低温是限制植物分布和生物产量的一个重要环境因子。低温的危害也是农业生产上经常遭受的主要自然灾害之一。改良作物的抗寒性是植物科学研究的一个重要课题。植物基因工程的兴起为此目的提供了有力的手段。 大量的研究证明,低温对植物的伤害,首先是使生物膜发生相变和相分离。因此,保持低温下生物膜功能性的液晶态是抗寒的重要机制。研究表明,生物膜这种具流动性的功能态的保持,是与其组成上的膜脂脂肪酸的不饱和度成正相关的。 已有几个关于通过提高膜脂脂肪酸不饱和度的基因操作而增加植物抗寒性的报道。在众多的植物脂肪酸去饱和酶中,硬脂酰ACP去饱和酶(SAD)是最为关键的酶之一。它催化脂肪酸的第一步去饱和反应:18:0-18:l¨。多不饱和脂肪酸是在1 8:1 中由其他去饱和酶再加入双键而生成的。因此,SAD的活性水平是决定植物膜脂不池和度的一个关键因素。 本研究以酸酚法提取的菠菜总RNA为材料,采用反转录一PCR的方法,克隆得到SAD基因,经定向缺失法获得一套该基因的缺失突变体后,用DNA 自动测序仪和双脱氧链终止法测序。获得的SAD基因序列与Nishlda(1992)等发表的菠菜sADcDNA核苷酸序列比较。两者的编码SAD的ORF都为ll97bp,核苷酸差异仅为8bp。但令人惊奇的是我们克隆到的基因,其5‘端上游还存在-个小的ORF,长30bp.编码10个氨基酸。其他报道的SAD基因中都没有这个ORF。 将克隆到的SAD基因构建成两个植物双元表达载体:正义的pB112-13和反义的pB112-6。用叶盘法转化烟草。DNA点杂交和Southern杂交筛选出转基因植株。抗寒性测定表明:当植株置于6'C40小时,转基因植株和对照的相对电导率比较一致,无明显改变;而在88小时,pB121-6转化植株和对照的相对电导率明显升高,以pB1121-6植株升高更多,但pB1121-13转化植株的相对电导率始终保持在较低水平。短暂冰冻处理(-20'C,4O分钟)后置室温下4天,pBl121-6转化烟草总叶绿素含量损失最多,对照次之,而pB1121-13转化烟草中多数植株总叶绿素损伤量都低于其他两种烟草。从这两个抗寒性测定实验,可判定pBl l 2 l—I 3烟株最抗寒,对照次之,而pB1121-6烟株最不抗寒。 由于pB1121-13是增强转基因烟草中SAD活性,而pB1121-6是削弱SAD基因的表达,因此.本研究首次证明通过SAD的基因工程可以改变植物的抗寒性。

