无根萍属(Wolffia )隶属于浮萍科天南星目,是世界上最小的被子植物。该属植物繁殖速度快;易于培养;结构简单,只具有一个雄蕊和一个雌蕊;自然状态下通常为克隆繁殖,遗传结构高度一致,具备特定研究目的模式植物的特点,正在或已经成为一些实验室研究光合作用、生物反应器、毒理学、生态修复和环境监测等的重要模式生物材料;同时还被作为建造航天生活仓和地外生命支撑系统的首选植物。该属植物蛋白含量高且氨基酸组分平衡,营养价值可与大豆相媲美。但该属植物一直是分类学界的疑难类群,不同的学者对该属的分类处理比较混乱;其次,对该属的生物地理研究也很不够,尤其是对国产类群的研究;另外,W. globosa 作为该属中国分布的物种,其生理学特性和形态结构发育还缺乏研究。为此,本文通过mat K 基因测序、RAPD 标记等手段,结合野外和室内的长期观测,对其分类和中国的地理分布以及生理学特性进行了研究。针对浮萍科植物作为水生植物,其对重金属和芳香烃衍生物的耐受逆境能力大小,和对淡水水体环境生态的指示作用。本文研究了W. globosa 具解毒功能的谷胱甘肽转硫酶的活性;最后,探索了从黄鳝(Monpterus albus Zuiew )中分离纯化GSTs 的技术与方法并对maGST 的部分特性进行了研究。主要研究结果如下: 1. Wolffia 系统分类学研究 前人认为,Wolffia 柱头的颜色是重要的分组、分种检索性状。我们对其长期、活体、原位、实时跟踪观测结果表明,柱头颜色是Wolffia 个体发育上的变化过程,不是一个稳定性状,用作Wolffia subgroup 内组的划分特征和种的鉴别特征是不适合的。在此基础上我们重新修定了该属的分种检索表。利用形态分类学性状——气孔、长/宽、高/宽以及最大宽度在水面上还是水面下等性状,认为中国分布的类群应是W. globosa,但亦有W. neglecta 存在的证据。mat K 基因片段结果支持形态学的结论。通过广泛的野外采集,在我国北京、河北和吉林发现Wolffia 的新分布。 2. 中国Wolffia 居群遗传学研究 以RAPD 分子标记对广泛分布的居群遗传多样性研究表明,无根萍属植物主要以无性繁殖方式繁育,居群主要由单一克隆后代组成,如海河流域以及松花江流域居群;但一些居群亦兼有性繁殖方式,并具较高的遗传多样性,如武汉、海南居群。利用MVSP, Popgene 和Ntsys 等分析方法探讨了中国产Wolffia 居群遗传多样性和地理分布格局间关系。 3. W. globosa 的生理学研究 建立了较为完善的W. globosa 的无菌培养和保存体系。W. globosa 在逆境中,会形成休眠体;同时,发现不同居群甚至不同克隆系之间其抗逆性和生长速度存在着显著差异,差异最大的如海南文昌居群的生长速率,是长春居群的4.19 倍;不同的时间统计生长周期存在着不同结果,生长节律每天有两个生长高峰呈双“S”型;W. globosa 的耐受温度范围和pH 范围广;低浓度的IAA,GA,6-BA, EDDHA-Fe 以及EDTA 等物质具有促进W. globosa 生长的特性;但是,所有这些处理均没能促使W. globosa 从营养生长转入生殖生长。 4. W. globosa 的解剖学研究 W. globosa 通常是进行克隆繁殖,通过组织切片发现无性分枝子体还未伸出母株之前就已经完成分化,与此同时分枝子体中又分化出新的子体,分枝呈聚伞状,子体生长方向彼此相对;另外,生殖生长结构的分化也是在母体中完成的;生殖生长点与营养生长点不是同一生长点。 5. 浮萍科植物的毒理学研究 以重金属Cr3+和芳香烃衍生物CDNB 溶液处理Wolffia,Spirodela 和Lemna, 三种水生生物,结果表明Wolffia 比Spirodela, Lemna 对重金属和芳香烃衍生物有更强的抗逆能力,如在同等条件下对于Cr3+Wolffia 的半致死剂量800GB(≈ 80mg/L),而Spirodela 和Lemna 则分别为10mg/L;20mg/L;表明W. globosa 是一个优良的生态环境修复植物。与此同时,研究了W. globosa 中具有解毒功能的GSTs 粗酶液活力在不同浓度的重金属离子(Cu2+和Cd2+)以及芳香烃衍生物(CDNB 和NBD-Cl)随处理时间的变化情况。 6. 从水生生物黄鳝Monpterus albus Zuiew 中分离纯化GSTs 的研究GSTs 活性的变化是环境监测的一个Biomarker,为此研究从M. albus 中分离 纯化GSTs 的技术与方法。经GSH 亲和层析纯化的酶活力为粗酶液的207 倍,进而鉴定了maGST 的部分特性。SDS-PAGE 电泳和MALDI-TOF/MS 表明MaGST 为同源二聚体,分子量约为52kDa,单亚基分子量约为26 kDa。maGST 酶动力学表明对CDNB 为13.07 ± 0.37 微摩尔每分钟每毫克蛋白;对NBD-Cl 为5.54 ± 微摩尔每分钟每毫克蛋白;对ECA、4-NPA 几乎没有活性。在GSH 底物饱和,CDNB 的Km 值和Vmax 分别为0.32 mM 和16.19 微摩尔每分钟每毫克蛋白;CDNB 底物饱和,GSH 的Km 值和Vmax 分别为0.44 mM 和28.83 微摩尔每分钟每毫克蛋白。maGST 的酶活性pH 值较宽,温度范围广:在pH7.0-7.5 具有最大速度,在pH6.5 和pH8.5 时分别具有65%和72%的酶活力;在45℃时具有最大活性,30℃和55℃时为最大活力的80%,60℃几乎完全丧失酶活力。

