Photoluminescence of GaAs0.973Sb0.022N0.005 is investigated at different temperatures and pressures. Both the alloy band edge and the N-related emissions, which show different temperature and pressure dependences, are observed. The pressure coefficients obtained in the pressure range 0-1.4GPa for the band edge and N-related emissions are 67 and 45 meV/GPa, respectively. The N-related emissions shift to a higher energy in the lower pressure range and then begin to redshift at about 8.5 GPa. This redshift is possibly caused by the increase of the X-valley component in the N-related states with increasing pressure.

P-selectin, a 70-nm-long cellular adhesive molecule, possesses elastic and extensible properties when neutrophils roll over the activated endotheliam of blood vessel in inflammatory reaction. Transient formation and dissociation of P-selectin/ligand bond on applied force of blood flow induces the extension of P-selectin and relevant ligands. Steered molecular dynamics simulations were performed to stretch a single P-selectin construct consisting of a lectin (Lec) domain and an epithelial growth factor (EGF)-like domain, where P-selectin construct was forced to extend in water with pulling velocities of 0.005-0.05 nm/ps and with constant forces of 1000-2500 pN respectively. Resulting force-extension profiles exhibited a dual-peak pattern on various velocities, while both plateaus and shoulders appeared in the extension-time profiles on various forces. The force or extension profiles along stretching pathways were correlated to the conformational changes, suggesting that the structural collapses of P-selectin Lec/EGF domains were mainly attributed to the burst of hydrogen bonds within the major beta sheet of EGF domain and the disruptions of two hydrophobic cores of Lee domain. This work furthers the understanding of forced dissociation of P-selectin/ligand bond.

The density fluctuations below the onset of convection in the Rayleigh-Benard problem are studied with the direct simulation Monte Carlo method. The particle simulation results clearly show the connection between the static correlation functions of fluctuations below the critical Rayleigh number and the flow patterns above the onset of convection for small Knudsen number flows (Kn=0.01 and Kn=0.005). Furthermore, the physical nature for no convection in the Rayleigh-Benard problem under large Knudsen number conditions (Kn>0.028) is explained based on the dynamics of fluctuations.

原产于亚马逊河流域的水生植物凤眼莲(Eichhornia crassipes)在花部结构上有三种花型(L、M和S),故其配殖系统(mating system)为典型的三型花柱(tristyly)。但在入侵地区,它却常只有M和L两种花型,其中尤以M型占据绝对优势,使有性繁殖水平大为下降。为了解释凤眼莲在入侵过程中,花型频率为何发生变化以及该变化对其在入侵地的适应性进化上有何影响,作者在中国西南的两个居群中连续两年开展了野外生长和人工授粉实验,对比分析水面和泥地两类不同生境中M和L在克隆生长、生物量积累以及有性繁殖水平等方面的异同,并利用RAPD、 ISSR片段比较了具M型和L型花植物个体间的遗传变异水平。对三型花柱这一长期引人注目的遗传模式的分子位点也尝试了连锁片段的克隆和测序。 克隆生长的对比实验发现,在2004年,漂浮生长的M个体平均克隆分株数为25.37,L为21.20,前者显示较强的克隆生长能力(t=2.252, P< 0.05);2005年的实验再次证实M(每个个体19.83个分株)比L(每个个体15.53分株)表现出了显著较强的克隆优势(t=2.631,P<0.001)。M较L多产生克隆约24%。 但在岸边泥地上固着生长时,2004年L个体平均克隆分株为16.20,M 为10.17个分株。L表现出了更强的克隆生长能力(t=4.788,p<0.001),与漂浮生长状况下的情形恰恰相反,暗示M与L个体可能存在一定的生态位分化。这种分化可能是在入侵过程中形成的,也可能反映了原产地亚马孙流域旱涝交替造成的固着生长与漂浮生长交替发生的种内适应性。 生物量(干重)的对比分析发现,M个体在漂浮生长和固着生长的情况下都比L有着更高的生物量积累(漂浮生长实验:t=6.173,p<0.005(2004年);t=6.99,p<0.001(2005年)。固着生长实验:t=4.029,p<0.001)。生物量对凤眼莲的竞争和过冬有着一定的作用,因此较大的生物量积累可能是M个体在当地气候和环境中逐渐适应的一个结果。 有性繁殖的实验包括了实验个体的花序数与花朵数、自交与异交人工授粉的结实情况以及种子萌发率等方面的对比分析。结果表明,虽然M和L个体在花序数和花朵数上不存在显著差异,但是M个体在自交和异交的种子产量上比L略高,尤其是自交的种子产量,M显著高于L(M平均每个蒴果的自交种子产量为139.8,L为76.2)。以各100粒种子进行萌发实验,发现两花型之间在种子萌发率上不存在显著差异。对于M而言,其自交种子产量远大于异交种子产量 (139.8 vs. 93.3),且种子萌发率也略大于异交的种子萌发率(自交种子萌发率45.47%,异交种子萌发率 30.73%)。结合以上的实验结果,本文认为M基因型优势的形成可能与M个体与当地环境长期适应导致的生长与繁殖 上的优势有关。M的本地适应性是建立在特定的遗传背景上时,异交反而会破坏这种遗传组合,造成远交衰退。 对40个M和30个L个体进行20个RAPD引物和20个ISSR引物的遗传分析时发现,所有个体在RAPD表型上没有区别,但是在M中出现了3个ISSR表型,在L中出现了2个ISSR表型。本文还尝试利用RAPD技术扫描与三型花柱的遗传位点连锁的DNA片段。在146个RAPD随机引物中,初步发现两个候选片段,一个750bp,另一个2 000bp;已对它们进行了克隆、测序。 这些初步实验表明凤眼莲在我国的入侵可能伴随着基因型的差异表现和居群遗传分化,这种基因型的差异表现对该植物的成功入侵具有作用。其中,花型为M的个体的优势生长解释了该花型在中国分布区内的主导地位。推测该生长优势的遗传基础可能来源于基因组内较高的杂合子水平和在入侵地较长的适应历史,但最终结论尚有待进一步的实验证据。

