川西北高寒草甸位于青藏高原东部地区,是我国四大牧区之一,也是长江和黄河等江河源区的重要水源涵养功能区。近几十年来,大量牦牛粪便被牧民作为生物能源、肥料或者食用菌产业的原料而利用,为草原生态系统的养分平衡增加了生态风险。鉴于在青藏高原地区针对牦牛粪便的相关研究尚未见报道,本文从粪便和土壤养分相互变化趋势的角度出发,研究了粪便在夏秋两季的分解状况和粪便其下及其周围土壤的养分变化。通过此研究,将有助于了解牦牛粪便在自然状态下的分解速率以及粪便对土壤养分及土壤微生物生物量的影响,为粪便对草地土壤生态系统的贡献提供概念性认识,同时也为高寒草甸草地这种脆弱生态系统的可持续管理提供理论依据。 针对牦牛粪便自身养分释放及其对土壤在时间和空间尺度上的影响,本文通过模拟牛粪堆积,在不同时间和固定区域内对牛粪和土壤进行了夏秋两季的采样测定,分析了牛粪及土壤NO3--N、NH4+-N、速效K、无机P、有机C、全N 和全P 含量随时间的变化趋势。得到如下结果: 1. 在研究区域内,牛粪对草地生态系统具有较强的养分(N、P)贡献能力。据初步统计,其估值大致为氮素699~932 kg ha-1,P 素为110~147 kg ha-1。牛粪(湿重、干重)在夏秋季节的分解速率具有较大差异,夏季显著快于秋季。夏季牛粪湿重、干重在2 个月左右之后分别降为初期的14%和24%,而秋季同期分别降为35%和52%。牛粪养分(NO3--N、NH4+-N、速效K、无机P、有机C、全N 和全P)的分解速率在夏季也要显著快于秋季。秋季经过2 个月左右的分解之后,牛粪以上含量分别降至初始态的32%、60%、36%、64%、58%、63%和43%,远高于夏季的同期水平。 2. 在不同季节,牛粪周围的土壤养分(NO3--N、NH4+-N、速效K 和无机P)含量变化随时间改变呈现相同的趋势。但是,牛粪周围不同远近下土壤养分随时间的动态变化幅度略有不同。粪下土壤养分含量随时间变化波动较大,距粪便越远,随时间变化的变幅越为平缓。总体来说,在夏季由于粪便分解较快,对土壤养分的持续作用时间不及秋季,秋季粪便分解变慢,表现出养分的缓释特征。其次,牛粪对粪下土壤影响的持续时间也长于对周围土壤的作用时间。 3. 粪便对土壤养分(NO3--N、NH4+-N、速效K、无机P)影响的范围在不同季节具有差异。夏季要高于秋季,但对周围土壤养分影响的持续时间低于秋季。在夏季短期内,牛粪对土壤NO3--N 和速效K 含量的影响范围能够超过30cm,而对NH4+-N 和无机P 的影响范围则介于10cm 和30cm 之间。在2 个月左右之后,牛粪对周围土壤养分的影响能力基本消失。在秋季,牛粪对周围土壤养分影响范围难以达到30cm 处。粪便在夏季对其下土壤和周围土壤的有机C、全N 和全P 含量并无显著影响,但在秋季能显著增加其下土壤有机C、全N 和全P 含量。 4. 牛粪在秋季对土壤SMB-C、SMB-N 和SMB-P 含量的影响能够持续2 个月以上,由于秋冬季节牛粪分解缓慢,因此推断这种效应持续时间至少能够1 年左右。另一方面,牛粪在秋季对土壤SMB-C、SMB-N 和SMB-P 含量的影响范围主要集中在其下土壤,而周围的影响效应并不明显。 The grassland on the eastern fringe of Qinghai-Tibetan Plateau was one of the four greatest pasture zones in our country and the main water conservation function zones in the hesastream of Yangtse River and Huanghe river. Rencent years, lots of dung in this area was used as biological energy, fertilizer or material of fungoid growing, leading to high risk of nutrient banlance in grassland ecosystem. In view of the researches on the impact of yak dung in this area are relatively rare, the present study focused on the relationship of dung and soil nutrient transformation in sunmer and autumn, which could profoundly illuminate the mechanism of dung decomposition and the effect of dung on soil chemical properties and soil microbe biomass. The present study also contributed to a basic understand and provided scientific management in the high-frigid ecosystem. Decomposition of yak dung and its effect on soil chemical properties in eastern grassland of Qinghai-Tibetan Plateau were determined. The study simulated the real dung pats, took dung and soil samples at different time and fixed-point in summer and autumn. The samples were analysed for NO3--N, NH4+-N, available K, inorganic P, total organic C (TOC), total N (TN), total P (TP). It was concluded that: 1. In study area, the yak dung supplied to ecosystem substantial nutrient. It is estimated that the N contribution of dung was approximately 699~932 kg ha-1, P contribution was approximately 110~147 kg ha-1. The rate of yak decomposition was more rapid in summer than autumn, the wet and dry weight of yak decreased to 14% and 24% respectively after 2 months when dung excreted in summer, with 35% and 52% in autumn. The content of NO3--N、NH4+-N、available K、inorganic P、TOC、TN and TP in dung decomposed more rapid in summer too. After 2 months when dung excreted in autumn, the content of above nutrient decreased to 32%、60%、36%、64%、58%、63% and 43% respectively,which were significantly higher than summer. 2. The content of NO3--N、NH4+-N、available K and inorganic P in soil around dung had the same transformation trend in each season, whereas it was distinguishing at different gradient of distance from dung, the nutrient in soil below dung had the most significant change while the more far from dung, the less change in soil. It was concluded that the yak dung had prolong impacts on soil in autumn compared with summer, besides, it aslo showed that the yak dung had protract effect on soil below dung compared with soil around dung. 3. The yak dung had expansive impact on soil around dung in summer whereas had relatively short effect compared with autumn. In short-term at summer, there was a significant increase about the content of NO3--N and available K around 30cm radius from dung pat while the content of NH4+-N and inorganic P between 10cm to 30cm. After 2 months, the impact almost disappeared. In autumn, the effect was hard to reach 30cm. The yak dung had no significant effects on the content of TOC、TN and TP in soil below or around dung in summer whereas there was a obvious increase in soil below dung pat in autumn. 4. The duration of effect of yak dung on soil microbial biomass(SMB) C、N and P was at least 2 months, maybe even more than 1 year. On the other hand, the impact of dung on SMB-C、SMB-N and SMB-P mainly acted on soil below dung while no obious effect on soil around dung.