Pigment epithelium-derived factor (PEDF) is acknowledged to be a non-inhibitory member of the serine protease inhibitor (serpin) superfamily, with antiangiogenesis, and neuroprotective and immumoregulatory function, mainly in the tissues of nervous system. Here, A PEDF gene homolog, Paralichthys olivaceus PEDF (PoPEDF), was isolated from flounder embryonic cells (FEC) treated with UV-inactivated Grass carp hemorrhage virus (GCHV) and subsequently identified as a differentially expressed gene. The full length of PoPEDF cDNA is 1803 bp with an open reading frame of 1212 bp encoding a 403-amino-acid protein. This deduced protein contains an N-terminal signal peptide, a glycosylation site, a consensus serpin motif, and a 34-mer and a 44-mer fragment, all of which are very conserved in the PEDF family. PoPEDF gene exhibits a conserved exon-intron arrangement with 8 exons and 7 introns. This conserved evolutionary relationship was further confirmed by a phylogenetic analysis, where fish PEDFs and mammalian members formed a well-supported clade. Constitutive expression of PoPEDF was widely detected in many tissues. In response to UV-inactivated GCHV or poly(I:C), PEDF mRNA was upregulated in FEC cells with time. Background: Subretinal microphotodiode array (MPDA) is a type of visual prosthesis used for the implantation in the subretinal space of patients with progressive photoreceptor cell loss. The present study aimed to evaluate the effect of materials for MPDA on the viability, apoptosis and barrier function of cultured pig retinal pigment epithelium (RPE) cells.Methods: Primary culture of pig RPE cells was performed and 24 pig eyes were used to start RPE culture. The third passage of the cultures was plated on different materials for MPDA and MPDAs. The tetrazolium dye-reduction assay (MTT) was used to determine RPE cell viability. Flow cytometry was measured to indicate the apoptosis rates of RPE cells on different materials. RPE cells were also cultured on microporous filters, and the transepithelial resistance and permeability of the experimental molecule were measured to determine the barrier function.Results: The data from all the methods indicated no significant difference between the materials groups and the control group, and the materials tested showed good biocompatibility.Conclusions: The materials for MPDA used in the present study had no direct toxicity to the RPE cells and did not release harmful soluble factors that affected the barrier function of RPE in vitro.

The photosynthetic performances of regenerated protoplasts of Bryopsis hypnoides, which were incubated in seawater for 1, 6, 12, and 24 h, were studied using chlorophyll (Chl) fluorescence and oxygen measurements. Results showed that for the regenerated protoplasts, the pigment content, the ratios of photosynthetic rate to respiration rate, the maximal photosystem II (PSII) quantum yield (F-v/F-m), and the effective PSII quantum yield (I broken vertical bar(PSII)) decreased gradually along with the regeneration progress, indicated that during 24 h of regeneration there was a remarkable reduction in PSII activity of those newly formed protoplasts. We assumed that during the cultivation progress the regenerated protoplasts had different photosynthetic vigor, with only some of them able to germinate and develop into mature thalli. The above results only reflected the photosynthetic features of the regenerated protoplasts at each time point as a whole, rather than the actual photosynthetic activity of individual aggregations. Further investigation suggested a relationship between the size of regenerated protoplasts and their viability. The results showed that the middle-sized group (diameter 20-60 mu m) retained the largest number of protoplasts for 24 h of growth. The changes in F-v/F-m and I broken vertical bar(PSII) of the four groups of differently sized protoplasts (i.e. < 20, 20-60, 60-100, and > 100 mu m) revealed that the protoplasts 20-60 mu m in diameter had the highest potential activity of the photosynthetic light energy absorption and conversion for several hours.