元素化学计量学指从化学计量学的角度出发,通过分析比较生命物质不同结构层次(分子、细胞、器官、机体、种群、群落等)或生态系统中元素的相对比值,来研究各层次相互之间以及生态学过程中元素之间的关系。生态化学计量学研究可以把生态实体的各个层次存元素水平上统一起来,足近年来新兴的一个生态学研究领域,广泛应用于生态学研究中。C,N,P是生物地球化学循环中的重要元素,在生态系统中占有重要地位,许多环境问题都与它们有关,由此这三种元素的化学计量学受到生态学家们的普遍关注。C:N:P化学计量学在水生生态系统中研究较为深入,目前已发展到染色体水平,而在陆地生态系统中的研究较为匮乏近年来由于人类活动的强烈影响,这三种元素的循环在速度和规模上都发生了前所未有的改变,导致一系列环境问题的出现,因此C:N:P化学计量学在陆地生态系统中的研究就显得尤为重要。作为地球生命存在基础的绿色植物,在地球上已有数亿年的演化历史,研究陆地植物的元素化学计量学不仅有助于深入了解植物存在于地球上的内在机制,而且可以为许多环境问题的解决提供理论依据。本文首先建立中国不同地区植物氮磷含量数据库,通过数据分析找出一般规律,并进一步揭示植物不同进化阶段N:P化学计量比的变化规律。在此基础上,通过在内蒙古羊草草原设立不同施肥样地来模拟自然界不同氮磷环境,从试验水平上研究不同施肥处理及施肥梯度下生态系统中氮、磷、有机质的变化规律,并从化学计量学角度研究其内在机制。 利州新建成的中国维管植物数据库(包含1603种植物)研究了不同进化水平以及不同功能群(生活型)间植物N:P比的变化规律,并沿胡焕庸线(胡线)把中团分为为东西两部分,从总体水平上对比了东、西部间叫植物氮磷含量以及N:P的异同。结果表明:l)从演化水平来看,尽管氮磷含量表现出极大的差异,除豆科植物外,植物N:P基本保持稳定水平;2)木本植物与草本植物的N:P比差异.显著,木本植物之间(常绿乔木,常绿灌木,落叶乔木,落叶灌木)N:P不具有显著差异;3)中国东西部植物养分含量和N:P比表现出极显著差异,东部的养分含量低于西部,而N:P显著高于西部。 在内蒙古羊草草原两块永久实验样地(样地A:1980年围封样地和样地B:1999年田封样地)进行了为期两年的N素和P素添加试验。氮素添加梯度为0,5,15,30,50,80 g NHN03.m.2.yr-1。P素添加梯度为0,2,4,8,16,and 32 g P2Osm-2 yr-1(仪分析了羊草器官的结果)。分别从植物器官、物种、功能群水平研究了N素添加对N:P化学计量学的影响,此外还研究了土壤和凋落物C:N:P化学计量学对N素添加的响应。结合生物量的变化趋势,探讨了元素化学计量学对养分状况的指示作用。 1.羊草器官对施肥的响应结果表明,添加N素可以显著提高羊草器官中的含N量,p素可以显著提高器官中的含P量:除2001年样地A中的根茎外,根茎中的含P量基本不受N素添加的影响;茎中的含P量同样表现出不受N素添加影响的趋势(2000年B区茎除外):N素添加可以显著增大羊草叶片中的含P量(B区2000年叶片除外)。P素添加对羊草器官的含N量没有影响;羊草器官中的氮磷含量施肥处理下表现出显著的正相关关系(N素添加下B区叶片除外)。N素添加对羊草器官的N:P比没有显著影响(A区茎2000年和B区叶片2000年除外):P素添加显著降低了羊草器官中的N:P比。 2.四种优势植物(羊草、羽茅、针茅和苔草)地上生物量和N:P化学计量学对氮肥的响应研究发现,四个物种的氮磷含量均具有极显著相关关系;氮肥可以显著提高样地A中的羽茅生物量,降低苔草的生物量,而使样地B中的羊草生物量增大;两块样地中,四个物种的氮磷含量及N:P比均随N素水平的增高而增大(样地A中的羽茅N:P比除外)。 3.