Hybridization between yak Poephagus grunniens and taurine Bos taurus or indicine B. indicus cattle has been widely practiced throughout the yak geographical range, and gene flow is expected to have occurred between these species. To assess the impact of cattle admixture on domestic yak, we examined 1076 domestic yak from 29 populations collected in China, Bhutan, Nepal, India, Pakistan, Kyrgyzstan, Mongolia and Russia using mitochondrial DNA and 17 autosomal microsatellite loci. A cattle diagnostic marker-based analysis reveals cattle-specific mtDNA and/or autosomal microsatellite allele introgression in 127 yak individuals from 22 populations. The mean level of cattle admixture across the populations, calculated using allelic information at 17 autosomal microsatellite loci, remains relatively low (mY(cattle) = 2.66 +/- 0.53% and Q(cattle) = 0.69 +/- 2.58%), although it varies a lot across populations as well as among individuals within population. Although the level of cattle admixture shows a clear geographical structure, with higher levels of admixture in the Qinghai-Tibetan Plateau and Mongolian and Russian regions, and lower levels in the Himalayan and Pamir Plateau region, our results indicate that the level of cattle admixture is not significantly correlated with the altitude across geographical regions as well as within geographical region. Although yak-cattle hybridization is primarily driven to produce F-1 hybrids, our results show that the subsequent gene flow between yak and cattle took place and has affected contemporary genetic make-up of domestic yak. To protect yak genetic integrity, hybridization between yak and cattle should be tightly controlled.

In 6 Chinese yak (Bos. grunniens) populations including 177 yaks, 34 blood protein loci were studied by horizontal starch gel electrophoresis, four of these loci (AKP: ALB, LDH-1, TF) were found to be polymorphic. The percentage of polymorphic loci(P) is 0.118, the mean individual heterozygosity(H) is 0.015, which means a low level of genetic diversity in the whole Chinese yak population. The coefficient of gene differentiation (G(ST)) is 0.0625, which indicated an almost-indistinguishable divergence among different populations at the level of blood protein electrophoresis.

