Active transport of substrates across cytoplasmic membranes is of great physiological, medical and pharmaceutical importance. The glycerol-3-phosphate (G3P) transporter (GlpT) of the E. coli inner membrane is a secondary active antiporter from the ubiquitous major facilitator superfamily that couples the import of G3P to the efflux of inorganic phosphate (Pi) down its concentration gradient. Integrating information from a novel combination of structural, molecular dynamics simulations and biochemical studies, we identify the residues involved directly in binding of substrate to the inward-facing conformation of GlpT, thus defining the structural basis for the substrate-specificity of this transporter. The substrate binding mechanism involves protonation of a histidine residue at the binding site. Furthermore, our data suggest that the formation and breaking of inter- and intradomain salt bridges control the conformational change of the transporter that accompanies substrate translocation across the membrane. The mechanism we propose may be a paradigm for organophosphate:phosphate antiporters.

A tricyclic core structure related to gelsemine was synthesized from an oxabicyclo[3.2.1]octanone by a three-step bridge swapping strategy involving elimination of the bridging ether oxygen and intramolecular Michael addition of a tethered cyanoacetamide unit.

Carbon fiber reinforced polymer (CFRP) bars were prestressed for the structural strengthening of 8 T-shaped reinforced concrete (RC) beams of a 21-year-old bridge in China. The ultimate bearing capacity of the existing bridge after retrofit was discussed on the basis of concrete structures theory. The flexural strengths of RC beams strengthened with CFRP bars were controlled by the failure of concrete in compression and a prestressing method was applied in the retrofit. The field construction processes of strengthening with CFRP bars—including grouting cracks, cutting groove, grouting epoxy and embedding CFRP bars, surface treating, banding with the U-type CFRP sheets, releasing external prestressed steel tendons—were introduced in detail. In order to evaluate the effectiveness of this strengthening method, field tests using vehicles as live load were applied before and after the retrofit. The test results of deflection and concrete strain of the T-shaped beams with and without strengthening show that the capacity of the repaired bridge, including the bending strength and stiffness, is enhanced. The measurements of crack width also indicate that this strengthening method can enhance the durability of bridges. Therefore, the proposed strengthening technology is feasible and effective.

In the investigation of real loading capacities in concrete bridge deck slabs, the study of this type of structure was carried out with consideration of compressive membrane action.A series of experimental test of steel-concrete bridge structures was developed with the analysis of influences from the varying of structural parameters on loading capacities, including reinforcement percentages, supporting beam sizes and concrete compressive strength.Through the study of the experimental results,it was found that the real structural loading capacities are larger than those predicted by current design methods. Therefore, based on the previous research,a prediction method for loading capacities of concrete bridge deck slabs was established with consideration of CMA,which was built based on the plastic ultimate analysis. In this method, the lateral restraint stiffness subjected by concrete bridge deck slabs was provided. The proposed theoretical model is capable of predicting the loading capacities of this type of structure accurately with comparison of results from several bridge deck experimental tests.

Sustainability is now recognised as a key issue that must be addressed in the design, construction and lifelong maintenance of civil engineering structures. This paper briefly discusses the generic aspects of sustainability, but the main focus is its application to bridges. Motorway bridges built in the 1960s and 1970s had design lives of 120 years; many, however, were showing signs of deterioration after only 20–40 years. This led to much (ongoing) debate on the issue of initial versus full life-cycle costing. In order to address the highly complex issue of the sustainability of bridges, this paper considers the following specific areas that impinge on this important subject: the impact on sustainability of different forms of bridge construction and maintenance/repair/replacement strategies; the utilisation of innovative in situ testing equipment for assessing the long-term durability of concrete; the development of innovative structural designs for bridges that inherently have greatly extended lives at minimal, if any, additional cost.