基于生活型划分的功能群(多年生根茎禾草,多年生丛生禾草,豆科植物,多年生杂类草,一二年生植物,灌木和半灌木)对N素添加的响应研究表明:施N可以提高样地A中的多年生丛生禾草的生物量,而使样地B中的多年生根苇禾草增加;多年生杂类草的相对多度在两个样地中均随施氨水平的增加而显著 降低:在样地B中,施氮可以显著提高不同功能群的氮磷含量;在样地A中,功能群N、P含量对施肥的响应并没有一致的变化规律,添加N素可以显著提高不同功能群的含N量(豆科植物除外),多年生根茎禾草和多年生杂类草的P含量有显著增大的趋势(P < 0.005),而其它功能群(豆科植物、灌木和小半灌木、多年生杂类草和~二年生杂类草)的P含量基本恒定(P>0.05);在样地A中,多年生丛生禾草,多年生杂类草,一二年生植物,灌木和半灌木的N:P比随施氨水平的增加而显著增大,多年生根茎禾草和豆科植物的N:P比基本不变;在样地B中,多年生丛生禾草的N:P比随施氨水平的增加而显著增大,多年生根茎禾草、多年生杂类草和…二年生杂类草不受施氨水平的影响。 4.添加N素对根实验结果表明:两块样地中,上层根(0-10 cm)的生物量仅在施N后第一年显著增加,而下层根(10-20 cm)的地下生物量在两年的施N处理下均不受施肥梯度的影响i在样地A中,施肥后第一年对根的N、P含量影响不显著,施肥后第二年可以显著增大上层根的N、P含量;在样地B中,添加N素后第一年可以显著增大根的含P量; 在两个样地中,两年的N肥处理对根的N:P比没有显著影响:在施氨处理中,根的N、P含量及N:P比在施肥第一年的响应要高于第二年。 5.所有处理中,上层土壤(O-lO cm)养分含量(有机碳,全氮,全磷)均高于下层土壤(10-20 cm);在样地A中,氮素添加对r十壤有机碳没有显著性作用,在施肥第一年可以显著增加上层土壤的N、P含量,而在施肥后第二年对土壤N、P含量没有显著影响;在样地B中,添加N素对两年的土壤养分均没有显著影响:养分添加两个样地土壤中的元素比值(C:N比,C:P比,N:P比)没有显著影响;土壤养分对施N一年后的响应要高于第二年。 6.养分添加对凋落物化学特征及化学计量学特征的影响研究结果表明:凋落物现存量不受施肥的影响;2001年凋落物现存量与2000年和2001年的地上生物量相关关系不显著;添加N素可以显著提高凋落物的N含量,而对有机碳含量和P含量没有显著影响;凋落物C:N比随施肥梯度的增大而显著降低,N:P比显著增高,而C:P比没有明显变化。 以上研究结果表明,不同植物功能群的N:P比存在差异,人类活动强烈影响自然植被中的植物N:P比;但植物的N:P比不受植物进化的影响(豆科植物除外);由于植物已有数亿年的演化历史,同时N与P在植物的结构和功能上具有密切的联系,在生物地球化学循环中办存在耦合作用。因此植物N:P比值恒定可能是一普适性规律。 N素添加试验表明,在植物根、地上器官、物种和功能群水平上N与P均呈显著正相关关系,反映了植物体内的氮磷含量具有协同作用。共存种对N肥的响应不同,表明物种受不同元素的制约。因此除非把生态系统中所有物种对施肥的响应刻画清楚,笼统的认定生态系统缺乏某种元素是不适当的。 施肥试验表明,两种围封时间样地的主要限制性元素不同。极度退化(样地B)植物生长主要缺N,而在保护较好样地(样地A),P也逐步成为一种限制因素。反映了随着保护时间的增加,植物生长逐渐由N限制型向N、P共同限制型过渡。添加的养分要么被植物吸收,挥发到大气中,或以凋落物的形式返还到土壤表层。但是对养分的预算有待于进一步研究。土壤中的养分含量对N素添加有…个滞后效应,而植物响应较为迅速。功能群N:P比在施肥实验中不能保持恒定,可能是由于实验时间较短的缘故;化学计量学的研究表明羊草草原分解凋落物的微生物受P素的制约,可能是由于植物体内具有高的P素再转运机制。这一结论有待于进一步的验证。氮肥降低了凋落物的C:N比,因此凋落物的分解速率可能将要发生改变。这需要进一步开展C循环的研究。

