A number of acrosome reaction (AR) initiators have been found to be effective in inducing AR of human, laboratory and domestic animal sperm. Using an improved simple fluorescence microscopy, effects of gamma-aminobutyric acid (GABA), progesterone and ionophore A23187 on sperm AR of tree shrew, a useful animal model in biomedical research, have been investigated. Spontaneous AR in 4.92-7.53% of viable sperm was observed. Complete AR in 10.31-18.25% of viable tree shrew sperm was obviously induced by 5 mu M and 10 mu M calcium ionophore A23187, 1 mM GABA, and 5 mu M progesterone, and there were no significant differences between their abilities to initiate complete AR. No significant differences of AR percentages between 1- and 2-h treatments with A23187, progesterone and/or GABA were observed. These results suggested that the responses of tree shrew sperm to these AR initiators are similar to that of human and other mammalian sperm. (C) 1997 Elsevier Science B.V.

The present study aimed to evaluate the effect of dietary linolenic acid (LNA)linoleic acid (LA) ratio on growth performance, hepatic fatty acid profile and intermediary metabolism of juvenile yellow catfish Pelteobagrus fulvidraco. Six isonitrogenous and isolipidic diets were formulated to contain incremental levels of LNA from 0 to 5% at the expense of corn oil (rich in LA), resulting in six dietary treatments with LNA to LA ratios ranging from 0.35 to 14.64. The experiment continued for 7 weeks. Best growth and feed intake were obtained in the fish fed the diets containing the LNA/LA ratios of 1.17 and 2.12 (P<0.05). In contrast, feed conversion ratio was the lowest for fish fed the diets containing the LNA/LA ratios of 1.17 and 2.12 (P<0.05). Dietary LNA to LA ratios significantly influenced viscerosomatic index and hepatosomatic index (P<0.05), but not condition factor (P>0.05). Body composition was also significantly influenced by dietary LNA to LA ratios (P<0.05). Generally, liver FA compositions reflected dietary FA profiles. Declining LA and increasing LNA contents in liver were observed with the increasing dietary LNA/LA ratios (P<0.05). Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) increased with the increasing LNA to LA ratios, suggesting that yellow catfish could elongate and desaturate C18 polyunsaturated fatty acids into highly unsaturated fatty acids. As a consequence, the n-6 fatty acids (FA) declined, and total n-3 FA and n-3/n-6 ratios increased with the dietary ratios of LNA/LA (P<0.05). Dietary LNA to LA ratios significantly influenced several enzymatic activities involved in liver intermediary metabolism (P<0.05), such as lipoprotein lipase, hepatic lipase, pyruvate kinase, succinate dehydrogenase, malic dehydrogenase and lactate dehydrogenase, suggesting that dietary LNA/LA ratios had significant effects on nutrient metabolism in the liver. To our knowledge this is the first demonstration of the effects of dietary LNA to LA ratios on the enzymatic activities of liver in fish, which provides information on diet quality and utilization, and can also be used as an indicator of the nutritional status of this fish. Human cerebral cortical function degrades during old age. Much of this change may result from a degradation of intracortical inhibition during senescence. We used multibarreled microelectrodes to study the effects of electrophoretic application of gamma-aminobutyric acid (GABA), the GABA type a (GABAa) receptor agonist muscimol, and the GABAa receptor antagonist bicuculline, respectively, on the properties of individual V1 cells in old monkeys. Bicuculline exerted a much weaker effect on neuronal responses in old than in young animals, confirming a degradation of GABA-mediated inhibition. On the other hand, the administration of GABA and muscimol resulted in improved visual function. Many treated cells in area V1 of old animals displayed responses typical of young cells. The present results have important implications for the treatment of the sensory, motor, and cognitive declines that accompany old age.