Adaptation to hypoxia is regulated by hypoxia-inducible factor 1 (HIF-1), a heterodimeric transcription factor consisting of an oxygen-regulated a-subunit and a constitutively expressed beta-subunit. How animals living on Qinghai-Tibetan plateau adapt to the extreme hypoxia environment is known indistinctly. In this study, the Qinghai yak which has been living at 3000-5000 m attitude for at least two millions of years was selected as the model of high hypoxia-tolerant adaptation species. The HIF-1 alpha ORFs (open reading frames) encoding for two isoforms of HIF-1 alpha have been cloned from the brain of the domestic yak. Its expression of HIF-1 alpha was analyzed at both mRNA and protein levels in various tissues. Both its HIF-1 alpha mRNA and protein are tissue specific expression. Its HIF-1 alpha protein's high expression in the brain, lung, and kidney showed us that HIF-1 alpha protein may play an important role in the adaptation to hypoxia environment. (c) 2006 Elsevier Inc. Six domestic yaks aged 6 years, comprising three culling male and three culling female yaks, from the Qinghai Lake area and three male semi-wild yaks aged 6 months from Datong Yak Farm were used to detect the levels of amino acids, mineral elements, residual heavy metals and pesticides in yak meat. The results showed that there was little difference in amino acid levels for different types of yak by age and sex. The meat of the adult male yak was 7.35 mg/kg higher in calcium (P < 0.05) and 124 mg/kg higher in phosphorus (P < 0.01) than that of the adult female yak. There was an obvious difference in zinc content, and there was also a significant difference for copper and sulfur between adult male and female yaks (P < 0.01). Iron was 8.85 mg/kg higher in semi-wild yaks than in domestic yaks, but there were no differences for other minerals. The levels of residual heavy metals and residual pesticides were well within those allowed by the National Standard of China. Furthermore, the data showed that introducing wild yak's blood did not result in a quality change in the domestic yak meat. Yak meat from Qinghai Plateau has the characteristics of being non-polluted and rich in amino acids in comparison with local yellow cattle meat.

Forty-five male yaks (born April 2001) were studied to determine how seasonal changes on the Qinghai-Tibetan plateau affected BW and body composition. Thirty yaks were weighed monthly from birth to 26 mo of age to determine seasonal changes in BW. The remaining 15 yaks were allocated randomly to five groups (three yaks per group), designated for slaughter at 13, 15, 18, 22, and 25 mo to measure seasonal effects on body chemical composition. All yaks were grazed on the alpine-meadow grassland of the plateau without any supplementation. All BW and body composition data were calculated on an individual basis. Body weight and body composition data were both compared across seven growth periods spanning 2 yr and defined by season. From April (birth) to December 2001 of the first growing season, yak BW increased (P < 0.01); however, during the subsequent cold season (December 2001 to May 2002), BW decreased (P < 0.01). The second growing season ran from May 2002 (13 mo of age) to October 2002 (18 mo of age), and the second live weight-loss season ran from October 2002 until May 2003. The weight loss experienced by yaks during the first weight loss season was 25.64% of the total weight gain in the first growing season. The weight loss experienced by yaks during the second weight-loss season was 29.73% of the total weight gain in the second growing season. Energy retention in the second growing season was 291.07 MJ, 50.8% of which was consumed during the subsequent cold season. Energy accumulation in the summer (from May to July) and fall (from July to October) of the second growing season did not differ (5.01 and 6.30 MJ/kg of EBW gain, respectively; P = 0.63). The energy mobilized during the second winter (from October 2002 to February 2003) was 16.49 MJ/kg of EBW, and in the second spring (from February to May 2003), it was 9.06 MJ/kg of EBW. These data suggest that the decrease in grazing yak BW during the first cold season is much less than during the second cold season, and that the energy content per unit of BW mobilized is greater (P = 0.02) in winter than in spring. Results from this study demonstrate highly efficient compensatory growth in grazing yaks following the first weight loss period during the first cold season. This benefit could be exploited by herders to improve yak production. Yaks may have developed a type of self-protection mechanism to overcome the long cold seasons in the Qinghai-Tibetan plateau.

It can be served as the theory basis from which crossbreed improvement of yak and government decision could be drawn.

Wheat straw was treated with microwave for 4 min and 8 min at a power of 750 W and frequency of 2,450 MHz. Chemical compositions of untreated, 4 min treated and 8 min treated Straws were analyzed and in sacco degradabilities of all these straws in yak rumens were measured. Microwave treatment didn't significantly (p > 0.05) affect the chemical composition of the straw. In sacco dry matter (DM) degradability of the straw after 18 h incubation in rumen was significantly (p < 0.01) improved by microwave treatment. In sacco crude protein (CP) degradability of the straw was not (p > 0.05) affected by microwave treatment. In sacco organic matter (OM) degradability of the straw was increased (p < 0.01) by around 20% for both the 4 min and 8 min microwave treatment, that of acid detergent fibre (ADF) was increased (p < 0.01) by 61.6% and 62.8%, and that of ash free ADF was enhanced by 72.1% and 69.6% for the 4 min and 8 min microwave treatment respectively. No significant difference was observed between the 4 min and 8 min microwave treatment on the degradability of DM, OM, CP, ADF and ash-free ADF of the straw.