用ApaⅠ、AvaⅠ、BamHⅠ、BclⅠ、BglⅠ、BglⅡ、ClaⅠ、DraⅠ、Eco RI、Eco RV、HaeⅡ、HincⅡ、HindⅢ、HpaⅠ、KpnⅠ、PstⅠ、PvuⅠ、PvuⅡ、SacⅠ、SalⅠ、ScaⅠ、SmaⅠ、StuⅠ、XbaⅠ和XhoⅠ等25种识别6个碱基对的限制性内切酶分析了浙江地方品种猪、浙江野猪等30个个体的线粒体DNA,共检出30种限制性态型(morph),可归结成3种单倍型,其中BamHⅠ-B、BclⅠ-B、DraⅠ-B和XbaⅠ-C为首次发现;各单倍型间平均遗传距离(p)为0.005,群体的平均多态度(#pi#)为0.0008,均处于遗传多样性贫乏范围。

采用蛋白电泳和线粒体DNA RFLP分析方法,研究了浙江七个地方品种猪及浙江野猪的遗传多样性状况。从分析的30个遗传座位、99个地方品种个体中发现MDH,PEPB,6PGD,AK,ADA,PHI六个座位具有多态性,其中AK,MDH多态座位在家猪中首次发现。浙江地方猪种的亲缘关系很近。在mtDNA RFLP水平,25种限制性内切酶在27个地方品种个体及3个野猪个体中共检出30种限制性态型,可归结成三种单倍型。各单倍型间平均遗传距离(P)为0.005,群体平均多态度(#pi#))为0.000 8,处于遗传多样性贫乏范围。

Background: The Galliformes is a well-known and widely distributed Order in Aves. The phylogenetic relationships of galliform birds, especially the turkeys, grouse, chickens, quails, and pheasants, have been studied intensively, likely because of their close association with humans. Despite extensive studies, convergent morphological evolution and rapid radiation have resulted in conflicting hypotheses of phylogenetic relationships. Many internal nodes have remained ambiguous. Results: We analyzed the complete mitochondrial (mt) genomes from 34 galliform species, including 14 new mt genomes and 20 published mt genomes, and obtained a single, robust tree. Most of the internal branches were relatively short and the terminal branches long suggesting an ancient, rapid radiation. The Megapodiidae formed the sister group to all other galliforms, followed in sequence by the Cracidae, Odontophoridae and Numididae. The remaining clade included the Phasianidae, Tetraonidae and Meleagrididae. The genus Arborophila was the sister group of the remaining taxa followed by Polyplectron. This was followed by two major clades: ((((Gallus, Bambusicola) Francolinus) (Coturnix, Alectoris)) Pavo) and (((((((Chrysolophus, Phasianus) Lophura) Syrmaticus) Perdix) Pucrasia) (Meleagris, Bonasa)) ((Lophophorus, Tetraophasis) Tragopan))). Conclusions: The traditional hypothesis of monophyletic lineages of pheasants, partridges, peafowls and tragopans was not supported in this study. Mitogenomic analyses recovered robust phylogenetic relationships and suggested that the Galliformes formed a model group for the study of morphological and behavioral evolution.