The effects of aniracetam on extracellular amino acid levels in the hippocampus of conscious gerbils, with or without transient cerebral ischemia/reperfusion, were measured by microdialysis and reverse phase-high performance liquid chromatography. Increased extracellular levels of aspartate and glutamate that were observed in the hippocampus of conscious gerbils during transient global forebrain ischemia were reversed by aniracetam. In contrast, the level of extracellular gamma-aminobutyric acid was increased, while taurine was maintained at a higher level than other amino acids by administration of aniracetam (100 mg/kg, p.o.) 60 min before ischemia. Further, in contrast to ischemic animals, administration of aniracetam (100 mg/kg, p.o.) enhanced the release of glutamate and aspartate in the normal gerbil hippocampus. The results suggest that these effects might be due to a partial calcium agonist activity of aniracetam, and that the effects of aniracetam on amino acid levels might be a mechanism of protection against delayed neuronal death in the ischemic hippocampus, thereby improving memory dysfunction induced by ischemia/reperfusion. The feasibility of biodiesel production from tung oil was investigated. The esterification reaction of the free fatty acids of tung oil was performed using Amberlyst-15. Optimal molar ratio of methanol to oil was determined to be 7.5:1, and Amberlyst-15 was 20.8wt% of oil by response surface methodology. Under these reaction conditions, the acid value of tung oil was reduced to 0.72mg KOH/g. In the range of the molar equivalents of methanol to oil under 5, the esterification was strongly affected by the amount of methanol but not the catalyst. When the molar ratio of methanol to oil was 4.1:1 and Amberlyst-15 was 29.8wt% of the oil, the acid value decreased to 0.85mg KOH/g. After the transesterification reaction of pretreated tung oil, the purity of tung biodiesel was 90.2wt%. The high viscosity of crude tung oil decreased to 9.8mm(2)/s at 40 degrees C. Because of the presence of eleostearic acid, which is a main component of tung oil, the oxidation stability as determined by the Rancimat method was very low, 0.5h, but the cold filter plugging point, -11 degrees C, was good. The distillation process did not improve the fatty acid methyl ester content and the viscosity.

1 初级视觉皮层功能,GABA系统功能在衰老过程中的变化 本章首先对衰老过程中神经形态学和神经电生理学上的研究进行了综述,然后报道了作者的博士学位论文研究工作。实验采用神经电生理的手段,探讨初级视觉皮层(primary visual cortex;V1)功能,以及GABA(gamma-aminobutyric acid)系统功能在衰老过程中的变化。 实验1和2均采用单细胞记录技术,检测了中年猴V1细胞的方位选择性、方向选择性、自发放和最大反应,并与年轻和老年猴进行对比;比较了年轻和老年猴V1细胞的感受野外周抑制能力。在实验3中,我们记录了年轻和中年大鼠在给予GABA直接或间接的激动剂,戊巴比妥钠或氯胺酮{通过拮抗NMDA(N-methyl-D-aspartate)受体}后,其皮层的EEG(electroencephalogram)活动,并分析与年龄相关的差异。结果如下: 实验1:中年猴V1细胞的方向选择性和自发放介于年轻猴和老年猴之间,而方位选择性和最大反应与年龄之间没有相关性。 实验2:感受野外周区的最优刺激明显降低了年轻和老年猴具有高方位选择性细胞的比例。同时,年轻猴所有细胞,以及老年猴高方位选择性细胞具有较高的最大抑制比,与它们相比,老年猴无明显方位偏好细胞的最大抑制比显著降低; 实验3:戊巴比妥钠注射后,在年轻和中年大鼠上,alpha (8-12 Hz) 和beta (12-20 Hz) 频段EEG功率增加,theta (4-8 Hz) 功率减少,这些变化在中年大鼠上较为明显。氯胺酮注射后,中年大鼠theta功率比年轻大鼠具有更大幅度的降低。 我们的结果表明,视觉皮层功能的下调在衰老早期就已发生,其机制可能与抑制系统功能普遍降低有关. 2 奖赏机制,极低频磁场的生物学效应研究 本章首先对自然奖赏和药物成瘾机制、极低频磁场生物学效应,以及极低频磁场对奖赏系统的影响进行了综述,然后报道了作者的博士论文研究工作。实验目的是探讨大鼠眶额叶皮质(orbitofrontal cortex;OFC)活动与食物奖赏刺激的相关性,以及极低频磁场对小鼠空间认知能力的影响。 实验1采用EEG记录技术,检测了大鼠OFC在食物奖赏和渴求过程中EEG各频段的功率变化。在实验2中,使用了一种探索型Y-迷宫实验范式,它仅依赖于啮齿类动物天生的探索欲望,避免了奖赏效应的干扰,利用此新型迷宫,我们检测了25和50 Hz磁场对小鼠空间识别记忆能力的影响。其结果如下: 实验1:大鼠OFC的delta频段(2-4 Hz)EEG活动与食物刺激显著相关,其相对功率在食物渴求时下降,在食物奖赏时升高。 实验2:与短时照射相比,长时的50 Hz磁场照射降低了小鼠对新异臂的探索能力,而25和50 Hz磁场暴露都不影响小鼠的活动力。 本研究表明,食物奖赏与OFC的delta频段EEG活动密切相关,而我们以前发现,大鼠和猴OFC的gamma(20-100 Hz)活动与吗啡成瘾相关,提示了OFC在自然奖赏和药物成瘾中具有不同的作用;另外,本实验首次证明,极低频磁场损害了小鼠不依赖于奖赏系统的空间认知能力,而我们先前发现,极低频磁场可以强化吗啡诱导的条件化位置偏好,从而说明极低频磁场对吗啡成瘾具有独特的生物学效应。