Mammalian cells subjected to conditions of spaceflight and the microgravity environment ofspace; manifest a number of alterations in structure and function. Among the most notable changes incells flown on the Space Shuttle are reduced growth activation and decline in growth rate in the totalpopulation. Other changes include chromosomal aberrations, inhibited locomotion, alteredcytokine production, changes in PKC distribution, and increased apoptos.

A visual observation of liquid-gas two-phase flow in anode channels of a direct methanol proton exchange membrane fuel cells in microgravity has been carried out in a drop tower. The anode flow bed consisted of 2 manifolds and 11 parallel straight channels. The length, width and depth of single channel with rectangular cross section was 48.0 mm, 2.5 mm and 2.0 mm, respectively. The experimental results indicated that the size of bubbles in microgravity condition is bigger than that in normal gravity. The longer the time, the bigger the bubbles. The velocity of bubbles rising is slower than that in normal gravity because buoyancy lift is very weak in microgravity. The flow pattern in anode channels could change from bubbly flow in normal gravity to slug flow in microgravity. The gas slugs blocked supply of reactants from channels to anode catalyst layer through gas diffusion layer. When the weakened mass transfer causes concentration polarization, the output performance of fuel cells declines.

Cell adhesion, mediated by specific receptor-ligand interactions, plays an important role in biological processes such as tumor metastasis and inflammatory cascade. For example, interactions between beta(2)-integrin ( lymphocyte function-associated antigen-1 and/or Mac-1) on polymorphonuclear neutrophils (PMNs) and ICAM-1 on melanoma cells initiate the bindings of melanoma cells to PMNs within the tumor microenvironment in blood flow, which in turn activate PMN-melanoma cell aggregation in a near-wall region of the vascular endothelium, therefore enhancing subsequent extravasation of melanoma cells in the microcirculations. Kinetics of integrin-ligand bindings in a shear flow is the determinant of such a process, which has not been well understood. In the present study, interactions of PMNs with WM9 melanoma cells were investigated to quantify the kinetics of beta(2)-integrin and ICAM-1 bindings using a cone-plate viscometer that generates a linear shear flow combined with a two-color flow cytometry technique. Aggregation fractions exhibited a transition phase where it first increased before 60 s and then decreased with shear durations. Melanoma-PMN aggregation was also found to be inversely correlated with the shear rate. A previously developed probabilistic model was modified to predict the time dependence of aggregation fractions at different shear rates and medium viscosities. Kinetic parameters of beta(2)-integrin and ICAM-1 bindings were obtained by individual or global fittings, which were comparable to respectively published values. These findings provide new quantitative understanding of the biophysical basis of leukocyte-tumor cell interactions mediated by specific receptor-ligand interactions under shear flow conditions.

New methods of surface modification of transparent silicone substrate were developed, and a new set of cell culture devices that provide homogeneous substrate strain was designed. Using the new device, effects of cyclic substrate strain on bone marrow mesenchymal stem cells(MSCs) were studied. It was found that cyclic strain influenced proliferation and differentiation of bone marrow MSCs in different ways.

Dynamic properties of proteins have crucial roles in understanding protein function and molecular mechanism within cells. In this paper, we combined total internal reflection fluorescence microscopy with oblique illumination fluorescence microscopy to observe directly the movement and localization of membrane-anchored green fluorescence proteins in living cells. Total internal reflect illumination allowed the observation of proteins in the cell membrane of living cells since the penetrate depth could be adjusted to about 80 nm, and oblique illumination allowed the observation of proteins both in the cytoplasm and apical membrane, which made this combination a promising tool to investigate the dynamics of proteins through the whole cell. Not only individual protein molecule tracks have been analyzed quantitatively but also cumulative probability distribution function analysis of ensemble trajectories has been done to reveal the mobility of proteins. Finally, single particle tracking has acted as a compensation for single molecule tracking. All the results exhibited green fluorescence protein dynamics within cytoplasm, on the membrane and from cytoplasm to plasma membrane.

