Os efetivos autóctones de pequenos ruminantes têm vindo a diminuir,em parte devido ao seu baixo potencial produtivo. A necessidade de encontrar formas mais expeditas de aumentar o potencial produtivo das nossas raças, e assim promover a sua manutenção bem como a sustentabilidade dos sistemas extensivos onde são explorados, levou à procura de marcadores moleculares, nomeadamente no gene da hormona de crescimento (GH), associados com a produção e qualidade do leite em pequenos ruminantes. Nas raças ovinas Churra da Terra Quente, Merino da Beira Baixa, Saloia e Serra da Estrela e caprinas Algarvia e Serrana verificou -se que o gene da GH é muito polimórfico, tendo sido encontrados polimorfismos específicos em algumas das raças. Os resultados sugerem que os polimorfismos do gene da GH, entre outros (e.g., nas caseínas), poderão vir a ser utilizados na seleção assistida por marcadores genéticos, de modo a melhorar da produção de leite sem afetar a sua qualidade. Contudo, a resposta à seleção será sempre condicionada pela prática de um correto maneio alimentar dos animais.

Numa Europa caracterizada pelos excedentes de produção agrícola, a qualidade das produções é um factor determinante da rentabilidade das explorações agrícolas. Podemos dizer mesmo que, nos dias de hoje, é mais importante produzir com qualidade que produzir em quantidade. A qualidade de um fruto cítrico é determinada por uma série de parâmetros (tamanho, características organolépticas, cor externa e interna, ausência de lesões, facilidade de descascar, etc.) que analisaremos a seguir e que dependem do cultivar e das técnicas culturais utilizadas.

Gilthead seabream is the most important farmed species in the Mediterranean, and knowledge on how common farming practices impact its quality is limited. As such, this Thesis aimed to evaluate how gilthead seabream flesh quality is affected by some of these practices. In Chapter 2, the influence of nutritional factors was evaluated, specifically the high replacement of traditional marine-derived ingredients, both fishmeal and fish oil, with vegetable sources. We have seen that the vegetable-based diets tested did not greatly impact seabream flesh quality, although some alterations were seen in the fatty acid profile of the muscle. However, and despite having caused no alterations in flesh texture, vegetable ingredients reduced the amount of sulphated glycosaminoglycans in the extracellular matrix, affected muscle pH and reduced the activity of proteolytic enzymes. Throughout this Thesis, we measured for the first time the activity of proteolytic enzymes in seabream muscle, and cathepsin B was found to play a pivotal role in post-mortem muscle degradation. In Chapter 3, we evaluated the effect of harvesting and slaughter stress on seabream quality, and contrary to what is seen in most farmed species, our results show that gilthead seabream muscle structure is highly resistant to changes caused by stressful events. Nonetheless, considering that welfare is an increasingly important quality criterion, the use of a zero-withdrawal anaesthetic as a rested harvest technique or even slaughter method could prove valuable to the industry. In Chapter 4, we used maslinic acid as a dietary supplement, to modulate the muscle’s energetic status pre-mortem. As a finishing strategy, maslinic acid failed to increase levels of glycogen and ATP in the muscle. However, supplementation resulted in higher muscle fibre diameter and lower cathepsin B activity, and maslinic acid is likely to be useful to promote growth in this species. In general our Thesis has generated new knowledge to a major challenge facing the aquaculture industry, which is to find a compromise between the trends towards intensive rearing and consumer demand for healthy, high quality seafood being ethically acceptable and having a low impact on the environment.