Order-disorder transition (ODT) behavior in eicosylated polyethyleneimine (PEI20C) comblike polymer obtained by grafting n-eicosyl group on polyethyleneimine backbone was systematically investigated by the combination of differential scanning calorimetry (DSC), wide-angle X-ray diffraction (WAXD), Fourier transform infrared (FTIR) spectroscopy as well as solid-state high resolution nuclear magnetic resonance (NMR) spectroscopy. DSC investigations showed two obvious transitions, assigned to the transitions (1) from orthorhombic to hexagonal and (2) from hexagonal to amorphous phase, respectively. These transitions are induced by the variations of alkyl side chain conformation and packing structure with temperature changing, which consequently lead to the destruction of original phase equilibrium. The ODT behavior can also be confirmed by spectroscopic methods like WAXD, FTIR and NMR. The ordered structure and the transition behavior of the alkyl side chains confined by the PEI backbone are obviously different from those of pristine normal alkanes. The transition mechanism of ODT and the origin of the phase transition behavior in PEI20C comblike polymer were discussed in detail in this paper.

A series of branched poly(ethyleneimine) (PEI) derived polymers with different lengths of n-alkyl side chains, denoted as PEI(n)Cs (n = 12, 14, 16, 18, 20, number of carbon atoms in alkyl side group), have been prepared by a N-alkylation method, and systematically characterized by differential scanning calorimertry (DSC) and wide-angle X-ray diffraction (WARD) as well as Fourier transform infrared spectroscopy (FTIR). The side chains grafted on these comblike polymers are long enough to form crystalline phase composed of paraffin-like crystallites. The crystallization of the side chains forces the branched poly(ethyleneimine) molecules to pack into layered structure, between which the crystallites are located. The melting temperatures of the side chain crystallites increase from -12.36 to +51.49 degreesC with increasing the length of the side chains from n. = 12 to n = 20, which are a little bit lower than the corresponding pristine n-alkanes. PEI18C was taken as an example in this work for the investigation of phase transition and conformational variation of the side chains with temperature changing.

The responses to rapid application of gamma-aminobutyric acid (GABA) and the GABA receptor characteristics of MTXO neurosecretory cells in the eyestalks of Chinese mitten-handed crab (Eriocheir sinensis) were examined by whole-cell patch clamp. Under current clamp mode, the depolarization and hyperpolarization were evoked from the three types of neurosecretory cells in response to the GABA (0.1 mmol/L) depending on the Nernst Cl- potential. Under voltage clamp mode, the inward Cl- channel currents (I-GABA) were resolved from all three types of neurosecretory cells in response to GABA (0.01similar to5 mmol/L). The GABA currents were activated within 1 200 ms and peaked within 800 ms. No obviously desensitization was observed during GABA application. The dose-response curve showed usual S-shape, with a just-discernible effect at 0.01 mmol/L and near-saturation at 0.5 mmol/L. The GABA currents had reversal potentials that followed Nernst Cl- potentials when [Cl-] was varied. The pharmacological results revealed that the GABA receptor of the crab neurosecretory cells was sensitive to the Cl- channel blockers picrotoxin and niflumic acid (0.5 mmol/L), insensitive to GABA(A) receptor antagonist bicuculline and GABA(C) receptor agonist cis-4-aminocrotonic acid (CACA 1 mmol/L) and trans-4-aminocrotonic (TACA 1 mmol/L).

