyner-Ziv video coding does not incur in any compression performance penalty regarding the more traditional predictive coding paradigm (at least for certain conditions). In the context of Wyner-Ziv video codecs, the so-called side information, which is a decoder estimate of the original frame to code, plays a critical role in the overall compression performance. For this reason, much research effort has been invested in the past decade to develop increasingly more efficient side information creation methods. This paper has the main objective to review and evaluate the available side information methods after proposing a classification taxonomy to guide this review, allowing to achieve more solid conclusions and better identify the next relevant research challenges. After classifying the side information creation methods into four classes, notably guess, try, hint and learn, the review of the most important techniques in each class and the evaluation of some of them leads to the important conclusion that the side information creation methods provide better rate-distortion (RD) performance depending on the amount of temporal correlation in each video sequence. It became also clear that the best available Wyner-Ziv video coding solutions are almost systematically based on the learn approach. The best solutions are already able to systematically outperform the H.264/AVC Intra, and also the H.264/AVC zero-motion standard solutions for specific types of content. (C) 2013 Elsevier B.V. In distributed video coding, motion estimation is typically performed at the decoder to generate the side information, increasing the decoder complexity while providing low complexity encoding in comparison with predictive video coding. Motion estimation can be performed once to create the side information or several times to refine the side information quality along the decoding process. In this paper, motion estimation is performed at the decoder side to generate multiple side information hypotheses which are adaptively and dynamically combined, whenever additional decoded information is available. The proposed iterative side information creation algorithm is inspired in video denoising filters and requires some statistics of the virtual channel between each side information hypothesis and the original data. With the proposed denoising algorithm for side information creation, a RD performance gain up to 1.2 dB is obtained for the same bitrate.

Low-density parity-check (LDPC) codes are nowadays one of the hottest topics in coding theory, notably due to their advantages in terms of bit error rate performance and low complexity. In order to exploit the potential of the Wyner-Ziv coding paradigm, practical distributed video coding (DVC) schemes should use powerful error correcting codes with near-capacity performance. In this paper, new ways to design LDPC codes for the DVC paradigm are proposed and studied. The new LDPC solutions rely on merging parity-check nodes, which corresponds to reduce the number of rows in the parity-check matrix. This allows to change gracefully the compression ratio of the source (DCT coefficient bitplane) according to the correlation between the original and the side information. The proposed LDPC codes reach a good performance for a wide range of source correlations and achieve a better RD performance when compared to the popular turbo codes.

Evolution by natural selection is driven by the continuous generation of adaptive mutations. We measured the genomic mutation rate that generates beneficial mutations and their effects on fitness in Escherichia coli under conditions in which the effect of competition between lineages carrying different beneficial mutations is minimized. We found a rate on the order of 10–5 per genome per generation, which is 1000 times as high as previous estimates, and a mean selective advantage of 1%. Such a high rate of adaptive evolution has implications for the evolution of antibiotic resistance and pathogenicity.