As conceções clássicas sobre as origens do perfeccionismo e os principais modelos explicativos do seu desenvolvimento assentam no pressuposto de que a relação parental constitui um importante fator na compreensão deste traço de personalidade nos filhos. Apesar do suporte teórico e conceptual, os resultados da investigação empírica são parciais e pouco conclusivos. Neste contexto, definimos como objetivos principais analisar a influência do Perfeccionismo parental no desenvolvimento do Perfeccionismo dos filhos jovens adultos e compreender o papel dos Estilos Parentais e dos Padrões de Vinculação. Procurou-se ainda averiguar se o efeito destas variáveis era moderado pelo sexo do progenitor ou pelo sexo do filho. Os resultados encontrados permitem um conhecimento mais aprofundado sobre o papel destes fatores parentais. A perceção de um Estilo Parental Autoritário e de uma Vinculação pouco Segura aos pais parece contribuir marcadamente para o desenvolvimento do Perfeccionismo Desadaptativo. Por outro lado, um Estilo Parental Democrático e uma Vinculação Segura parecem apenas influenciar uma parte do Perfeccionismo o Adaptativo. O estudo do contributo diferencial de cada um dos progenitores no desenvolvimento do perfeccionismo em função do sexo do filho permitiu testar as duas hipóteses descritas na literatura sobre o papel moderador do sexo. Os dados sugerem que os filhos desenvolvem tendências perfeccionistas pela identificação com o progenitor do mesmo sexo (hipótese do cuidador do mesmo sexo), enquanto a componente desadaptativa do perfeccionismo parece desenvolve-se tanto nos filhos do sexo masculino como nas filhas quando a mãe manifesta também níveis elevados de Perfeccionismo Desadaptativo (hipótese do cuidador principal). Os resultados apoiam ainda o Modelo da Aprendizagem Social e o Modelo das Expectativas Sociais formulados por Flett, Hewitt, Oliver e Macdonald (2002). Por último, são discutidas as implicações destes resultados na prevenção e na intervenção psicológica com pais e filhos e apontadas algumas das limitações do presente trabalho que poderão ser colmatadas em futuras investigações.

Com o objectivo de determinar o crescimento de larvas de sardinha foram realizadas experiências em condições controladas. Um cardume de sardinhas adultas foi mantido em cativeiro e a desova induzida por meios naturais (aumento da disponibilidade alimentar, regulação da temperatura para cerca de 15º C e salinidade para ~35‰). Quando ocorreram desovas com mais do que 500 ovos num determinado dia, estes foram colocados a eclodir em condições controladas. As larvas de sardinha apresentaram um desenvolvimento morfológico normal (p.e. pigmentação dos olhos e desenvolvimento da boca entre o 3º e 4º dia). Diferentes tipos e combinações de alimentos foram experimentados (nauplios de Acartia grani, rotíferos e Gymnodinium sp.) e analisou-se a condição nutricional das larvas amostradas. Foram efectuadas experiências de inanição para se poder calibrar os resultados obtidos. As taxas de crescimento variaram entre 0.12 e 0.36 mm.dia-1, sendo a mais elevada correspondente a uma experiência de inanição. Observou-se, através da histologia, que dietas ricas em Acartia grani são mais favoráveis para um melhor desenvolvimento larvar. As análises da razão RNA/DNA sugerem que há uma forte possibilidade de os efeitos parentais terem bastante influência no desenvolvimento e crescimento das larvas de sardinha nas duas primeiras semanas de vida, mas não na sua sobrevivência, que dependeu da quantidade e qualidade de alimento fornecida.

Cardiogenesis is a delicate and complex process that requires the coordination of an intricate network of pathways and the different cell types. Therefore, understanding heart development at the morphogenetic level is an essential requirement to uncover the causes of congenital heart disease and to provide insight for disease therapies. Mouse Cerberus like 2 (Cerl2) has been defined as a Nodal antagonist in the node with an important role in the Left-Right (L/R) axis establishment, at the early embryonic development. As expected, Cerl2 knockout mice (Cerl2-/-) showed multiple laterality defects with associated cardiac failure. In order to identify the endogenous role of Cerl2 during heart formation independent of its described functions in the node, we accurately analyzed animals where laterality defects were not present. We thereby unravel the consequences of Cerl2 lossof- function in the heart, namely increased left ventricular thickness due to hyperplasia of cardiomyocytes and de-regulated expression of cardiac genes. Furthermore, the Cerl2 mutant neonates present impaired cardiac function. Once that the cardiac expression of Cerl2 is mostly observed in the left ventricle until around midgestration, this result suggest a specific regulatory role of Cerl2 during the formation of the left ventricular myoarchitecture. Here, we present two possible molecular mechanisms underlying the cardiac Cerl2 function, the regulation of Cerl2 antagonist in activation of the TGFßs/Nodal/Activin/Smad2 signaling identified by increased Smad2 phosphorilation in Cerl2-/- hearts and the negative feedback between Cerl2 and Wnt/ß-catenin signaling in heart formation. In this work and since embryonic stem cells derived from 129 mice strain is extensively used to produce targeted mutants, we also present echocardiographic reference values to progressive use of juveniles and young adult 129/Sv strain in cardiac studies. In addition, we investigate the cardiac physiology of the surviving Cerl2 mutants in 129/Sv background over time through a follow-up study using echocardiographic analysis. Our results revealed that Cerl2-/- mice are able to improve and maintain the diastolic and most of systolic cardiac physiologic parameters as analyzed until young adult age. Since Cerl2 is no longer expressed in the postnatal heart, we suggest that an intrinsic and compensatory mechanism of adaptation may be active for recovering the decreased cardiac function found in Cerl2 mutant neonates. Altogether, these data highlight the role of Cerl2 during embryonic heart development in mice. Furthermore, we also suggest that Cerl2-/- may be an interesting model to uncover the molecular, cellular and physiological mechanisms behind the improvement of the cardiac function, contributing to the development of therapeutic approaches to treat heart failures.

