Studies of HeLa cells and serum- and glucocorticoid-regulated kinase 1 (SGK1) knockout mice identified threonine residues in the n-myc downstream-regulated gene 1 protein (NDRG1-Thr(346/356/366)) that are phosphorylated by SGK1 but not by related kinases (Murray et al., Biochem J 385:1-12, 2005). We have, therefore, monitored the phosphorylation of NDRG1-Thr(346/356/366) in order to explore the changes in SGK1 activity associated with the induction and regulation of the glucocorticoid-dependent Na+ conductance (G (Na)) in human airway epithelial cells. Transient expression of active (SGK1-S422D) and inactive (SGK1-K127A) SGK1 mutants confirmed that activating SGK1 stimulates NDRG1-Thr(346/356/366) phosphorylation. Although G (Na) is negligible in hormone-deprived cells, these cells displayed basal SGK1 activity that was sensitive to LY294002, an inhibitor of 3-phosphatidylinositol phosphate kinase (PI3K). Dexamethasone (0.2 mu M) acutely activated SGK1 and the peak of this response (2-3 h) coincided with the induction of G (Na), and both responses were PI3K-dependent. While these data suggest that SGK1 might mediate the rise in G (Na), transient expression of the inactive SGK1-K127A mutant did not affect the hormonal induction of G (Na) but did suppress the activation of SGK1. Dexamethasone-treated cells grown on permeable supports formed confluent epithelial sheets that generated short circuit current due to electrogenic Na+ absorption. Forskolin and insulin both stimulated this current and the response to insulin, but not forskolin, was LY294002-sensitive and associated with the activation of SGK1. While these data suggest that SGK1 is involved in the control of G (Na), its role may be minor, which could explain why sgk1 knockout has different effects upon different tissues.

The network is currently not only widely used for performative actions, but also extensively theorised. However, the artistic strategies in networked environments and the ways in which these new performance practices and cultural contexts can give rise to new design approaches have not been equally explored. This article therefore investigates the notion of dramaturgy as it provides a useful framework for addressing design strategies and performative relationships in networked environments. It is argued that dramaturgies suggest a robust method for understanding artistic practices in the network. The author refers to two different viewpoints that emerge in networked practice, which are entitled ;views from within' and ;views from without'. The article gives a brief overview of the history and the theories of the network. It then looks at the histories of dramaturgies in order to better understand artistic strategies, and in order to enhance collaborative work in network performance environments.

The present study examines those features which promote bat feeding in agricultural riparian areas and the riparian habitat associations of individual species. Activity of Nathusius' pipistrelle (Pipistrellus nathusii), common pipistrelle (Pipistrellus pipistrellus), soprano pipistrelle (Pipistrellus pygmaeus), Leisler's bat (Nyctalus leisleri), and Myotis species (Myotis sp.) were recorded, and their habitat associations both "between" and "within" riparian areas were analyzed. General feeding activity was associated with reduced agricultural intensity, riparian hedgerow provision, and habitat diversity. Significant habitat associations for P. pipistrellus were observed only within riparian areas. Myotis species and P. pygmaeus were significantly related to indices of landscape structure and riparian hedgerow across spatial scales. Myotis species were also related to lower levels of riffle flow at both scales of analysis. The importance of these variables changed significantly, however, between analysis scales. The multi-scale investigation of species-habitat associations demonstrated the necessity to consider habitat and landscape characteristics across spatial scales to derive appropriate conservation plans.

Teaching on social work values is centrally important in social work education as a core aspect of underpinning knowledge in preparing students for practice. This paper describes an innovative project occurring within the first year of the degree in social work, where service users and carers have assisted students with their understanding of social work values. The positive contribution of service users and carers in facilitating students to make links between theory and practice is now well documented. Applying this user perspective to the educational domain of values, however, is relatively uncharted territory given the challenges that have traditionally accompanied the teaching of values. Importantly, this paper describes the ‘value talk’ which occurred when first-year students sought further meaning from service-user and carer groups in their community settings following classroom teaching on values. The paper not only discusses the detailed preparations involved in the project but also the learning which resulted, drawing on the evaluation findings from the students and participating groups. Whilst the findings show that the students' understanding of social work values has been most significantly influenced by the contributions from service users and carers, it is recognized that further research is needed to monitor the longer term impact on social work students' practice after they qualify.