水母雪莲(Saussurea medusa Maxim.)为多年生菊科植物,是我国珍稀药用资源。所含的主要生物活性成分是黄酮类物质,具有抗炎、镇痛、免疫抑制及抗氧化等功效。但由于水母雪莲生长环境特殊,生长缓慢,人工引种困难;加上长期掠夺性采挖,造成其野生药用资源短缺,已经不能满足市场的需求。近年来,世界上掀起了植物药开发的热潮,植物药以其天然低毒的特点倍受关注,而黄酮类化合物更是以其广谱的药理作用引人瞩目。 黄酮类化合物的合成代谢途径在植物界进化过程中很保守,黄酮类生物合成途径中的相关酶也已得到确证并进行了系统的研究。二氢黄酮醇-4-还原酶(Dihydroflavonol-4-reductase, DFR)是一个处于花色素或者原花色素合成途径中的关键酶,它与黄酮合成途径中的黄酮醇合成酶(flavonol synthase)竞争底物。本研究以水母雪莲为研究对象,根据近缘物种DFR基因的保守核苷酸序列设计兼并引物,通过PCR技术,从已经建立的水母雪莲红色愈伤组织cDNA文库中筛选到一个编码该酶的cDNA序列,该序列全长1166个碱基对。根据生物信息学分析,此cDNA编码342个氨基酸;Blastp分析结果显示,该氨基酸序列与同科植物翠菊(Callistephus chinensis)的相似性最高,达87%;SWISSMODEL软件预测其蛋白的三级结构与葡萄(Vitis vinifera)的十分相似,活性中心的关键氨基酸残基也完全一致。据此可以断定,我们所得到的cDNA为编码二氢黄酮醇还原酶的基因,并命名为水母雪莲二氢黄酮醇还原酶基因(SmDFR)。为了得到SmDFR的DNA序列,我们又设计特异引物,从水母雪莲的基因组中扩增出了由1871个碱基对组成的DNA序列,该序列包含五个内含子和六个外显子。 为了提高水母雪莲和大苞雪莲中黄酮类物质的含量,我们构建了SmDFR的反义植物表达载体,利用根癌农杆菌介导进行基因转化。通过改变影响农杆菌转化的实验条件包括外植体来源、农杆菌菌株、细菌浓度、外植体预培养时间、侵染时间和乙酰丁香酮的浓度进行转基因试验,目前尚未得到转基因植株。另外,我们构建了SmDFR正义植物表达载体,通过对拟南芥(Arabidopsis thaliana) DFR基因突变体和矮牵牛(Petunia hybrida)进行基因转化,来验证SmDFR的功能;目前,此实验尚在进行之中。

Junggar Basin has a large amount of recoverable reserves, However, due to the unfavorable factors, such as bad seismic data quality, complex structure with many faults and less wells, the exploration of oil and gas is still relatively limited, so advanced theoretical guidance and effective technical supports are desirable. Based on the theories of sedimentology, as well as comprehensive studies of outcrops, seismic data, drilling data and setting of this area, the paper establishes the isochronous correlation framework, and analyzes the sedimentary facies types and provenance direction, and obtains the profile and plain maps of the sedimentary facies combined with the logging constrained inversion. Then the paper analyzes the reservoir controlling factors, reservoir lithology attribute, 4-property relationship and sensibility based on the sedimentary facies research, and sets up a 3D geological model using facies controlled modeling. Finally, the paper optimizes some target areas with the conclusions of reservoir, structure and reservoir formation.Firstly, the paper establishs the isochronous correlation framework by the seismic data, drilling data and setting of this area. The sedimentary facies in Tai13 well block are braided river and meandering river according to the analysis of the lithology attribute, logging facies and sedimentary structure attribute of outcrop. The concept of “wetland” is put forward for the first time. The provenance direction of Badaowan and Qigu formation is obtained by the geology setting, sedimentary setting and paleocurrent direction. The paper obtains the profile and plain maps of the sedimentary facies from the sand value of the wells and the sand thickness maps from the logging constrained inversion. Then, this paper takes characteristics and control factors of the Jurassic reservoirs analysis on thin section observation, scanning transmission electron microscope observation and find out the petrology characteristics of reservoir, space types of reservoir and lithofacies division. In this area, primary pores dominate in the reservoir pores, which believed that sedimentation played the most important roles of the reservoir quality and diagenesis is the minor factor influencing secondary porosity. Using stochastic modeling technique,the paper builds quantitative 3-D reservoir Parameter. Finally, combined the study of structure and reservoir formation, the reservoir distribution regularity is concluded: (a) structures control the reservoir formation and accumulation. (b) Locating in the favorable sedimentary facies belt. And the area which meets these conditions mentioned above is a good destination for exploration.

So far the studies on personality are based on indirect way due to the complexity and limitation. Present research attempt to find another route to approach personality through olfaction. The framework of present research is on account of a modern personality theory model produced by the British psychologist Eysenck. The aim of present research is to prove a correlation between olfaction and personality because sensation and perception of the olfaction on special part of brain closely related to emotional arousal. This research was carefully designed and performed by both psychological questionnaire and olfaction examination. The results indicated some good correlation between smell sense and personal character, which can by applied into personality and behavior analysis.