Bone morphogenetic proteins (BMPs) are multifunctional growth factors belonging to the transforming growth factor β (TGFβ) superfamily with a central role in bone formation and mineralization. BMP2, a founding member of this family, has demonstrated remarkable osteogenic properties and is clinically used to promote bone repair and fracture healing. Lack of basic data on factors regulating BMP2 expression and activity have hampered a better understanding of its role in bone formation and bone-related diseases. The objective of this work was to collect new functional data and determine spatiotemporal expression patterns in a fish system aiming towards a better understanding of BMP2 function and regulation. Transcriptional and post-transcriptional regulation of gilthead seabream BMP2 gene was inferred from luciferase reporter systems. Several bone- and cartilage-related transcription factors (e.g. RUNX3, MEF2c, SOX9 and ETS1) were found to regulate BMP2 transcription, while microRNA 20a was shown to affect stability of the BMP2 transcript and thus the mineralogenic capacity of fish bone-derived host cells. The regulation of BMP2 activity through an interaction with the matrix Gla protein (MGP) was investigated in vitro using BMP responsive elements (BRE) coupled to luciferase reporter gene. Although we demonstrated the functionality of the experimental system in a fish cell line and the activation of BMP signaling pathway by seabream BMP2, no conclusive evidence could be collected on a possible interaction beween MGP and BMP2. The evolutionary relationship among the members of BMP2/4/16 subfamily was inferred from taxonomic and phylogenetic analyses. BMP16 diverged prior to BMP2 and BMP4 and should be the result of an ancient genome duplication that occurred early in vertebrate evolution. Structural and functional data suggested that all three proteins are effectors of the BMP signaling pathway, but expression data revealed different spatiotemporal patterns in teleost fish suggesting distinct mechanisms of regulation. In this work, through the collection of novel data, we provide additional insight into the regulation, the structure and the phylogenetic relationship of BMP2 and its closely related family members.

Gla-rich protein (GRP) is a vitamin K-dependent protein related to bone and cartilage recently described. This protein is characterized by a large number of Gla (γ-carboxyglutamic acid) residues being the protein with the highest Gla content of any known protein. It was found in a widely variety of tissues but highest levels was found in skeletal and cartilaginous tissues. This small secreted protein was also expressed and accumulated in soft tissues and it was clearly associated with calcification pathologies in the same tissues. Although the biological importance of GRP remains to be elucidated, it was suggested a physiological role in cartilage development and calcification process during vertebrate skeleton formation. Using zebrafish, an accepted model to study skeletal development, we have described two grp paralog genes, grp1 and grp2, which exhibited distinct patterns of expression, suggesting different regulatory pathways for each gene. Gene synteny analysis showed that grp2 gene is more closely related to tetrapod grp, although grp1 gene was proposed to be the vertebrate ortholog by sequence comparison. In addition, we identified a functional promoter of grp2 gene and using a functional approach we confirmed the involvement of transcription factors from Sox family (Sox9b and Sox10) in the regulation of grp2 expression. In an effort to provide more information about the function of grp isoforms, we generated two zebrafish transgenic lines capable to overexpress conditionally grp genes and possible roles in the skeleton development were studied. To better understand GRP function a mammalian system was used and the analysis of knockout mice showed that GRP is involved in chondrocyte maturation and the absence of GRP is associated to proteoglycans loss in calcified articular cartilage. In addition, we detected differences in chondrogenesis markers in articular chondrocyte primary culture. Overall, our data suggest a main role for GRP on chondrocyte differentiation.