Herein batch and continuous mesophilic anaerobic digestion of grass silage liquor was studied. The continuous process was carried out in Armfield digesters with an OLR ranging from 0.851 to 1.77 kg COD m-3 day-1. The effect of recirculation of effluent from the digester was investigated using different OLRs of grass silage liquor feed. These results showed that as the OLR increased, the methane yield decreased for the reactor with no recycle and increased for the reactor with recycle. However, the COD removal for both digesters was nearly the same at the same OLR. Overall these studies show that grass silage liquor can produce a high quality methane steam between 70% and 80% and achieve methane yields of 0.385 m3 kg-1 COD.

Many different immunochemical platforms exist for the screening of naturally occurring contaminants in food from the low cost enzyme linked immunosorbent assays (ELISA) to the expensive instruments such as optical biosensors based on the phenomenon of surface plasmon resonance (SPR). The primary aim of this study was to evaluate and compare a number of these platforms to assess their accuracy and precision when applied to naturally contaminated samples containing HT-2/T-2 mycotoxins. Other important factors considered were the speed of analysis, ease of use (sample preparation techniques and use of the equipment) and ultimately the cost implications. The three screening procedures compared included an SPR biosensor assay, a commercially available ELISA and an enzyme-linked immunomagnetic electrochemical array (ELIME array). The qualitative data for all methods demonstrated very good overall agreements with each other, however on comparison with mass spectrometry confirmatory results, the ELISA and SPR assay performed slightly better than the ELIME array, exhibiting an overall agreement of 95.8% compared to 91.7%. Currently, SPR is more costly than the other two platforms and can only be used in the laboratory whereas in theory both the ELISA and ELIME array are portable and can be used in the field, but ultimately this is dependent on the sample preparation techniques employed. Sample preparative techniques varied for all methods evaluated, the ELISA was the most simple to perform followed by that of the SPR method. The ELIME array involved an additional clean-up step thereby increasing both the time and cost of analysis. Therefore in the current format, field use would not be an option for the ELIME array. In relation to speed of analysis, the ELISA outperformed the other methods.

The chromium bearing wastewater in this study was used to simulate the low concentration discharge from a major aerospace manufacturing facility in the UK. Removal of chromium ions from aqueous solutions using raw dolomite was achieved using batch adsorption experiments. The effect of; initial Cr(VI) concentration, amount of adsorbent, solution temperature, dolomite particle size and shaking speed was studied. Maximum chromium removal was found at pH 2.0. A kinetic study yielded an optimum equilibrium time of 96 h with an adsorbent dose of 1 g/L Sorption studies were conducted over a concentration range of 5-50 mg/L Cr(VI) removal decreased with an increase in temperature (q(max): 20 degrees C = 10.01 mg/g; 30 degrees C = 8.385 mg/g; 40 degrees C = 6.654 mg/g; and 60 degrees C = 5.669 mg/g). Results suggest that the equilibrium adsorption was described by the Freundlich model. The kinetic processes of Cr(VI) adsorption onto dolomite were described in order to provide a more clear interpretation of the adsorption rate and uptake mechanism. The overall kinetic data was acceptably explained by a pseudo first-order rate model. Evaluated Delta G degrees and Delta H degrees specify the spontaneous and exothermic nature of the reaction. Data regarding the total number of anti-asthma drugs dispensed via community pharmacies within Northern Ireland from 1980 to 1997 were collected and analysed. The use of anti-asthma drugs within this population increased markedly over the study period from 19.84 DDDs/1000/day to 84.07 DDDs/1000/day. With the exception of the non-selective beta(2)-agonists, there has been an overall increasing trend in the utilization of all of the anti-asthma drugs during the study period. Most of the increase is attributable to the increase in prescribing of the selective beta(2)-agonists and the glucocorticoids, This increase in the utilization of anti-asthma drugs may be explained by an increasing prevalence of the condition, increased adherence to asthma management protocols or to the prescribing of more intensive drug therapies.