Macrophage differentiation antigen associated with complement three receptor function (Mac-1) belongs to beta(2) subfamily of integrins that mediate important cell-cell and cell-extracellular matrix interactions. Biochemical studies have indicated that Mac-1 is a constitutive heterodimer in vitro. Here, we detected the heterodimerization of Mac-1 subunits in living cells by means of two fluorescence resonance energy transfer (FRET) techniques (fluorescence microscopy and fluorescence spectroscopy) and our results demonstrated that there is constitutive heterodimerization of the Mac-1 subunits and this constitutive heterodimerization of the Mac-1 subunits is cell-type independent. Through FRET imaging, we found that heterodimers of Mac-1 mainly localized in plasma membrane, perinuclear, and Golgi area in living cells. Furthermore, through analysis of the estimated physical distances between cyan fluorescent protein (CFP) and yellow fluorescent protein (YFP) fused to Mac-1 subunits, we suggested that the conformation of Mac-1 subunits is not affected by the fusion of CFP or YFP and inferred that Mac-1 subunits take different conformation when expressed in Chinese hamster ovary (CHO) and human embryonic kidney (HEK) 293T cells, respectively. Integrins alpha(M)beta(2) plays important role on leukocytes, such as adhesion, migration, phagocytosis, and apoptosis. It was hypothesized that homomeric associations of integrin subunits provide a driving force for integrins activation, and simultaneously inducing the formation of integrins clusters. However, experimental reports on homomeric associations between integrin subunits are still controversial. Here, we proved the homomeric associations of the isolated Mac-1 subunits in living cells using three-channel fluorescence resonance energy transfer (FRET) microscopy and FRET spectra methods. We found that the extent of homomeric associations between beta(2) subunits is higher than alpha(M) subunits. Furthermore, FRET imaging indicated that the extent of homomeric associations of the Mac-1 subunits is higher along the plasma membrane than in the cytoplasm. Finally, we suggested that homomeric associations of the transmernbrane domains or/and cytoplasmic domains may provide the driving force for the formation of constitutive homomeric associations between alpha(M) or beta(2) subunits. The interaction between integrin macrophage differentiation antigen associated with complement three receptor function (Mac-1) and intercellular adhesion molecule-1 (ICAM-1), which is controlled tightly by the ligand-binding activity of Mac-1, is central to the regulation of neutrophil adhesion in host defense. Several "inside-out" signals and extracellular metal ions or antibodies have been found to activate Mac-1, resulting in an increased adhesiveness of Mac-1 to its ligands. However, the molecular basis for Mac-1 activation is not well understood yet. In this work, we have carried out a single-molecule study of Mac-1/ICAM-1 interaction force in living cells by atomic force microscopy (AFM). Our results showed that the binding probability and adhesion force of Mac-1 with ICAM-1 increased upon Mac-1 activation. Moreover, by comparing the dynamic force spectra of different Mac-1 mutants, we expected that Mac-1 activation is governed by the downward movement of its alpha 7 helix. (c) 2007 Elsevier Inc. The effect of alcohol solution on single human red blood Cells (RBCs) was investigated using near-infrared laser tweezers Raman spectroscopy (LTRS). In our system, a low-power diode laser at 785 nm was applied for the trapping of a living cell and the excitation of its Raman spectrum. Such a design could simultaneously reduce the photo-damage to the cell and suppress the interference from the fluorescence on the Raman signal. The denaturation process of single RBCs in 20% alcohol solution was investigated by detecting the time evolution of the Raman spectra at the single-cell level. The vitality of RBCs was characterized by the Raman band at 752 cm(-1), which corresponds to the porphyrin breathing mode. We found that the intensity of this band decreased by 34.1% over a period of 25 min after the administration of alcohol. In a further study of the dependence of denaturation on alcohol concentration, we discovered that the decrease in the intensity of the 752 cm(-1) band became more rapid and more prominent as the alcohol concentration increased. The present LTRS technique may have several potential applications in cell biology and medicine, including probing dynamic cellular processes at the single cell level and diagnosing cell disorders in real time. A single-cell diagnostic technique for epithelial cancers is developed by utilizing laser trapping and Raman spectroscopy to differentiate cancerous and normal epithelial cells. Single-cell suspensions were prepared from surgically removed human colorectal tissues following standard primary culture protocols and examined in a near-infrared laser-trapping Raman spectroscopy system, where living epithelial cells were investigated one by one. A diagnostic model was built on the spectral data obtained from 8 patients and validated by the data from 2 new patients. Our technique has potential applications from epithelial cancer diagnosis to the study of cell dynamics of carcinogenesis. (c) 2006 Optical Society of America.