Bat flight poses intriguing questions about how flight independently developed in mammals. Flight is among the most energy-consuming activities. Thus, we deduced that changes in energy metabolism must be a primary factor in the origin of flight in bats. The respiratory chain of the mitochondrial produces 95% of the adenosine triphosphate (ATP) needed for locomotion. Because the respiratory chain has a dual genetic foundation, with genes encoded by both the mitochondrial and nuclear genomes, we examined both genomes to gain insights into the evolution of flight within mammals. Evidence for positive selection was detected in 23.08% of the mitochondrial-encoded and 4.90% of nuclear-encoded oxidative phosphorylation (OXPHOS) genes, but in only 2.25% of the nuclear-encoded nonrespiratory genes that function in mitochondria or 1.005% of other nuclear genes in bats. To address the caveat that the two available bat genomes are of only draft quality, we resequenced 77 OXPHOS genes from four species of bats. The analysis of the resequenced gene data are in agreement with our conclusion that a significantly higher proportion of genes involved in energy metabolism, compared with background genes, show evidence of adaptive evolution specific on the common ancestral bat lineage. Both mitochondrial and nuclear-encoded OXPHOS genes display evidence of adaptive evolution along the common ancestral branch of bats, supporting our hypothesis that genes involved in energy metabolism were targets of natural selection and allowed adaptation to the huge change in energy demand that were required during the origin of flight.

非人灵长类动物模型在艾滋病(AIDS)发病机制、传播途径、疫苗和药物等方面的研究中具有重要作用。树突状细胞(DC)作为最重要连接先天免疫与获得性免疫的抗原递呈细胞,在AIDS发病进程中扮演着重要的角色。研究非人灵长类AIDS动物模型中DC亚群数量、表型以及功能的变化,对揭示AIDS发病机制具有十分重要的意义。该文将重点总结近些年来DC亚群在AIDS动物模型发病机制中的作用研究进展,为以后的研究提供思路。

Somatic cell nuclear transfer (SCNT) has been successfully used in many species to produce live cloned offspring, albeit with low efficiency. The low frequency of successful development has usually been ascribed to incomplete or inappropriate reprogramming of the transferred nuclear genome. Elucidating the genetic differences between normal fertilized and cloned embryos is key to understand the low efficiency of SCNT. Here, we show that expression of HSPC117, which encodes a hypothetical protein of unknown function, was absent or very low in cloned mouse blastocysts. To investigate the role of HSPC117 in embryo development, we knocked-down this gene in normal fertilized embryos using RNA interference. We assessed the post-implantation survival of HSPC117 knock-down embryos at 3 stages: E9 (prior to placenta formation); E12 (after the placenta was fully functional) and E19 (post-natal). Our results show that, although siRNA-treated in vivo fertilized/produced (IVP) embryos could develop to the blastocyst stage and implanted without any difference from control embryos, the knock-down embryos showed substantial fetal death, accompanied by placental blood clotting, at E12. Furthermore, comparison of HSPC117 expression in placentas of nuclear transfer (NT), intracytoplasmic sperm injection (ICSI) and IVP embryos confirmed that HSPC117 deficiency correlates well with failures in embryo development: all NT embryos with a fetus, as well as IVP and ICSI embryos, had normal placental HSPC117 expression while those NT embryos showing reduced or no expression of HSPC117 failed to form a fetus. In conclusion, we show that HSPC117 is an important gene for post-implantation development of embryos, and that HSPC117 deficiency leads to fetal abnormalities after implantation, especially following placental formation. We suggest that defects in HSPC117 expression may be an important contributing factor to loss of cloned NT embryos in vivo. (C) 2010 Elsevier Inc. All rights reserved.

A total of 36 compounds (1-36) were obtained from the stem bark of Poncirus trifoliata including three new prenylated flavonoids, (-)-5,4'-dihydroxy-7,8-[(3 '',4 ''-cis-dihydroxy-3 '',4 ''-dihydro)-2 '',2 ''-dimethylpyrano]-flavone (1), (-)-5,4'-dihydroxy-7,8-[(3 ''-hydroxy-4 ''-one)-2 '',2 ''-dimethylpyrano]-flavone (2), and (-)-5,4'-dihydroxy-7,8-[(cis-3 ''-hydroxy-4 ''-ethoxy-3 '',4 ''-dihydro)-2 '',2 ''-dimethylpyrano]-flavone (3). The new structures were elucidated by means of spectroscopic methods. Compounds 1-20 were evaluated for their anti-human immunodeficiency virus-1 (HIV-1) activity, in which 2 showed significant anti-HIV-1 activity with high therapeutic index (T1) of 143.65.

HIV/AIDS的流行趋势没有减弱的迹象,人们迫切需要新的预防HIV传播的手段。杀微生物剂旨在通过局部用药于阴道或直肠,从而阻止HIV的传播。鉴于目前有大量的杀微生物剂候选物,亟待能够有效评价其有效性及安全性的动物模型。通过比较非灵长类小型动物模型与非人灵长类动物模型在评价HIV杀微生物剂的有效性及安全性上的重要作用,该文总结了评价杀微生物剂有效性及安全性的动物模型的优缺点,同时指出了杀微生物剂研究与发展的方向和建议,希望能够对杀微生物剂的研发有所帮助。