用超临界萃取技术对产于青藏高原上的糖茶藨种子中脂肪酸进行萃取,其萃取率为12%。用毛细管气相色谱对萃取的脂肪酸进行了分离和分析。结果表明不饱和脂肪酸的质量分数在90%以上。其中α-亚麻酸(α-linolenic acid)为27.4%,У-亚麻酸(У-linolenic acia)为2.70%。亚油酸(1inoleic acid)40.0%,油酸(oleic acid)21.0%。[著者文摘]

近二十年生物化学,药理学,营养学的研究表明:多烯脂肪酸(PUFA)尤其是ω-3系列,具有特殊的生理功能。例如α-亚麻酸(α-Linolenic acid,ALA),EPA(Eicosapentaenoic aciod)具有降低血脂,预防心血管疾病发生的功效。而DHA(Docosahexaenoic acid)对大脑发育及视网膜的形成有着重要的促进作用^[1,2],自然界中,紫苏油中富含亚麻酸,鱼油中含有较多的EPA与DHA,将其富集提纯,可以提高产品的生理活性,成为目前保健品及医药开发的热点。

MoNi/gamma-Al2O3 catalysts were prepared by the impregnation method. The catalyst samples were characterized by XRD and TPR. The effects of Mo promoter content and the catalyst reducing temperature Oil hydrotreatment activity of the catalyst were studied under 200 degrees C and 3 MPa hydrogen pressure using acetic acid as the model compound. The XRD results indicate that the addition of Mo promoter is beneficial to the uniformity of nickel species on the catalyst and decreases the Interaction between nickel species and the support Which results in the decrease the of NiAl2O4 spinel formation. The addition of Mo promoter also decreases the reducing temperature of the catalyst. After the catalyst of 0.06 MoNi/gamma-Al2O3 being reduced Under the atmosphere of H-2/N-2(5/95, V/V), nickel oxide was reduced to Ni-0. The reaction was promoted obviously upon the addition of the MoNi/gamma-Al2O3 catalyst reduced at 600 degrees C. The Mo-modified Ni/gamma-Al2O3 catalyst reduced at 600 degrees C displayed the highest activity during the reaction, the conversion of acetic acid reached the highest point of 33.2%. The products included ethyl acetate and water.

A novel structural triblock copolymer of poly(gamma-benzyl-L-glutamic acid)-b-poly(ethylene oxide)-b-poly(epsilon-caprolactone) (PBLG-PEO-PCL) was synthesized by a new approach in the following three steps: (1) sequential anionic ring opening polymerization (ROP) of ethylene oxide and epsilon-caprolactone with an acetonitrile/potassium naphthalene initiator system to obtain a diblock copolymer CN-PEO-PCL with a cyano end-group; (2) conversion of the CN end-group into NH2 end-group by hydrogenation to obtain NH2-PEO-PCL; (3) ROP of gamma-benzyl-L-glutamate-N-carboxyanhydrides (Bz-L-GluNCA) with NH2-PEO-PCL as macroinitiator to obtain the target triblock copolymer. The structures from CN-PEO precursor to the triblock copolymers were confirmed by FT-IR and H-1 NMR spectroscopy, and their molecular weights were measured by gel permeation chromatography. The monomer of Bz-L-GluNCA can react almost quantitatively with the amino end-groups of NH2-PEO-PCL macroinitiator by ROP.

A biodegradable two block copolymer, poly(epsilon-caprolactone)-b- poly(gamma-benzyl-L-glutamic acid) (PCL-PBLG) was synthesized successfully by ring-opening polymerization of N-carboxyanhydride of gamma-benzyl-L-glutamate (BLG-NCA) with aminophenyl-terminated PCL as a macroinitiator. The aminophenethoxyl-terminated PCL was prepared via hydrogenation of a 4-nitrophenethoxyl-teminated PCL, which was novelly obtained from the polymerization of c-caprolactone (CL) initiated by amino calcium 4-nitrobenzoxide. The structures of the block copolymer and its precursors from the initial step of PCL were confirmed and investigated by H-1 NMR, FT-IR, GPC, and FT-ICRMS analyses and DSC measurements.

CdS nanocrystals were synthesized through AOT/heptane/H2O reverse micelles. New stable reverse mikelles were obtained by adding an appropriate amount of acrylic. acid monomer, CdS nanocrystal-poly(acrylic acid) composites were synthesized by gamma-radiation with a reverse mi'celle route at room temperature. The US nanocrystals with narrow size distribution were, found to be dispersed homogeneously in the poly(acrylic acid) matrix. (C) 2002 Elsevier Science B.V. All rights reserved.