A feeding trial A as conducted at the farm of Qinghai Academy of Animal and Veterinary Science, Xining, China during 1996 - 1997 with three dry yak cows (initial body weight 163 - 197 kg, age 5 - 6 years) by using 3 x 3 Latin Square Design to determine the effect of levels of feed intake on digestion, nitrogen balance and purine derivative excretion in urine of yak cows. The animals were fed oat hay (nitrogen 13.5 g/kg dry matter (DM), metabolisable energy 8.3 MJ/kg DM), i.e., 0.3, 0.6 and 0.9 of voluntary intake (VI). Each intake treatment lasted for 17 days and the samples (feeds, faeces and urine) were collected during last 7 days of each period. The results indicate that digestibility of dietary DM, OM, NDF and ash declined when intake levels increased from 0.3 to 0.9 VI [DM, from 66.1% to 59.1% (P < 0.05); OM, from 68.1% to 59.9% (P < 0.05); NDF, from 62.1% to 54.3% (P < 0.05); and ash, from 33.9% to 11.8% (P < 0.05)]. Around 0.10 g N/kg W-0.75 was deficient daily in yak cows at 0.3 VI, and positive N balances were observed at 0.6 and 0.9 VI. Intake levels significantly (P < 0.05) affected total PD excretion in yak urine. The proportion of allantoin increased (P < 0.05) and uric acid decreased (P < 0.05) as intake level of feed increased. Using three different types of yaks, 30 head of 1-yr-old calves (liveweight 60.7 +/- 6.3 kg), 30 head of 2-yr-old calves (97.7 +/- 10.2 kg) and 30 head of yak cows (160.2 +/- 15.1 kg) were randomly selected from the same yak herds. Each type of yak herd was divided into control (C) and trial (T) groups using a completely randomized design, with 10 and 20 animals in the C and T groups, respectively. The animals in the C group were grazed on natural rangeland, and the animals in the T group were supplemented with urea multinutritional molasses blocks (UMMB), together with grazing on natural rangeland from January to May of 1998. The objective was to determine the effect of UMMB on productive performance of yak calves and yak cows in the cold season. Live weight loss of 1-yr-old calves, 2-yr-old calves and yak cows was reduced by 1.2, 8.3 and 7.9 kg after UMMB supplementation (P < 0.01). The 1-yr-old calves gained the most in the first month of supplementation, but the 2-yr-old calves and yak cows gained the most both in the first and last supplementation months. Daily milk yield of yak cows increased by 0.21 kg d(-1) when the lactating animals were supplemented with UMMB (P < 0.01), although there was no effect (P < 0.01) of UMMB supplementation on hair and downy hair production. Supplementation with UMMB also improved reproductive performance of yak cows, with 8.8 and 30.9% increments in pregnancy rate and newborn weight, respectively. We conclude that the benefit of UMMB supplementation the 1-yr-old calves was not economical, with only 0.3:1 output to input ratio, but supplementation of the 2-yr-old calves and yak cows may be economical, with 1.8:1 and 1.4:1 output to input ratios, respectively.

Ten restriction endonucleases were used to investigate the mitochondrial DNA restriction fragment length polymorphism (mtDNA RFLP) of 11 native cattle breeds and one cultivated cattle breed in South China. Twenty-three restriction morphs were detected, which can be sorted into five haplotypes, A phylogenetic tree of the haplotypes was constructed by using the 'upgma' method. Our study showed that haplotype I and II are identical to the zebu (Bos indicus) and taurine (Bos taurus) haplotypes, respectively. Zebu and taurine were the two major origins of cattle populations in South China, and the zebu probably had more influence on the native cattle population than taurine did. Haplotype III is identical to haplotype I of yak (Bos grunniens), which was only detected in the Diqing cattle breed. Haplotype IV was detected for the first time. This haplotype, found only in Dehong cattle, might be from an independent domestication event, probably from another Bos indicus population. Divergence of haplotypes I and IV occurred about 268,000-535 000 years ago, much earlier than the 10,000-year history of cattle husbandry. Our results also suggest a secondary introgression of mtDNA from yak to Diqing cattle.
